Oxygen Consumption and Ammonia Excretion in Cultured Soft-Shelled Turtle, Pelodiscus sinensis Exposed Vibration Stress (진동 스트레스에 따른 양식 자라, Pelodiscus sinensis의 산소소비 및 암모니아 배설)

  • We examined the effects of the vibration stress on oxygen consumption and ammonia excretion of cultured soft-shelled turtle, Pelodiscus sinensis. For the stressed group vibration of $45{\sim}78\;dB$ (V) from electric vibrator was turned on for 15 min with 2-h intervals during daytime hours ($08:00{\sim}18:00$) for 28 days. Two different sizes of soft-shelled turtle, large (carapace length: $12{\sim}16\;cm$, body weight: $300{\sim}600\;g$, large size turtle: LST) and middle ($16{\sim}20\;cm,\;700{\sim}1,000\;g$, middle size turtle: MST) were used. The oxygen consumption of LST the beginning of the experiment (1 day) was 124.30 mg $O_2/kg/hr$, and after 7, 14, 21 and 28 days of stress decreased by 47.2, 71.6, 79.1 and 86.0%, respectively. In MST, oxygen consumption of beginning day was 66.04 mg $O_2/kg/hr$, and after 28 days of stress decreased by 76.5%. Ammonia nitrogen excretion showed a similar pattern to that of oxygen consumption. Ammonia nitrogen excretion of beginning of the experiment was 0.537 mg N/kg/hr, and after 28 days of stress decreased by 75.4% for LST, and by 74.3% for the middle ones. From in this study it was showed that the physiological activity was reduced by 50% following 8.4 days of vibration stress for LST, and 15.5 days for MST.

  • The purpose of this study is to investigate daily lives of the intellectually disabled living in residential facilities. The daily time use method that monitors and records 24 hours of the subject was used for the analysis of daily lives of the institutionalized disabled and verbal interaction patterns within each facility. In particular, the analysis focused on 'frequency of activities' and 'lives', with the method of time budget. Based on the activity category table of the 2004 nationwide Time Use Survey, the analysis aimed to identify differences and similarities between the non-disabled and the disabled in terms of daily time use. The interaction pattern analysis method on grounds of the Flanders Category system(1963) indicates the level, the type, and the full details of interaction between the institutionalized disabled and the living rehabilitation teachers. This study is all the more significant in that it monitored the daily lives of the institutionalized disabled for the first time to allow in-depth understanding about the daily lives of the disabled; analyzed interaction patterns between the institutionalized disabled and living rehabilitation teachers; applied time budget study method, a frequently used one in adjacent fields, and therefore should serve as a valuable source material for future studies.

Beneficial Role of Ginseng Saponin on Hemodynamic Functions of Porcine Blood Vessel

  • The previous reports have showed that ginseng saponins, which are the active ingredients of Panax ginseng, cause the relaxation of artery that are contracted due to a various of hormones or potassium ($K^+$). Recently, we also showed that ginsenosides differentially regulate channel activity. The purpose of this study was to examine whether ginseng saponins affect contraction induced by $K^+$, serotonin (5-HT), or acetylcholine (Ach) in porcine coronary vessel. Treatment with concentrations of ginseng saponins caused a relaxation of 25 mM KCl-induced porcine coronary artery contraction. Also, ginseng saponin induced a significant dose-dependent relaxation of $3\;{\mu}M$ 5-HT-induced porcine coronary artery with the endothelium. In the porcine artery with the endothelium, ginseng saponins induced a relaxation by $3\;{\mu}M$ 5-HT in a concentration-dependent pattern. Ginseng saponins induced relaxation of both 25 mM KCl- and $3\;{\mu}M$ 5-HT-induced coronary artery contraction in the absence and presence of the endothelium. In contrast, treatment with $100\;{\mu}g/mL$ ginseng saponin did not induce relaxation in coronary artery contraction induced by Ach ($0.01\;{\mu}M$ to $30\;{\mu}M$) in the presence of the endothelium, but did cause significant relaxation of coronary artery contractions by Ach ($0.01\;{\mu}M$ to $30\;{\mu}M$) in the absence of the endothelium. These findings indicate that ginseng saponin (> $100\;{\mu}g/mL$) significantly inhibits porcine coronary artery contractions caused by $K^+$, 5-HT, and Ach. Therefore, in this study, we demonstrated that ginseng saponin may show beneficial roles on abnormal coronary contraction.

