고지방식이를 섭취한 흰쥐에서 고추장의 체중 및 지방조직과 혈청내의 지질감소효과 (Decreasing Effect of Kochujang on Body Weight and Lipid Levels of Adipose Tissues and Serum in Rats Fed a High-Fat Diet)

  • 고추장은 체중감소 효과를 보였는데 흰쥐를 이용한 식이섭취 실험에서 발효 숙성된 전통고추장이나 공장식 상품고추장은 고지방 식이로 인한 체중 증가를 감소시켜 정상에 가까운 체중을 유지하도록 하였다. 반면에 발효되지 않은 고추장의 경우 고춧가루군과 거의 비슷한 감소효과를 나타내었으며 발효된 전통고추장이나 공장식 상품고추장에 비해서 체중감소효과가 그리 크지 않았다. 발효된 전통고추장은 높은 체중 감소효과와 더불어 부고환 지방 및 신장 주위 지방조직의 총지방, TG, 콜레스테롤의 함량도 낮추는 효과를 나타내었으나 발효되지 않은 고추장식이군은 감소효과가 크지 않아 발효의 유무가 체중감소, 지방조직감소에 큰 변수임을 알 수 있었다.

Antiobesity Effect of Major Korean Spices (Red Pepper Powder, Garlic and Ginger) in Rats Fed High Fat Diet

  • Commonly used spices in Korean cooking (red pepper powder, garlic, ginger) were evaluated for anti-obesity properties and effects on triglyceride (TG) and cholesterol in blood and adipose tissues in rats fed a high fat (20%) diet. SD rats were raised for four weeks on either a normal diet (ND, based on the AIN-93M diet), high fat diet (HFD, supplemented with 16% lard oil in the ND), or diets containing 5% red pepper powder (RPP), garlic or ginger. These spices significantly decreased weight gain compared to HFD, but garlic and ginger showed a greater effect on reducing weight gain than RPP. The weights of liver and epididymal and perirenal fat pads in garlic and ginger diet groups were lower than those of the HFD groups (p < 0.05). The garlic and ginger also decreased triglyceride and cholesterol contents in liver and epididymal and perirenal fat pad, reversing the higher levels seen in HFD. RPP, garlic and ginger supplemented diets were effective in lowering serum triglyceride and cholesterol levels (p< 0.05). These results indicated that garlic and ginger more effectively suppressed the effects of HFD on body fat gain and lipid values of adipose tissues and serum than RPP.

The Inhibiton Effects of Hypercholesterolemia and Platelet in Fermented and Non-Fermented Preparation of Garlic

  • This Dietary cholesterol augments lipid profile and primes production and activation of platelets, leading to development of atherosclerosis which produce several detrimental effects on cardiovascular health. Ethnomedicine and Mediterranean diet are natural sources and cost effective modes against several ailments including cardiovascular diseases while fermented foods have gained interest due to their increased nutrient profile, enhanced bioavailability and efficacy. Garlic has been known to reduce cholesterol and inhibit platelet activation. We examined whether fermented garlic ameliorates effects of hypercholesterolemia and platelet functions in rats. Methodology: Male SD rats were fed with hypercholesterolemia diet and treated with spirulina, fermented and non-fermented preparations of garlic for one month. Platelet aggregation and granule secretion were assessed to evaluate platelet activation. Liver and kidney weights, lipid and enzymatic profile of serum and whole blood analysis was performed. Expressions of SREBP, ACAT-2 and HMG-CoA were assessed using RT-PCR while liver and adipose tissues were analyzed for histological changes. Both fermented and non-fermented garlic inhibited platelet aggregation and granule secretion while fermented garlic showed greater inhibitor tendency. Fermented garlic significantly reduced liver weight and triglycerides concentrations than non-fermented garlic. Similarly, fermented garlic greatly abrogated the detrimental effects of steatosis on liver and adipose tissues. Fermented garlic significantly improved lipid profile and modulated platelet functions, thereby inhibiting atherosclerosis and platelet related cardiovascular disorders.

