The antioxidant activity was analyzed in gamma-irradiated cooking juices. Because the activities of antioxidants have been attributed to various mechanisms, different assay methods including 1,1-diphenyl-2-picryl-hydrazyl (DPPH) radical scavenging activity and ferric reducing antioxidant power (FRAP), have been conducted and compared. All of these antioxidant assay showed that the antioxidant activity of cooking juice was increased by gamma-irradiation. To investigate this increase of antioxidative activity, the protein was extracted from cooking juices and its antioxidant activity was measured. From the results, it was thought that the modification of protein in cooking juiced by irradiation caused the increase of antioxidant activity of cooking juice. Therefore, gamma irradiation could be an promising method for a sterilization of cooking juice with increased antioxidant activity.
The rhizome of Phragmites communis Trin. is used for vomiting and belching by clearing stomach and the sprout is used as tea. Phragmites japonica is similar with P. communis except the color of sheath is purple. This study is aimed to compare the in vitro antioxidant activity, total polyphenol and flavonoids contents of P. communis and P. japonica. The antioxidant activities of fractions from the two Phragmites plants were evaluated by 1,1-diphenyl-2- picrylhydrazyl (DPPH) and 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging assay. The antioxidant activity varied with plant parts and extract solvents. The fractions of leaf extract from the two Phragmites plants (4.06±1.32-16.47±1.28%) showed higher antioxidant activity by DPPH assay compared with rhizome fractions of two Phragmites plants (0.00±0.00-14.15±0.07%), these are lower compared with ascorbic acid and butylhydroxyanisole (BHA). The highest ABTS radical scavenging activity was found for rhizome ether fraction, namely 74.95±0.56% and 73.04±1.85% for P. communis and P. japonica, these are higher than BHA. The total polyphenol and flavonoids contents were different with plant parts and extract solvents, likewise antioxidant activity. A significant correlation was shown between DPPH and ABTS radical scavenging activity. Considering the results of this study, the leaves and stems of P. communis and P. japonica are expected to be used as natural antioxidants.
본 연구는 만형자(Vitex rotundifolia)의 세포독성을 확인하기 위한 목적으로 methanol, ethanol, acetone으로 추출한 뒤, 인간 유래의 대장암 세포주인 HT-29에 처리하여 MTT reduction assay를 이용하여 실험을 하였다. 그 결과, 모든 만형자 추출물에서 농도 의존적으로 성장 저해 효과가 나타났으며, 그 중에서도 acetone 추출물을 50 ${\mu}g/ml$ 농도로 처리 시 18%로 가장 높은 항암 효과를 보였다. 또한 각 용매별 추출물을 50 ${\mu}g/ml$를 처리하여 HT-29 세포주의 성장을 관찰한 결과, acetone 추출물은 대조구에 비하여 암세포의 수축 등 형태변화가 두드러지게 나타났으며, Hoechst 33342 staining을 통하여 apoptotic body가 형성된 것을 확인 하였다. 그리고 가장 활성이 좋은 acetone 추출물을 용매 분획하여 얻은 분획물 중, n-hexane 분획물에서 농도 의존적으로 세포독성을 나타냄을 알 수 있었다. 또한 만형자의 n-hexane 분획물은 다른 대장암 세포주 SW620에 대해서도 50 ${\mu}g/ml$ 농도에서 대조군에 비하여 7배 정도의 높은 세포독성을 나타내고 있음을 MTT reduction assay와 LDH release assay를 통하여 확인 할 수 있었다.