Molecular genetic analysis of phytochelatin synthase genes in Arabidopsis

  • This study has investigated the biosynthesis and function of the heavy metal binding peptides, the phytochelatins, in plants. PCs are synthesised enzymatically from glutathione by the enzyme PC synthase in the presence of heavy metal ions. Using Arabidopsis thaliana as a model organism cadmium-sensitive, phytochelatin-deficient mutants have been isolated and characterised in previous studies. The cadl mutants have wildtype levels of glutathione, are PC deficient and lack PC synthase activity. Thus, the CADl gene has been proposed to encode PC synthase. The CADl gene was isolated by a positional cloning strategy The gene was mapped and a candidate identified. Each of four cadl mutants had a single base pair change in the candidate gene and the cadmium-sensitive, cadl phenotype was complemented by the candidate gene. This demonstrated the CADl gene had been cloned. A homologous gene in the fission yeast, Schizosaccharomyces pombe was identified through database searches. A targeted-deletion mutation of this gene was constructed and the mutant, like cadl mutants of Arabidopsis, was cadmium-sensitive and PC-deficient. A comparison of the redicted amino acid sequences reveals a highly conserved N-terminal region Presumed to be the catalytic domain and a variable C-terminal region containing multiple Cys residues proposed to be involved in activation of the enzyme by metal ions. Similar genes were also identified in animal species. The Arabidopsis CADl/AtPCSl and S. pombe SpbPCS genes were expressed in E. coli and were shown to be sufficient for glutathione-dependent, heavy metal activate PC synthesis in vitro, thus demonstrating these genes encode PC synthase enzymes. Using RT-PCR, AtPCSl expression appeared to be independent of Cd exposure. However, at higher levels of Cd exposure a AtPCSl-CUS reporter gene construct appeared to be more highly expressed. Using the reporter gene construct, AtPCSl was expressed most tissues. Expression appeared to be greater in younger tissues and same higher levels of expression was observed in some regions, including carpels and the base of siliques. AtPCS2 was a functional gene encoding an active PC synthase. However, its Pattern of expression and the phenotype of a mutant (or antisense line) have not been determined. Assuming the gene is functional then it has clearly been maintained through evolution and must provide some selective advantage. This implies that, at least in some cells or tissue, it is likely to be the dominant PC synthase expressed. This remains to be determined

Effect of Composts Fermented with Korean Medicinal Herb Wastes on Physiological Activity of Rubus coreanus Miquel (Bokbunja) (한약탕제찌꺼기 발효퇴비 처리에 따른 복분자과실의 생리활성)

  • Fermented compost made from medicinal herb wastes (MHWC) as an environment-friendly manure resource was applied to determine physiological activities of Rubus coreanus Miquel (Bokbunja). MHWC, poultry manure compost (PMC), and MHWC+PMC (1:1, w/w) were applied to 2 year-old Bokbunja plant at levels of 0, 20, 40 Mg/ha, respectively. Physiological activities of Bokbunja were investigated at 15, 20 and 25 days after flowering (DAF). Content of total phenolics in fruit was high in order of 25 > 20 > 15 DAF, showing highest value in fruit treated with MHWC at 40 Mg/ha. The flavonoid level showed same pattern to total phenolics. Electron donation abilities at fruits at 15 and 20 DAF were higher than at 25 DAF. Nitrite scavenging ability (NSA) was increased on in order of pH 1.2 > 4.2 > 6.0. NSA in fruit at 15 and 20 DAF was higher than at 25 DAF. Tyrosinase inhibition ability at fruits treated with MHWC showed the highest value among all composts. Xanthine oxidase inhibition ability in fruit treated with MHWC at 25 DAF was the highest. In conclusion, physiological activities of Rubus coreanus Miquel (Bokbunja) were improved in MHWC group.

