Starters of lactic acid bacteria(LAB) were isolated from the commercial yoghurt products and the four isolates have been studied on their identification and some physiological characteristics. For the purpose of identification, microscopic examination, API test, and 16s rRNA gene sequencing were conducted. Isolate A from a yogurt product of local dairy company A was shown to be Gram-positive rod-shaped bacterium. All strains isolated were turned out to be as Lactobacillus paracasei by using a API 50 CHL kit. In contrast, isolate A was identified as a strain of Lactobacillus helveticus based on the 16S rRNA sequencing data, and L. casei ssp. casei for both B and D and L. paracasei for C. All the isolates survived the simulated gastric juice, pH 2.0 within 3 hours and sharply decreased in viability so that no viable cell was observed after 4.5 hours incubation. In addition, the four isolated strains were almost identical in antibiotic susceptibility to six different kinds of antibiotics including erythromycin ($15\;{\mu}g$), ampicillin ($10\;{\mu}g$), gentamycin ($10\;{\mu}g$), neomycin ($30\;{\mu}g$), but rather resistant to colistin ($10\;{\mu}g$) and streptomycin ($10\;{\mu}g$). It was noteworthy that four isolates were confirmed to produce antibacterial substance against foodborne pathogens of Gram-positive Staphylococcus aureus and Gram-negative Escherichia coli 0157:H7 as test organisms based on the inhibitory zones on an MRS soft agar medium. At presence, the inhibitory factor is unknown so that further studies are required to ascertain the active factor responsible for the inhibitory activities.

As nuclear industry has been developed, a various types of radiological contamination has occurred. After 9.11 terror in U.S.A., it has been concerned that terrorists' active area has been enlarged to use nuclear or radioactive substance. Recently, the most powerful earth-quake stroke, which triggered a massive tsunami in Japan and then Fukushima nuclear power plant reactor has suffered from a serious accident in history. The Fukushima reactor accident has occurred an anxiety of radiation leaks and about 170,000 people have been evacuated from the accidental area near the nuclear power plant. For these reasons, a social chaos can be occurred if radiological contamination occurs to the supply system for the drinking water. As such, the establishment of the radiation monitoring system for the city main water system is compelling for the national security. In this study, a feasibility test of radiation monitoring system which consists of unified hybrid-type radiation detectors was experimented for multi detection system by using gamma-ray imaging. The hybrid-type radiation sensors were fabricated with CsI(Tl) scintillators and photodiodes. A preamplifier and amplifier was also fabricated and assembled with the sensor in the shielding case. For the preliminary test of detection of radiological contamination in the river, multi CsI(Tl)-PIN photodiode radiation detectors and $^{137}Cs$ gamma-ray source were used. The DAQ was done by Linux based ROOT program and NI DAQ system with Labview program. The simulated contamination was assumed to be occurred at Gapcheon river in Daejeon city. Multi CsI(Tl)-PIN photodiode radiation detectors were positioned at the Gapcheon river side. Assuming that the radiological contaminations flows in the river the $^{137}Cs$ gamma-ray source has been moved and then, the contamination region was reconstructed.
Crude extract of Coptis japonica root-stem was evaluated for antifungal activity against Phytophthora capsici, Fusarium oxysporum, Colletotrichum dematium, Colletotrichum truncatum, Botrytis cinerea, Botryosphaeria dothidea and Alternaria porri, and antifungal active compound from the extract was identified. In addition, the usefulness of the extract for some plants disease control was investigated. Crude extract of C. japonica root-stem exhibited antifungal activity against P. capsici, F. oxysporum, C. dematium, B. cinerea, B. dothidea and A. porri. Antifungal activity of the substance isolated from C. japonica root-stem was similar to a standard chemical berberine-Cl. Red-pepper fruit rot, sesame stem rot and welsh-onion alternaria leaf spot were effectively controlled by the crude extract of C. japonica root-stem. Phytotoxicity was not observed in the red-pepper and welsh-onion leaves and red-pepper and strawberry fruits with exogenous foliage application of the crude extract. Seeds germination and radicle growth of red-pepper and sesame were inhibited by the crude extract of C. japonica root-stem. 4.24g of yellowish compound per 100g of C. japonica root-stem was obtained. The compound was identified as berberine-Cl by HPLC.
This objective of this study was to investigate the degradation characteristics of phenol, a refractory substance, by using a submerged dielectric barrier discharge (DBD) plasma reactor. To indirectly determine the concentration of active species produced in the DBD plasma, the dissolved ozone was measured. To investigate the phenol degradation characteristics, the phenol and chemical oxygen demand (COD) concentrations were evaluated based on pH and the discharge power. The dissolved ozone was measured based on the air flow rate and power discharged. The highest dissolved ozone concentration was recorded when the injected air flow rate was 5 L/min. At a discharge power of 40W as compared to 70W, the dissolved ozone was approximately 2.7 - 6.5 times higher. In regards to phenol degradation, the final degradation rate was highest at about 74.06%, when the initial pH was 10. At a discharged power of 40W, the rate of phenol decomposition was observed to be approximately 1.25 times higher compared to when the discharged power was 70W. It was established that the phenol degradation reaction was a primary reaction, and when the discharge power was 40W as opposed to 70W, the reaction rate constant(k) was approximately 1.72 times higher.

