Oral Acute and Subacute Toxicity Studies of Decursin and Decursinol Angelate of Angelica gigas Nakai

  • In this study, we assessed the acute and subacute toxicity of Angelica gigas Nakai (A. gigas Nakai) extracts, which are comprised of decursin and decursinol angelate (D/DA) in rats. For the oral acute toxicity test, Sprague-Dawley (SD) male and female rats were gavaged with two doses of D/DA (200 and 2,000 mg/kg body weight) and then observed for any toxic symptoms for 2 weeks. The LD$_{50}$ value for the rats was greater than 2,000 mg/kg body weight for both male and female rats, which indicates that there were no toxic symptoms induced by doses of up to 2,000 mg/kg body weight. For the subacute toxicity study, rats were treated with D/DA at doses of 2 and 20 mg/kg body weight once a day for 30 days. There were no significant changes in body weight and food intake observed during the subacute toxicity study. In addition, no differences were observed between the control and treated groups when urinalysis was conducted or when hematology and biochemical parameters were evaluated. Finally, histopathological examination of the organs did not reveal any lesions in the control or treated groups. Taken together, these findings indicate that D/DA is safe and non-toxic.

Acute pancreatitis in hand, foot and mouth disease caused by Coxsackievirus A16: case report

  • Coxsackievirus A16 (CA16), which primarily causes hand, foot, and mouth disease (HFMD), is associated with complications, such as encephalitis, acute flaccid paralysis, myocarditis, pericarditis, and shock. However, no case of pancreatitis associated with CA16 has been reported in children. We report a case of CA16-associated acute pancreatitis in a 3-year-old girl with HFMD. She was admitted because of poor oral intake and high fever for 1 day. Maculopapular rashes on both hands and feet and multiple vesicles on the soft palate were observed on physical examination. She was treated conservatively with intravenous fluids. On the fourth hospital day, she had severe abdominal pain and vomiting. The serum levels of amylase and lipase were remarkably elevated (amylase, 1,902 IU/L; reference range, 28-100 IU/L; lipase, >1,500 IU/L; reference range, 13-60 IU/L), and ultrasonography showed diffuse swelling of the pancreas with a small amount of ascites. The real-time reverse transcription polymerase chain reaction result from a stool sample was positive for CA16. CA16 can cause acute pancreatitis, and should be considered in the differential diagnosis of abdominal pain in children with HFMD.

Acute beetroot juice supplementation does not attenuate knee extensor exercise muscle fatigue in a healthy young population

  • [Purpose] The effect of acute nitrate supplementation on muscle fatigue is largely unknown. This study aimed to evaluate the effect of acute nitrate supplementation on muscle fatigue. [Methods] Thirty-five recreationally active subjects consumed 140 ml of beetroot (BR) juice (nitrate: 8 mmol·d-1) or placebo (PL) 12 and 2.5 hours before two exercise sessions. Peak torque was measured during 50 repetitions, at maximal effort, and during concentric knee extensions at 90°·s-1. Blood pressure (BP) was recorded pre- and post-exercise. [Results] Peak torque, maximum work, rate of fatigue, and rate of work fatigue were similar between the BR and PL conditions. Post-exercise diastolic BP (BR: 67.2 ± 9.8 vs. PL: 64.5 ± 7.9 mmHg, p < 0.05) and mean arterial pressure (BR: 91.6 ± 9.3 vs. PL: 88.8 ± 8.2 mmHg, p < 0.05) were higher with BR supplementation. [Conclusion] These findings suggest that the acute intake of BR juice had no effect on knee extensor muscle strength or fatigue but increased BP in a healthy recreationally active population.

THE BIDAS-2007: BIOASSAY DATA ANALYSIS SOFTWARE FOR EVALUATING A RADIONUCLIDE INTAKE AND DOSE

  • Bioassay data analysis software (BiDAS-2007) has been developed by KAERI, which adds several new functions to its previous version. New functions of the BiDAS-2007 computer code enable the user not only to do a simultaneous analysis by using two or more types of bioassay for the best internal dose evaluation, but also to do a continual internal dose evaluation from a change of the internal exposure conditions such as an intake type (acute, chronic), an intake pathway (inhalation, ingestion), an absorption type (Type F, M, S), and a particle size (AMAD, activity median aerodynamic diameter), and also to estimate the intakes in various conditions of an internal exposure at a time. The values calculated by the BiDAS-2007 code are consistent and in good agreement with those values by IMIE-2004 code by Berkovski and IMBA code by Birchall. The BiDAS-2007 computer code is very useful and user-friendly to estimate the radionuclide intakes and committed effective doses of a radiation worker.

