Short-term acute toxicity of synthetic detergent(LAS) to larvae of loach, Misgurnus angillicaudatus was examined by static bioassay. The larvae were exposed to 15 different concentration of synthetic detergent for 16, 48, 72, 96 and 120 hours in order to determine median lethal concentration($LC_{50}$). The $100\%$ mortarlity of larvae was showed within 120, 96, 48 and 16 hours for 6, 18, 30 and 38 ppm, respectively. The median lethal concentration values of the larvae were 12.59 ppm for 48 hours, 4.00 ppm for 96 hours and 1.02 ppm for 120 hours. The permissible toxicant concentration of acute toxicity to larvae was $0.37{\sim}0.43$ ppm, and application factor of the synthetec detergent was $0.093{\sim}0.108$. The median lethal time($LT_{50}$) for different concentration also was determined. The $LT_{50}$ of 0.2 ppm was found within 165.1 hours and 2 ppm was 106.2 hours, while the $LT_{50}$ of 8 ppm was 60.3 hours and that of 38 ppm was 23.5 hours.

Background: Citric acid (CA) and sodium hypochlorite (NaOCl) have been used to disinfect animals to protect them against avian influenza and foot-and-mouth disease. Objectives: We performed a good laboratory practice (GLP)-compliant animal toxicity study to assess the acute toxic effects of CA and NaOCl aerosol exposure in Sprague-Dawley rats. Methods: Groups of five rats per sex were exposed for 4 h to four concentrations of the two chemicals, i.e., 0.00, 0.22, 0.67, and 2.00 mg/L, using a nose-only exposure. After a single exposure to the chemicals, clinical signs, body weight, and mortality was observed during the observation period. On day 15, an autopsy, and then gross findings, and histopathological analysis were performed. Results: After exposure to CA and NaOCl, body weight loss was observed but recovered. Two males died in the CA 2.00 mg/L group and, two males and one female died in the 2.00 mg/L NaOCl group. In the gross findings and histopathological analysis, discoloration of the lungs was observed in the CA exposed group and inflammatory lesions with discoloration of the lungs were observed in the NaOCl exposed group. These results suggest that the lethal concentration 50 (LC50) of CA is 1.73390 mg/L for males and > 1.70 mg/L for females. For NaOCl, the LC50 was 2.22222 mg/L for males and 2.39456 mg/L for females. Conclusions: The Globally Harmonized System is category 4 for both CA and NaOCl. In this study, the LC50 results were obtained through a GLP-based acute inhalation toxicity assessment. These results provide useful data to reset safety standards for CA and NaOCl use.

Recent studies indicated the utility of whole effluent assessment as a measure to control discharge of toxic effluents to receiving water in Korea. However, most studies have been focussed on acute lethal effects of toxic wastewater with little consideration of chronic sublethal impacts which are of growing concern in protecting aquatic ecosystem. We conducted acute and chronic toxicity tests with effluents discharged from five different dyeing plants in Gyeong-gi province using a marine bacterium Vibrio fischeri and a freshwater macroinvertebrate Daphnia magna to demonstrate the importance of assessing chronic sublethal effects. Various levels of acute and chronic toxicities were observed in many samples tested in this study. In 21-d chronic toxicity tests using D. magna all samples showed effects on reproduction and growth. Notable mortalities were also noted in three out of five effluents. The result of the Microtox assay indicated that acute microbial toxicity existed in effluents from two out of five plants and acute daphnid toxicity was observed in only one effluent. The result of this study clearly suggests chronic toxicity tests are more suitable to assess biological effects of effluents because it was shown from this study that even an effluent with no acute toxicity could cause chronically lethal and/or sublethal adverse effects on aquatic biota which may affect the population dynamics in aquatic ecosystem.
This study evaluated acute nitrate nitrogen ($NO_3-N$) toxicity in juvenile blackhead seabream Acanthopagrus schlegelii. Seventy juveniles (Trial A, $7.1{\pm}0.6g$) and nine juveniles (Trial B, $71.3{\pm}3.5g$) per 70 L tank were exposed to $NO_3-N$ concentrations of 0, 500, 1500, 2500, 3500, and 4500 mg/L and 0, 600, 1200, 1800, 2400, and 3000 mg/L, respectively, in triplicate for 7 days. In Trial A, all fish exposed to 3500 and $4500mg\;NO_3-N/L$ died within 48 h; in Trial B, all fish exposed to $3000\;NO_3-N/L$ died after 120 h. The $96\;LC_{50}$ and $168\;LC_{50}$ were 2505 and $1806mg\;NO_3-N/L$, respectively, in Trial A, and 2663 and $2377mg\;NO_3-N/L$ in Trial B. Large juveniles were more resistant to $NO_3-N$ than small juveniles. The results of acute $NO_3-N$ toxicity studies provide important data for subsequent chronic toxicity studies.

