Four phenylpropanoid glycosides and a monoterpene glycoside were isolated from the flower of Clerodendrum trichotomum. Structures of the isolated compounds were identified as acteoside (1), martynoside (2), leucosceptoside A (3), isoacteoside (4) and neohancoside A (5) by spectroscopic analysis. Compounds 1-4 were isolated from the flower of C. trichotomum for the first time. Compound 5 was first obtained from genus Clerodendrum as well as family Verbenaceae. The antioxidant activity of compounds 1-5 were evaluated by the DPPH free radical scavenging assay. Compounds 1-4 exhibited strong antioxidant activity.
To develop potential cosmetic ingredients with antimicrobial and antioxidant activities of 4 Korean bamboo species (P. bambusoides, P. nigra var. henonis, P. pubescens and Sasa coreana) using three different extraction methods-water, ethanol and supercritical fluid extraction. Antimicrobial activities and DPPH assay have been examined. Among the antimicrobial activities against two test strains, Escherichia coli and Staphylococcus aureus, ethanol extracts of 3 bamboo trees, P. bambusoides, P. nigra var. henonis, and P. pubescens, showed stronger than those of supercritical extracts. However, 4 bamboo supercritical extracts showed dose-dependent increase in antioxidant activity by DPPH assay. These results suggest that water fraction of bamboo extracts may be useful for the cosmetic ingredient with low cytotoxicity.
We have employed a DNA-native-polyacrylamide gel electrophoresis (DNA-native-PAGE) assay system to characterize the enzyme activity of the endonuclease secreted by human B lymphoblastic IM9 cells. Experimental results clearly demonstrated that the endonuclease activity of IM9 cell culture medium is distinct from that of DNase I in the DNA-native-PAGE assay system. Immunoprecipitation analysis using anti-DNase I antibody showed that the secreted endonuclease is not recognized by the antibody. The secreted endonuclease was estimated using supercoiled plasmid DNA as a substrate. The pH optimum required for the catalytic activity was determined to be in the range of pH 6.6-7.4. No significant difference in the endonuclease secretion was observed by stimulation of the IM9 cells with interferon-${\gamma}$ or interleukin-$1{\beta}$.
The purpose of this study was to confirm the anti-tumor activity of an ethanol extract of Saussurea laniceps against pancreatic tumor and to isolate the active compound from S. laniceps extract. Treatment with S. laniceps extract and hispidulin inhibited proliferation of pancreatic cell lines, such as Capan-1, Capan-2, Panc-1 and S2-013 in a dose-dependent manner using the hollow fiber assay. Hispidulin showed typical hallmarks of apoptotic cell death a significant anti-tumor activity on Capan-2 cells at a dose of 100 mg/kg and 200 mg/kg. S. laniceps has potential cytotoxic and apoptotic effects on human pancreatic carcinoma cells. Its mechanism of action might be associated with the apoptotic cell death through DNA fragmentation.

Seven flavonoids were isolated from the flower of Clerodendrum trichotomum. Their structures were identified as apigenin (1), genistein (2), chrysoeriol (3), genistein 7-O-glucoside (4), kaempferol 3-O-glucoside (5), isorhamnetin 3-O-glucoside (6) and apigenin 7-O-glucoside (7) on the basis of spectral data. These compounds were isolated from C. trichotomum for the first time. The antioxidant activity of compounds 1-7 were evaluated by the ORAC (oxygen radical absorbance capacity) assay, and the ORAC values were expressed as relative trolox equivalent. All isolated compounds exhibited antioxidant activity.
To investigate the effect on the antimicrobial activity against S.aureus ATCC 6538, E.coli KCTC 1039 and cell toxic level against transformed mouse fibroblast L929 in formula added with various concentrations of UVB blockers commonly used in cosmetic products, these experiments were carried out by preservative efficacy testing methods and in vitro cytotoxicity methods. The results obtained were as follow ; 1) Octyl Dimethyl PABA had a broad antibacterial spectrum against the Gram (+) and the Gram(-) bacteria at 5.84 % concentration, but not Octyl Methoxycinnamate. 2) Antibacterial activity was decreased in a combined UVB blocker system of squalane base. Three different methods of bacterial viability monitoring were compared to detect chemically or thermally inactivated Escherichia coli. Direct colony enumeration, live/dead bacterial cell staining with a fluorescent dye, and the dehydrogenase activity assay were compared with respect to their ease of use and time required to perform the three different tests. The green (live cell)/red (dead cell) ratio obtained from the fluorescent bacterial cell staining approach showed a linear relationship with the colony forming units; the result obtained with dehydrogenase was similar to those. The sensitivity of the monitoring methods to detect bacterial deactivation varied with different disinfection conditions. After thermal treatment, the sensitivity of the staining approach was lower, while that of the dehydrogenase activity assay was the highest. After chemical treatment, the sensitivity of detection for both methods was similar.
Four anthocyanins were isolated from the acidic alcoholic extract of Syzygium cumini fruits growing in Egypt: Pelargonidin-3-O-glucoside, pelargonidin-3,5-O-diglucoside, cyanidin-3-O-malonyl glucoside, and delphenidin-3-O-glucoside. They were identified by the chromatographic, TLC and PC, and spectral analyses, UV, $^1$H-NMR and FAB/MS. The fruits were found to contain 0.03 gm % anthocyanins calculated on fresh weight basis calculated by spectrophotometric assay. Cytotoxic activity of total alcoholic extract of the fruits was performed against several types of tumor cell lines using the SRB assay. The tested extract exhibited significant cytotoxic activity for MCF7 (breast carcinoma cell line) (IC$_{50}$= 5.9 ${\mu}$g/mL), while the IC$_{50}$ was > 10 ${\mu}$g/mL for both Hela (Cervix carcinoma cell line), HEPG2 (liver carcinoma cell line), H460 (Lung carcinoma cell line) and U251 (Brain carcinoma cell line).
Bacterial components and their derivatives have been reported to mediate various immunomodulating activities and to activate immune cells including macrophage. In this study, the secretory and cellular macrophage response to teichoicated fragments (TFs) from pneumococcal cell wall subcomponent were examined. Tumoricidal activity was measured by MTT assay and secretory molecules were assessed by biological assay. After stimulation of macrophages with various doses of TFs for 18hrs, secretion of TNF-$\alpha$, nitrite and $H_2O$$_2$ were significantly increased as compared to medium-treated control. In addition, tumorcidal activity of TFs-treated macrophages was enhanced, whereas production of IL-1 and IL-6, and phagocytic activity were not induced. These data suggest that TFs is a potent inducer of macrophage secretory and cellular activities.
Glycosaminoglycans(PT -Gag) were isolated from the porcine testis. From the PT -Gag, we obtained two different types of Gag fractions using Dowex macro porous Resin MSA-1 column, PT -Gag-1.5% NaCl and PT -Gag-16% NaCl. Various biological activities of the GAGs were examined in aspect of anticoagulant and immunomodulating activity. The anticoagulant activity of the GAGs was evaluated by activated partial thromboplastin time (aPTT ) assay and thrombin time (TT) assay. The GAGs of porcine testis markedly incresed the clotting times of both of aPTT and TT. showing that PT-Gag-16% NaCl was more effective than PT-Gag-1.5% NaCl. The immunomodulating activityof the GAGs was examined in relation to regulation of xytoxine prodution of murine peritoeal maerophages. Taken together. GAGs isolated from porcine testis possess bilolgical functions such as anticoagulant and immunomodulating activity.
