Comparison Task-Oriented Training according to the Applicable Blocked Practice and Random Practice: Chronic Hemiplegic Patients

  • Purpose: The purpose of this study was to compare the blocked practice and random practice of task-oriented training in patients with chronic stroke to determine the effect of lower extremity muscle activity and balance ability. Methods: The thirty participants were randomly assigned to either the block practice group (BP) group or the random practice group (RP) and received the training three times per week, 30 minutes per day, for six weeks. Surface electromyography was used for measurement of lower extremity muscle activity. Static balance was to measured the stability index (SI) and weight distribution index (WDI) using the Tetrax. The four square step test (FSST) was used to measure dynamic balance. The paired t-test was used for determination of differences before and after intervention, and the independent t-test was used for determination of differences between groups. Results: Lower extremity muscle activity, RA and GCM was improved in the RP group after intervention and between groups. TA was significantly improved in the RP group compared with the BP group. In comparison of before and after interventions, SI was reduced in BP and RP. WDI in OS was reduced in comparison of BP and RP before and after intervention. CS was reduced in BP and RP. The OS and CS was improved in RP compared with BP. In comparison of before and after intervention, FSST was improved in BP and RP. Conclusion: Task-oriented training methods using random practice was found to be effective in promoting lower extremity muscle activity and balance ability in chronic stroke patients.

산림토양내 carboxymethylcellulase의 분포와 미생물의 생장 및 활성과의 상관에 대하여 (Distribution of abiontic carboxymethylcellulase in relation to microbial growth and activity in forest soils)

  • Seasonal and vertical variations of abiontic soil carboxymethylcellulase (CMCase) activities were assessed every other month for a year in two contrasting forest soils and evaluated the relationships between soil CMCase activity and environmental parameters. In climax deciduous soil, variations in CMCase activities caused by differences in sampling time were greater than those caused by differences in soil depth. On the other hand, counter phenomenon was obserned in coniferous soil at the stage of development. Correlation analyses showed that soil CMCase activities were significantly (p>0.01) correlated with microbial respiration rates ($O_2$ uptake) and all of the microbial population sizes. From these results, it is suggested that determination of abiontic soil CMCase activity is an useful additional index for evaluating the overall microbial growth and activity in soils.

향신료의 약물대사효소 CYP3A4 저해효과 (Inhibitory Effect of a Drug Metabolizing Enzyme CYP3A4 on Spices)

  • For the determination of inhibiting cytochrome P450(CYP)3A4 activity, an improvement HPLC method was established by using a new internal standard and solvent system. Moreover, CYP3A4 amount for a optimum reaction of enzyme was determined by a comparative study with a variety concentration of enzyme. Using a established method, inhibitory effect of CYP3A4 that is drug metabolizing enzyme Investigated on EtOAc extracts of 5-class spices. As a result of experiment, EtOAc extract of white pepper (Piper nigrum L.) showed strong inhibitory activity. On a continuous experiment, the fraction 2, 4 and 5 of while pepper extract showed remarkable inhibitory activity. Pipeline, a main constituent of pepper was not included in these fraction. It is suggested that major compounds for the inhibitory activity of white pepper may be other ingredient that is not piperine.

N-Acetylglycine Side Chain is Critical for the Antimicrobial Activity of Xanthostatin

  • This study was carried out to elucidate the mode of bacteriostatic property of xanthostatin (XS), a novel depsipeptide antibiotic with an N-acetylglycine side chain and selective antimicrobial activity against Xanthomonas spp. Two biotransformed XSs were isolated by the treatment of XS with the cell lysate of Xanthomonas campestris pv. citri, a solvent partition, preparative TLC, and HPLC. Structure determination of those two biotransformed XSs demonstrated deletion of the N-acetylglycine side chain. Noteworthily, they showed no antimicrobial activity against Xanthomonas spp. This result suggests that the N-acetylglycine side chain plays a critical role in the antimicrobial activity of XS, and that the bacteriostatic property of XS is due to susceptibility of the ester bond between the hexadepsipeptide nucleus and the N-acetylglycine side chain to hydrolytic enzyme(s) produced by Xanthomonas spp.

Estimation of Death Time by Changes of Postmortem Xanthine Oxidase Activity in Rats

  • To evaluate the postmortem changes in activities of oxygen free radical metabolizing enzymes, the rats were sacrificed with cervical dislocation and were kept in an incubator at $25^{\circ}C$, 70% of humidity for 12 hours. The activities of aniline hydroxylase, catalase, glutathione-S-transferase and superoxlde dismutase were decreased with the time. On the other hand, the activity and type conversion ratio (type D ${\to}$type O) of hepatic xanthine oxidase (XO) were gradually increased. From these changes of XO, the estimation of death time (mathematical equation) could be determined with the least square method. To clarify the cause of increasing XO activity, enzyme kinetics were examined. The Km values of XO were decreased with the time. In conclusion, the determination of liver XO activity might be used for the estimation of death time in the early postmortem period.

