Adipocyte differentiation inhibition, whitening, antibacterial and antioxidant activities of extracts from Aloe vera by-product (알로에 베라 가공 부산물 추출물의 지방세포 분화억제, 미백, 항균 및 항산화 활성)

  • Aloe has been widely used as a cosmetic and medicinal plant. Until now, several effects such as antioxidant, anti-cancer, anti-diabetic, immunity and whitening of aloe gel extract have been reported, but research on aloe by-products occurring in food processing has not been actively conducted. In this study, we investigated whether the aloe by-product extract from food processing could be used as a functional biomaterial. Cytotoxicity was not seen in both the mixer and press extracts. Inhibition of 3T3-L1 adipocyte differentiation was detected only in the mixer extract and not in the press. It was confirmed that hyaluronic acid accumulation and tyrosinase inhibition increased according to the treatment concentration of the mixer extract. The antimicrobial activity of the mixer extract was observed in the Porphyromonas gingivalis strain, but not in the Streptococcus mutans strain. Antioxidant activity through DPPH and SOD analysis increased with the concentration of the mixer extract. In summary, it was confirmed that the mixer extract of aloe by-products has the effect of inhibiting adipocyte differentiation, moisturizing, whitening, and antioxidant, suggesting the possibility of using it as a functional bio-material for health drinks or beauty masks.

Effect of Boehmeria nivea on Adipocyte Differentiation and Angiogenesis (모시풀 추출물이 지방세포분화와 혈관신생에 미치는 영향)

  • Boehmeria nivea (L.) Gaud., a flowering plant, has been widely cultivated in Asian countries including Korea. It has been reported that B. nivea exhibits health beneficial effects for the prevention of inflammation, oxidative stress, and virus-related diseases. In this study, we evaluated the inhibitory effect of B. nivea on adipocyte differentiation and angiogenesis. DPPH radical scavenging activities of 70% ethanol extract of B. nivea (EBN) and water extract of B. nivea (WBN) were $90.8{\pm}1.1%$ and $20{\pm}6.9%$, respectively. EBN was also effective in the reduction of adipocyte differentiation in 3T3-L1 cells. We next examined the transcriptional activity of peroxisome proliferator-activated receptor gamma ($PPAR-{\gamma}$), a pivotal target for anti-obesity. We found that treatment with rosiglitazone induced the transactivation of $PPAR-{\gamma}$. Under the same condition, $800{\mu}g/mL$ EBN reduced the transactivation of $PPAR-{\gamma}$ in rosiglitazone-induced cells. These results demonstrate that EBN-inhibited adipocyte differentiation was accompanied by $PPAR-{\gamma}$ inhibition. The study also tested whether EBN exhibits an anti-angiogenic effect by inhibiting tube formation in HUVECs. We found that EBN effectively inhibits tube formation, suggesting that EBN exhibited an anti-angiogenic effect. Taken together, B. nivea can be used as a functional food for the prevention of obesity and angiogenesis-related diseases including cancer.

Inhibitory Effect of Ethyl Acetate Extract of White Peach Pericarp on Adipogenesis of 3T3-L1 Preadipocyte Cells

  • In order to determine whether peach contains compounds to regulate adipocyte differentiation, extracts of flesh/pericarp of yellow/white peach were prepared in water, ethyl acetate (EtOAc), or n-butanol solvent and determined for effects on adipocyte differentiation in C3H10T1/2 or 3T3-L1 cells. Interestingly, none of peach extracts has statistically significant stimulatory effect on the adipocyte differentiation in C3H10T1/2. Furthermore, the presence of EtOAc extract of white peach pericarp (WPP) was found to inhibit lipid accumulation in 3T3-L1 cells both by microscopic examination of Oil Red O-stained lipid droplets and by spectrophotometric quantification of extracted stain, indicating a significant inhibitory effect on adipocyte differentiation. The inhibition of lipid accumulation was accompanied by a significant decrease in the expression levels of adipocyte molecular markers-peroxisome proliferator-activated receptor $\gamma$, CAAT enhancer binding protein $\alpha$, and fatty acid-binding protein. Thus, this study determined that WPP EtOAc extract contains the inhibitory compound(s) on adipogenesis.

