Objectives : The purpose of this study is to investigate whether Haenggan(LR2) Reduction in Acupuncture affects cerebral hemodynamics〔regional cerebral blood flow(rCBF), mean arterial blood pressure(MABP)〕in normal rats, and to make manifest whether Haenggan(LR2) Reduction in Acupuncture is mediated by cyclooxygenase or guanylate cyclase. Methods : This experiments was to investigate at the other changes of rCBF and MABP at Haenggan(LR2) Reduction in Acupuncture in normal rats, pretreated rats with indomethacin(1 mg/kg, i.v.) and pretreated rats with methylene blue$(10\;{\,u}g/kg,\;i.v.)$. Results : 1. Haenggan(LR2) Reduction in Acupuncture was significantly increased rCBF during acupuncture and after withdrawing of the needle. 2. Haenggan(LR2) Reduction in Acupuncture was decreased MABP during acupuncture, but Haenggan(LR2) Reduction in Acupuncture was increased MABP in compared with normal condition. 3. Pretreatment with indomethacin(1 mg/kg, i.v.) was significantly inhibited Haenggan(LR2) Reduction in Acupuncture induced increase of rCBF, but was increased Haenggan(LR2) Reduction in Acupuncture induced increase of MABP. 4. Pretreatment with methylene blue$(10\;{\mu}g/kg,\;i.v.)$ was significantly decreased Haenggan(LR2) Reduction in Acupuncture induced increase of rCBF and MABP. This results suggest that Haenggan(LR2) Reduction in Acupuncture increased rCBF by dilating pial arterial diameter, and the mechanism of Haenggan(LR2) Reduction in Acupuncture is mediated by guanylate cyclase.
Experimental Study of Taegye($KI_3$) Taebaek($SP_3$) Draining Acupuncture on the Improvement of Cerebral Blood flow and Arterial Blood Pressure. Objectives : This study was designed to investigate the effects of acupuncture on Taegye($KI_3$) Taebaek($SP_3$) draining and determine the mechanism of action of PCBL by measuring the changes of regional cerebral blood flow (rCBF) and mean arterial blood pressure (MABP) in normal rats. Methods : This study was designed to investigate whether acupuncture on Taegye($KI_3$) Taebaek($SP_3$) draining affect Regional cerebral blood flow(rCBF), Mean arterial blood pressure(MABP) in normal rats. To make manifest whether acupuncture on Taegye($KI_3$) Taebaek($SP_3$) draining was mediated by inhibitor of cyclooxygenase or guanylate cyclase which diate arterial diameter. Results 1. Acupuncture on Taegye($KI_3$) Taebaek($SP_3$) draining significantly increased rCBF but decreased MABP. This result suggests that Acupuncturing on Taegye($KI_3$) Taebaek($SP_3$) draining might significantly increase rCBF by dilating arterial diameter. 2. Acupuncture on Taegye($KI_3$) Taebaek($SP_3$) draining induced increase of rCBF was significantly inhibited by pretreatment with indomethacin (1 mg/kg, i.p.), an inhibitor of cyclooxygenase. 3. This result suggests that the action of Acupuncture on Taegye($KI_3$) Taebaek($SP_3$) draining might be mediated by cyclooxygenase.
The purpose of this study was finding the pain inhibitory effect of acupuncture at $LI_4$. The pain at dentes incisivi was evoked by noxious electric stimulation and EMG changes based on the interval were measured. To do this, the opioid antagonist was administered intraperitoneally. Following are some of results. 1. The pain inhibitory effect of acupuncture at $LI_4$ was 94.35%, 84.56%, 57.62%, 54.40%, at 10, 20, 30 and 40 minutes respectively. All the data were calculated based on 100% at 0 minute. The effect was very significant at 40 minutes. 2. The continuous effect of acupuncture at $LI_4$ after taking out the needle was 58.58%, 57.62%, 66.22%, 73.18%, 83.70%, 92.68%, at 50, 50, 70, 80, 90 and 100 minutes respectively. The overall continuous effect was shown its maximum 20 minutes after taking out the needle and reached to the initial value at 60 minutes. 3. The pain inhibitory effect following the naloxone administration and acupuncture application at $LI_4$ was 95.96%, 96.04%, 94.86%, 94.92% at 10, 20, 30 and 40 minutes respectively. All values showed similar tendency to the initial data. 4. The continuous effect of acupuncture at $LI_4$ after taking out the needle which was preceded by the naloxone administration was 94.48%, 96.02%, 96.02%, 98.00%, 98.46%, 97.18% at 50, 60, 70, 80, 90 and 100 minutes respectively. This trend was not a significant fluctuation. It was concluded that effect of acupuncture at $LI_4$ was shown in conjunction with secretion of analgesic substance. Therefore it is implied that the acupuncture application will play a major role in treating many diseases with more revelation of scientific acupuncture mechanism. Further studies of acupuncture manipulation are needed in the future based on our study.