  • 본 연구에서는 인체 내에서 방사성핵종의 거동에 관하여 국제방사선방호위원회에서 권고한 최근 호흡기 모델, 소화기 모델 및 생체동역학 모델을 사용하여 생물학적분석 자료로부터 섭취량과 예탁유효선량을 평가하기 위한 BIDAS 프로그램을 개발하였다. 프로그램은 생물학적분석 자료를 관리하는 데이터베이스 모듈, 각 방사성 핵종에 대한 예측 생물학적분석 양을 내장하고 있는 모듈, 측정된 생물학적분석 양에 근거하여 급성 및 만성 피폭으로부터 섭취량과 선량을 평가하는 계산모듈 등으로 구성되어 있다. 본 논문은 프로그램의 특성과 검증결과에 대해 기술한다.

Short- and Long-term Effects of Potassium on Renin-Aldosterone System in Hypertensive Rats Fed with Different Amounts of Sodium

  • To evaluate the acute and chronic effects of potassium on aldosterone response to different amounts of sodium intake, two series of experiments were conducted. In the first series of experiments, when the plasma K level was increased acutely by KCI infusion $(20\;{\mu}g/kg/min\;for\;20 min)$, plasma aldosterone concentration increased in both low Na and high Na groups. However, the aldosterone response to K infusion was significantly greater in the low Na than in the high Na groups. In the second series of experiments, rats fed a high K diet chronically showed a significantly higher plasma K level than those fed a low K diet. However, plasma Na level was maintained relatively constant independent of the Na intake. Both the plasma renin activity (PRA) and aldosterone levels were inversely related to the Na intake. There was a highly positive correlation between aldosterone level and PRA over a wide range of sodium intakes. However, the slope of the correlation line was distinctly steeper in the K-repleted than in the K-depleted rats. The above results indicate that the adrenotropic action of acute K load was augmented in the presence of high plasma renin levels. However, when plasna K level was elevated chronically by a high-K diet, aldosterone secretion was markedly stimulated, although the plasma lenin levels were suppressed.

Impact of Clinical, Laboratory and Fluid Therapy Variables on Hospital Length of Stay for Children with Acute Pancreatitis

  • Purpose: There have been many efforts to develop generalizable severity markers in children with acute pancreatitis (AP). Expert opinion panels have developed consensus guidelines on management but it is unclear if these are sufficient or valid. Our study aims to assess the effect of clinical and laboratory variables, in addition to treatment modality on hospital length of stay (LOS) as a proxy variable for severity in pediatric patients admitted with AP. Methods: We conducted a retrospective chart review of patients between ages of 0-18 years, who were admitted with AP at 2 institutions between 2013-2018, John R. Oishei Children's Hospital (Buffalo, NY, USA) and Medical University of South Carolina Children's Hospital (Charleston, SC, USA). We constructed three linear regression models to analyze the effect of clinical signs of organ dysfunction, laboratory markers and fluid intake on hospital LOS. Results: Ninety-two patients were included in the study. The mean age was 12 years (range, 7.6-17.4 years), 55% were females, and median LOS was 3 days. The most frequent cause of AP was idiopathic. Our study showed that elevated blood urea nitrogen (BUN) on admission (p<0.005), tachycardia that lasted for ≥48 hours (p<0.001) and need for fluid resuscitation were associated with increase LOS. Total daily fluid intake above maintenance did not have a significant effect on the primary outcome (p=0.49). Conclusion: Elevated serum BUN on admission, persistent tachycardia and need for fluid resuscitation were associated with increase LOS in pediatric AP. Daily total fluid intake above recommended maintenance did not reduce LOS.

