  • Puerariae Radix is one of the medicinal plants used in oriental medicine for hangover, It has been claimed for several pharmacological effects including anti-alcohol abuse, antidipsotropic activity and anti-alcohol intoxication. In connection with Puerariae Radix effects, an activity-guided purification of active substance on alcohol dehydrogenase (hnH) was carried-out. The most active compound was isolated as puerarin (C$_{21}$H$_{20}$ O$_{9}$ ), molecular weight 416. Puerarin inhibited ADH noncompetitively against ethanol or NAD$^{+}$./.

A preliminary study of the anti-inflammatory activities of the Japanese oak silk moth, Antheraea yamamai

  • The present study aimed to determine whether a hemolymph prepared from Antheraea yamamai larvae had the same biological activities using a Bombyx mori hemolymph prior to exposure to lipopolysaccharide (LPS) in order to induce an inflammatory response. The effects of the hemolymph were determined using a reverse transcription-quantitative polymerase chain reaction to assess the expression of pro-inflammatory molecules. The A. yamamai hemolymph exerted anti-inflammatory effects on LPS-activated human monocytic leukemia cells via Toll-like receptor (TLR) 4-mediated suppression, similar to the B. mori hemocyte extract. Treatment with the A. yamamai hemolymph significantly suppressed LPS-induced upregulated inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) mRNA expression at all tested concentrations compared with the control, similar to the B. mori immune-challenged hemolymph. Finally, the A. yamamai hemolymph, like the B. mori immune-challenged hemolymph, suppressed all of these concentrations in a dose-independent manner. These results demonstrate that the hemolymph of A. yamamai exhibited important biologically active substances. Further in-depth functional studies are required to fully understand the mechanisms underlying the biological activities of wild-type silkworm hemolymphs.

급성 직업성 농약중독 및 손상에 대한 감시체계: 미국의 경험 (Surveillance of acute Occupational Pesticide-related Illness: The US Experience)

  • Pest control is required for protecting the food supply and for controlling disease vectors. Unfortunately, there is no perfectly safe form of pest control. Pesticides are commonly used for pest control. Pesticides are defined under the US Federal Insecticide Fungicide and Rodenticide Act(FIFRA) as any substance or mixture of substances intended to prevent, destroy, repel, or mitigate pests, and any substance or mixture of substances intended for use as a plant regulator, defoliant, or dessicant(40 CFR Part 152). Currently in the United States, there are 890 active ingredients registered as pesticides. Approximately one billion pounds of active ingredient are used in the US per year. Unlike most chemicals(anti-neoplastic and anti-micobial medications are the principal exceptions), pesticides are specifically designed to kill and cause harm. Because society allows these chemicals to be disseminated into the environment, it is important to monitor the health effects associated with these releases. This represents an important justification for establishing and maintaining surveillance systems for acute pesticide-related illness and injury. A comprehensive, national surveillance system for acute pesticide-related illness and injury does not currently exist in the US. Although the United States has several surveillance systems for this condition, none provide a complete understanding of the problem of acute pesticide-related illness and injury. The Toxic Exposure Surveillance System(TESS) and Bureau of Labor Statisitics(BLS) are useful for assessing magnitude and trends. The state-based surveillance systems are more useful for timely identification of outbreaks and emerging problems. Efforts are underway to increase the number of states that conduct surveillance, and to broaden the use of the standardized case definition to facilitate aggregation of data across states. Through such efforts, a comprehensive, national surveillance system may be attainable.

Extraction and Application of Bulk Enzymes and Antimicrobial Substance from Spent Mushroom Substrates

  • Pleurotus ostreatus, P. eryngii, and Flammulina velutipes are major edible mushrooms that account for over 89% of total mushroom production in Korea. Recently, Agrocybe cylindracea, Hypsizygus marmoreus, and Hericium erinaceu are increasingly being cultivated in mushroom farms. In Korea, the production of edible mushrooms was estimated to be 614,224 ton in 2013. Generally, about 5 kg of mushroom substrate is needed to produce 1 kg of mushroom, and consequently about 25 million tons of spent mushroom substrate (SMS) is produced each year in Korea. Because this massive amount of SMC is unsuitable for reuse in mushroom production, it is either used as garden fertilizer or deposited in landfills, which pollutes the environment. It is reasonably assumed that SMS includes different secondary metabolites and extracellular enzymes produced from mycelia on substrate. Three major groups of enzymes such as cellulases, xylanases, and lignin degrading enzymes are involved in breaking down mushroom substrates. Cellulase and xylanase have been used as the industrial enzymes involving the saccharification of biomass to produce biofuel. In addition, lignin degrading enzymes such as laccases have been used to decolorize the industrial synthetic dyes and remove environmental pollutions such as phenolic compounds. Basidiomycetes produce a large number of biologically active compounds that show antibacterial, antifungal, antiviral, cytotoxic or hallucinogenic activities. However, most previous researches have focused on therapeutics and less on the control of plant diseases. SMS can be considered as an easily available source of active compounds to protect plants from fungal and bacterial infections, helping alleviate the waste disposal problem in the mushroom industry and creating an environmentally friendly method to reduce plant pathogens. We describe extraction of lignocellulytic enzymes and antimicrobial substance from SMSs of different edible mushrooms and their potential applications.

