Evaluation of Arabinofuranosidase and Xylanase Activities of Geobacillus spp. Isolated from Some Hot Springs in Turkey

  • Some hot springs located in the west of Turkey were investigated with respect to the presence of thermophilic microorganisms. Based on phenotyping characteristics and 16S rRNA gene sequence analysis, 16 of the isolates belonged to the genus Geobacillus and grew optimally at about $60^{\circ}C$ on nutrient agar. 16S rRNA gene sequence analysis showed that these isolates resembled Geobacillus species by ${\ge}97%$, but SDS-PAGE profiles of these 16 isolates differ from some of the other species of the genus Geobacillus. However, it is also known that analysis of 16S rRNA gene sequences may be insufficient to distinguish between some species. It is proposed that recN sequence comparisons could accurately measure genome similarities for the Geobacillus genus. Based on recN sequence analysis, isolates 11, IT3, and 12 are strains of G stearothermophilus; isolate 14.3 is a strain of G thermodenitrificans; isolates 9.1, IT4.1, and 4.5 are uncertain and it is required to make further analysis. The presence of xylanase and arabinofuranosidase activities, and their optimum temperature and pH were also investigated. These results showed that 7 of the strains have both xylanase and arabinofuranosidase activities, 4 of them has only xylanase, and the remaning 5 strains have neither of these activities. The isolates 9.1, 7.1, and 3.3 have the highest temperature optima ($80^{\circ}C$), and 7.2, 9.1, AO4, 9.2, and AO17 have the highest pH optima (pH 8) of xylanase. Isolates 7.2, AO4, AC15, and 12 have optimum arabinofuranosidase activities at $75^{\circ}C$, and only isolate AC15 has the lowest pH of 5.5.

  • A GABA synthesizing enzyme, glutamate decarboxylase, has been purified from bovine brain by several chromatographic procedures. The preparation appeared homogeneous on SDS-PAGE. The enzyme is a homodimeric protein with a molecular mass of 120 kDa. The activation of glutamate decarboxylase by ginesenosides from Panax ginseng C.A. Meyer has been studied. Preincubation of the enzyme with total ginsenoside, $Rb_2$ and Rc ginsenosides, increased glutamate decarboxylase activities in a dose-dependent manner. There was a reproducible decrease in $K_m$, in addition to a increase in $V_{max}$, in response to increasing concentrations of the Rc ginsenoside fraction. Upon addition of the ginsenoside to the enzyme, a decrease in flurorescence intensity was discernible, together with an increase in emission anisotropy. Judging from the anisotropy values, the ginsenoside is rapidly trapped by the protein matrix. Total ginsenoside was administered to rats and the rat brains were removed for the measurement of the changes of GABA shunt regulating enzyme activities. Among the GABA shunt regulating enzymes, only the glutamate decarboxylase activities were increased after ginsenoside treatment. Therefore, it is suggested that the ginsenosides may elevate the GABA level in brain by activation of glutamate decarboxylase and the enzymatic activation might be due to the conformational change induced by binding of ginsenoside to the enzyme.

우리나라 최초의 보건간호학 교과서 "공중위생간호학" 연구 (A Study on the Gongjungwisaeng Kanhohak, the First Korean Textbook for Public Health Nursing)

  • Purpose: The purpose of this study is to extend the knowledge about the textbook, Public Health Nursing Textbook (in Korean, Gongjungwisaeng Kanhohak) which is published in 1933 by the Nurses' Association of Korea. Methods: Public Health Nursing Textbook was investigated from the cover page to the last page and compared with two original books and other nursing books published in modern Korea. Results: Public Health Nursing Textbook was the forth published Korean nursing textbook and the first published Korean public health nursing textbook. Rosenberger, a missionary nurse from USA, Lee Keumjeon, Korean public health nurse who studied in Canada, and other Koreans were in charge of translation, etc. It is a user - friendly book written mainly in Korean (Hangul) with Chinese characters and included the English table of contents, preface and back cover. Most of its contents were extracted and translated from two original books, one is Public Health Nursing by Mary Gardner and the other is Personal Hygiene Applied by Jesse Williams. It is worth noting that the book not only introduced the public health nurses's activities across the country, but also focused on the information necessary for common health problems at that time. Conclusion: From the Public Hygiene and Nursing, it can be seen that public health nurses had been independently educated since Imperial Japan's colonial period, and public health nursing was growing as a independent field in Korea.

Enzyme Activities and Substrate Degradation by Fungal Isolates on Cassava Waste During Solid State Fermentation

  • The growth and bioconversion potential of selected strains growing on cassava waste substrate during solid state fermentation were assessed. Rhizopus stolonifer showed the highest and the fastest utilization of starch and cellulose in the cassava waste substrate. It showed 70% starch utilization and 81% cellulose utilization within eight days. The release of reducing sugars indicating the substrate saccharification or degradation potential of the organisms reached the highest value of 406.5 mg/g by R. stolonifer on cassava waste during the eighth day of fermentation. The protein content was gradually increased (89.4 mg/g) on the eighth day of fermentation in cassava waste by R. stolonifer. The cellulase and amylase activity is higher in R. stolonifer than A. niger and P. chrysosporium. The molecular mass of purified amylase and cellulase seemed to be 75 KDal, 85 KDal respectively.