꾸지뽕나무(Cudrania tricuspidata)는 뽕나무과(Moraceae)에 속하는 낙엽활엽 소교목이다. 우리나라와 중국, 일본과 같은 동아시아에 주로 분포하며 척박한 땅에서도 잘자라고 병충해에 강하다고 알려져 있다. 예로부터 꾸지뽕나무는 항암, 간보호, 눈을 밝게하는 작용이 있다고 동의보감과 신농본초경에 기록되어 있다. 우리나라는 오랫동안 된장 등 발효식품을 자주 접하고 섭취하여 왔다는 점을 고려하여 본 연구를 수행하였다. 발효균을 접종하면 항암활성, 면역체계 개선 등 다양한 생리활성 물질이 증가한다고 알려져 있다. 본 연구는 가시가 없고 잎이 커 작업성이 용이한 대품 품종을 2018년 9월에 전남 신안군에서 채취하여 분석 시료로 사용하였다. 항산화활성 측정은 프리라디칼(DPPH, ABTS) 소거능을 측정하여 농도(EC50)별 측정결과 $100{\mu}g/mL$ ext. 이하로 항산화 활성이 열매보다 잎이 높다는 것을 확인하였다. 황국균(Aspergillus oryae)을 꾸지뽕나무 잎과 열매에 접종시켜 페놀성화합물을 스크리닝 한 결과 기존에 발견되지 않은 Salicylic acid, Naringenin, Vanilic acid, Oxyresveratrol 등 기능성 물질이 발견되었고, 잎의 경우 36시간 발효물(355mg/g)은 무처리군(179mg/g)에 비해 2배정도 상승하였다. 열매의 경우 48시간 발효시켰을 경우(472mg/g)으로 무처리군(156mg/g)보다 3배정도 상승하였다. 발효를 통해 꾸지뽕나무 잎과 열매의 최적의 추출조건을 확립하고 생리활성 물질 분석을 이용한 효능탐색 등을 진행하였다. 향후 꾸지뽕나무를 활용한 식품 소재개발 등 사업화에 기초적인 자료를 제공하여 임업인의 새로운 소득품목 육성에 기어코자 한다.

Background: Korean ginseng (Panax ginseng Meyer) contains a variety of ginsenosides that can be metabolized to a biologically active substance, compound K. Previous research showed that compound K could be enriched in the red ginseng extract (RGE) after hydrolysis by pectinase. The current study investigated whether the enzymatically hydrolyzed red ginseng extract (HRGE) containing a notable level of compound K has cognitive improving and neuroprotective effects. Methods: A scopolamine-induced hypomnesic mouse model was subjected to behavioral tasks, such as the Y-maze, passive avoidance, and the Morris water maze tests. After sacrificing the mice, the brains were collected, histologically examined (hematoxylin and eosin staining), and the expressions of antioxidant proteins analyzed by western blot. Results: Behavioral assessment indicated that the oral administration of HRGE at a dosage of 300 mg/kg body weight reversed scopolamine-induced learning and memory deficits. Histological examination demonstrated that the hippocampal damage observed in scopolamine-treated mouse brains was reduced by HRGE administration. In addition, HRGE administration increased the expression of nuclear-factor-E2-related factor 2 and its downstream antioxidant enzymes NAD(P)H:quinone oxidoreductase and heme oxygenase-1 in hippocampal tissue homogenates. An in vitro assay using HT22 mouse hippocampal neuronal cells demonstrated that HRGE treatment attenuated glutamate-induced cytotoxicity by decreasing the intracellular levels of reactive oxygen species. Conclusion: These findings suggest that HRGE administration can effectively alleviate hippocampus-mediated cognitive impairment, possibly through cytoprotective mechanisms, preventing oxidative-stress-induced neuronal cell death via the upregulation of phase 2 antioxidant molecules.
Objectives : In this study, we investigated the neuroprotective effects of ethanol extract of Lonicera japonica flower buds (EELJ) on glutamate-induced neurotoxicity in mouse hippocampus-derived neuronal HT22 cells. Methods : After analyzing the cytoprotective effect of EELJ on glutamate in HT22 cells, the inhibitory effect of apoptosis was studied using flow cytometry. In order to analyze the antioxidant efficacy of EELJ, the levels of reactive oxygen species (ROS) and glutathione (GSH) were investigated, and the effects on the activities of superoxide dismutase (SOD) and catalase (CAT) were also analyzed. Furthermore, the effect of EELJ on the expression of apoptosis regulators such as Bax and Bcl-2 in glutamate-treated HT22 cells was investigated. Results : According the current results, pretreatment with EELJ significantly reduced glutamate-induced loss of cell viability and release of lactate dehydrogenase. EELJ also markedly attenuated glutamate-induced generation of intracellular ROS, which was associated with increased levels of GSH, and activity of SOD and CAT in glutamate-stimulated HT22 cells. In addition, EELJ was strikingly inhibited glutamate-induced apoptosis in HT22 cells. Furthermore, the expression of pro-apoptotic Bax was increased and the expression of anti-apoptotic Bcl-2 was decreased in glutamate-treated HT22 cells, while in the presence of EELJ, their expressions were maintained at the control levels. Conclusions : These findings indicate that EELJ protects glutamate-induced cytotoxicity in HT22 hippocampal neurons through antioxidant activity. Therefore, although identification of biologically active substances of EELJ and re-evaluation through animal experiments is necessary, this natural substance is a promising candidate for further research in preventing and treating oxidative stress-mediated neurodegenerative diseases.