Effects of Hormones on the Proliferation of Stromal Vascular Cells from Hanwoo Cattle Adipose Tissues

  • This study was designed to determine the effects of the insulin-like growth factor (IGF-1) and estradiol $17-{\beta}$ on the in vitro proliferation of stromal vascular cell from Hanwoo omental, subcutaneous, intermuscular and intramuscular adipose tissues. Cells were cultured in M199+20% newborn calf serum and the proliferation of cells was measured by direct microscopic cell counting and change of genomic DNA amount. Cell numbers increased slightly over the first 72 hour of culture and then increased greatly, regardless of adipose tissue depots. In IGF-1 treatment, the number of omental preadipocytes maintained highest level from the beginning to the 20th day of culture. However, in estradiol-$17{\beta}$ treatment, those tended to be lower than the control from the beginning of culture and significantly lower at the 24th day. When IGF-1 was added to subcutaneous preadipocytes, the numbers of cells were higher from 11th day than those from other treatments, although there was no statistical significance. For intermuscular preadipocytes treated with IGF-1, its numbers were significantly (p<0.05) higher at 11th day, and in the other days it showed a similar tendency to those of the subcutaneous tissue. In this experiment, preadipocytes were taken from 24 month old fully matured steers and the highest proliferation rate was shown in intramuscular tissue followed by those of subcutaneous preadipocytes. Addition of $5{\mu}M$ estradiol-$17{\beta}$ to the growth medium failed to promote the replication of Hanwoo preadipocytes, as indicated by direct cell counts and total genomic DNA content. As the culture period proceeded, the amounts of DNA were increased, but the patterns of increment were not consistent with the results of cell numbers.

고지방 식이 비만 모델 마우스의 체중 및 혈장 지질 대사에 미치는 한방보조식품의 영향 (Effects of Natural Dietary Supplement on Body Weight and Lipid Metabolism in High Fat Diet-induced Obese Mice)

  • Objective : Recently, obesity has increased at an alarming rate and is now a worldwide health problem. Natural dietary supplement, $CharmSlim^{(R)}$(C), is mixed pills, which consists of beans, black sesames, kelps, onions, adlay seeds, anchovies, pyogo mushrooms, green tea. Our study was performed to determine the weight-loss effects of natural dietary supplement, based on natural ingredients, in mice fed high-fat diet with an abundance of carbohydrates. Methods : Four groups of male ICR mice were fed different diets during 6 weeks: normal diet(NOR), high-fat (15%, w/w) diet(HF), high-fat supplemented with natural dietary supplement powdered 5%(C5) and 10%(C10) groups. We measured the changes of food intake, body weight and adipose tissues weights. Also we examined levels of lipid profiles in serum. Results : Body weight, liver weight and food efficiency ratio were significantly higher in the HF group than in NOR group. After 6 weeks of treatment, body weight, liver weight and adipose tissues weights (epididymal, perinephric, visceral and BAT) were significantly decreased in the C10 group when compared to the control HF group. The C10 group had markedly lower serum levels of total cholesterol and triglyceride when compared with HF group. Conclusion : These results demonstrate that this dietary supplement can suppress the increased lipid profiles, body weight and fat gains, thus could be considered as an effective adjunct in the treatment of obesity.

Relationship Between Nutrient Supply to Muscle and Adipose Tissues and Nitrogen Retention in Growing Wethers on Forage Based Diets Fed with Different Forage Sources

  • Three growing wethers were used to investigate the differences in nitrogen (N) retention, blood plasma metabolite concentration and energy-yielding nutrient supply to muscle and adipose tissue. The wethers were fed one of three diets: timothy hay with concentrate (THD), Italian ryegrass with concentrate (IRD), and rice straw with concentrate (RSD) for 11 days. The experimental diets were adjusted to the animals to provide 100 g of daily gain. The triglyceride (TG) concentration of blood plasma in arterial and portal veins was higher with THD and IRD than with RSD. Conversely, the available amount of TG in tissues was higher with IRD. The daily amount of glucose and non-esterified fatty acids (NEFA) supplied to muscle tissue and adipose tissue was numerically higher with THD than IRD or RSD. Although N retention did not differ among the diets, it was numerically higher with THD than with IRD or RSD. The results suggest that the difference in the amount of glucose and NEFA delivered to muscle tissue may reflect the N retention in response to forage based diets.

