A Study on the Azolla imbricata using as a Cosmetic Active Ingredient (물개구리밥(Azolla imbricata)을 이용한 주요 활성성분의 분리 및 화장품 소재 개발 연구)

  • The water fern Azolla belongs to the Azollaceae and forms a symbiotic association with a $N_2$-fixing cyanobacterium, referred to as Anabaena azollae, and this association has currently been demonstrated to have potential as a nitrogen source for rice production. Because of that, Azolla fern has been used not only as organic manure in southern China and northern Vietnam for a long time but also as food for animate creatures in the underwater and decontaminant in the water. However, the phenolic compounds and active materials of Azolla have not been examined in detail in the past studies. In the present study, anti-oxidant ability test and experiment to find a particular active material of Azolla imbricata and Azolla imbricata fraction (AIF) were performed. In anti-oxidant test such as DPPH test and lipoxygenase inhibition test, the value of test represented high activities compared with authentic sample - green tea and NDGA (nordihydroguaiaretic acid). In MMP-1 test, related to collagen protection and elasticity of skin, its inhibitory effect was measured over 75 %, and the phenolic compounds of AIF related with this activity were confirmed luteolin derivatives by using FT-IR spectroscopy, element analyzer (EA) and Liquid chromatography-MASS spectroscopy.

Antimetastatic Effects of Jipae-san by Inflammation Control and Activation of Innate Immune System (지패산(芷貝散) 추출물의 염증억제와 선천면역 활성에 의한 항암 효과)

  • Objectives: This study was designed to investigate the anti-tumor metastasis by anti-inflammatory and innate immunomodulating effects of extracts of Jipae-san on cancer cells. Methods: Antimetastatic experiments were conducted in vivo mouse model by using 4T1 mouse mammary carcinoma cells. Cell viability of Jipae-san was tested with 4T1 mouse mammary carcinoma cells, colon 26-M3.1 carcinoma cells and macrophage. In addition expression of $TNF-{\alpha}$ and NO induced by LPS was measured after treating with Jipae-san. To observe innate immunomodulating effects of Jipae-san on macrophage, we measured $TNF-{\alpha}$, IL-12, IL-6 and MCP-1, respectively. Cell cytotoxicity was tested with the macrophage stimulated with Jipae-san and we evaluated the activation of $TNF-{\alpha}$ and NO. And the effect of Jipae-san on metastasis was measured without NK-cell using GM1 serum. Results: Intravenous inoculation of Jipae-san significantly inhibited metastasis of 4T1 mouse mammary carcinoma cells. In an in vitro cytotoxicity analysis, cell growth are closer to 100% less than $1,000{\mu}g/ml$ concentration. The expression of $TNF-{\alpha}$ and NO induced by LPS after treating Jipae-san was down regulated in dose-dependent manner. Level of cytokines such as $TNF-{\alpha}$, IL-12, IL-6 and MCP-1 of Jipae-san group were up regulated in compared to the control group. The macrophage stimulated with Jipae-san significantly inhibits the cancer cell at ratio of 10:1, 20:1. The activation of NO was significantly up regualted in a group of 5:1, 10:1, 20:1. The depletion of NK-cells by anti-asialo GM1 serum partly abolished the inhibitory effect of Jipae-san on tumor metastasis. Conclusions: Jipae-san appears to have considerable activity on the anti-metastasis by inflammation control and activation of innate immune system.

Studies on the Composition and Antioxidative Effect of Leaves form Korean Rosa davurica Pall. (한국산 생열귀나무(Rosa davurica Pall.) 잎의 성분 및 항산화 효과에 관한 연구)

  • The objective of this study was carried out to investigate nutritional characteristics and biological activities effects of Korean leaf of Rosa davurica Pall. in vitro. They were extracted with methanol and then further fractionated to n-hexane, chloroform, ethyl-acetate, n-butanol and water from methanol extracts. Methods of the antigenotoxic used in this experiment were UVA/UVB absorption property and DPPH radical scavenge. The proximate compositions of leaves of Rosa dauvrica Pall were 67.5% of crude Moisture, 0.7% of crude fat, 6.8% of crude protein, 6.1% of crude ash, and 20.8% of crude fiber. The major minerals were K (1637.2 mg%), Ca (219.5 mg%), P (182.1 mg%), and Mg (135.1 mg%). Most of the fractions of methanol extract which leaves of Rosa dauvrica Pall. have strong absorbency at UVB region (308 nm) and UV A region (350nm). These fractions have a good absorbency property as synthetic filter and could be served as substitutes for synthetic UV sunscreen agents. All fractions (n-hexane, ethyl-acetate, n-butanol and water) from methanol extracts except chloroform fraction exhibited DPPH radical scavenging activity with IC$\_$50/ of 35.3, 6.0, 14.0, and 18.0 $\mu\textrm{g}$/mL.