Effect of acute heat stress on feed intake and plasma concentrations of tumor necrosis factor-α and free amino acids in growing pigs

  • Exposure to heat stress (HS) has negative effects on pig production and health. Plasma concentrations of amino acids (AAs) can be used as indicators of HS. Therefore, this study was conducted to evaluate the effect of acute HS on feed intake, water drink, and plasma AAs in pigs. A total of 6 growing pigs (n = 6, 3 boars and 3 gilts) were raised in thermal neutral (TN; 25℃) conditions for the 5-d adaptation period as a control. After the adaptation, pigs were exposed to HS at 33℃ (HS33) for 24 h. All the pigs were fed the same diet formulated to meet or exceed predicted requirements during the whole experimental period. Blood samples were collected after the adaptation and heat treatment to verify the AAs. Measurements were the average daily feed intake (ADFI), average daily water intake (ADWI), water loss, tumor necrosis factor (TNF)-α, nitrate, total nitric oxide, and AAs in the plasma samples. Data were analyzed using the PROC GLM of SAS. HS33 had a lower ADFI (p < 0.05) and a tendency for an increased ADWI and water loss compared to TN. The level of TNF-α was significantly decreased (p < 0.05) in HS33. HS33 had a lower concentration of histidine and sarcosine compared to TN; however, there were no differences in the levels of nitrates and total nitric oxide. In conclusion, the results of this study show the differential effect of HS on the plasma concentration of AAs. We expect that the changed AAs could be potential indicators of HS.

뇌졸증 후 급성기 운동이 뇌허혈 유발쥐의 뒷다리근 질량에 미치는 영향 (Effect of exercise during acute stage of stroke on affected and unaffected hindlimb muscle mass of cerebral ischemic rat)

  • The purpose of this study was to identify the effect of cerebral ischemia on affected(Lt) and unaffected(Rt) side of soleus, plantaris and gastrocnemius muscle mass and determine the effect of exercise on affected and unaffected side on soleus, plantaris and gastrocnemius muscle mass during acute stage of stroke. Sixteen male Sprague-Dawley rate with 200-270g body weight were randomly divided into three groups: control, stroke, and exercise after stroke(St+Ex) group. The control group received sham operation and the stroke group and St+Ex group received transient right MCA(middle cerebral artery) occlusion operation. The St+Ex groups ran on a treadmill for 20min/day at 10m/min and $10^{\circ}$ grade for 6days. During the experimental period body weight and diet intake was measured every morning. On the 7th day after operation, muscles were dissected from both affected and unaffected side of hindlimb. Cerebral infarction of stroke and St+Ex groups were identified by staining with TCC for 30 minutes. The data were analyzed by Kruskal-Wallis test and Mann-Whitney U test using the SPSSWIN 9.0 program. Significance was accepted at the level of p<0.05. The results were summarized follows : 1) There were no significant difference of the body weight on the first day of experiment among 3 groups. Whereas on the 7th day, the body weight of both stroke group and St+Ex group were significantly smaller than that of control group. Body weight of St+Ex group on the 7th day tended to be larger than that of stroke group. 2) Total diet intake of both stroke group and St+Ex group were also significantly smaller than that of control group. While total amount of diet intake in St+Ex group tended to be larger than that of stroke group. 3) The weight of gastrocnemius muscle of affected side in stroke group significantly decreased compared to that of control group and the weight of soleus and plantaris muscle of affected side in stroke group tended to decrease compared to that of control group. 4) The weight of plantaris muscle of unaffected side in stroke group significantly decreased compared to that of control group and the weight of soleus and gastrocnemius muscle of unaffected side in stroke group tended to decrease compared to those of control group. 5) The weight of gastrocnemius muscle of affected side in stroke group significantly decreased compared to that of unaffected side and there was no significant difference of the weight of soleus and plantaris muscle in stroke group between affected side and unaffected side. 6) The weight of soleus, plantaris and gastrocnemius muscle of both affected side and unaffected side in St+Ex group had a tendency of increase compared to those of stroke group. The relative weight of soleus and gastrocnemius muscle of affected side and soleus muscle of unaffected side in St+Ex group had a tendency to increase compared to those of stroke group. Based on these results, exercise during acute stage of stroke might attenuate muscle atrophy of both affected and unaffected side of hindlimb muscles.

Preliminary pharmacological evaluation of an Ayurvedic formulation Dasamularista

  • In this study the Ayurvedic formulation Dasamularista was studied for its preliminary pharmacological properties using laboratory mice. Dasamularista showed a decrease in food intake and stool formation, while the water content of stool and water intake was higher and the volume of the urine was less. Dasamularista in a slight extent reduced the intestinal motility. This constipating effect was further supported by the significant anti-diarrhoeal property of the formulation in castor oil induced dairrhoea. The tested formulation markedly increased the latent period of diarrhoea and reduced the purging index value. Dasamularista did not alter the acetic acid induced abdominal writhing. Significant reduction on the onset of sleeping time and increased duration of sleep was observed in pentobarbital induced sleeping time test.