Species Difference in the Inhibition of Alcoholdehydrogenase by cArnitine and Acetylcarnitine

  • Acetylcarnitine, a metabilite of carnitine, has been porven to be a potent inhibitor of ethanol oxidation in hepatocytes. It inhibits the activity of alcohol dehydrognase (ADH), but not the microsomal ethanol oxidizing system. which was significatly inhibited by acetylcarnitine at NAD ; acetylcarnitine $\leq$1. the main objectives of his study were to ascertain the interaction between acetylcarnitine and NAD on ADH activity and to elucidate whether different species have different effects. Tehpost-mocrosomal supernatant (PMS) was prepared from normal rat, guinea pig, mouse and broilers by differential centrifugation . Horse and yeast ADH were purchased from the Sigma Chemical Co. Prepared and purchased ADH are used for determination of ADH activity in the presence or absence of carnitine and acetylcar- nitine. Binding studies showed that acetylcarnitine did bind to ADH in a dose realted manner when low NAD ; acetylcar- nitine ratio was provided. It was found that the inhibitionof ADH activity occurred only when NAD concentration was less than the inhibitor concentration . Crystalline and crude ADH preparation from different vertebrate species wer inhibited by acetylcarnitine, whereas the yeast ADH was not affected by acetylcarnitine.

보골지 및 염초보골지의 proteasome 저해 작용 (Protesome Inhibition Activity of Psoraleae Semen and Processed Psoraleae Semen)

  • Ubiquitin-proteasome proteolytic system plays an important role in selective protein degradation and regulates cellular events including apoptosis. Cancer cells have been shown to be more sensitive to the proapoptotic effects of proteasome inhibition than normal cells. Thus, proteasome inhibitor can be potential anticancer agent. Since the MeOH extracts of psoraleae semen and processed psoraleae semen showed potent proteasome inhibition activity, the fractions of the extracts were evaluated on the activity to screen the proteasome inhibitors. The $CHCl_3$ fr. of the processed psoraleae semen showed the most potent activity, of which chemical investigation led to two coumarins, psoralen and isopsoralen. Their structures were determined by spectroscopic methods such as $^1H-NMR$ and EIMS spectra.

  • Hypocholesterolemic peptide isolated from glycimin (11S protein) hydrolyzate by trypsin was purified and identified as LPYP and IAVPGEVA. To investigate the effects of phyiscal properties of side chains of the hypocholesterolemic activity, some of mutant peptides were designed and synthesized chemically. The structure related structures of each peptide were simulated and constructed and their conformations were observed by using spectropolarimeter. The hypocholesterolemic activities were monitored by assaying the inhibition of 3-hydroxy-3-methylglutaryl CoA reductase (HMG-CoA reductase) in vitro and by the determination of cholesterol content in mice serum. For LPYP derivatives, Hypocholesterolemic activity was lost when hydrophobic leucine residue at N-terminus was not so critical for maintaining hypocholesterolemic activity. For idealogical design of hypocholesterolemic peptides, the structure of HMG-CoA reductase are shown and inhibition mechanism of some peptides or inhibitors will be presented. For IAVPGEVA derivative inhibition of HMG-CoA reductase has been studied. For detail study of hypocholesterolemic activity, kinetic study of inhibition of peptides on HMG-CoA reductase and structural view of ligand binding should be investigated.

In vitro anti oxidant activity of methanol extract of Clerodendrum infortunatum Linn

  • The antioxidant potency of methanolic extract Clerodendrum infortunatum Linn. (MECI), which are widely used in the Indian indigenous system of medicine for different purposes, was studied. The antioxidant potential was evaluated using different established in vitro antioxidant tests viz. determination of total amount of polyphenolics compounds, DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging, nitric oxide scavenging, superoxide anion radical scavenging, hydroxyl radical scavenging and reductive power assay. It was found that MECI contain a high amount of polyphenolics and possesses significant free radical scavenging activity in all the assay. The higher activity was may be due to presence of richest amount of polyphenolics and flavonoids in it.

Antifungal Activity of Valinomycin, a Cyclodepsipeptide from Streptomyces padanus TH-04

  • Using antifungal activity-guided fractionation, an actinomycete, Streptomyces padanus strain TH-04, which was isolated from mummified peaches infected with Monilinia fructicola, was found to produce the valinomycin (1). The structure of 1 was established using spectroscopic data, which including one- and two-dimensional NMR experimental and mass spectroscopy. Valinomycin (1) showed antifungal activity against Phytophthora capsici, with an IC$_{50}$ value of 15.9 ${\mu}$g/mL.