Effects of Fermentation Pine Needle Extract on the Quality of Plain Bread (솔잎 발효액이 식빵의 품질에 미치는 영향)

  • In efforts to use pine needle extract as a substitute for sugar, fermented pine needle extract syrup was added to the wheat flour nea in the manufacture of white bread The extract was added to levels of 8.3%, 11% and 18% of total weight based on the Brix degree of the dough The cohesion of each dough was checked, and the physical properties and storage stabilities of the baked breads were analyzed during storage at mom temperature. Analysis of the cohesion of each dough by farinogram showed that dough with pine needle extract was better than dough with sugar, in terms of both stability and durability of mixing. The pH of dough with syrup was steady during storage at pH 5.4 5.8, which is favorable for yeast activity. The dough with syrup also showed low firmness and good extensibility, both of which would favorably affect gas retention on fermentation. Increases in syrup addition resulted in higher product volumes. Bread with syrup was slow to increase in hardness during storage, suggesting that higher syrup concentrations inhibited development of staleness. The addition of syrup also inhibited the growth of aerobic bacteria and mold on the bread surface. The addition of syn did not cause any negative effect. The use of pine none extract syrup may thus contribute to improvements the physical properties, the storage stability, and the hygienic quality of bread.

Nutritional Components and Antioxidative Activities of Jujube (Zizyphus jujuba) Fruit and Leaf (대추 열매와 잎의 영양성분 및 항산화 활성)

  • The nutritional components and antioxidative activities of jujube fruit and leaf were investigated and analyzed to provide basic data for functional food materialization and processing. The nitrogen-free extract contents of the fruit and leaf were 71.92% and 41.51%, respectively. The mineral components of the fruit and leaf were rich in Ca (72.14 and 3,252.09 mg/100 g), K (899.82 and 1,708.12 mg/100 g), and P (172.11 and 286.28 mg/100 g), respectively. The major free sugars of the fruit were glucose (13.01 %) and fructose (7.35%); and of the leaf, sucrose (3.94%) and fructose (0.75%). The ascorbic acid contents were higher in fruit (135.73 mg/100 g) than in the leaf (100.43 mg/100 g). The analysis of the component amino acid showed a relatively high ratio of glutamic acid, aspartic acid, proline, and essential amino acids of leucine, but a low methionine and cystine content. The ABTS and FRAP assays indicated that the butanol fraction of the leaf was a more potent radical scavenger and reducing agent than the other five solvent fractions. The butanol fraction of the leaf also presented inhibitory effects against lipid peroxidation in a dose-dependent manner. Therefore, this study verified that the butanol fraction of the leaf has strong antioxidative activities that are correlated with its high level of phenolics, particularly rutin and quercitrin. These phenolics of jujube leaf can be utilized as effective and safe functional food substances, i.e., natural antioxidants.

Application of Electrical Resistivity Measurement to an Evaluation of Saline Soil in Cropping Field (염류집적 농경지에서 전기비저항 탐사기법의 활용성)

  • Salinity of soil under the plastic film houses in Korea is known as a significant factor to lower the crop production and to hamper the sustainable agricultural land management. In this study we propose a field monitoring technique to examine the methods applied to minimize the adverse effect of salts in soil based on the relationship between soil electrical characteristics and soil properties. Field experiments for 4 different treatments (water only, fertilizer only, DTPA only, and DTPA and fertilizer together) were conducted on soils at the plastic film house built for cultivating a cucumber plant located at Chunan-si, Chungchungnam-do in Korea. The electrical resistivity was measured by both a dipole-dipole and wenner multi-electrodes array method. After the electrical resistivity measurement we also measured the soil water content, temperature, and electrical conductivity on surface soil. The resulted image of the interpreted resistivity by the inversion technique presented a unique spatial distribution depending on the treatment, implying the effect of the different chemical components. It was also highly suspected that resistivity response changed with the nutrients level, suggesting that our proposed technique could be the effective tool for the monitoring soil water as well as nutrient during the cropping period. Especially, subsoils under DTPA treatment at 40 to 60 cm depth typically presented lower soil water accumulation comparing to subsoils under non-DTPA treatment. It is considered that DTPA resulted in increase of a root water uptake. However, our demonstrated results were mainly based on qualitative comparison. Further experiments need to be conducted to monitor temporal changes of electrical resistivity using time lapse analysis, providing that a plant root activity difference based on changes of soil water and nutrients level in time.

