Molecular Biological Study of Anti-cancer Effects of Bee Venom Aqua-acupuncture (봉독약침(蜂毒藥鍼)의 항암효과(抗癌效果)에 대한 분자생물학적(分子生物學的) 연구(硏究))

  • To study anti-cancer effect and molecular biological mechanism of bee venom for aqua-acupuncture, the effects of bee venom on cell viability and apoptosis were analyzed using MTT assay, tryphan blue assay, $[^3H]$thymidine release assay, flow cytometric analysis, and activity of caspase-3 protease activity assay. To explore whether anti-cancer effects of bee venom are associated with the transcriptional control of gene expression, quantitative RT-PCR analysis of apoptosis-related genes was performed. The obtained results are summarized as follows: 1. The MTT assay demonstrated that cell viability was decreased by bee venom in a dose-dependant manner. 2. Significant induction of apoptosis was identified using tryphan blue assay, $[^3H]$thymidine release assay, and flow cytomet1 ric analysis of sub $G_1$ fraction. 3. In analysis of caspase-3 protease activity, the activity had increased significantly, in a dose-dependant manner. 4. Quantitative RT-PCR analysis of the apoptosis-related genes showed that Bcl-2 and Bcl-$X_L$ were down-regulated whereas Bax was up-regulated by bee venom treatment.

  • Acoording to improvement of HTOS (high throughput organic synthesis) and HTS (high throughput screening) technique, the CoMFA (comparative molecular field analysis), CoMSIA (comparative molecular similarity indeces analysis) and molecular HQSAR (hologram quantitative structure-activity relationship) analysis techniques as methodology of computer assisted molecular design (CAMD) were introduced generally and summarized for some application cases.

  • The purpose of this paper was the determination of the relationship between the chaotic charateristics and various levels of brain activities. Assuming that EEG(eletroencephalogram), which is generated by a nonlinear electiecal behavior of billions of neurons in the brain, has chaotic characteristics, it was confirmed by frequency spectrum analysis, log frequency spectrum analysis, correlation dimension analysis and Lyapunov exponents analysis. Chaotic characteristics are related to the degree of brain activity. The slope of log frequency spectrum increased and the correlation dimension decreased with respect to the brain activities, while the lagrest Lyapunov exponent has some rough correlation.

  • The purpose of this thesis is to provide basic information of efficient flow line arrangement, which results from examining the factors of route choice with a focus on the store' space elements and pedestrian activity patterns in the outlets. The route tracking is applied to grasp pedestrian activity patterns, therefore, a concrete analysis into influential factors of the space structure arrangement and forms on route choice. The route tracking is a means to understand pedestrian activity patterns by establishing an unrecognizable space and examining every route in the investigating area for pedestrian activity pattern research. Three sample stores have different systems in the ground plan structures, the escalator location and the directions. The analysis focuses on the booth arrangement and shopping patterns. In conclusion, route choice of the customers and the structure of the space are quite closely related and affect the quality of shopping. This can suggest evidence for the need of the space structure to meet the pedestrian activity patterns.

  • Proteases are one of the most abundant classes of enzymes in living organisms and have been considered major targets for drug development. However, despite the ability to specifically cleave their substrates, many attempts to assay protease activity have generally relied upon the use of gel zymography or fluorophore-labeled peptide substrates, which is limited in rapid and multiplex analysis. Here we review the recent advances in nanoparticle (NP)-utilized assays of protease activity focused on in vitro and cell imaging-based approaches. Owing to large surface area and unprecedented physical properties of NPs, these approaches are anticipated to facilitate many applications related to protease activity-based disease diagnosis and drug discovery.

Chemical Analysis and Biological Activity of Endotoxin from Vibrio vulnificus

  • Vibrio vulnificus endotoxin was extracted, analyzed the chemical composition, tested its biological activity, and compared to those of Escherichia coli and Salmonella typhimurium. The major fatty acid of three endotoxins were different each other; V. vulnificus endotoxins were different each other; V. vulnificus endotoxin was myristric acid (C14:0), E. coli was lauric acid (C12:0), S. typhimurium was capric acid (C10:0). The biological activities of V. vulnificus endotoxin were similar to those of E. coli and S. typhimurium in terms of the gelation activity of the Limulus amebocyte lysate and the lethal toxicity. But the result of enzyme (AST, ALT, and LDH) analysis showed that the enzyme activity of V. vulnificus endotoxin was similar to that of E. coli, but smaller than that of S. typimurium.

  • The purpose of this study was to investigate the influence of physical activity on communication and adaptation to school life of elementary school students. Seventy-two students (male 37; female 35) from two classes participated in the experiment. One class served as an experimental group and the other as a control group. The program consists of 16 sessions. Participants completed the Communication Scale and School Adjustment Scale. The collected data were analyzed by using t-test and covariance analysis. The results of the analysis showed that there were statistically significant differences between elementary school students who participated in physical activity and ones who did not in terms of communication and school life adaptation. Based upon these results, it is concluded that physical activity is effective for communication and school life adaptation of elementary school students.

  • Since 2005, POSCO has been conducting continuous innovation activities using its own innovation methodology, QSS + (Quick Six Sigma Plus). Recently, the effectiveness of QSS + has been decreased owing to the different needs and purposes for each group. The purpose of this study is to compare the weights of the QSS + factors using AHP analysis. QSS + factors consist of two levels. The high level consists of 7 items : 'Master Plan, Waste Zero Activity, Daily Improvement Activity, Talent Development, Project Activity, Initiative/Encouragement activity'. The low level consists of 3 items '5S, My Machine, My M&S' for Daily Improvement Activity. As a result, we found that there was a difference between the weights of the four groups. We suggest the effective QSS + directions to improve the quality of POSCO.

  • Sulfated polysaccharides from plants or seaweed are well known for antioxidant, anticoagulant activity or other biological activities. The aim of the present study is to optimize the condition for the isolation of water soluble polysaccharides (WSP) and evaluate the biological activity. WSP was isolated from defatted microalgae B. braunii using chlorosulfonic acid or sulfuric acid. Among WSP isolated using sulfuric acid showed the lowest the content of total carbohydrates (g/L), whereas showed the highest antioxidant activity. Also the preliminary structural analysis of WSP was performed by FT-IR spectroscopy analysis.

A Study on Good Use of Leisure Among the Retired Elderly in Korea (우리나라 노인의 은퇴 후 여가선용에 관한 연구)

  • The purposes of this study were to investigate the actual conditions of leisure life of the retired elderly in Korea and to analyze the difference of their leisure activity types by socio-demographic characteristics, the factors which have an effect on their satisfaction of leisure life. This study was secondary analysis of the national data; from The Social Statistics Survey; collected by the Korea National Statistical Office in 2000. The major findings are as follows. 1) The types of leisure activity of the retired elderly included watching television and doing nothing. 2) There were significant differences in the types of leisure activity by sex, age, education degree, having spouse or not, having house or not, and residential area. 3) Results of logistic regression analysis indicated that the most effective factors of the leisure life satisfaction were sex, age, having spouse or not, having house or not, residential area and the types of leisure activity.