Effects of Mahuang-Chuanwu(Mahwang-Cheonoh) Pharmacopuncture Solution on Adipocyte Differentiation and Gene Expression in 3T3-L1 Adipocytes (마황천오 약침액이 3T3-L1 지방세포 분화 및 유전자발현에 미치는 영향)

  • Objectives : Mahuang-Chuanwu(Mahwang-Cheonoh) Pharmacopuncture(MCP) has been used to treat obesity in Clinical Korean Medicine. MCP solution(MCPS) is also expected to have strong anti-obesity activities. However, little is known about the mechanisms of its inhibitory effects on adipocyte differentiation and lipogenesis. Methods : In the present study, we examined the effects of MCPS on differentiation and lipogenesis of 3T3-L1 adipocytes. To elucidate the mechanism of the effects of MCPS on lowering lipid content in 3T3-L1 adipocytes, we examined whether MCPS modulates the expressions of transcription factors to induce lipogenesis and adipogenic genes related to regulate the accumulation of lipids. Results : Our results showed that MCPS significantly inhibited differentiation and lipogenesis of 3T3-L1 adipocytes in a dose-dependent manner. MCPS suppressed the mRNA expressions of cytidine-cytidine-adenosine-adenosine-thymidine(CCAAT)/enhancer binding proteins ${\alpha}$($C/EBP{\alpha}$), C/EBP ${\beta}$, $C/EBP{\delta}$, and peroxisome proliferator-activated receptor ${\gamma}$($PPAR{\gamma}$) genes related to the induction of adipose differentiation. MCPS inhibited the mRNA expressions of adipose-specific aP2, adipsin, lipoprotein lipase(LPL), CD36, TGF-${\beta}$, and leptin genes related to the fat formation. MCPS downregulated the mRNA expressions of liver X receptor(LXR) ${\alpha}$ and fatty acid synthase(FAS) genes related to the induction of lipogenesis. In addition, MCPS reduced the production of adipocyte-induced pro-inflammatory cytokines. Conclusions : MCPS could regulate the accumulation of lipids and expression of adipogenic genes via inhibition of transcript factors related to induction of adipose differentiation.

Inhibition of Adipocyte Differentiation and Adipogenesis by the Extract from Sophora japonica Fruit (회화나무 열매 추출물에 의한 지방세포 분화 및 지방생성 억제)

  • The world-wide rate of obesity is increasing continuously, representing a serious medical threat since it is associated with a variety of diseases including type 2 diabetes, cardiovascular disease, and numerous cancers. Sophora japonicais used as a traditional herb for medicinal purposes in eastern Asia. However, the anti-obesity effects of S. japonicafruit have not been explored. The aim of this study is to investigate the inhibition of adipocyte differentiation and adipogenesis by an ethanol extract of S. japonicafruit (EESF) in 3T3-L1 pre-adipocytes. Our results demonstrate that EESF suppressed the terminal differentiation of 3T3-L1 pre-adipocytes in a dose-dependent manner, as confirmed by a decrease in lipid droplet number and lipid content through Oil Red O staining. EESF significantly reduced the accumulation of cellular triglyceride, which was associated with a significant inhibition of the levels of pro-adipogenic transcription factors, including PPARγ, C/EBPα and C/EBPβ. In addition, EESF potentially down regulated the expression levels of adipocyte-specific proteins, including aP2 and leptin. In particular, EESF treatment effectively enhanced the activation of the AMPK signaling pathway; however, the co-treatment with compound C, an inhibitor of AMPK, significantly restored the EESF-induced inhibition of pro-adipogenic transcription factors and adipocyte-specific genes. These results indicate that EESF may exert an anti-obesity effect by controlling the AMPK signaling pathway, suggesting that the fruit extract of S. japonica may be a potential anti-obesity agent.

Effects of Betaone Extract on the Inhibition of Adipocyte Differentiation in High-fat Diet-induced Obesity (고지방 식이 유발 비만에서 베타원 추출물이 지방세포 분화 억제에 미치는 영향)

  • Obesity is a disease in which an abnormally large amount of fat accumulates in the body. Various diseases such as type 2 diabetes, dyslipidemia, high blood pressure, fatty liver, gallbladder disease, and coronary artery disease are induced. In this study, we investigated the effect of betaone, a type of barley, on obesity suppression. After the betaone extract was treated with 3T3 L1 adipocytes, the effect on adipocyte formation was investigated through Oil Red O staining. It was observed that differentiation was inhibited without affecting the viability of 3T3 L1 adipocytes. The effect of betaone extract on obesity inhibition in a mouse model was investigated. As a result of administering betaone extract after a high-fat diet, it was confirmed that the level of blood sugar and body weight was decreased, and glucose uptake ability was improved in a glucose tolerance test. The formation of mouse adipose tissue was suppressed, and the expression of genes involved in the formation and degradation of obesity in liver tissue was improved. These results suggest that betaone extract is a useful substance for improving obesity and is an excellent material for health functional food.

