Genetic Diversity and Characterization of DPE1 Gene in Rice Germplasm

  • Disproportionating Enzyme 1 (DPE1) is an a-1,4-D-glucanotransferase that cleavages the a-1,4-glucosidic bonds and transfers glucosyl groups. In rice endosperm, it participates in starch synthesis by transferring maltooligosyl groups from amylose and amylopectin to amylopectin. Here, we investigated the haplotype variations and evolutionary indices (e.g., genetic diversity and population structure) for the DPE1 gene in 374 rice accessions representing seven subgroups (wild, indica, temperate japonica, tropical japonica, aus, aromatic, and admixture). Variant calling analysis of DPE1 coding regions leads to the identification of six functional haplotypes representing/occupying 8 nonsynonymous SNPs. Nucleotide diversity analysis revealed the highest pi-value in wild group (0.0556) compared to other cultivated groups, of which temperate japonica showed the most reduction of genetic diversity value (0.003). A significant positive Tajima's D value (1.6330) of admixture highlights sudden population contraction under balancing selection, while temperate japonica with the lowest Tajima's D value (-1.3523) showed a selection signature of DPE1 domestication which might be the cause of excess of rare alleles. Moreover, these two subpopulations exhibits a greater differentiation (FST=0.0148), indicating a higher genetic diversity. Our findings on functional DPE1 haplotypes will be useful in future breeding programs, and the evolutionary indices can also be applicable in functional studies of the DPE1 gene.

Evolutionary Analyses of SSII-1 Gene Provides Insight into Its Domestication Signatures in Collected Rice Accessions

  • Starch synthase proteins (SSI, SSII and SSIII) in rice are mainly involved in amylopectin synthesis mediating its chain elongation, and the functional loss of SSII can increase amylose accumulation through decreasing of amylopectin chain proportions. For purposes of identifying functional haplotypes and evolutionary analyses of this gene, SSII-1, we investigated 374 rice accessions belonging to different subgroups of origins. We subsequently performed bioinformatic analyses on their variations through haplotyping, resequencing and structuring based on different classified populations. Haplotyping of cultivated rice accessions using genetic variations within SSII-1 genomic region of chromosome 10 revealed a total of 8 haplotypes, representing 6 functional haplotypes by 4 non-synonymous SNPs of three different exons (1, 4 and 10), which effect on protein structure. Higher nucleotide diversity value was found in wild group (0.0055) compared to any of cultivated subpopulations, of which aus showed the most reduction of diversity value (0.0003). Tajima's D analysis exhibits the most Tajima's D value only in admixture group (0.3600) which appears to be the cause of a sudden population contraction by rare alleles scarcity. A clear separation of some wild accessions from the admixed cultivated subpopulations was observed in PCA and phylogenetic analysis. Similar admixed pattern of population structure was estimated with an increased K values of 2 to 8 where genetic components of almost all cultivated subpopulations were shared with the wild which can also be subsequently estimated by very low FST-values by -0.011 (wild-aromatic) and -0.003 (wild-admixture).

아파트 난방용 배관의 부식원인 규명 (Corrosion Analysis of the Pipeline for Heating System in a Apartment)

  • The corrosion failure analysis for the heating pipeline of apartment was performed. Usually, the heating pipe was layed in the concrete. However, it was known that the corrosion problems could be induced by admixture containing chlorine such as set accelerant. antifreezer and so on. Therefore the new construction method using stone ash instead of concrete was attempt to solve these problems. However water leaking accidents by corrosion occurred after 1 month from the completion of the apartment. In this paper, the reason of corrosion failure was investigated by the field study and laboratory analysis in many aspects.

