Phaeophyta Extracts Exhibit Antiviral Activity against Feline Calicivirus

  • The objective of this study was to evaluate the antiviral activity of Phaeophyta extracts against feline calicivirus (FCV), used as a norovirus surrogate. A bioassay-guided cytotoxicity and virus infectivity assay revealed that methanolic extracts of Phaeophyta possessed significant antiviral activity against FCV. Among them, Eisenia bicyclis extract exhibited the highest antiviral activity against FCV. The 50% effective concentration of the extract ($EC_{50}$) inhibiting FCV viral replication by 50% was $80{\mu}g/mL$. The extract also showed the highest selectivity index, calculated from the ratio of the median cellular cytotoxicity concentration ($CC_{50}$) and $EC_{50}$, indicating antiviral efficacy against FCV. In addition, significant interruption of FCV infection was observed by pretreatment of host Crandall-Reese feline kidney cells with the E. bicyclis extract ($200{\mu}g/mL$) prior to virus infection, in a dosedependent manner.

Characteristics of $\beta$-Galactosidase with High Transgalactosylation Activity Produced by Penicillium sp. KFCC 10888. (Penicillium sp. KFCC 10888이 생산하는 갈락토스 전이활성이 우수한 $\beta$-Galactosidase의 특성)

  • A Penicillium strain which produces $eta$-galactosidase with high transgalactosylation activity, was isolated from soil and registered as Penicillium sp, KFCC 10888. When $eta$-galactosidase from Penicillium sp. KFCC 10855 reacted with 40% lactose, transgalactosylation ratio reached up to 70% at the 73% conversion of initial lactose. The biosynthesis of the enzyme in Penicillium sp. KFCC 10888 was not induced by lactose. The soybean meal was an effective component of the culture medium. The optimum pH and temperature for transgalactosylation were 4.0 and 55$^{\circ}C$, respectively. The production of galactooligosaccharides was in proportion to the initial lactose concentration. When the enzyme reacted with 40% lactose (pH 4.0) at 55$^{\circ}C$, the concentration of galactooligosaccharides increased up to 40% of total solid concentration.

Analysis of the Taste Components and Antioxidant Properties of Cheonggukjang Containing Korean Red Ginseng

  • This study was performed to investigate the taste composition and antioxidant properties of cheonggukjang containing Korean red ginseng (RGC), as compared to either general cheonggukjang (GC) or non-fermented boiled soybeans (BS). Amylase activity was the highest (576.7 unit/g) in RGC, whereas protease activity was the highest (326.0 unit/g) in GC. The total soluble sugar contents of BS, GC, and RGC were 2,027.5, 905.5, and 837.5 mg/100 g, respectively. RGC had the highest amount of total amino acids (2,127.4 mg/100 g) and essential amino acid (50.9%) among the samples. The ratio of sweet to bitter components was higher in RGC than in GC. Although the extracts of RGC had higher radical scavenging activity for 1,1-diphenyl-2-picrylhydrazyl (DPPH) than BS or GC, regardless of the extract concentration, the ethanol extract of RGC showed the highest scavenging ability (92.4%) at 2.0 mg/mL. The chloroform extracts from GC and RGC showed their greatest superoxide dimutase-like activities at 17.2 and 19.7% at a concentration of 2 mg/mL, respectively. Regardless of the samples, the nitrite scavenging ability was positively correlated to the extract concentration, and RGC had highest ability among samples under the same extract concentrations.

Antitumor and Immunomodulatory Activity of Lycoperdon pedicellatum (긴꼬리말불버섯 (Lycoperdon pedicellatum)의 항암 면역활성)

  • Protein-polysaccharide fractions separated from nine Korean wild mushrooms were subjected to an in vitro screening test for lymphoblastogenic activity. Of these, PPLP, the protein-polysaccharide fraction of Lycoperdon pedicellatum, showed the most potent activity and were further investigated for its antitumor activity. When intraperitoneally injected into ICR mice once daily for six days at a dose of 30 mg/kg, PPLP strongly inhibited the growth of sarcoma 180 tumor cells, showing the inhibition ratio of 97.6%. PPLP also showed in vitro inhibitory activity on sarcoma 180 or leukemia L1210 at the concentration of 500 $\mu\textrm{g}$/$m\ell$ or higher. These results strongly suggest that PPLP might exert its antitumor activity through immunostimulation as well as inhibitory activity on the tumor cells.

  • The color of textile-wastewater hindered spectrometric measurements of $H_2O_2$ and enzyme activity during enzymatic decoloring. By using ABTS, we developed a new method for measuring peroxidase activity and $H_2O_2$ concentration. The ratio of enzyme and $H_2O_2$ was optimized as 1:150 by investigating the effects of $H_2O_2$ on enzymatic decoloring. Pulse feeding of $H_2O_2$, upon depletion, significantly increased the decoloring of Congo Red.

Production and Characterization of Selenium Peptide from Saccharomyces Cerevisiae (효모를 이용한 selenium peptide 생산 및 특성 연구)

  • Selenium containing peptide was produced by culturing yeast with selenium, Selenium was broadly incorporated in the various size of proteins based on the GPC analysis of the total yeast protein. The ratio of selenium to protein increased with the concentration of added selenium in the culture medium. Antioxidant activity (glutathione peroxidase-like activity) was proportional to the concentration of selenium concentration in the peptide. Different size of proteins were obtained by hydrolyzing the total yeast protein by protease XIV. Average molecular weight of selenium peptide was analyzed by GPC. Glutathione peroxidase (GPx) activity of the selenium peptide increased as the size of peptide decreased. Sodium selenite had strong inhibition on the yeast growth than sodium selenate. The ratio of selenium to protein was higher with sodium selenate than with sodium selenite. These results showed the potentials of selenium peptide production by yeast cultivation.