Objectives : The aim of the present study is to observe effects of acupuncture related with the nerve system, Inferior mesenteric nerve activity(IMNA) was examined on BL60 acupuncture closely connected with the bladder and HT8 with little tied. Materials and methods : Acupunctures were performed during 100 seconds after stabilization. IMNA was measured by micromanipulator, preamplifier, mean arterial pressure(MAP) and heart rate were observed by blood pressure transducer, 4-channel transducer amplifier in anesthetized rat. Results : On BL60 acupuncture, the heart rate was not significantly decreased but IMNA, MAP were significantly decreased and Maximum peak of IMNA was increased during inserted acupuncture, decreased after ejected acupuncture respectively. On HT8 acupuncture, IMNA was decreased during acupuncture than before acupuncture but the others did not showed signigicant difference. Conclusions : Our results demonstrated a meridian interaction between BL60 acupoint in the bladder channel and Inferior mesenteric sympathetic nerve related to the bladder. This fact suggest that BL60 acupuncture have effect on the bladder through the nerve system. Meanwhile, IMNA was decreased during HT8 acupuncture. This result is supposed HT8 to have not meridian function but acupoint function by another mechanism. It needs to be closely examined other meridians and nerve by the optimal approaching method.
The frequency of the hypertension is increasing as the life level is improved and an average span of the life is extended since we approached modern stage. The hypertension is also dangerous disease which raises fatal complication for example with the bleeding aproplexy and the ischemic attack. The medicinal treatment about the hypertension is required patients to take continually. The acupuncture have been introduced because a medicine-chemical treatment hasn't good influence on the human body. It recently has been processed that studies acupuncture effect for blood pressure and have been found out that blood pressure go down. The object of this study observe the effect of LR3 acupuncture on hypertension in Renal Hypertension RAT induced by 2K1C. The 2K1C model was based on renin-angiotensin system. We put the silver clip in renal artery to induce renal hypertension. We try to observe that LR3 acupuncture influence on the blood pressure and c-fos expression in CVLM, NTS, RVLM. In results, the blood pressure was decreased during acupuncture than before acupuncture, after acupuncture. The heart rate was also decreased during acupuncture than before acupuncture, after acupuncture. The LR3 acupuncture significantly effects on blood pressure and heart rate (P<0.05). The increased expression of c-fos was shown in CVLM, NTS, but not in RVLM. In conclusion, LR3 was relived the action of control upon the hypertension and related with medulla, particularly CVLM, NTS. It needs to be closely examined pharmacological mechanism and studied combination with other acupoints.
Objective : The purpose of this study is to examine the tendency of experimental studies on the physiological effects of acupuncture at $ST_{36}$(Zusanli). Methods : We investigated 69 theses (10 Korean and 59 international) which were searched by the keyword 'Zusanli' through PubMed website, and that were experimented with manual acupuncture or electroacupuncture on healthy human subjects or normal animals. Results : The 69 theses were classified into iou groups based on the main topic which is related with the influence of acupuncture stimulation at $ST_{36}$(Zusanli), such as digestive system, nervous system, immune system, and cardiovascular system. The main results found in this study are : 1. Acupuncture at $ST_{36}$(Zusanli) increases gastric mobility. And such effect is related with vagal nerve and opioid pathway. 2. Acupuncture at $ST_{36}$(Zusanli) is valuably related with cerebral cortex. And it influences on the cerebrum activities. 3. Acupuncture at $ST_{36}$(Zusanli) has noticeable analgesic effect, which is related with Opioid mechanism, Gate control and SP 4. Acupuncture at $ST_{36}$(Zusanli) increases immunity Conclusion . Acupuncture at $ST_{36}$(Zusanli) has many effects on digestive system, nervous system, immune system, and cardiovascular system.