Isolation of a sleep-promoting compound from Polygonatum sibiricum rhizome

  • The aim of this study was to identify sleep-promoting substance from Polygonatum sibiricum rhizome extract (PSE) with the regulation of sleep architecture. PSE showed a decrease in sleep latency time and an increase in the sleeping time. In the electroencephalography analysis of rats, PSE (150 mg/kg) showed an increase of non-rapid eye movement by 38% and a decrease of rapid eye movement by 31% compared to the control. This sleep-promoting activity was found to be involved in the $GABA_A$-BDZ receptor. The chemical structure of the pure compound was determined by the $^1H$ and $^{13}C$ nuclear magnetic resonance spectroscopy and gas chromatography mass spectrometry analysis; active compound was glyceryl-1-monolinoleate. The commercial standard glyceryl-1-monolinoleate showed a similar inhibitory concentration on [$^3H$]-flumazenil binding to $GABA_A$-BDZ receptors with final active fraction of PSE. The results indicate that glyceryl-1-monolinoleate is a major active compound responsible for the PSE-derived sleep promotion.

Rahnella aquatilis AY 2000과 Streptomyces griseus의 공배양 상등액의 항암활성 (Anti-cancer Activity of Supernatant of Rahnella aquatilis AY 2000 Cocultured with Streptomyces griseus)

  • 미생물들 사이에 존재하는 수평적 유전자 전달을 이용하여 Jurkat T cell에 대하여 새로운 항생물질을 생산하기 위해 토양박테리아 AY2000과 여러 종류의 항생물질 생산 균주인 Streptomyces griseus의 공배양을 수행하였다. MTT assay를 수행하여 세포 독성을 실험을 하였을 때 공배양 상등액은 각각 배양 하였을 때보다 높은 세포독성을 보였고 또한 48시간 배양 하였을 때 가장 높은 활성을 나타내는 것으로 나타났다. 더욱이 DAPI 염색을 하였을 때 Jurkat T cell 세포의 세포핵의 변화도 관찰 되었다. 이런 결과는 공배양 상등액에 새로운 항생물질이 생성되었음을 보여주었고 이런 방법으로 새로운 항생물질 생산에 이용되어 질수 있음을 의미한다.

  • 땅콩껍질에 함유된 항미생물 활성물질의 탐색을 위해 땅콩껍질을 MeOH로 추출하고 이 MeOH 추출물을 용매분획하여 EtOAc 중성획분을 얻었다. 이 획분에 함유된 항미생물 활성물질을 silica gel adsorption column chromatography, Sephadex LH-20 column chromatography, ODS column chromatography로 정제한 다음 HPLC를 이용하여 1종의 화합물을 단리하였다. $^1H-NMR$, MS 및 NOESY 분석에 의해 땅콩껍질로부터 분리된 항미생물 활성물질은 pratensein으로 동정되었다.

Verlamelin, an Antifungal Compound Produced by a Mycoparasite, Acremonium strictum

  • A strain of Acremonium strictum, the mycoparasite of Botrytis cinerea, showed strong antifungal activities both in vitro and in vivo against several phytopathogenic fungi. An antifungal substance was purified from the liquid cultures of A. strictum and identified as verlamelin by instrumental analyses. Verlamelin exhibited in vitro antifungal activity against some phytopathogenic fungi such as Magnaporthe grisea, Bipolaris maydis, and Botrytis cinerea, while it was net active against all the bacteria tested. In viva, verlamelin exhibited strong protective and curative activities, particularly against barley powdery mildew. At 100 μg/ml, it inhibited the development of barley powdery mildew with control values of more than 90% in 7-day protective and 2-day curative applications. This is the first report on the production of verlamelin by Acremonium species.

Purification and Characterization of Antistaphylococcal Substance from Pseudomonas sp. KUH-001

  • A bacterium producing unique antistaphylococcal substance (ASS) was isolated from soil samples. The isolated strain KUH-001 was identified to belong to Pseudomonas species from the characteristic properties of its fluorescence and cellular 3-hydroxy fatty acid composition, etc. The ASS component was purified by procedures employing activated carbon adsorption, column chromatography with silica gel, preparative TLC and HPLC. This compound could also be purified mainly by repeating of trituration and precipitation with chilled ether. Purified ASS with a m.p. value of $140~142^{\circ}C$ showed marked stability at high temperature (at $121^{\circ}C$ for 10 min) and extreme pHs (in 1N HC1 and 1N NaOH for 1 day) without significant loss of antibiotic activity. From spectral data of UV, IR, NMR, and FAB-MS, the compound was elucidated as 2-heptyl-4-hydroxyquinoline N-oxide (HHQO). Under the conditions employed, HHQO exhibited a narrow antimicrobial spectrum. active particularly against Staphylococcus aureus including the methicillin resistant strain. Moreover, it did not induce resistance, and besides, interacted synergistically with certain antibiotics such as vancomycin or erythromycin.

Determination of Silybin of Nutraceutical Herbal Preparations Using HPLC-PDA

  • Silymarin is an antihepatotoxic substance isolated from the fruits of silybum mariamum. Possibly due to their antioxidant and membrane stabilizing properties, the compounds was shown to protect various organs and cells against a number of insults (Kvasnicka et al., 2003). Among the main silymarin components, [silybin($SB_A,\;SB_B$, isosilybin ($ISB_A,\;ISB_B$) silydianin (SD) and silychristin (SC)], silybin is the major pharmacologically active compound. Korean Pharmaceutical Codex (2nd ed.) describes silybin as the main substance of Cardus Marianus extract as supportive treatment of chronic inflammatory liver disorders. The aim of this work was to analyze silybin from various preparations containing cardus marianus extract, nicotinamide, and riboflavin (CNR). Nine commercial products were tested using reversed-phase HPLC-PDA assay. The limits of detection and quantification were $0.2\;{mu}g/ml$ and $1\;{mu}g/ml$, respectively. Calibration curve showed a good linearity ($r^2$=1.00000) in the range of $1{\sim}500\;{\mu}g/ml$ of silybin standard solutions.
