Purification and gene cloning of .alpha.-amylase of neurospora crassa (Neurospora crassa에서 알파아밀라제의 정제 및 유전자의 클로닝)

  • $\alpha$-Amylase (EC.3.2.1.1) of Neurospora crassa (ATCC9279) was cloned in E. coli HB101 using shotgun method, and the enzymes isolated from both N. crassa and E. coli were compared. Chromosomal DNA isolated from the spores of N. crassa was partially digested with PstI restriction endonuclease and rejoined to pBR322 which had been digested with the same enzyme. The resulting recombinant DNA were introduced into E. coli HB101 which had competancy by treating with $CaCl_{2}$. As the result, about 8000 colonies which showed tetracycline resistance were selected and two of the colonies which had 13.5Kb recombinant plasmid exhibit starch degrading activity on starch-containing plate when treated with D-cycloserine. $\alpha$-Amylases from both N.crassa and E. coli were isolated by using ammonium sulfate precipitation, DEAE-cellulose ion exchange column chromatography and Bio-Gel P150 gel foltration column. As the result, about 81.3 fold and 5.6 fold purifications in specific activities were obtained respectively, and specific activities of the gel filtrates were 6.1u/mg and 85u/mg respectively. The properties of both enzymes were compared and they showed quite the similar patterns in optimal temperature, optimal pH and had same molecular weight about 100,000 daltons on gel filtration method. Optimal temperatures for both enzymes were $70^{\circ}C$ and optimal pH were about 6 and 10.

Hypothermia Regulates Endoplasmic Reticulum (ER) Stress through the X-box Binding Protein-1 (XBP1) Gene Expression in PC12 Cells

  • Endoplasmic reticulum (ER) stress induces unfolded protein response (UPR) via inositol-requiring enzyme 1 (IRE1) activation, which sends a molecular signal for X box-binding protein 1 (XBP1) mRNA splicing in the cytosol. IRE1 endoribonuclease activity induces cleavage of XBP1 mRNA. The XBP1 mRNA is then ligated by an uncharacterized RNA ligase and translated to produce spliced XBP1 by 23 nt removed in which contains the PstI restriction enzyme site. The splicing of XBP1 mRNA can be detected by semiquantitative RT-PCR, and then splicing of XBP1 is a useful tool to measure the genetic variability in ER stress. In this study, we have estimated IRE1-dependent splicing of XBP1 mRNA under conditions of various hypothermia. The results indicated that hypothermia regulated ER stress. This study demonstrated that hypothermia is closely related to ER stress and may be useful for early diagnosis of ER-associated disease.

A Comparative Study on the Effects of GMFM and ICF Sub-item Function on the Sub-item Activity and Participation Restriction of the ICF (GMFM과 ICF의 하위영역 기능이 ICF의 하위영역 활동과 참여문제에 미치는 영향 비교)

  • Background: The purpose of this study is to explore the effect of the functions of GMFM and ICF-CY on the activities and participation of ICF-CY sub-items. Design: Cross-sectional study. Method: This study compared and analyzed 95 children with cerebral palsy [type of CP: spasticity 86 (90.5%), hypotonia 4 (4.2%), mixed 5 (5.3%); type of palsy: quadriplegia 13 (13.7%), diplegia 71 (74.7%), hemiplegia 11 (11.6%)] using sub-items of functions, activities and participation from GMFM and ICF-CY. Result: The results show that the activities and participation of ICF-CY (9 sub-items) have significant effect on the functions of GMFM and ICF-CY (8 sub-items) (p<0.05). Conclusion: It is intended to provide data to establish practical therapeutic goals and interventions for functions, activities and participation, which are sub-categories of ICF-CY in cerebral palsy.

  • In the past the pattern of business down-trend usually appeared in the form of, first, decrease in facility investment, then decrease in inventory level, followed by reduced level of consumption. But the pattern nowadays is becoming just the opposite, that is, first, consumption decrease, then inventory level increase, followed by restriction of facility investment. Also in the past, the greater effort was placed in strengthening of hardware areas through optimization and modernization of production means on the premise of sales. But lately software areas take most of the main effort to establish production mean with sales as its objective. Under these circumstances one of the real problems facing production activities today is the conflicting relationship between sales and production functions. This occurs due to differences of their view points. Then, in order to achieve maximum profit at the least cost, which is the ultimate objective of a production activity, the need arises to effectively coordinate sales demand and plant production capacity. For this purpose strong control means and function must be devised. In our case study example we illustrate a management technique for a combined planning function, of optimal coordination of product mixes utilizing a computerized linear programming model as control means of attaining maximum profit. It is hoped that this example help achieve some of corporate objectives.

The Effect of Father's Parenting Behavior, Parenting Involvement and Father-Child Communication on Children's Multiple Intelligence (아버지의 양육행동, 양육참여도 및 아버지-자녀간 의사소통이 아동의 다중지능에 미치는 영향)

  • The purpose of this study was to find the effects of parenting behavior, parenting involvement and father-child communicationon on children's multiple intelligence. For this purpose, 182 children selected from three elementary schools and their fathers participated in the study. The findings of this study are as follows : First, there were some significant differences in children's multiple intelligence according to the children's grade, gender, father's educational background and income. Second, there were some significant differences in children's multiple intelligence according to the parenting behavior, warmth acceptance, rejection restriction and permissiveness nonintervention behaviors, and to leisure activity, life guidance, study guidance of parenting involvement and to father-child open communication, problematic communication. Third, It was also found that children's grade, gender, father's educational background, warmth acceptance behavior, permissiveness nonintervention behaviors and open communication, problematic communication were all significant predictors of the children's multiple intelligence. In order to increase the children's multiple intelligence, parents should be warmer and more accepting and have open commnication with their children.