We applied several machine learning methods for developing QSAR models for prediction of acute toxicity to fathead minnow. The multiple linear regression (MLR) and artificial neural network (ANN) method were applied to predict 96 h $LC_{50}$ (median lethal concentration) of 555 chemical compounds. Molecular descriptors based on 2D chemical structure were calculated by PreADMET program. The recursive partitioning (RP) model was used for grouping of mode of actions as reactive or narcosis, followed by MLR method of chemicals within the same mode of action. The MLR, ANN, and two RP-MLR models possessed correlation coefficients ($R^2$) as 0.553, 0.618, 0.632, and 0.605 on test set, respectively. The consensus model of ANN and two RP-MLR models was used as the best model on training set and showed good predictivity ($R^2$=0.663) on the test set.
The plant of Pedicularis resupinata var. oppositifolia(Scrophulariaceae) is described to be used for treatment of rheumatoid arthritis, malignant abscess (tumor), urolith and diuretics in oriental medicinal books. This study is concerned with some general pharmacological activities. It was observed that the methanol extract of the aerial part of this plant showed some effects on acute toxicity, carrageenin induced edema in rat paw, bile juice secretion and antibacterial effect. From this result it was obtained that the methanol extract revealed the antiinflammatory activity and bile juice secretion increasing effect. Thus, it was fractionated and buthanol fraction has obvious inhibition effect on edema and bile juice secretion increasing effect. The total extract showed low acute toxicity i.e., the minimum lethal dose was more than 8000mg/kg in oral administration in mice.
Objectives : Kaempferia parviflora Rhizome is black ginger indigenous to Laos and Thailand. It has been used as a folk medicine to improve blood flow and promote vitality and longevity with good health and well being. For these reasons, Kaempferia parviflora Rhizome has been focused on developing it as a food or food supplement. In addition, Kaempferia parviflora Rhizome could be under consideration of new prescription based on its characteristic compounds, polymethoxyflavonoids. However, it needs to be certified as safe before it can be used. Here, a single-oral toxicity test and safety classification was carried out to identity acute information of the toxicity of Kaempferia parviflora Rhizome powder and to make sure of its safety in clinical applications. Methods : Test substance was orally administered to male and female SD-rat at dose levels of 5000 mg/kg to estimate approximate lethal dose(ALD). Based on the acute information of the toxicity, the safety classification was estimated using the HED(human equivalent dose)-based MOS(margin of safety). Results : At 14 days after treatment with test substance. there were no of test substance related with mortalities and clinical signs. In addition, no changes in the body or organ weights and no gross or histopathological findings were observed. Thus, the ALD of Kaempferia parviflora Rhizome powder was considered over 5,000 mg/kg in both female and male mice. Conclusions : Based on the single oral toxicity test using the highest and limit dose, 5,000 mg/kg and the decision guideline for safety classification based on HED-based MOS, it was estimated that Kaempferia parviflora Rhizome powder is classified as "Specified class B" indicating that clinical dose is not limited to patients as safe as food.
This study was conducted to obtain acute information of the oral dose toxicity of DHU001, a polyherbal formula in male and female mice. In order to calculated 50% lethal dose ($LD_{50}$) and approximate lethal dose (LD), test material was once orally administered to male and female ICR mice at dose levels of 2000, 1000, 500, 250 and 0 (vehicle control) ml/kg (body weight). The mortality and changes on body weight, clinical signs, gross observation, organ weight and histopathology of principle organs were monitored 14 days after treatment with DHU001. We could not find any mortalities, DHU001 treatment-related clinical signs, changes on the body and organ weights, gross and histopathological findings. The results obtained in this study suggest that $LD_{50}$ and approximate LD in mice after single oral dose of DHU001 were considered over 2000 mg/kg in both female and male mice.
To screen biologically active components of the higher fungi of Korea, the dried carpophores of Auricularia polytricha were extracted with water. The extract was examined for acute toxicity in ICR mice. A low molecular weight toxin of this fungus was purified by a acetone precipitation followed by cellulose, silica gel and Sephadex LH-20 column chromatography. Major symptoms of this toxin were decreasing of normal motility, eye extrusion, hair erection, shivering, trembling of head, paralysis, rapid running or moving before death and depression of respiration. The median lethal doses of the total extract were 1. 28 g/kg and 4. 31 g/kg by i.p. and p.o. administrations, respectively. The amounts of one mouse lethal unit of the total extract and final fraction that killed a 20 g mouse within 30 minutes were 28.5 and 12.0 mg/mouse, respectively.
Objectives This study aims to evaluate the size-dependent toxicity of spherical silver nanoparticles (Ag NPs) to an endemic benthic organism, Glyptotendipes tokunagai. Methods Ag nanoparticles of three nominal sizes (50, 100, and 150 nm) capped with polyvinyl pyrrolidone (PVP-Ag NPs) were used. Their physicochemical properties, acute toxicity (48 hours), and bioaccumulation were measured using third instar larvae of G. tokunagai. Results The aggregation and dissolution of PVP-Ag NPs increased with exposure time and concentration, respectively, particularly for 50 nm PVP-Ag NPs. However, the dissolved concentration of Ag ions was not significant compared with the median lethal concentration value for $AgNO_3$ (3.51 mg/L). The acute toxicity of PVP-Ag NPs was highest for the smallest particles (50 nm), whereas bioaccumulation was greatest for the largest particles (150 nm). However, larger PVP-Ag NPs were absorbed and excreted rapidly, resulting in shorter stays in G. tokunagai than the smaller ones. Conclusions The size of PVP-Ag NPs significantly affects their acute toxicity to G. tokunagai. In particular, smaller PVP-Ag NPs have a higher solubility and stay longer in the body of G. tokunagai, resulting in higher toxicity than larger PVP-Ag NPs.