Design of Compound Knowledge Repository for Recommendation System (추천시스템을 위한 복합지식저장소 설계)

  • The article herein suggested a compound repository and a descriptive method to develop a compound knowledge process. A data target saved in a compound knowledge repository suggested in this article includes all compound knowledge meta data and digital resources, which can be divided into the three following factors according to the purpose: user roles, functional elements, and service ranges. The three factors are basic components to describe abstract models of repository. In this article, meta data of compound knowledge are defined by being classified into the two factors. A component stands for the property about a main agent, activity unit or resource that use and create knowledge, and a context presents the context in which knowledge object are included. An agent of the compound knowledge process performs classification, registration, and pattern information management of composite knowledge, and serves as data flow and processing between compound knowledge repository and user. The agent of the compound knowledge process consists of the following functions: warning to inform data search and extraction, data collection and output for data exchange in an distributed environment, storage and registration for data, request and transmission to call for physical material wanted after search of meta data. In this article, the construction of a compound knowledge repository for recommendation system to be developed can serve a role to enhance learning productivity through real-time visualization of timely knowledge by presenting well-put various contents to users in the field of industry to occur work and learning at the same time.

A Study on Comparative Analysis of Socio-economic Impact Assessment Methods on Climate Change and Necessity of Application for Water Management (기후변화의 사회경제적 영향평가 방법론 비교분석과 물관리 부문 적용 필요성에 관한연구)

  • Although it is uncertain that the cause of changed pattern of the natural disaster related to water (i.e. flood and drought) is due to excessive carbon dioxide yielded from economic activity or the increased number of sunspots, it is apparent that there have been unusual climate change that directly affects the water resource management. Due to such a frequent unusual weather activities, there have been increased natural disaster and the most direct and major reason is considered as climate change. As we see, the climate change necessarily causes social costs. Especially, the effects on the water resource due to flood and drought take the considerable part of such costs. Therefore, this study is basic work to develop a new economic analysis technique to be used in pursuing appropriate adaptation project in field of the amount of cost damage through analysis of the effects of the climate change on the water resource. The models appeared in many reports for cost assessment of climate change were various (e.g., PAGE, DICE, AIM, IMAGE, MERGE, and etc.) and this report summarizes general characteristics of each model. To assess the effects of climate change of the water management, we defined the field of the water management on climate change. The results help post-study in field of the climate change's social-economic effect assessment, can be employed for the prioritizing process of the national fund's investment.

Daily Water Intake and Exposure Parameters Related to the Multi-route Exposure in Drinking Water (음용수의 섭취량 및 다경로 노출평가를 위한 노출변수 조사연구)

  • Human exposure to volatile compounds in tap water can occur from inhalation and dermal absorption as well as direct ingestion. The relative contribution to total human exposure from these pathways has been considered to be important especially for VOC's (volatile organic compounds). In an attempt to reduce the uncertainty of the risk assessment, it has been suggested that the exposure assessment process could be significantly improved by adopting Monte-Carlo simulation. However, there is no actual data in Korea for each exposure parameter to determine the level of exposure, and the distributional pattern. Therefore, we surveyed water use patterns and behavior related to multi-route exposure to VOC's in household tap water in Korea, and compared these values to the those in western countries. In the first survey, we calculated daily water intake using data from a sample of 1322 persons of several cities in Korea. In the second survey, we obtained questionnaire data on exposure time for showering, bathing and household activities, and tap water intake from 851households in Korea. In the last survey, we measured the exposure parameters (exposure time, water use rate etc.) related to showers, baths, toilets, dish washing, washing and cooking, and tap water intake was surveyed. Also, the subjects were measured their body weight, height and tidal volume, etc. A diary, a flow meter and a measuring cup were used to measure these values as precisely as possible. Average daily water intake was ranged 0.79-1.71 L/day for adults in three surveys. Tap water intake measured by log-sheet during one week in third survey was 1.26 (average), 1.98 L/day (90 percentlie), respectively. These results were comparable with results from EPA (1.4, 2L/day). The average amount of water used by housewives in the third survey was 515.0 $\pm$ 564.6L/day. In usual activity, the amount of water used in the bathroom, the laundry and the kitchen was 140.0 $\pm$ 538.9, 148.0 $\pm$ 174.5, 229.3 $\pm$ 205.4 L/day, respectively. Exposure parameters such as water intake rate, exposure duration, body weight, inhalation rates in surveyed data of Korean people differed from those published from western countries. This could be attributed to variations in lifestyle, dietary habits and physiological characteristics.