시멘트 페이스트의 유동 특성에 관한 평가방법 비교연구 (A Comparison Study Between Evaluation Methods on the Rheological Properties of Cement Paste)

  • This study investigates the influence of various blending factors on cement paste fluidity and find out the most effective evaluation method of cement paste flow, comparing flow test apparatuses such as ring flow(R-F), flow cone(F-C) and mini slump(M-S). A viscometer also measures the rheology coefficients to secure faithful numerical data. Firstly, series I examines cement paste, affected by several cement products and mineral admixture types in the range of W/B 40%, ordinary fluidity, and W/B 30%, high fluidity. In this series, the three types of cement product depended on companies, are randomly used and the mineral admixture, such as fly ash, blast furnace slag and silica fume, are incorporated in the cement paste, in response to the ratio of 10, 20, to 30%, respectively. In addition, series II studies various chemical admixture types, affecting the cement paste. This series is carried out with manufacturing companies and component types in the range of W/C 30%, high fluidity. For the manufacturing companies, randomly four products are used and for the component types, polycaboxylate, melamine, naphthalene and lignosulfonate type are chosen. Test results showed that in the fluidity test of cement paste considering various types of blending factors, R-F exhibited similar tendency with F-C and M-S. In the analysis of consistency curves measured by viscometer, the fluidity evaluation method using flow test apparatuses was significantly effective, except for the some of the low fluidity specimens. In conclusion of this study, R-F was the most convenient, faithful and effective fluidity evaluation method of cement paste.

시멘트 혼합재의 입도분포와 페이스트 유동특성 (The Particle Size distribution of Cement Binder and Rheological Properties of Paste)

  • 본 연구에서는 현재 시멘트 콘크리트 혼합재로서 널리 사용되고 있는 고로수쇄 슬래그와 애쉬의 페이스트 유동특성을 파악하였다. 그 방법으로 혼합재 단독을 사용한 페이스트의 유동특성을 파악하여, 아래와 같은 결론을 얻을 수 있었다. 혼합재 단독의 페이스트 유동특성을 시험하기 위하여, 3종류의 분쇄기를 사용하여 3가지 입도로 분쇄된 분말을 제조하였다. 이 분말을 Rosin-Rammler 분포식을 사용하여 얻은 계수 n 값과 De 값을 유동특성과 비교 분석한 결과에서, 일반적으로 같은 입경 크기 일 때 Ash 분말이 Slag 분말보다 소성점도 및 항복응력이 높았으며, 또한 동일 n 값에서 Ash 분말이 Slag 분말 보다 비교적 높은 소성점도와 항복응력을 나타내었으나, 입도분포 폭의 변화에는 Slag 분말이 Ash 분말 보다 민감한 유동특성의 변화 경향을 나타내었다.

초미분말 서냉 슬래그를 혼화재로 사용한 콘크리트의 기초적 특성 (Basic Properties of Concrete with Ultrafine-Blaine Air Cooling Slag as Admixture)

  • In this study, a test has been carried out to solve the problem with ground granulated blast-furnace slag, low early strength & lack of supply and to find out a way to use as concrete admixture of the ultrafine blaine air cooling slag which is all disposed as the by product of air cooling slag and its test was conducted to the replacement rate of ultrafine blaine air cooling slag & mixing condition of every concrete admixtures by type for the purpose of obtaining later a basic data for practical use of the cement that used ultrafine blaine air cooling slag by conducting comparative analysis. If ultrafine-blaine air cooling slag is used to the concrete following the results, a high efficiency water reducing agent won't be needed much for flow acquisition due to a high increase in flow, and the stripping time of concrete form will be shortened thanks to the acquisition of early strength, And though, it has the problems with long term strength which is similar or a little lower than the 3 types of ground granulated blast-furnace slag, it's still applicable as the substitute materials for 3 types of ground granulated blast-furnace slag at 10, 15% replacement rate of ultrafine-blaine air cooling slag, at which it shows higher activation index than 3 types of ground granulated blast-furnace slag.

광물질 혼화재 사용 초고강도 모르타르 및 콘크리트에 CASB 화합 고성능감수제의 효과분석 (Analysis of the Effect of Superplasticizer combined CASB on Ultra High Strength Mortar and Concrete Using Mineral Admixture)

