Alzheimer's Disease 병태모델에서 녹용대보탕(鹿茸大補湯)의 신경세포 보호효과 (The neuroprotective effects of Nokyongdaebo-tang(Lurongdabutang) treatment in pathological Alzheimer's disease model of neural tissues)

  • Objectives : Alzheimer's disease(AD) is the most common form of dementia, which is characterized by progressive deterioration of memory and higher cortical functions that ultimately results in total degradation of intellectual and mental activities. Nokyongdaebo-tang(Lurongdabutang) has been usually used for the treatment for the deficiency syndrome dementia and amnesia. This experiment was designed to investigate the effect of the Nokyongdaebo-tang(Lurongdabutang) hot water extract on pathological AD model. Methods : The effects of the Nokyongdaebo-tang(Lurongdabutang) hot water extract on cultured spinal cord cells induced by ${\beta}$-amyloid were investigated. The effects of the Nokyongdaebo-tan(Lurongdabutang) hot water extract on the memory deficit mice induced by scopolamine were investigated. Results : 1. ${\beta}$-amyloid treatment on cultured spinal cord cells increased both GFAP-staining intensity of astrocytes and caspase 3 immunoreactivity on cultured cells. Then, Nokyongdaebo-tang(Lurongdabutang) treatment reduced the labeling intensity for both GFAP and caspase 3 proteins in culture cells. 2. Scopolamine treatment into mice increased levels of GFAP-positive astrocytes and caspase 3-labeled cells of the hippocampal subfields dentate hilar region, CA3 and CA1 area. In vivo administration of Nokyongdaebo-tang(Lurongdabutang) attenuated labeling intensity for those two proteins in the same hippocampal areas. Similar effects were observed by the treatment of galanthamine, an inhibitor of acetylcholinesterase. Conclusions : This experiment shows that the Nokyongdaebo-tang(Lurongdabutang) may play a protective role in damaged neural tissues. Since neuronal damage seen in degenerative brains such as AD are largely unknown, the current data may provide possible insight into therapeutic strategies for AD treatments. Nokyongdaebo-tang(Lurongdabutang) might be effective for the prevention and treatment of AD.

Anti-Inflammatory Herbal Extracts and Their Drug Discovery Perspective in Atopic Dermatitis

  • Atopic dermatitis (AD) is an allergic disorder characterized by skin inflammation. It is well known that the activation of various inflammatory cells and the generation of inflammatory molecules are closely linked to the development of AD. There is accumulating evidence demonstrating the beneficial effects of herbal extracts (HEs) on the regulation of inflammatory response in both in vitro and in vivo studies of AD. This review summarizes the anti-atopic effects of HEs and its associated underlying mechanisms, with a brief introduction of in vitro and in vivo experiment models of AD based on previous and recent studies. Thus, this review confirms the utility of HEs for AD therapy.

DNCB로 유발된 생쥐의 아토피 피부염에 대한 지실(枳實)의 효능 (Effects of Aurantii Immaturus Fructus (AI) on Atopic Dermatitis (AD) Induced by DNCB in Mice)

  • Objectives The purpose of this study was to investigate the effects of AI on AD induced by DNCB in mice. AI has antiallergic property that is useful in treating allergy-related-diseases, such as asthma, anaphylactic shock, acute bronchitis and skin diseases, skin pruritus from gastrointestinal diseases. However, AI has not been studied intensively yet regarding anti-inflammatory effect on AD. Therefore, this study was conducted on 2,4-dinitrochlorobezene (DNCB)-induced mice to investigate effects of AI in AD. Methods In the experiment, we divided mice into four groups: a normal group (NOR), a control group (CON), an AI spread group (AI spread), and an AI spread and feeding group (AI spread & feeding). Then examined the changes in the body weight, weights of spleen and ear, thickness of dorsum skin and ear skin, clinical aspects on dorsum skin, historical assessments, proliferation of splenocytes in vitro and in vivo, and cytokine (TNF-${\alpha}$, IL-10). Results From the experiment, the ear weight of AI spread & feeding group was significantly dropped and the ear thickness of both AI spread and AI spread & feeding were decreased significantly. Dorsum skin thickness was also decreased significantly in both AI spread and AI spread & feeding group. Also, AI treatment improved the symptoms of AD, such as coloration, erythema and desquamation and had a better effect on AI spread & feeding group. In histopathological observation, thickened epidermis, hyperkeratosis, pigmentation, hypergranulosis, parakeratosis were diminished as well in both AI spread and AI spread & feeding group. In vitro, we could observe when AI was increased as proliferation rate of splenocytes were increased, too. Conclusions In conclusion, these data suggest that AI can decrease symptoms of AD and show AI can be useful herbal therapy for AD.