Characteristics of the Extracellular Proteolytic Enzyme Produced by Candida lipolytica FM5 Isolated from Mackerel (Scomber japonicus) (Candida lipolytica FM5가 생산하는 균체외 단백분해효소의 특성)

  • Candida lipolytica(C. lipolytica) FM5 was selected as one of the strong saprophytic yeasts isolated from mackerel (Scomber japonicus). The selected strain could produce extracellular proteolytic enzyme. The effective medium for production of proteolytic enzyme by C. lipolytica FM5 was TPPY broth containing Bacto-tryptone $0.5\%$, proteose peptone $0.5\%$, yeast extract $0.25\%$, NaCl $0.5\%$ and $CaCl_2\;0.2\%$. The pattern of proteolytic enzyme production by C. lipolytica FM5 was the almost same as that of growth curve of the strain. Namely, the enzyme production was begun from the early stage of exponential phase and it was reached the highest at the begining of the stationary phase of the yeast growth. The optimum toeperature of the produced proteolytic enzyme was $35^{\circ}C$ and its activity was not significantly changed by pH between 6.5$\~$9.0 and also it was not significantly affected by several kinds of cations such as $Ca^{2+},\;Cu^{2+},\;Fe^{2+}$ and $Mg^{2+}$ but it was affected negatively by some cations such as $Zn^{2+},\;Mn^{2+}$ and $K^+$.

The Cytotoxicity of Artemisia fukudo Extracts Against HL-60 Cells (큰비쑥(Artemisia fukudo) 추출물의 암세포 증식 억제효과)

  • The present study describes the cytotoxic effect of Artemisia fukudo extracts. The extract from A. fukudo by 80% ethanol was fractionated with n-hexane, dichloromethane, ethylacetate, and butanol in serial. The cytotoxicity of A. fukudo extracts was examined for its effect on the growth of HL-60 cells by the colorimetric 3-(4,5-dimethylthiazol)-2,5-diphenyltetrazolium bromide (MTT) assay. In addition, we used the HL-60 cells to see what effects the A. fukudo extracts will have on apoptosis of cancer cells. We checked the cell activity, cell morphologic changes, DNA fragmentation, and DNA content after 24 hr incubation with administering 25 ${\mu}g/mL$ of the A. fukudo extracts. In the treatment of the low concentration of n-hexane and dichloromethane fractions, the survival rate of HL-60 cells is lower than that of the control. The laddering-pattern DNA fragmentation was observed in the treatment with n-hexane and dichloromethane fractions. The DNA content of the cells apoptosis measured as the density of sub-$G_{1}$ hypodiploid cells by flow cytometric analysis. The number of sub-$G_{1}$ hypodiploid cells increased in the treatment with n-hexane and dichloromethane fractions. These fractions obstructed the cell cohesion and caused the blebbing of the cell membrane and fragmentation of the nucleus, both of which are symptoms of apoptosis. These results suggest that A. fukudo has a great potential value as food additives, medicinal supplements for patients with chronic diseases, and preventive measures against cancer.