  • 본 연구는 실리카퓸 사용 여부로 서로 다른 광물질 혼화재를 사용하는 초고강도 모르타르 및 콘크리트에 CASB 화합 고성능감수제를 적용함으로서 CASB의 효과를 분석하고자 실시한 것으로, 그 결과를 요약하면 다음과 같다. 굳지않은 모르타르 및 콘크리트의 특성으로 모르타르의 미니 슬럼프 플로 및 콘크리트의 슬럼프 플로는 2성분계 배합의 경우에는 B2-CASB가 B2-PC 보다 CASB 혼입에 의한 점성증가에 기인하여 작게 나타났지만, 3성분계인 배합의 경우에는 B3-CASB가 B3-PC 보다 CASB의 입도분포 개선 효과로 구속수가 감소한 것에 기인하여 크게 나타났다. 공기량은 콘크리트 보다 특히, 모르타르의 경우 2성분계 보다 3성분계 결합재에서 PC보다 CASB에서 결합재간 공극 충전 효과에 기인하여 감소하는 경향이었다. 단, 단위용적질량은 공기량과 반비례의 경향으로 나타났다. 경화 모르타르 및 콘크리트의 특성으로 압축강도는 2성분계 보다 3성분계 결합재에서, PC보다 CASB에서 높게 나타났는데, 이는 광물질 혼화재인 SF의 포졸란 반응에 의해 생성된 규산칼슘수화물로 조직이 치밀해진 것과 CASB의 모세관 공극 충전 효과에 기인한 결과로 분석된다. 이상을 종합하면 나노물질인 CASB를 고성능감수제와 화합하여 사용할 경우 실리카퓸 사용 3성분계 초고강도 모르타르 및 콘크리트의 유동성 및 강도 측면에서 품질향상이 가능할 것으로 사료된다.

  • In South Korea, about 900 metal mines have been abandoned, and about 88 million-t metal mine wastes have been discarded in recent years. The treatment of the tailings which are the main wastes in the abandoned metal mines becomes a social problem because they cause environmental pollution such as acidic waste water generation, groundwater contamination, and dust generation. Since almost whole quantities of the tailings have disposed by landfill now, the development of effective recycling methods for the tailings are strongly requested. It is expected that the fine tailings obtained by centrifugal separation process among the tailings can be utilized as admixture for cement. The purpose of this study is to evaluate the micro-structure of cement mortar admixed with fine tailing. Various admixtures were made of Fine tailings and 2 Types of OPC, fly-ash and blast furnace slag. The hydration reactivity of cement mortar with FT was examined by Porosity, XRD and SEM morphology analysis. The anolytical result about hardened hydrates shows that waste fine tailing help hydrates none densified due to it,s filling-space, These densified effect is concluded with improving the resistance to attack of cement mortar including waste fine tailing.

  • As super-high-strength concrete uses a large amount of binder, there is an autogenous shrinkage strain larger than dry shrinkage and it degrades the quality of structures. Thus, we need a technology to minimize the shrinkage strain of super-high-strength concrete. Accordingly, the present study prepared super-high-strength concrete with design strength of over 100MPa and, using an embedded gauge, measured the shrinkage strain of free shrinkage specimens for super-high-strength concrete containing expansion agent. According to the results of this study, the expansion rate of concrete increased in the early stage due to the admixture of expansion agent, but the shrinkage rate went down with the lapse of time. The effect of the admixture of expansion agent on compressive strength appeared insignificant. Further research shall be made on different kinds of expansion agents and various mixture ratios for basic analysis to reduce autogenous shrinkage of super-high-strength concrete.

3성분계 고성능 콘크리트 개발을 위한 기초 특성 분석 (The Analysis of Fundamental Property for Developing High Performance Concrete of Ternary System)

  • 본 연구에서는 BS와 FA를 치환한 3성분계 고성능 콘크리트의 개발을 위해 기초적 특성을 분석하였는데, 그 결과를 요약하면 다음과 같다. 굳지않은 콘크리트 특성으로, 유동성은 혼화재 치환율이 증가할수록 OPC보다 증가하는 것으로 나타났고, 공기량은 전반적으로 혼화재 치환율이 증가할수록 감소하는 경향을 나타내었다. 경화 콘크리트 특성으로, 압축강도는 BS와 FA의 치환율이 증가할수록 초기재령에서는 OPC보다 저하하는 것으로 나타났으나, 재령 28일에서는 OPC과 유사하거나 그 이상으로 증가하는 것으로 나타났다. 특히 B30F15일때 재령 28일에서 OPC보다 약 15% 정도 높은 최대 압축강도 값을 발휘하였다.