Ethanolic extract of Red Sweet Pepper (Capsicum annuum L.) regulates the skin inflammation in vitro and in vivo

  • Allergic inflammatory disease has been increased by abnormal lifestyle and food habits. Especially, prevalence of atopic dermatitis (AD) has been elevated and treatment of AD has not been unclear. Red sweet pepper (RSP), named as Capsicum annuum L, has been known as having pharmacological effects such as antioxidant, detoxification and antibacterial effects. However, the beneficial effect of ethanolic extract of RSP on AD has not been partly examined yet. Therefore, the aim of this study was to investigate anti-inflammatory effects of RSP on AD in vitro and in vivo models. The treatment of RSP inhibited the secretion of inflammatory cytokine such as interleukin (IL)-6 and IL-8 in tumor necrosis factor (TNF)-${\alpha}$ and interferon (IFN)-${\gamma}$-stimulated human keratinocyte (HaCaT cell). Also, RSP extract regulated 2,4-dinitroflorobenzene (DNFB)-induced AD-like skin lesions in BALB/c mice. Oral administration of RSP ameliorated DNFB-induced AD-like symptoms. In presented results indicated that RSP inhibited inflammatory cytokines in HaCaT cell and ameliorated AD-like skin lesion through suppression of symptom of DNFB-induced skin inflammation. Thus, RSP might be a potential therapeutic agent for AD.

The Effect of Microbial Extracts on the Cell Activation and Inhibition Associated with Atopic Dermatitis

  • Atopic dermatitis (AD) is an inflammatory, relapsing, chronic skin disease and lesions in AD are frequently colonized with Staphylococcus aureus (S. aureus). Activation of T cells and IgE production by staphylococcal enterotoxins B (SEB) plays a crucial role in the pathogenesis of AD. Enterococcus faecalis (E. faecalis) is a nonpathogenic bacterium and produces the probiotic products that have been shown to have inhibitory effects on inflammatory responses. In present study, we carried out to assess the anti-inflammatory role of lyzed E. faecalis against the damaging effects of SEB on AD related immune responses. Furthermore, we attempted to determine whether the co-cultured lyzed E. faecalis can influence the colonization of S. aureus. As a result, we identified the effect of E. faecalis lysate as a potent therapeutic agent for atopic dermatitis (AD). E. faecalis lysate reduces the productions of total IgE and cytokines of AD-related immune cells in response to SEB stimulation. The proliferation of S. aureus was also inhibited by E. faecalis lysate. In conclusions, E. faecalis lysate may improve the skin-defense system disturbed by atopic condition, and may prevent subsequent secondary infection of S. aureus and development of AD.

Anti-atopic dermatitis effects of Parasenecio auriculatus via simultaneous inhibition of multiple inflammatory pathways

