SVE 및 생물학적 공법을 이용한 TPH 오염토양처리에 관한 연구 (The Study on the Remediation of Contaminated Soil as TPH using SVE and Bioremediation)

  • This study examined the contaminated soils with an indicator of TPH using SVE (Soil Vapor Extraction) and biological treatments. Their results are as follows. Water content in the polluted soils slowly decreased from 15% during the initial experimental condition to 10% during the final condition. Purification of polluted soils by Bioventing system is likely to hinder the microbial activity due to decrease of water content. Removal rate of TPH in the upper reaction chamber was a half of initial removal rate at the 25th day of the experiment. The removal rate in the lower reaction chamber was 45% with concentration of 995.4 mg/kg. When the Bioventing is used the removal rate at the 14th day of the experiment was 53%, showing 7 day shortenting. Since the Bioventing method control the microbial activity due to dewatering of the polluted soil, SVE method is likely to be preferable to remove in-situ TPH. The reactor that included microbes and nutrients showed somewhat higher removal rate of TPH than the reactor that included nurtients only during experimental period. In general, the concentration showed two times peaks and then decreased, followed by slight variation of the concentration in low concentration levels. Hence, in contrast to SVE treatment, the biological treatment tend to show continuous repetitive peaks of concentration followed by concentration decrease.

산초 정유성분의 식중독균에 대한 항균 활성 (Antibacterial Activities of Essential Oil from Zanthoxylum schinifolium Against Food-Borne Pathogens)

  • In this study, the antibacterial activities of essential oil from Zanthoxylum schinifolium against four Gram-positive bacteria and six Gram-negative bacteria were investigated. The antibacterial activity of the oils was determined using the agar-well diffusion assay, MIC (minimum inhibitory concentration) and MBC (minimum bactericidal concentration). In particular, essential oil from Z. schinifolium showed higher antibacterial activity against Gram-positive bacteria than against Gram-negative bacteria. Essential oil from Z. schinifolium displayed large inhibition zones especially against Bacillus cereus (31 mm). At concentrations between 0 and $20\;{\mu}g/mL$ the oils showed an antibacterial effect against both Gram-negative and Gram-positive bacteria. The minimum inhibitory concentration (MIC) values against nine bacteria ranged from 1.25 to $5\;{\mu}g/mL$. The minimum bactericidal concentration (MBC) values against eight bacterial ranged from 2.5 to $20\;{\mu}g/mL$, except Shigella sonnei. Furthermore, our finding on the antibacterial activities of essential oils from Zanthoxylum schinifolium validated the use of this plant for medical purposes.

The development of a portable MO4- (M = 188Re or 99mTc) concentration device for extending the lifetime of RI generators

  • The activities per volume of $^{188}Re$ and $^{99m}Tc$ from their generators are dependent on the specific activity of their mother nuclides $^{188}W$ and $^{99}Mo$ respectively. After a particular lapse of time, the eluted RI activity is exponentially reduced and thus cannot satisfy the needs of clinical application. The purpose of this study is to develop a $^{188}Re$ and $^{99m}Tc$ concentration device with a compact size that can extend the period of use as well as conveniently concentrate the RI. We designed the concentration module by including two-different check valves that do not required any manual on-off operations. In these concentration process, cation exchange resin embedded with Ag and anion exchange resins were used. After completing the concentrating step, the recovering yield was identified to be more than 93% for $^{188}Re$ generators and 88% for $^{99m}Tc$ generators. Moreover, all these procedures were done within 5 min.