Cloning, Expression, and Regulation of Bovine Cellular Retinoic Acid-binding Protein-II (CRABP-II) during Adipogenesis

  • The mammalian cellular retinoic acid-binding proteins, CRABP-I and CRABP-II, bind retinoic acid which acts as an inducer of differentiation in several biological systems. To investigate a possible role for CRABP-II in bovine adipogenesis, we have cloned bovine CRABP-II cDNA and the coding region for CRABP-I. The predicted amino acid sequences of CRABP-II were highly conserved among several animal species (human, mouse, and rat at 97%, 93%, and 93%, respectively). The expression pattern of bovine CRABP-II was examined in greater details by applying RT-PCR to various bovine tissues. CRABP-II mRNA was expressed in most adipose-containing tissues. Moreover, the expression of CRABP-I and -II mRNA dramatically increased during the differentiation of adipocytes from bovine intramuscular fibroblast-like cells. The effects of retinoic acid on adipocyte differentiation of bovine intramuscular fibroblast-like cells were concentration-dependent. Retinoic acid activated the formation of lipid droplets at a level of 1 nM, whereas inhibition was observed at a level of $1{\mu}M$. CRABP-I gene was up-regulated and CRABP-II gene down-regulated by retinoic acid during adipocyte differentiation. These results suggest that CRABPs may play an important role in the regulation of intracellular retinoic acid concentrations during adipogenesis.

The Study on anti-obesity of Myrrh ethanol extract (몰약(沒藥) 에탄올 추출물의 항비만에 관한 연구)

  • Objectives : The objective of this study was to investigate the effect of Myrrh 80% ethanol extract on adipocyte differentiation and adipogenesis in 3T3-L1 cell.Methods : Myrrh was prepared by extracting with 80% ethanol. Cell viability was assessed by MTT assay using 3T3-L1 cells. Anti-obesity activity was measured in lipid droplets and triglyceride (TG) accumulation in 3T3-L1 cells. We also analyzed the expression of C/EBPβ, C/EBPα, PPARγ, SREBP1c, and aP2 by reverse transcriptase polymerase chain reaction (RT-PCR). In addition, we observed the production of fatty acid, acetyl-CoA carboxylase and Oil-red O stainingResults : No cytotoxicity from Myrrh 80% ethanol extracts was observed at the concentration of 1, 10, 100 (㎍/㎖) in 3T3-L1 cells. Treatment with Myrrh significantly suppressed the terminal differentiation of 3T3-L1 in a dose-dependent manner, as confirmed by a decrease in triglyceride and Fatty acid and Acetyl-CoA carboxylase. Also, Myrrh exhibited potential adipogenesis inhibition and downregulated the expression of pro-adipogenic transcription factors, such as sterol regulatory element binding protein-1c (SREBP-1c), peroxisome proliferator-activated receptor-γ (PPARγ), CCAAT/enhancer binding proteins α (C/EBPα) and C/EBPβ, and adipocyte expressed genes, such as adipocyte fatty acid binding protein (aP2) and Fas. In addition, lipid accumulation determined by Oil-red O staining showed that Myrrh extract had inhibitory effects on lipid accumulation in 3T3-L1 cells.Conclusions : These results suggest that Myrrh suppresses obesity factors in 3T3-L1 cells. Myrrh may be a useful medical herbs for attenuating metabolic diseases such as obesity.

Inhibition of Adipogenesis in 3T3-L1 Adipocytes with Magnolia officinalis Extracts (후박 추출물의 지방세포 분화 억제 효능에 관한 연구)

  • Magnolia extract, prepared from the Chinese herb Magnolia officinalis, is known for its potent anti-oxidative and anti-inflammatory effects. In this report, we showed that Magnolia extract inhibits adipocyte differentiation, as evidenced by reduced triglyceride (TG) accumulation. Also, Magnolia extract increased hormone sensitive lipase (HSL) protein level, and decreased the adipogenic transcription factor peroxisome proliferator activated receptor (PPAR)-${\gamma}$ protein and their corresponding mRNA. Our results suggest a potential apllication of Magnolia extract as anti-obesity agents inhibits adipocyte differentiation through the down-regulation of adipogenic transcription factors and other adipocyte-specific genes.