  • The treatment of atopic dermatitis (AD) is challenging due to its complex etiology. From epidermal disruption to chronic inflammation, various cells and inflammatory pathways contribute to the progression of AD. As with immunosuppressants, general inhibition of inflammatory pathways can be effective, but this approach is not suitable for long-term treatment due to its side effects. This study aimed to identify a plant extract (PE) with anti-inflammatory effects on multiple cell types involved in AD development and provide relevant mechanistic evidence. Degranulation was measured in RBL-2H3 cells to screen 30 PEs native to South Korea. To investigate the anti-inflammatory effects of Parasenecio auriculatus var. matsumurana Nakai extract (PAE) in AD, production of cytokines and nitric oxide, activation status of FcεRI and TLR4 signaling, cell-cell junction, and cell viability were evaluated using qRT-PCR, western blotting, confocal microscopy, Griess system, and an MTT assay in RBL-2H3, HEK293, RAW264.7, and HaCaT cells. For in vivo experiments, a DNCBinduced AD mouse model was constructed, and hematoxylin and eosin, periodic acid-Schiff, toluidine blue, and F4/80-staining were performed. The chemical constituents of PAE were analyzed by HPLC-MS. By measuring the anti-degranulation effects of 30 PEs in RBL-2H3 cells, we found that Paeonia lactiflora Pall., PA, and Rehmannia glutinosa (Gaertn.) Libosch. ex Steud. show an inhibitory activity of more than 50%. Of these, PAE most dramatically and consistently suppressed cytokine expression, including IL-4, IL-9, IL-13, and TNF-α. PAE potently inhibited FcεRI signaling, which mechanistically supports its basophil-stabilizing effects, and PAE downregulated cytokines and NO production in macrophages via perturbation of toll-like receptor signaling. Moreover, PAE suppressed cytokine production in keratinocytes and upregulated the expression of tight junction molecules ZO-1 and occludin. In a DNCB-induced AD mouse model, the topical application of PAE significantly improved atopic index scores, immune cell infiltration, cytokine expression, abnormal activation of signaling molecules in FcεRI and TLR signaling, and damaged skin structure compared with dexamethasone. The anti-inflammatory effect of PAE was mainly due to integerrimine. Our findings suggest that PAE could potently inhibit multi-inflammatory cells involved in AD development, synergistically block the propagation of inflammatory responses, and thus alleviate AD symptoms.

독활지황탕(獨活地黃湯)이 ${\beta}A$로 유도된 Alzheimer's Disease 병태 모델에 미치는 영향 (The Effects of Dokhwaljihwang-tang(Duhuodihuangtang) on the Alzheimer's Disease Model Induced by ${\beta}A$)

  • Objectives: This research investigates the effect of the DHJHT extract on Alzheimer's disease. Specifically, the effects of the DHJHT extract on (1) the behavior (2) the infarction area of the hippocampus, and brain tissue injury in AD mice induced with ${\beta}A$ were investigated. Methods: The effects of the DHJHT extract on the proinflammation cytokines mRNA expression and production of BACE, APP and ${\beta}A$ in in BV2 microglial cell line treated by lipopolysacchaide(LPS) plus ${\beta}A$ were investigated. The effects of the DHJHT extract on the behavior of the memory deficit mice induced by scopolamine were investigated. Results: 1. The DHJHT extract suppressed the expression of IL-$1{\beta}$, IL-6, TNF-$\alpha$, COX-2, and NOS-II, BACE and APP mRNA in BV2 microglial cell line treated by LPS plus ${\beta}A$. 2. The DHJHT extract suppressed the expression of ${\beta}A$ production in BV2 microglial cell line treated with LPS plus ${\beta}A$. 3. The DHJHT extract showed significantly inhibitory effect on the scopolamine-induced impairment of memory in the experiment of Morris water maze. 4. The DHJHT group suppressed the expression of IL-$1{\beta}$, TNF-$\alpha$, MDA, and CD68+/CD11b+ in the brain tissue of the mice with AD induced by ${\beta}A$. 5. The DHJHT group reduced the infarction area of hippocampus, and controlled the injury of the brain tissue in the mice with AD induced by ${\beta}A$. 6. The DHJHT group reduced tau protein, and GFAP in the brain tissue of the mice with AD induced by ${\beta}A$. Conclusions: These results suggest that the DHJHT group may be effective for the treatment of AD. Thus, DHJHT could be considered among the future therapeutic drugs indicated for the treatment of AD.

생료사물탕(生料四物湯)이 DNCB로 유발된 생쥐의 아토피 피부염에 미치는 영향 (Effects of SaengRyo-SaMul-tang on Atopic Dermatitis Induced by DNCB in Mice)