Effects of Dietary Zinc and Phytic Acid Levels on Enzyme Activity and Lipid Metabolism of Rats (아연과 Phytic Acid 수준별 식이가 흰쥐의 효소활성 및 지질대사에 미치는 영향)

  • In order to investigate the effect of dietary zinc and phytic acid levels on enzyme activity and lipid metabolism in rats, male Sprague-Dawley rats, weighing approximately 60-74g, were fed different diets which contained 0, 0.35 or $1.05\%$ phytic acid each at 3 levels of zinc (0, 30 and 1500ppm zinc) for 28 days. Body weight gain, food consumption, and food efficiency ratio were lower in the rats fed a zinc deficient diet (0ppm zinc) than those consuming 30 or 1500ppm dietary zinc. The activities of GOT, GPT and alkaline phosphatase were lower in the rats consuming 30ppm zinc than those fed 0 or 1500ppm zinc diet. The activity of GOT was increased in rats consuming $0.35\%$ phytic acid, whereas that of alkaline phosphatase was decreased in the rats fed phytic acid-containing diet. The concentration of phospholipid in serum was higher in rats fed $0.35\%$ dietary phytic acid, whereas that of liver phospholipid was higher in zinc deficient groups, and increased by addition of dietary phytic acid. The concentration of triglyceride in serum from rats fed 30ppm zinc was lower than those fed 0 or 1500ppm zinc On the other hand, liver triglyceride was higher in both the rats fed 30ppm zinc and $0.35\%$ phytic acid. The concentration of serum total cholesterol was lower in the rats fed 30ppm zinc diet, and it was increased by addition of dietary phytic acid. But liver total cholesterol was higher in 30ppm zinc group. HDL-cholesterol in serum was the highest in both rats consuming 30ppm zinc and $0.35\%$ dietary phytic acid, and the ratio of HDL-cholesterol to total cholesterol was higher in rats consuming 30ppm zinc diet. In conclusion, we suggest that coronary heart disease or liver disease can be prevented with phytic acid in rats which are fed the high zinc diet.

Changes in Physiological and Psychological Conditions of Humans to Color Stimuli of Plants

  • This study investigates the color stimuli of two varieties of foliage plants by extracting electroencephalogram, electrocardiogram and physiology activity data from 30 participants in their 50s or older. Changes in the physiological activity of subjects against six color stimuli were examined. The stimulus to real green plants 'Silver Queen' was set as the control group, and was compared with other groups including the stimulus to real 'Angel' plants and four stimuli to artificial colors (two color images and color schemes of the same green and red plants). Compared to the five groups, the relative theta power spectrum (RT) and the ratio of alpha to high beta (RAHB) increased in the subjects exposed to real green plants. This result demonstrates that the green plant ('Silver Queen') increases the stability, relaxation, and internal concentration of subjects in a proper state of awakening. The result of this experiment showed a statistically significant difference in the level of RT when subjects were exposed to the groups of real green and red plants. This finding indicates that the green plant increases internal concentration more than the red plant. RT and the relative low beta power spectrum (RLB) in the groups of natural colors were higher than the groups of artificial colors when subjects focused their mind on the two types of real plants. However, the level of relative mid beta power spectrum (RMB), ratio of SMR to theta (RST), ratio of mid beta to theta (RMT), relative high beta power spectrum (RHB), and spectral edge frequency 95% were higher when subjects were exposed to the photos and colors scheme of plants than when they were exposed to real plants. The subjects experienced more "comfortable" emotions when they were looking at plants with green colors. Overall, it is recommended to use the natural colors of real plants in places where which stability and relaxation are required. On the contrary, the artificial colors of plants such as their photos and color schemes are useful in places where a high level of concentration is required in a short period of time.

  • The present study was carried out to investigate the effect of Ni on germination, cell elongation, ${\alpha}-amylase$ activity, contents of chlorophyll and protein in radish were determined in the water culture. As the concentration of Ni was increased in the water culture, germination of radish was 55% by Ni 10 mg/kg and 30% by Ni 20 mg/kg. The ratio of cell elongation injury was 50%, by two days after Ni 20 mg/kg treatment. The injury ratio of ${\alpha}-amylase$ activity was 45% in the same condition and as the time goes on, inhibition of ${\alpha}-amylase$ activity were slightly decreased. Contents of chlorophyll a and b were decreased two days after treatment and chlorophyll a was more inhibited than chlorophyll b. Also changes of the protein contents was slightly decreased. Activity of ${\alpha}-amylase$ was decreased at germination stage, contents of chlorophyll a and b were decreased at growing stage.

  • The effect of cyclic-AMP on the induction of acid phosphatase activity in barley aleurone layers was examined. Tannic acid was used as a inhibitor. Decursinol and coumarin were also used as a comparison. Maxiumu promotion of the enzyme activity was obtained with 10-5M cyclic-AMP, but this promotion was lower than that of 10-5M GAS induced enzyme activity in incubation medium. The inhibition rate in the addition of tannic acid was shown 17% and 63% at a ratio to GAs (by weight) of 10 : 1, and 58% and 94% at a ratio of 100 : 1 treated with GAs, and cyclic-AMP, respectively. The most potentiation of 10-6M GAS effect was induced by the additiion of suboptimal concentration (10-6M) of cyclic-AMP. Additional GAs and cyclic-AMP were shown the recovery of the enzyme activity inhibited by tannic acid. The combination with cyclic-AMP and theophylline enhanced the enzyme activity, too. Any other nucleotides tested except cyclic-AMP didn't show the action. There were no differences in acid phosphatase isozyme patterns by polyacrylamide disc electrophoresis, in conjunction with the different additions but the size of bands showed great differences. Especially, the 3rd band and the 5th band group were remarkable.