Oxyfluorfen에 대한 내성(耐性) 및 감수성(感受性) 수도품종(水稻品種)의 전기영동(電氣泳動) 표현형(表現型) 차이(差異) (Difference in Electrophoretic Phenotypes of Rice Cultivars Selected to Oxyfluorfen)

  • Oxyfluorfen에 대표적(代表的)인 내성군(耐性群)(Chokoto, Aichiasahi, Agabyeo, IR 3941, Tabali)과 감수성품종군(感受性品種群)(Mushakdanti, Weld pally, HP 1033, HP 857, HP 907)을 공시(供試)하여 종실단백질(種實蛋白質)(SDS-PAGE 법(法))과 동위효소(同位酵素)(acid phosphatase, peroxidase: 7% PAGE 법(法))의 전기영동(電氣泳動) pattern을 분석(分析)하고 oxyfluorfen $10^{-4}M$ 처리후(處理後)의 동위효소(同位酵素)(malate dehydrogenase, peroxidase, esterase) pattern의 특이성(特異性)을 확인(確認)하므로써 oxyfluorfen 내성차이(耐性差異)와 이들 전기영동(電氣泳動) pattern 차이(差異)가 갖는 연관성(聯關性)을 알고저 하였다. 결과(結果)를 요약(要約)하면 다음과 같다. l. 종실단백질(種實蛋白質)의 전기영동(電氣泳動)에서는 19종(種) band를 찾아볼 수 있었고, 두 품종군간(品種群間)에 비유사성(非類似性)이 있는 것으로 clustering 되었다. 이는 감수성군(感受性群)에서 G band가 없고, H, N, O, P, Q, R, S band에서 활성(活性)이 낮았던 데 기인한다. 2. 품종군간(品種群間)의 동위효소(同位酵素) 분석결과(分析結果), 4개(個)의 acid phosphatase band 가운데 내성군(耐性群)은 B band의 활성(活性)이 낮으나 특이적(特異的)으로 C band를 갖는 공통점(共通點)이 있었다. Peroxidase의 4개(個) minor band에 대하여는 내성군(耐性群)이 A, C, D band의 활성(活性)이 높고 B band의 활성(活性)은 거의 없었던 특이성(特異性)이 있었다. 3. Oxyfluorfen 처리(處理)에 기인한 동위효소(同位酵素)의 경시적(經時的) 변화(變化)로 보아, 내성(耐性)인 Chokoto에서는 malate dehydrogenase의 3개(個) band 가운데 A band의 활성(活性)은 거의 없었고 B 및 C band의 활성(活性)은 지속적(持續的)으로 높게 유지(維持)되는 특이성(特異性)이 있었다. Peroxidase의 5개(個) band 가운데 B band는 내성(耐性)인 Chokoto에서 발견되지 않았으나 A, C, D, E band는 경시적(輕視的)으로도 소실경향(消失傾向)이 크지 않았다. Esterase는 대략 4개(個)의 band로 분리(分離)되었는데 내성(耐性)인 Chokoto는 A, C, D band가 높은 활성(活性)을 갖고 지속적(持續的)으로 유지되는 특이성(特異性)을 나타내었다.

  • 카카오엔터테인먼트는 영상 및 음악 콘텐츠를 맡고 있는 카카오M과 웹툰, 웹소설을 담당하고 있는 카카오페이지가 합병을 통해 탄생된 대형 콘텐츠 기업이다. 참고로, 카카오M은 15개가 넘는 콘텐츠 및 매니지먼트 기획사와 4개의 음반 레이블을 보유하고 있으며 카카오페이지 역시 8,500개가 넘는 콘텐츠 IP를 축적했다. 본 연구는 두 기업의 합병을 통해 탄생된 카카오 엔터테인먼트 사례연구를 토대로 흡수역량을 확보하기 위한 M&A 활동과 경계관리 활동의 폭을 규정할 수 있는 콘텐츠 가치사슬에 관한 관계를 세밀히 분석하였다. 카카오엔터테인먼트 사례연구를 통해 본 연구가 제안한 주요 결과는 다음과 같다. 카카오의 인수합병 활동은 엔터테인먼트 산업에서 흡수역량을 축적하는 핵심 요인이며 동시에 경쟁우위를 가질 수 있는 여러 자산과 강력한 네트워크를 형성하는 데 효과적이다. 이와 함께 콘텐츠 비즈니스의 경쟁 강도가 점점 높아짐에 따라 카카오는 지속적으로 산업 경계선을 넘나들며 경계관리 활동을 토대로 창의적인 기회를 포착할 필요가 있다. 카카오엔터테인먼트가 인수합병과 가치사슬을 통해 확보한 흡수역량과 경계관리 활동의 활성화는 엔터테인먼트 산업의 지배 전략에 가장 적합하다고 볼 수 있다.