본 연구는 사방오리 추출 분획물의 항산화 활성을 검증하기 위하여 시행되었다. 사방오리는 메탄올(MeOH) 분획에 가장 높은 총 페놀화합물($452.80{\pm}7.01{\mu}g$ gallic acid equivalents/mg)을 그리고 에틸에세테이트(EA) 분획 에 가장 높은 총 플라보노이드 함량($112.29{\pm}11.14{\mu}g$ rutin equivalents/mg)과 가장 높은 총 항산화 활성($936.23{\pm}0.07{\mu}g$${\alpha}$-tocopherol equivalents/mg)을 가지고 있었다. 사방오리의 용매 추출분획물에 대한 항산화 활성은 아스코빅산 등 표준 물질과 비교하여 1,1-diphenyl-2-picrylhydrazyl (DPPH) 라디칼 소거능 측정, 슈퍼옥사이드 음이온 라디칼 소거능 측정, ${\beta}$-carotene-linoleate 에세이, 환원력 측정, ferric thiocyanate법으로 지질과산화 억제력 측정 그리고 금속 킬레이팅능 측정 등 다양한 항산화 활성을 측정하였다. $250{\mu}g/ml$ 농도의 사방오리 EA와 MeOH 분획은 DPPH 라디칼 소거능 측정에서 각각 92.43와 89.20% 항산화 활성을 가지고 있었다. $50{\mu}g/ml$의 EA분획은 72.49% 슈퍼옥사이드 음이온 라디칼 소거능을 보였으며 이는 gallic acid의 소거능(60.88%)보다 조금 높았다. 또한 ${\beta}$-carotene-linoleic acid 시스템에 의한 과산화 억제능 조사에서 0.5과 1 mg/ml 농도의 EA 분획이 각각 73.45와 73.29% 억제능을 보였다. 항산화 활성은 EA 분획 > BuOH 분획 > DCM분획 > HX분획 순 이였다. 이상의 본 연구 결과에서 사방오리는 천연 항산화제제 개발을 위한 용이하게 접근 가능한 잠재적인 자원으로 판단된다.
Roots of Scutellaria baicalensis have been used for fever remedy; diuresis, antiphlogistic. For the investigation of the active component from aerial parts of Scutellaria baicalensis, MeOH extracts from aerial parts of Scutellaria baicalensis were suspended with $H_2O$, and partitioned by $CHCl_3$. In order to investigate the efficacy of antioxidative activity the activity guided fraction and isolation of physiologically active substance were peformed. Its $H_2O,\;30\%,\;60\%$ MeOH and MeOH fractions were examined on antioxidative activity using DPPH method and TBARS assay; It was revealed that $30\%\;and\;60\%$ MeOH fractions have significant anti-oxidative activity. its fractions testing type I allergy, compound 48/80 induced systemic anaphylaxis was applied. As a result, compared with reference (cromolygate), these fraction significantly inhibited systemic anaphylaxis by $71\%\;and\;57\%$, respectively. From $30\%,\;60\%$ MeOH fraction, five compounds were isolated and elucidated apigenin 6-C-${\alpha}$-L-arabinopyranosyl-8-C-${\beta}$-D-glucopyranoside (isoschaftside, I), scutellarein 7-O-${\beta}$-D-glucuronopyranoside (scutellarin, II), apigenin 7-O- ${\beta}$-D-glucuronopyranoside (III), isoscutellarein 8-O-${\beta}$-D-glucuronopyranoside (IV), kaempferol 3-O-${\beta}$-D-glucopyranoside (V) through their physicochemical data and spectroscopic methods. We measured radical scavenging activity with DPPH method and anti-lipid peroxidative efficacy on human LDL with TBARS assay. [$I] showed antioxidant activities in order. Type I allergy compound 48/80 induced systemic anaphylaxis was applied. $[V inhibited systemic anaphylaxis in order.

Objectives : ROS are involved in a wide spectrum of diseases including chronic inflammation and cancer. S.chinensis Baill, a perennial herb commonly called Chinese lizard's tail or Sam-baek-cho in Korea, is used for the treatment of edema and inflammatory diseases in the Oriental folk medicine. In this study, we investigated the antioxidant activities and anti-inflammatory effects of the two extracts, water(WE) and ethyl acetate(EAE) from S.chinensis Baill. Methods : Anti-oxidant activity was evaluated using Fe2+ chelating and hydroxyl radical scavenging assay. DNA cleavage assay, and western blot and immunostaining for phospho-p65 were performed to evaluate anti-oxidative effect. Anti-inflammatory effect was performed using NO generation assay and western blot in LPS-stimulated RAW264.7 cell. Results : In Fe2+ chelating activity and hydroxyl radical scavenging activity, WE showed more strong scavenging activity for hydroxyl radical than EAE. WE scavenged hydroxyl radical by 12% at 3.2 ${\mu}g/ml$, 21% at 16 ${\mu}g/ml$, 32% at 80 ${\mu}g/ml$, 66% at 400 ${\mu}g/ml$ and 82% at 2000 ${\mu}g/ml$, respectively. In addition, WE showed more strong chelating activity than EAE. WE chelated Fe2+ ion by 1.1% at 3.2 ${\mu}g/ml$, 8.2% at 16 ${\mu}g/ml$, 26.3% at 80 ${\mu}g/ml$, 72% at 400 ${\mu}g/ml$ and 89% at 2000 ${\mu}g/ml$, respectively. Also, WE inhibited oxidative damage via its anti-oxidant activity. In anti-inflammatory effect, EAE inhibited NO production and iNOS expression. In addition EAE suppressed the NF-${\kappa}B$ and MAPK signaling pathway in LPS-stimulated RAW 264.7 cells. Conclusions : Together, these data indicate that S. chinensis Baill, shows anti-oxidant activity and anti-inflammatory effect.