  • Objectives : Atopic Dermatitis (AD) is chronic skin disease characterized by allergic hypersensitivity reactions. Saeng-ryo-Samul-tang (SRSM) can treat skin disease by cooling down blood heat, cealering away congenital fever and detoxicating. This study was conducted to investigate the effects of SRSM on AD induced by 2,4-dinitrochlorobenzene (DNCB) in mice Methods : In this study, the effects of SRSM on changes in body weights, thicknesses of dorsum skin, thicknesses and weights of ear, changes of symptoms on the dorsum skin, histopathological degree of ear and dorsum skin, IL-10 and $TNF-{\alpha}$ in serum were observed. And the effects on the proliferation rates of splenocytes were also investigated in vivo and in vitro study. Results : In SRSM topical application (Topical) group, SRSM oral application (Oral) group and SRSM Combination (Combi) group thickness of dorsum skin decreased significantly. But in TPC, ORL and CBN group, weight of ear didn't show any changes, but thickness of ear decreased significantly. And TPC, ORL and CBN group showed meaningful effectiveness symptoms like desquamation and erythema on AD's clinical espect. In histopathological observation, spongiosis, edema and inflammatory cell infiltration of epidermal were remarkably diminished in TPC, ORL and CBN group. And SRSM diminished the proliferation rates of splenocytes in vivo and vitro study. Conclusions : The present study suggests that SRSM can significantly reduced symptoms of AD, therefore SRSM is effective to treatment of AD.

Galgeun-tang, an Herbal Formula, Ameliorates Atopic Dermatitis Responses in Dust Mite Extract-treated NC/Nga Mice

  • Objectives: Galgeun-tang (GGT, gegen-tang, kakkon-to), an herbal formula, is used to treat the common cold, fevers, headaches, hangovers and neck and upper back stiffness. The drugs currently used to treat atopic dermatitis (AD) are limited by the significant adverse effects associated with their long-term usage. The need to efficiently manage the AD response while reducing side effects has led to the development of alternative remedies. Methods: To assess the effects of GGT on AD, the anti-inflammatory and anti-AD properties of GGT were evaluated in both in vitro and in vivo systems. Results: Nitric oxide (NO) and histamine production on lipopolysaccharide (LPS)-treated RAW264.7 cells and phorbol-12 myristate 13-acetate (PMA)/A23187-treated MC/9 cells, respectively, were inhibited by GGT. GGT reduced thymus and activation-regulated chemokine (TARC/CCL17) release on TNF-${\alpha}$/IFN-${\gamma}$ stimulated HaCaT cells in a dose-dependent manner. GGT reduced both plasma levels of IgE and histamine and the dermatitis score in house dust mite induced atopic dermatitis-like lesions on NC/Nga mice. However, there were no significant histopathological differences observed between the GGT group and the AD-induced group, such as AD-like lesions in the dorsal skin or ear or mast cell infiltration in the dorsal skin. Conclusions: These results indicate that GGT inhibits chemokine production by keratinocytes and the atopic dermatitis response in NC/Nga mice, suggesting that GGT may be useful as a therapeutic remedy for treating AD and allergic inflammation-related diseases.

The Slough of Cicadidae Periostracum Ameliorated Lichenification by Inhibiting Interleukin (IL)-22/Janus Kinase (JAK) 1/Signal Transducer and Activator of Transcription (STAT) 3 Pathway in Atopic Dermatitis

  • It is known that animal-origin medicine could be one of effective treatment to remedy atopic dermatitis (AD) by controlling the cytokines. Cicadidae Periostracum (CP), the slough of Cryptotympana pustulata, has been frequently used for treating AD and skin affliction in traditional Korean Medicine. This study is aimed at investigating the ameliorating effects of CP on AD and its potential mechanism. The dinitrochlorobenzene sensitized mice were treated with CP for 2 weeks. The various biomarkers and the dermatitis scores presented that CP treatment can induce the visual and biological improvements of AD model. Pruritus, the most serious symptom of AD, which can cause repeated scratching behaviors and finally lead to lichenification, was reduced with CP treatment by regulating the inflammatory reactions. In addition, CP treatment diminished the number of mast cells that are known for causing inflammatory reactions. Moreover, it is proven that CP can decline secretion of interleukin-22, which means CP treatment has anti-inflammatory effects. CP treatment can correct the imbalance of helper T (Th)1 and Th2, downregulating thymic stromal lymphopoietin that leads to decrease of mRNA level of inflammatory cytokines. The crucial role of CP treatment is controlling of the Janus kinase 1/signal transducer and activator of transcription 3 pathway. In addition, CP treatment has the inhibitory effects on kallikrein related peptidase (KLK) 5 and KLK7. Taken together, CP treatment can ameliorate most symptoms and problems caused by AD disease, improving the AD patients' life quality.
