  • The concentration effect relationship (pharmacokinetic pharmacodynamic model, PKPD) of drugs used for Parkinson's disease is complex. The benefits and adverse effects of drug treatment have to be considered in terms of short term and long term effects. Acute effects, observed over hours and days, reflect symptomatic benefit while chronic effects, observed over months and years, also reveal influences on the progress of the disease. (omitted)

Effects of total body irradiation-based conditioning on allogeneic stem cell transplantation for pediatric acute leukemia: a single-institution study

  • Purpose: To evaluate the effects of total body irradiation (TBI), as a conditioning regimen prior to allogeneic stem cell transplantation (allo-SCT), in pediatric acute leukemia patients. Materials and Methods: From January 2001 to December 2011, 28 patients, aged less than 18 years, were treated with TBI-based conditioning for allo-SCT in our institution. Of the 28 patients, 21 patients were diagnosed with acute lymphoblastic leukemia (ALL, 75%) and 7 were diagnosed with acute myeloid leukemia (AML, 25%). TBI was completed 4 days or 1 day before stem cell infusion. Patients underwent radiation therapy with bilateral parallel opposing fields and 6-MV X-rays. The Kaplan-Meier method was used to calculate survival outcomes. Results: The 2-year event-free survival and overall survival rates were 66% and 56%, respectively (71.4% and 60.0% in AML patients vs. 64.3% and 52.4% in ALL patients, respectively). Treatment related mortality rate were 25%. Acute and chronic graft-versus-host disease was a major complication; other complications included endocrine dysfunction and pulmonary complications. Common complications from TBI were nausea (89%) and cataracts (7.1%). Conclusion: The efficacy and toxicity data in this study of TBI-based conditioning to pediatric acute leukemia patients were comparable with previous studies. However, clinicians need to focus on the acute and chronic complications related to allo-SCT.

The effects of immune response on maximal aerobic and anaerobic exercise (급성적인 최대 유·무산소 운동이 면역반응에 미치는 영향)

  • The purpose of this study is to examine the effect of the acute aerobic and anaerobic exercise on immune response. Fourteen university soccer players are sampled and divided into two groups -- one group of seven for aerobic exercise, the other for anaerobic exercise. The one group of aerobic exercise went through the Graded Exercise Test of Bruce protocol, using Treadmill, and were driven to be all-out ; the other of anaerobic exercise took the Wingate Test. Blood the respective periods of rest, post exercise, and recovery for ten minutes. The results are as follows. (1) The acute aerobic and anaerobic exercise don't affect monocyte and granulocyte. (2) The change in lymphocyte and NK cell resulting from the acute aerobic and anaerobic exercise shows a significant differences in the three different periods, but not significant differences between the two groups.

The Effect of a Pulsed Electromagnetic Field with Time on Pain in Muscle Crushed Rat Model

  • Acute injuries to skeletal muscles can lead to significant pain and disability. Muscle pain results in muscle weakness and range of motion (ROM) decreases. Pulsed electromagnetic fields (PEMF) promote tissue repair, healing rates and reduce musculoskeletal pain. The results of many previous studies suggest that PEMF can contribute to chronic pain reduction, particularly in musculoskeletal injurys. However, we do not have enough information of its effects compared to a placebo. The principal objective of this study was to investigate differences in acute pain induced by the direct destruction of muscle tissue (extensor digitorum) with varying times of the application of PEMF, measured through the expression of c-fos on the spinal cord. Significant reduction of pain was found in groups exposed to PEMF and the group exposed to PEMF immediately after muscle injury showed the most significant differences. In conclusion, PEMF may be a useful strategy in reducing acute pain in muscle injury.

  • Objectives: This study was to evaluate the effects of herbal medication and acupuncture therapy for children with acute appendicitis diagnosed by abdominal ultrasonography. Methods: Kamieuiyeeintang, a Korean Herb medicine, and acupuncture were given for four days. The abdominal ultrasonography and VAS scale were used to check the improvements. Results: Right lower abdominal pain, nausea and anorexia were totally gone after the treatments, and the abdominal ultrasonography showed normal. Conclusions: The result of this study showed that Kamieuiyeeintang medication and acupuncture treatment have cured acute appendicitis.

Nephroprotective Effects of Opi-san Aqueous Extracts on Cisplatin-induced Rat Acute Renal Failure (오피산(五皮散)이 Cisplatin으로 유발(誘發)된 흰쥐의 급성신부전(急性腎不全)에 미치는 영향(影響))

  • Purpose : This study was to observe the nephroprotective effects of the traditional prescription, Opi-san(OPS). OPS has been generally used for treating various edematous renal diseases, including acute renal failure. Methods : Three different dosages of OPS extracts were administered once a day for 28 days (5 mg/kg, single intraperitoneally administered). On the 23rd day after OPS extract treatment, cisplatin was treated. All the rats (6 groups of 8 rats each) were checked as follows. Changes of body weight, kidney weight, serum BUN and creatinine levels were observed, as well as changes to the kidney MDA and GSH contents. The results were compared with captopril 100 mg/kg, of which the effects on cisplatin-induced acute renal failures were already confirmed. Results : Cisplatin-induced ARF were induced by oxidative stress and related lipid peroxidation in the present study. However, these ARF and inhibition of antioxidant effects induced by cisplatin were dose-dependently reduced by treatment of OPS extracts except in the 100 mg/kg treated group. The effects of OPS extracts at 500 mg/kg were similar to those of 100 mg/kg of captopril. Conclusion : This study suggests that OPS extracts 500 mg/kg showed favorable effects on the cisplatin-induced rat's ARF as similar to 100 mg/kg of captopril.