In this study, degradation of estrone (E1) and $17{\alpha}$-ethynylestradiol (EE2) by gamma-irradiation and subsequent reduction of estrogenic activity as a function of absorbed dose were conducted using the yeast two-hybrid assay. Relative potency of E1 and EE2 compared to estrogenic activity of $17{\beta}$-estradiol (E2) was found to be 0.0144 and 0.1605, respectively. More than 90% of E1 and EE2 (both $5.0{\times}10^{-6}M$) was removed at an absorbed dose of 5 kGy, but more than 40% of estrogenic activity still remained. The addition of $TiO_2$ catalyst appeared to improve the removal efficiency of E1 and decrease estrogenic activity while there was no significant effect for EE2. Additionally, the calculated estrogenic activity of E1 and EE2 based on a regression model was well correlated with the observed activity.
This study was carried out to investigate the antibacterial activity against pathogens of acne and the anti-inflammatory effect of 75% ethanol extract and its fractions from the leaves of Prunus sargentii. In the antibacterial activity by the disc diffusion assay, the extract showed the highest effect against Propionibacterium acnes, Staphylococcus aureus and Staphylococcus epidermidis in 5 mg/disc. However, the ethyl acetate fraction showed the highest antibacterial activity in 1 mg/disc. On the other hand, the hexane and chloroform fraction showed strong nitric oxide (NO) production inhibitory effect in lipopolysaccharide (LPS)-stimulated Raw 264.7 cell. In the cell viability of Raw 264.7 by MTT assay, the extract and all fractions were exhibited normal viabilities as nontoxic result. Consequently, the extract from the leaves of P. sargentii and its ethyl acetate fraction could be applicable to functional materials for antibacterial activity related fields. Moreover, the hexane and chloroform fraction could be applicable to candidate materials as anti-inflammatory agent.
In this study, n-butanol and ethyl acetate extracts were prepared from raw yam (Dioscorea batatas DECNE.). Their antioxidative potencies were investigated employing various in vitro methods, such as ferrous ion chelating, ${\beta}$-carotene bleaching assay, lipid peroxidation inhibition and nitric oxide (NO) radical scavenging and nitrite scavenging activity. The n-butanol fraction was assayed to possess stronger antioxidant activity by ${\beta}$-carotene bleaching assay, lipid peroxidation inhibition and NO radical scavenging activity. However, ethyl acetate extract was more effective in chelating ferrous ion and scavenging nitrite. Based on the results obtained, yam is a potential active ingredient that could be applied in antioxidation as well as bio-health functional food to take a good part in prevention of human diseases and aging.
Screenings of potential antioxidant activities of Crataegus pinnatifida BUNGE Major. fruits extracted 80% methanol were performed using four antioxidant assays. Significant differences were observed both in total phenolic contents (TPC) and total flavonoid contents (TFC), DPPH radical scavenging activity, nitric oxide scavenging activity, ABTS radical scavenging assay, and reducing power assay. The total polyphenol content and total flavonoid content in the extract were measured to be $224.4{\pm}0.52$ mg GAE/100 g and $12{\pm}0.25$ mg QE/100 g, respectively. When the tested concentration was $500{\mu}g/mL$, DPPH and ABTS radical-scavenging activities of methanolic extracts were 84.15% and 88.8%, respectively. The reducing power and nitric oxide scavenging activity were increased at the manner of dose-dependently. These results suggest that methanolic extracts of Crataegus pinnatifida Bge. fruits possess excellent radical scavenging activities and may serve as a potential source of natural antioxidant.