Activity Concentrations of 137Cs and 90Sr in Seawaters of East Sea, Korea

  • Background: This study was a long-term evaluation of $^{137}Cs$ and $^{90}Sr$ activity concentrations in seawater samples from the East Sea, Korea, in order to establish current activity levels. Results and long-term monitoring trends will be useful in the future monitoring of environmental radioactivity. Materials and Methods: Surface seawater samples were collected quarterly from Guryongpo and Jangho in the East Coast between 1998 and 2010 and the quarterly deep seawater samples were collected from three sites in the sea adjacent to Ulleung-do between 2012 and 2015. The activity concentrations of $^{137}Cs$ were measured using a gamma-spectrometer. The activity concentrations of $^{90}Sr$ and $^{90}Y$ in a radioactive equilibrium state were measured using a gas flow proportional counter. Results and Discussion: We found the annual average activity concentrations of $^{137}Cs$ in the surface seawater was $1.66-2.89mBq{\cdot}kg^{-1}$ in Guryongpo and $1.68-2.43mBq{\cdot}kg^{-1}$ in Jangho. The annual average activity concentrations of $^{90}Sr$ in the surface seawater was $0.83-1.98mBq{\cdot}kg^{-1}$ in Guryongpo and $0.82-1.57mBq{\cdot}kg^{-1}$ in Jangho. The annual average activity concentrations of $^{137}Cs$ in the deep seawater sites were $1.51-1.73mBq{\cdot}kg^{-1}$, $1.19-1.60mBq{\cdot}kg^{-1}$ and $0.87-1.15mBq{\cdot}kg^{-1}$ in TH, JD, and HP. The annual average activity concentrations of $^{90}Sr$ in the same deep seawater sites were $1.00-1.94mBq{\cdot}kg^{-1}$, $0.82-1.26mBq{\cdot}kg^{-1}$, and $0.79-1.32mBq{\cdot}kg^{-1}$. The effective half-life was calculated by analyzing change over time in the activity concentration in the surface seawater. The effective half-life of $^{137}Cs$ was $15.3{\pm}0.1years$ in Guryongpo and $102{\pm}3years$ in Jangho. The effective half-life of $^{90}Sr$ was $28.3{\pm}4.3years$ in Guryongpo and $16.6{\pm}0.1years$ in Jangho. The ratio of the average activity concentration ($^{137}Cs/^{90}Sr$) was 1.72 in the surface seawater, which is similar to the reported ratio of the global radioactive fallout. The ratio in the deep seawater was 1.24, which is somewhat low compared to the global ratio (1.6, 1.8). Conclusion: Activity concentrations of $^{137}Cs$ and $^{90}Sr$ in the seawaters of the East Sea were similar to the previously reported activity levels in the East Sea and northwestern Pacific as a result of global radioactive fallout following atmospheric nuclear weapon tests.

용매와 추출조건에 따른 단삼 (Salvia mitiorrhiza) 추출물의 항균력 (Effects of Solvents and Extracting Condition on the Antimicrobial Activity of Salviae miltiorrhizae Radix (Saliva miltiorrhiza ) Extract)

  • In order to develop a natural food preservative, dried salviae miltiorrhizae radix (Salvia miltiorrhiza) was extracted with several solvents, and then antimicrobial activity was investigated. The optimum extracting condition for the antimicrobial sustance from the sample, minimal inhibitory concentration (MIC) of the extracted substance against microorganisms were also examined. Antimicrobial activity of the initial ethanol extract from the sample was the strongest compared to those of other solvent extracts such as n-hexane, acetone, butanol, methanol and water. the optimum extractingcondition for antimicrobial substance from the sample was shaking extraction for 2 hours at room temperature incase that 10 volumes of absolute ethanol was added to crushed Saliva Miltiorrhiza. The ethanol extract had strong growth inhibition activity against Gram-positive Bacteria (MIC, 3.13-50$\mu\textrm{g}$/ml) such as B. cereus, B, subtilis, L. minocytogenes, S. aureus, Sc. Mutans. Among Grampositive bacteria tested, Bacillus species was the most susceptibile to the extracted substance. The antimicrobial activity of the ethanol extract from the sample was weak to Gram -negative bacteria yeasts, for example MIC for Gram-negative bacteria and yeasts was 0.8mg/ml and 0.4-0.8mg/ml , respectively.

백일해(百日咳) 유발균(誘發菌) Bordetella pertussis에 대한 호장근(虎杖根)의 생육(生育) 억제(抑制) 효과(效果) (Effective Antibacterial Activity of Reynoutria Japonica Against Bordetella Pertussis ATCC 9797)

  • This study was designed to identify the effects of Reynoutria Japonica on antibacterial activity aganist Bordetella pertussis ATCC 9797 which is cause of whooping cough. The ethanol- and water-extracts of more than 80 oriental herbal medicine were Investigated by Kirby -Bauer method to determine their inhibitory effects on growth of B. pertussis ATCC 9797 in vitro. For that Reynoutria Japonica was selected. The ethanol-soluble extract of Reynoutria Japonica showed relatively high antivacterial activity against B. pertussis ATCC 9797. However, the water-soluble extract of Reynoutria Japonica showed no antibacterial activity. The ethanolic extract was further fractionated with organic solvents such as hexane, chloroform, and ethyl acetate, in that order. Among the fraction tested, the chloroform fraction showed the highest antibacterial activity when the ethanol-soluble extract of Reynoutria Japonica minial inhibitory concentration(MIC) was $25{\mu}g/m{\ell}l$. Results support a role far Reynoutria Japonica in inhibiting the cell growth of B. pertussis ATCC 9797, but further experimentation is required.