Antiarrhythmic effects of ginsenoside Rg2 on calcium chloride-induced arrhythmias without oral toxicity

  • Background: Malignant arrhythmias require drug therapy. However, most of the currently available antiarrhythmic drugs have significant side effects. Ginsenoside Rg2 exhibits excellent cardioprotective effects and appears to be a promising candidate for cardiovascular drug development. So far, the oral toxicity and antiarrhythmic effects of Rg2 have not been evaluated. Methods: Acute oral toxicity of Rg2 was assessed by the Limit Test method in mice. Subchronic oral toxicity was determined by repeated dose 28-day toxicity study in rats. Antiarrhythmic activities of Rg2 were evaluated in calcium chloride-induced arrhythmic rats. Antiarrhythmic mechanism of Rg2 was investigated in arrhythmic rats and H9c2 cardiomyocytes. Results: The results of toxicity studies indicated that Rg2 exhibited no single-dose (10 g/kg) acute oral toxicity. And 28-day repeated dose treatment with Rg2 (1.75, 3.5 and 5 g/kg/d) demonstrated minimal, if any, subchronic toxicity. Serum biochemical examination showed that total cholesterol in the high-dose cohort was dramatically decreased, whereas prothrombin time was increased at Day 28, suggesting that Rg2 might regulate lipid metabolism and have a potential anticoagulant effect. Moreover, pretreatment with Rg2 showed antiarrhythmic effects on the rat model of calcium chloride induced arrhythmia, in terms of the reduced duration time, mortality, and incidence of malignant arrhythmias. The antiarrhythmic mechanism of Rg2 might be the inhibition of calcium influx through L-type calcium channels by suppressing the phosphorylation of Ca2+/calmodulin-dependent protein kinase II. Conclusion: Our findings support the development of Rg2 as a promising antiarrhythmic drug with fewer side effects for clinical use.

Inhibitory Effects of Acute and Chronic Allergic Reaction by Anal Therapy of Gamiseunggal-tang (加味升葛湯의 肛腸療法에 의한 急慢性 알레르기 反應 抑制效果)

  • Gamiseunggal-tang(G-tang) has long been used for the treatment of acute and chronic allergic diseases(especially, urticaria) in oriental medicine. But, It's mechanism of action is not well investigated. Anal therapy is another way of taking medicine, which is not used in common situation but available for the treatment of colon and anal diseases. It is also used in GI tract diseases, Gynecological diseases, and pediatric diseases and so on. It is especially benefitable for patients who are in such situations as coma, severe vomiting, and swallowing difficulty. In this study, the author investigated the effects of G-tang by anal therapy on acute and chronic allergic reactions in murine model. The results obtained are as follows: 1. G-tang(0.01, 0.1 g/kg) inhibited the compound 48/80-induced ear swelling response in mice. Inhibitory effects of G-tang was significant (P<0.05) at the dose of 0.01 g/kg. 2. G-tang(0.001-0.1 g/kg) inhibited the cutaneous allergic reaction activated by anti-dinitrophenyl(DNP) IgE in rats. Especially, G-tang 0.01, 0.1 g/kg inhibited the cutaneous allergic reaction significantly. 3. G-tang(0.01-1 g/L) dose-dependently inhibited the compound 48/80-induced histamine release from the peritoneal mast cells. 4. G-tang(0.001-0.1 g/L) had a inhibitory effect on anti-DNP IgE-induced tumor necrosis factor-${\alpha}$ production. Above results indicate that anal therapy of G-tang may be beneficial in the treatment of acute and chronic allergic diseases.

Effects of Yuhyangjeongtong-san on the Carrageenin-induced Acute Inflammation and Adjuvant-induced Arthritis (유향정통산이 Carrageenin 유발 급성 염증과 Adjuvant 유발 관절염에 미치는 영향)

  • Objectives This study was to investigate the effects of Yuhyangjeongtong-san on the carrageenin-induced acute inflammation and adjuvant-induced arthritis in rats. Methods Acute inflammation was induced by injection of 2% carrageenin 0.15 ml into right hind foot of rats. Control group was carrageenin injected and taken distilled water, treated group was carrageenin injected and taken Yuhyangjeongtong-san by orally once. 2 hours after injection, plantar temperature and paw volume were measured. 3 hours after, counts of white blood cell (WBC) were performed. Arthritis was induced by injection of complete freund's adjuvant (CFA) into base of tail. Control group was CFA injected and taken distilled water, treated group was CFA injected and taken Yuhyangjeongtong-san by orally for 10 days. 0,5,10 day, body weight, thickness of ankle joint and paw edema were measured. 10 day, counts of WBC, $interleukin-1{\beta}$ ($IL-1{\beta}$) and tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) level were analysed. Histochemical study of NADPH-d and immunohistochemical study of c-fos were performed. Results In the carrageenin-induced acute inflammation, neutrophils of treated group were significantly decreased compared with control group. In the adjuvant-induced arthritis, paw edema, total counts of WBC and TNF-${\alpha}$ level in paw exudates were significantly decreased compared with control group, and the number of c-fos positive neurons of treated group was significantly decreased compared with control group. Conclusions On the basis of these results, we concluded that Yuhyangjeongtong-san have anti-arthritic and anti-inflammatory effects.