Anti-oxidative capacity of mulberry genetic resources (뽕나무 유전자원의 항산화능 비교)

  • Much attention has been focused on the activity of the natural antioxidants present in fruits and vegetables, because potentially these components may reduce the level of oxidative stress. Especially, mulberry leaves containing many natural components are considerable resource for natural antioxidants. The antioxidant capacity of mulberry leaves was investigated with minilum L-100 device and ARAW-KIT (anti-radical ability of water-soluble substance), in comparison to the ascorbic acid. The antioxidant capacity of 16 varieties was 3303.4 nmol at opening stage of five leaves in spring. The highest stage of antioxidant capacity (3708.0 nmol) and yield rate was just before the coloration stage with anthocyanin in fruits, whereas the lowest stage was middle of June (2231.6 nmol) and about two months growing stage after summer pruning (2064.6 nmol). But after summer pruning, the antioxidant capacity of mulberry leaves increased gradually until just before fallen leaves stage. Even if samples were same variety, antioxidant effect of those showed different results according to collected regions. Also, antioxidant effect of mulberry leaves were higher than that of branches. The antioxidant capacity of yield-type mulberry leaves and fruits (Morus alba L., M. bombycis Koidz, and M. Lhou (Ser.) Koidz) collected from In-je, Won-ju and Yang-yang regions, Kang-won province, Korea, was investigated. The results indicated that total antioxidant capacity of yield-type mulberry leaves was 2711.2 nmol. In the antioxidant capacity analysis of Jeollabuk-Do genetic resources, autumn's mulberry leaves showed higher antioxidant capacity than that of spring's it. To investigate the effect of tea on antioxidative capacity, five kinds of tea(coffee mix, green tea added brown rice, mulberry leaf tea, Polygonatum odoratum tea and black tea added lemon) were selected and analyzed. Their's anti-oxidative capacity were 2,531.01 nmol, 1,867.42 nmol, 1,053.72 nmol, 292.71 nmol and 188.91 nmol, respectively. The antioxidative capacity of drinking water soaked with mulberry leaf showed 891.96 nmol.

Isolation and purification of a cecropin-like antimicrobial peptide from the japanese oak silkworm, Antheraea yamamai (천잠 세크로핀 항균펩타이드 분리 및 정제)

  • Cecropin is a well-studied antimicrobial peptide that play important role as key factor in insect humoral immunity. In this study, cecropin-like antimicrobial peptide was isolated and purified from the larval haemolymph of immune-challenged japanese oak silkworm, Antheraea yamamai. To isolate antimicrobial peptide, we separated and compared acidic extracted hemolymph protein bends between control and immune-challenged larvae using SDS-PAGE analysis. In the immune hemolymph extract, but not of non-immune hemolymph, we detected differential expressed peptide band with molecular mass 4,223.01 Da. To understand this peptide better, we successfully purified this peptide using cation exchange chromatography and gel permeation chromatography. Its N-terminal amino acid sequence obtained by Edman degradation evidenced a significant degree of identity with other lepidopteran cecropins. The purified A. yamamai cecropin-like peptide showed a broad spectrum of activity against fungi, Gram-negative and Gram-positive bacteria.

Phosphate Solubilizing Activity of Pseudomonas sp. CL-1 and Kluyvera sp. CL-2 (Pseudomonas sp. CL-1 및 Kluyvera sp. CL-2 균주의 인산가용화 특성)

  • From the rhizoplane and rhizosphere of pepper, tomato, lettuce, pasture, and grass, unsoluble inorganic phosphate solubilizing bacterial strains were isolated using plate base assay on Pikovskaya's medium. Two strains, CL-1 and CL-2, which produced largest halo on plates (indicative of phosphate solubilization)were selected for further studies. Based on these biochemical and 16S rRNA analysis strains CL-1, CL-2 were found to be as species of Pseudomonas sp. and Kluyvera sp., respectively. In broth assay Pseudomonas sp. CL-1 and Kluyvera sp. CL-2 solubilized insoluble phosphate by 193.4 mg and $493.6P\;mg\;L^{-1}$, respectively after $3^{rd}$ day inoculation. These effecient phosphate solubilizing bacteria have a potential to be developed as microbial based fertilizer in future.

Analyzing the Occurrence Trend of Sediment-Related Disasters and Post-Disaster Recovery Cases in Mountain Regions in N orth Korea Based on a Literature Review and Satellite Image Observations (문헌 및 위성영상에 기초한 북한의 산지토사재해 발생경향 및 복구사례 분석)

  • This study investigated spatiotemporal trends of sediment-related disasters in North Korea from 1960 to 2019 and post-disaster recovery cases based on a literature review and satellite images. Results showed that occurrence status of sediment-related disasters was initially externally reported in 1995 (during the Kim Jongil era); their main triggering factor was heavy summer rainfall. Furthermore, forest degradation rate was positively correlated with population density (R2 = 0.4347, p = 0.02) and occurrence number of sediment-related disasters was relatively high on the west coast region, where both variables showed high values. This indicates that human activity was a major cause of forest degradation and thus, significantly affected sediment-related disasters in mountain regions. Finally, sediment- related disasters due to shallow landslides, debris flow, and slow-moving landslides were observed in undisturbed forest regions and human-impacted forest regions, including terraced fields, opencast mines, forest roads, and post-wildfire areas, via satellite image analysis. These disaster-hit areas remained mostly abandoned without any recovery works, whereas hillside erosion control work (e.g., treeplanting with terracing) or torrent erosion control work (e.g., check dam, debris flow guide bank) were implemented in certain areas. These findings can provide reference information to expand inter-Korean exchange and cooperation in forest rehabilitation and erosion control works of North Korea.