Background and objective: Artemisia argyi has a long history as an effective treatment for various diseases. The detection of environmental pollutant benzo(a)pyrene, a known human carcinogen, in the leaves of Artemisia argyi is cause for concern. For medicinal plant extracts, both a reduction of benzo(a)pyrene as well as the maintained effectiveness of the compound are important. Therefore, in this study, we propose an optimized process for the addition and filtration of activated carbon to reduce benzo(a)pyrene and change the contents of the indicating substance(jaceosidine and eupatilin). Methods: Artemisia argyi EtOH extract containing 36 ppb of benzo(a)pyrene was added to 0.1, 0.5, 1.0, and 1.5% (w/w) of activated carbon for 120 min and filtered using an activated carbon filter 1, 2, 3, and 5 times respectively. The content of benzo(a)pyrene and indicating substances in Artemisia argyi extract were then measured with high performance liquid chromatography (fluorescence and UV detectors). Results: As the amounts of activated carbon powder and filtering cycles increased, the content of benzo(a)pyrene in the Artemisia argyi extract decreased. However, when activated carbon powder 1.5% was added to the extract, and when the activated carbon filter was filtered five times, the results were reduced by 15% and 30~40% respectively. The optimal extraction condition for reducing benzo(a)pyrene was adding 1.5% of activated carbon powder. This resulted in reducing benzo(a)pyrene by 83% and indicating substances by about 4%. Conclusions: Here we present a process for reducing benzo(a)pyrene in Artemisia argyi extract using activated carbon to reduce toxicity and minimize the loss of active ingredients. This approach has potential application within a manufacturing process of various medicinal plant extracts.
Objectives : Decrease in muscle mass and loss of muscle function due to aging are associated with various diseases. As interest in healthy aging increases, efforts to prevent and treat muscle hypoxia as an illness are increasing. Considering the physical limitations, a pharmacologic approach to the treatment of myopenia is needed. Methods : Terminalia chebula Rets has a wide range of pharmacological effects and is used as a medicinal product in traditional medicine. However, the drug effect on the treatment of muscle disorders has not been revealed. The purpose of this study was to evaluate the value of water extract of Terminalia chebula (WETC) as a therapeutic agent to relieve symptoms of muscle hypoxia. Results : WETC showed strong radical scavenging ability. In addition, WETC increased cell activity of myoblast, and we observed that WETC induces myoblast differentiation by immunoblot analysis using differentiation protein markers as well as cell morphology of myoblast. Based on these results, we examined the effect of chebulic acid, chebulagic acid, gallic acid, geraniin, and punicalagin on cell activity and differentiation of myoblasts. Gallic acid significantly increased cell activity of myoblast, and it was found to be an effective substance which not only induces myoblast differentiation but also promotes proliferation. Conclusions : We suggest that the WETC with antioxidant effect and its indicator gallic acid on cell activity, proliferation and differentiation of myoblast can be studied and developed as a food and medicine for prevention and treatment of various muscle diseases.

This study provides an investigating the electrolyte reaction characteristics during thermal runaway of a lithium-ion battery(LIB). Dimethyl carbonate(DMC) is known as the main substance that makes up the electrolyte. The mono-molecular decomposition characteristics of DMC were derived through numerical analysis. Cobalt oxide can release oxygen under high temperature conditions. Also, DMC is converted to CH4, H2, CO, and CO2. Especially, it was found that the decomposition of the DMC begins at a temperature range of 340-350℃, which dramatically increases the internal pressure of the LIB. In the by-products gases, the molar ratio of CO and CO2 changed according to the molecular structure of DMC and temperature conditions. The correlation of the [CO]/[CO2] ratio according to the temperature during thermal runaway was derived, and the characteristics of the reaction temperature could be estimated using the molar ratio as an indicator. In addition, the oxidation and decomposition characteristics of DMC according to the residence time for each temperature were estimated. When DMC is exposed to low temperature for a long time, both oxidation and decomposition may occur. There is possibility of not only increasing the internal pressure of the LIB, but also promoting thermal runaway. In this study, internal environment of LIB was identified and the reaction characteristics between the active materials of the cathode and electrolyte were investigated.