Degradation of Polyvinyl Alcohol by Brevibacillus laterosporus: metabolic Pathway of Polyvinyl Alcohol to Acetate

  • Approximately 0.1 mg/ml of polyvinyl alcohol (PVA) was degraded by the growing cell, Brevibacillus laterospours, for 30 h, and 0.2 mg/ml of PVA was degraded by the cell-free extract that was isolated from Brevibacillus laterosporus. Approximately $0.29{\mu}g$/ml of acetic acid was produced from PVA by using the cell-free extract as a catalyst for 40 min. $V_{max}\;and\;K_m$ value of purified PAV-degradation enzyme was 3.75g/l and 2.75 g/l/min in reaction with EDTA and 3.99 g/l and 2.98 g/l/min in reaction without EDTA, respectively. Molecular weight of the purified enzyme determined by SDS-PAGE was 63,000 Da. Alcohol dehydrogenase and aldehyde dehydrogenase activities were qualitatively detected on a native acrylamide gel by an active staining method, indicating the existence of the metabolic pathway to use PVA as a substrate.

Antifungal, Nematicidal and Antioxidant Activity of the Methanol Extracts Obtained from Medicinal Plants

  • The nematicidal, antifungal and antioxidant activities of methanol extracts from six Vietnamese native medicinal plants were evaluated by various assays in vitro. Of the plant extracts tested, Terminalia nigrovenulosa was found to possess the highest activity when compared to the others. The leaves and bark of T. nigrovenulosa showed strong inhibitory activity against Meloidogyne incognita and Fusarium solani. The DPPH (1,1-Diphenyl-2-picryl-hydrazyl) radical scavenging, reducing power and total antioxidant activities of T. nigrovenulosa bark were higher than that of the remainder plant extracts. Chitinase activity of these plants was also investigated using SDS-PAGE. The results obtained in the present study indicate that T. nigrovenulosa leaf extracts are the greatest potential source as nematicides and fungicides for the control of M. incognita and F. solani. In particularly, T. nigrovenulosa bark extracts could be used as a potential source of commercially viable levels of natural antioxidant.

벼잎 절편에서 Polyamine이 엽록소 함량 및 Chloroplast Peroxidase활성에 미치는 영향에 관한 연구 (Studies on the Effect of Polyamine on Chlorophyll Contents and Chloroplast Peroxidase Activities in Rice Leaf Segments)

  • 호르몬이 결여되고 polyamine이 농도별로 처리된 M MS배지에서 10일 동안 발아생장(light, 16hrs: d dark, 8hrs) 시켜서 얻은 벼 잎 절편에서 엽록소 함 량은 polyamine 처리구가 대조구에 비해 높았으며, s spermine(O.OlmM, O.lmM, ImM) 이 가장 효과적이였다. 한편 chloroplast peroxidase의 활성은 polyamine 처리구가 대조구에 비해 전반적으로 높았으며, 특히 1mM spermidine 처리구는 약 100% 정도 활성을 증가시켰다. SDS-PAGE에 의해 chloroplast thylakoid membrane protein의 band를 조사한 결과 polyamine 처리구와 대조구에서 56, 2 25Kd의 major band를 얻었으며 이 band들의 total a area는 polyamine 처리구가 대조구 보다 더 높았다. 이러한 결과들은 벼 유식물에서 엽록체 발달에 p polyamine이 중요한 인자로 작용한 것으로 사료된다.

Purification and Characterization of Novel Bifunctional Xylanase, XynIII, Isolated from Aspergillus niger A-25

  • Three types of xylanases (EC 3.2.1.8) were detected in the strain Aspergillus niger A-25, one of which, designated as XynIII, also displayed ${\beta}-(l,3-1,4)-glucanase$ (EC 3.2.1.73) activity, as determined by a zymogram analysis. XynIII was purified by ultrafiltration and ion-exchange chromatography methods. Its apparent molecular weight was about 27.9 kDa, as estimated by SDS-PAGE. The purified XynIII could hydrolyze birchwood xylan, oat spelt xylan, lichenin, and barley ${\beta}-glucan$, but not CMC, avicel cellulose, or soluble starch under the assay conditions in this study. The xylanase and ${\beta}-(l,3-1,4)-glucanase$ activities of XynIII both had a similar optimal pH and pH stability, as well as a similar optimal temperature and temperature stability. Moreover, the effects of metal ions on the two enzymatic activities were also similar. The overall hydrolytic rates of XynIII in different mixtures of xylan and lichenin coincided with those calculated using the Michaelis-Menten model when assuming the two substrates were competing for the same active site in the enzyme. Accordingly, the results indicated that XynIII is a novel bifunctional enzyme and its xylanase and ${\beta}-(l,3-1,4)-glucanase$ activities are catalyzed by the same active center.