Cloning of the Alkaline Phosphatase Gene from Kluyveromyces fragilis

  • In order to clone the gene coding for alkaline phosphatase in the yeast Kluyveromyces fragilis, a genomic library was constructed using the yeast-E. coli shuttle vector pHN114 as a cloning vector. From the genomic library, a clone carrying the gene was isolated and the plasmid was designated as pSKH101. A restriction enzyme map was made using this plasmid. Subcloning experiments and complementation studies showed that alkaline phosphatase was active only in the original 3.1 kb insert. Southern hybridization analysis confirmed that the cloned DNA fragment was derived from K. fragilis genomic DNA. Using a minicell experiment, the product of the cloned gene was identified as a protein with a molecular weight of 63 KDa. A 0.6 kb HindIII fragment, which showed promoter activity, was isolated using the E. coli promoter-probe vector pKO-1.

Construction of a New Gene-Fusion Expression Vector, pMONSTER

  • The fur (ferric uptake regulation) expression vector pMON2064 was modified to produce a Fur-fusion expression vector. A kinker site, factor Xa cleavage site, and several restriction endonuclease sites were introduced to facilitate easy cloning and isolating of the fusion protein. The resulting fusion expression vector, pMONSTER, was then used to make fusion expression vector, pMONSTER, was then used to make fusion proteins with $\beta$-galactosidase and the protease of the human immunodeficiency virus type 1 (HIV-1 PR). Strain SW4020 harboring the Fur $\beta$-galactosidase fusion vector produced blue colonies on a 5-bromo-4-chloro-3-indolyl-$\beta$-D-galactoside plate and the resulting 133 kDa fusion protein reacted with an anti-Fur antibody. The strain harboring the Fur-HIV-1 PR fusion vector produced a 29 kDa fusion protein, which also reacted with an anti-Fur antibody. The Fur-HIV-1 PR fusion protein was purified by a single column application that was designed to isolate the Fur protein. The purified Fur-HIV-1 PR fusion protein digested with factor Xa cleaved a recombinant Gag protein to release smaller fragments, including a p24 capsid protein. The Fur-HIV-1 PR fusion protein itself did not exhibit any proteolytic activity.

A Design of Smart Cane for Visually Impaired Person (시각장애인을 위한 전자 지팡이의 설계)

  • An official report printed in June, 2005 represents that there are 180 thousands people with visual impairments in Korea, and 10 thousands of people with visual impairments are newly increased by every year since 2000. They are neglected from getting education, selecting job, and enjoying recreation activity. Above all, they have strong restriction when they are walking on the street; they are facing difficulty and inconvenience. Many smart cane products were introduced but expensive cost blocked popularization of them. In this study we have an object to development of smart cane that is chip and proper for korean street. For this, we suggest basic design of smart cane and experiment on ultrasonic sensors for visually impaired person.

Functional Status and Health Care Utilization among Elders with Hip Fracture Surgery from a Fall (낙상후 대퇴골절 수술 노인의 신체기능, 의료이용 및 낙상의 두려움)

  • Purpose: The purposes of this study were to examine 1) functional status at 2 months after hip fracture surgery 2) health care utilization after a fall episode and 3) fear of falling experienced during first 2 months after a fall episode. Method: With a convenient sample of 99 elderly from six university or general hospitals with hip fracture from a fall, data were collected at 2-3 days before discharge and at 2 months after hip fracture surgery. Result: 1) At 2 months after hip fracture from a fall, significant proportion (25.3%) of elderly was not able to walk indoors. 2) Average length of hospital stay was 27.6 days with a range of 8 to 86 days. About 51% subjects received physical therapy during hospital stay, and only 6.1% subjects received physical therapy following discharge from the hospital. 3) Significant proportion (72.7%) had fear of falling after the fall episode. About 51% reported that they restricted their activities because they had fear of falling. Conclusion: Fall is a dreaded event which result in loss of independence and restriction of activity. Development and application of fall prevention program is critical especially for those with risk factors of fall.

Wilms' tumor with polydipsia, polyuria, hyponatremic hypertension and congestive heart failure : a case report (다음, 다뇨, 저나트륨성 고혈압, 심부전증을 보이는 윌름씨 종양 1례)

  • A 3-year-old girl presented with polydipsia, polyuria, hyponatremia, hypertension and congestive heart failure. Her polyuria was unresponsive to water restriction and vasopressin challenge tests, and her blood pressure was not effectively controlled by antihypertensive drugs. Radiologic examinations revealed a Wilms' tumor in the right kidney. Her plasma renin activity and aldosterone concentration were greatly increased. After surgical removal of the tumor, the congestive heart failure disappeared. Congestive heart failure due to Wilms' tumor is very rare and we report here on such a case, with a brief review of the literature.