Objectives : Neurological disorders have been one of main therapeutic targets of acupuncture. The present study investigated the protective effects of Gwakhyangjeonggisan herbal acupuncture solution (GHAS). Methods : We performed 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay in glioblastoma cells, and did microarray analysis with cells exposed to reactive oxigen species (ROS) of hydrogen peroxide by 8.0 k Human cDNA, with cut-off level of 2-fold changes in gene expression. Results : MTT assay showed protective effect of GHAS on the glioblastoma cells exposed to hydrogen peroxide. When glioblastoma cells were exposed to hydrogen peroxide, 24 genes were downregulated. When the cells were pretreated with GHAS before exposure to hydrogen peroxide, 46 genes were downregulated. Many of the genes downregulated by hydrogen peroxide stimulation were decreased in the amount of downregulation or reversed to upregulation. Conclusions : The gene expression changes observed in the present study are supposed to be related to the protective molecular mechanism of GHAS in the glioblastoma cells exposed to ROS stress.
Background : Acupuncture has been used as a clinical treatment in Oriental medicine for various diseases including pain relief. The descending pain control system of periaqueductal gray (PAG) is a powerful pain control system in mammalians. Expression of c-Fos is used as a marker for stimuli-induced metabolic changes of neurons. Objective : In the present study, the effects of acupuncture on analgesic effect in visceral pain were investigated through the writhing reflex and c-Fos expression in ventrolateral PAG (vlPAG) area using immunohistochemistry in mice. Method : For the writhing test, mice were divided into five groups. Immediately after finishing the behavioral test, the animals were weighed and overdosed with Zoletil. After a complete lack of response was observed, the brains of the mice were dissected into serial coronal sections, and c-Fos immunohistochemistry was performed. Statistical analysis of all data was performed using one-way ANOVA. Result : The present results showed that acupuncture affected the writhing reflex and that Choksamni (zusnali) acupoint and aspirin significantlysuppressed acetic acid treatment-induced increased writhing reflex, and the expression of c-Fos in vlPAG was significantly increased in the acupunctured group. Conclusion : The present study suggests that acupuncture has an antinociceptive effect on acetic acid-induced visceral pain by increase of c-Fos expression in mice. Aspirin also showed analgesic effect, however the mechanism is different from the acupuncture.
Objective: The aim of this study was to investigate the effect of acupuncture on migraine prophylaxis. Method: Articles were searched from the online databases: PubMed, OASIS, NDSL, and RISS. The search keywords were "migraine prophylaxis" and "acupuncture." The inclusion criterion was randomized clinical trial. The exclusion criteria were in vivo, vitro, and review articles. Results: Eighty-nine articles were found among those published until March 2019. After reviewing the title, abstract, and original article, 10 articles were selected to show the effect of acupuncture for migraine prophylaxis. Conclusion: The effect of acupuncture for the frequency and number of days of a migraine and for the intensity of the pain seems to be effective, but the mechanism does not appear to be clear. Therefore, further research is needed in future acupuncture for migraine prevention.
Acupuncture has been used for treatment of numerous diseases, especially for pain control in the oriental culture. However, the mechanism of pain control by acupuncture was not clear so far. The present study was examined that the effects of electro-acupuncture (EA) applied to the acu-point of extra-segmental area on modulation of formalin induced pain in Sprague - Dawley rats. In order to apply EA to acu-points in the plantar area of right fore paws, a pair of teflon - coated stainless steel wires were implanted in HT 7 (shin-mun) and PC 7 (dae-neong) 5 days before behavioral test. A behavioral test was performed by means of video camera after injection of 5% formalin ($50{\mu}l$) into the lateral plantar region of left hind paw. EA was delivered by a constant current stimulator at 4~5 mA, 2 ms, and 10 Hz for 30 min. The electromyographic activities were recorded in the biceps femoris muscle under chloral hydrate anesthesia. Test stimuli with 1~9mA were applied to the sural nerve territory including the medial portion of the 4th toe and the lateral portion of the 5th toe. Behavioral responses including favoring, flinching and bitting were occured in the biphasic pattern, such as the lst phase (0~5 min) and the 2nd phase (20~45 min) after formalin injection. However, EA (4~5 mA, 2 ms, 10 Hz) significantly inhibited Che behavioral responses. EMG activities of flexor reflex had a latency of 100~300 ms and thresholds of test stimuli for EMG were 4~5 mA in normal rats. Injection of formalin decreased threshold of test stimuli and increased EMG activities for 2hrs after injection. However, EA significantly inhibited EMG activities of flexor reflex increased by formalin and recovered EMG evoked thresholds. These results suggest that contralateral extra-segmental EA inhibits the first and second phases of formalin induced pain but their mechanism be needed to examine additionally.