하고초 에탄올추출물이 유방암 예방효소계에 미치는 영향 (Effect of Ethanol Extract from Thesium chinense Tunczaninov on Chemopreventive Enzymes of Breast Cancer)

  • Ethanol extract from Thesium chinense Tunczaninov (TCTE) was tested for breast cancer chemopreventive activity by measuring 7,12-dimethylbenz[a]anthracene (DMBA) - induced cytochrome P450 1A1 activity, induction of quinone reductase and glutathione S-transferase, and glutathione level. TCTE significantly inhibited cytochrome P45O 1A1 activity at the concentration of 90 and 150 mg/ml. TCTE induced quinone reductase activity in a dose-dependent manner in a concentration range of 3-150 mg/ml. In addition glutathione S-transferase activity and glutathione level were increased with TCTE in cultured murine hepatoma Hepa1c1c7 cells. These results suggest that TCTE has breast cancer chemopreventive potential by inhibiting cytochrome P45O 1A1 activity, inducing quinone reductase and glutathione S-transferase activities, and increasing GSH level.

Chitosan-Based Film of Tyrothricin for Enhanced Antimicrobial Activity against Common Skin Pathogens Including Staphylococcus aureus

  • Chitosan-based film-forming gel is regarded as a promising vehicle for topical delivery of antimicrobial agents to skin wounds, since it protects from microbial infection and the cationic polymer itself possesses antibacterial activity. In this study, possible synergistic interaction against common skin pathogens between the cationic polymer and tyrothricin (TRC), a cyclic polypeptide antibiotic, was investigated, by determining the concentration to inhibit 90% of bacterial isolates (MIC). The addition of the polysaccharide to TRC dramatically reduced the MIC values of TRC by 1/33 and 1/4 against both methicillin-resistant and methicillin-susceptible Staphylococcus aureus, respectively. The synergism of TRC and chitosan combination against both strains was demonstrated by the checkerboard method, with a fractional inhibitory concentration index below 0.5. Moreover, co-treatment of TRC and chitosan exhibited antibacterial activity against Pseudomonas aeruginosa, due to the antibacterial activity of chitosan, whereas TRC itself did not inhibit the gram-negative bacterial growth. These findings suggested that the use of chitosan-based film for topical delivery of TRC could be an alternative to improve TRC antimicrobial activity against strains that are abundant in skin wounds.

Inhibitory effects of medicinal plants on elastase activity and biological activities in the active Plant extracts

  • Inhibitory effects of 200 medicinal plants on elastase activity were screened. Among the 200 plants, six plant extracts exhibited more than 65% of inhibition on elastase activity by their total methanol extracts at 1, 000$\mu\textrm{g}$/$m\ell$ as a final concentration. For six active plants including Areca catechu, Cinnamonum cassia, Myristica frograns, Curcumma longa, Alponia katsumadai, and Dryopteris crassirrhizoma, the inhibitory effects of their comsituents were examined on the activity of human leukocyte elastase, hyaluronidase and lipid peroxidation. In lipid peroxidation assay using TBA method, three of the 6 plants including Curcumma longa, Areca Catechu and Alponia katsumadai exhibited more than 70% of inhibition at the concentration of 1, 000$\mu\textrm{g}$/$m\ell$, but only one plant, Areca Ctechu showed high inhibitory effect on hyaluronidase activity. The results suggest that medicinal plants showing several biological activities may be potent inhibitors of anti-aging process on skin and that might be useful for application in cosmetics.

Effects of Insulin on Acetyl-CoA Carboxylase in Bovine Mammary Secretory Cells

  • Objectives of this study were to determine effects of insulin on acetyl-CoA carboxylase (ACC) activity and correlate this activity with relative amounts of ACC in MAC-T cells. MAC-T cells were grown in Medium 199 supplemented with fetal bovine serum (5%), cortisol ($1{\mu}g/ml$), and insulin ($1{\mu}g/ml$). At confuluence, the cells were transferred to $100mm^2$ culture dishes coated with the extracelluar matrix. After 10 h of incubation, the media were replaced with media without fetal bovine serum and the concentration of insulin was lowered to 5 ng/ml. After 24 h, the media were changed to contain the varying concentrations of insulin and incubations continued for 48 h. The addition of insulin resulted in increases in the specific activity of ACC. The maximal effects of insulin on the ACC activity occurred at concentrations of insulin, 1,000 ng/ml. In contrast, the relative change in lactate dehydrogenase (LDH) activity in response to increasing insulin concentration was minimal as compared to the effects of insulin on ACC. Transblot and enhanced chemiluminescence (ECL) analysis indicated that the increase in ACC activity in MAC-T cells caused by insulin were due to actual increases in amounts of enzyme.