Diphlorethohydroxycarmalol of Ishige okamurae and Caffeine Modified the Expression of Extracellular Fibrillars during Adipogenesis of Mouse Subcutaneous Adipose Derived Stem Cell

  • Although, one of the etiologies of localized lipodystrophy of the subcutaneous connective tissue (cellulite) is the histological alternation of adipose tissue, the characteristics of expression of the components of extracellular matrix (ECM) components during adipogenesis are not uncovered. In this study, the effects of caffeine and Ishige okamurae originated diphlorethohydroxycarmalol (DPHC) on the expression of extracellualr fibers was analyzed with quantitative RT-PCR during differentiation induction of mouse subcutaneous adipose derived stem cells (msADSC) into adipocyte. The expression levels of Col1a, Col3a1, and Col61a were decreased by the adipogenci induction in a time-dependent manners. However, Col2a mRNA and Col4a1 mRNA expressions were oposit to them. Caffeine and DPHC stimulated the changes of the expression of these collagens. Eln mRNA expression was increased by induction. DPHC stimulated the expression of it. Mfap5 mRNA expression was deceased in both adipogenic cell and matured adipocytes. Caffeine suppressed the expression of Mfap5 but the effect of DPHC was different by the concentration. The expression of bioglycan, decorin, and lumican were also modified by caffeine and DPHC in a concentration-dependent manner. Based on this study, we revealed firstly the effects of caffeine and DPHC on the expression of collagens, elastin, and glycoproteins during adipogenesis of msADSCs. Those results suggest that DPHC may have antiadipogenic effect and has more positive effets on normal adipose tissue generation and work as suppressor the abnormality of ECM structure. Such results indicate that DPHC can be applied in keeping the stability of the ECM of adipogenic tissues.

Cissus quadrangularis Extracts Decreases Body Fat Through Regulation of Fatty acid Synthesis in High-fat Diet-induced Obese Mice

  • The current study investigated the anti-obesity effect of Cissus quadrangularsis extracts (CQR-300) and its molecular action mechanism on obese mice induced high-fat diet (HFD). To induce the obesity, mice were fed a HFD for 6 weeks and then fed HFD only or HFD with CQR-300 at 50 and 200 mg/kg. Then, body weight gain and white adipose tissue weights were measured. We investigated the reduction in body fat and the regulation of fatty acid synthesis was measured by dual energy X-ray absorptiometry and real-time PCR with Western blot, respectively. In vitro study, CQR-300 inhibited pancreatic lipase activity. The CQR-300 treatment was significantly decreased the body weight gain and adipocytes size as well as white adipose tissues weights in HFD-induced obese mice. Furthermore, CQR-300 reduced the body fat and fat mass with regulating of adipose tissue hormones as leptin. Treatment with 50 mg/kg CQR-300 showed effectively lower expression levels of adipogenesis/lipogenesis related genes and proteins such as CCAAT/enhancer binding protein ${\alpha}$ ($C/EBP{\alpha}$), peroxisome proliferator-activated receptor ${\gamma}$ ($PPAR{\gamma}$), Sterol regulatory element binding protein-1c (SREBP-1c), and fatty acid synthase (FAS) in white adipose tissue (WAT) as compared with the HFD fed only mice. These results suggest that the CQR-300 has an anti-obesity effect via inhibition of lipase activity, decrease the body fat mass by regulating the adipogenesis and lipogenesis related genes and proteins in epididymal adipose tissue with evaluate body fat reduce in the HFD-induced obese mice.

The Relationship of a Combination of Human Adipose Tissue-Derived Stem Cells and Frozen Fat with the Survival Rate of Transplanted Fat

  • Background The survival rate of grafted fat is difficult to predict, and repeated procedures are frequently required. In this study, the effects of the freezing period of harvested adipose tissue and the addition of human adipose tissue-derived stem cells (ASCs) on the process of fat absorption were studied. Methods Adipose tissue was obtained from patients who underwent a lipoaspirated fat graft. The fat tissue was cryopreserved at $-20^{\circ}C$ in a domestic refrigerator. A total of 40 nude mice were used. The mice in the experimental group received three different subcutaneous injections in the back: an injection of fresh fat and ASCs, an injection of fat that had been frozen for one month and ASCs, and an injection of fat that had been frozen for two months and ASCs. The control mice received fat grafts without ASCs. The mice were sacrificed at four or eight weeks after the procedure, and the grafted fat tissues were harvested. The extracted fat was evaluated using photographic analysis, volume measurements, and histological examination. Results In the control group, the fat resorption rates four weeks after transplantation in the grafts of fresh fat, fat that had been frozen for one month, and fat that had been frozen for two months were 21.14%, 22.46%, and 42.56%, respectively. In the experimental group, the corresponding resorption rates were 6.68%, 13.0%, and 33.9%, respectively. Conclusions ASCs can increase the fat graft survival rate. The use of ASCs in fat grafting can reduce the need for repeated fat grafts and provide good long term results.
