A Streptomyces strain No. 182-27, which produced amino acid antimetabolite, was isolated from soil. During the course of screening for new amino acid antimetabolites from the culture broths of Actinomycetes, we found that the strain produced a substance active against Gram-positive bacteria and its activity was reversed by L-Ieucine on the synthetic minimal agar medium in the culture broth. The morphological and cultural characteristics serve to identify the producing organism strain 182-27 as the Streptomyces, although the species of this strain should be resolved in further studies. Fermentation was carried out in the synthetic medium at $28^{\circ}C$ for 78 hours. The fermentation yield reached about 2 mg per liter of the broth. Purification was done by ion exchange resin, active carbon, silica gel column chromatography and obtained 20 mg of pure active substance from the 20 $\ell$ culture broth. The 182-27 substance was obtained as white powder, mp 18SoC. From the physicochemical characteristics of the substance, it was amino acid like substance but unknown about its chemical structure. It is active against some Gram-positive bacteria and reversed by L-Ieucine.
Recovery component for cell division in UV-irradiated E. coli B was detected with use of the cell extract of E. coli B/r which is a resistant mutant of E. coli B against UV-irradiation. The active substance was non-dialyzable and increased the activity by adding B-NAD remarkably. One more factor for increasing or promoting the restoration recognized was magnesium. The active substance stimulating the filament cell may exist as a complex which is inactivated easily in the dissociated state ana requrie B-NAD or magnesium.
An actinomycete, AS-568, which produced an inhibitory substance against succinate dehydrogenase of Ehrlich ascites carcinoma and Sarcoma-180 cells of mouse, was isolated. The inhibitory activity was determined by SDI (Succinate Dehydrogenase Inhibition) method. The active substance was specific against carcinoma cells compared to normal cells in mouse; liver, kidney and brain. The inhibitory ratio was about 50% after one hr treatment at 37$^{\circ}C$ in vitro. Maximal productivity of active substance was recognized by 5 days culture in glucose-asparagine. The active component in cultural liquid was stable in neutral pH range and heat treatment reasonably, add it was recovered from precipitate by ammonium sulfate or non-dialyrable fraction in cellophane membrane as showing the behavior of high molecular substance.
A strain of Streptomyces sp. SK-862, isolated from soil in Wonju city, was able to prodce a biologically active substance that has a strong inhibitory activity against proteolsis by trypsin. The inhyibitory substance was extracted by n-butanol, and then purified by the adsorption chromatography followed by the reverse-phase high performacne liquid chromatography. The purified substance was stable over the pH range from 2 to 10, but was unstable when treated at 8$0^{\circ}C$ for 60 min. This substance was soluble in water, methanol, ethanol nd butanol, but insoluble in chlorofrom and ethylacetate. The Rf value of the purified substance on the thin layer chromatography were 0.56 in n-butanol : methanol : water(5 : 3 : 1v/v) solvent system compare dto 0.23 in ethanol : ammonium hydoxide : water(8 : 1 : 1v/v) solvent system. This substance has maximum absorption at 259 nm. The chemical reaction of the substance was negative for sugar but positive for ninhydrine and iodine reaction.
For the inactivation of venoms, the chemical methods are generally applied. In the chemical method many works have been carried out with the chemical reagents and immunological antiserums. However, all inhibitory effect of these chemicals acting on snake venomes may well be due not to the specific, but to the nonspecific inhibitory action. Therefore, it is necessary to separate venom into its compositional active proteins and develop specific inhibitor which acts on the each protein. Until now, there have not been any reports about the substance which acts on snake venom as a specific inhibitor. Recently in 1979, we had actually isolated a specific venom inhibitor(ISV) which has a strong inhibitory activity against the proteinase of snake venom of Colubridae. In our experiments described here, a strain of Aspergillus sp., isolated from soil, was able to produce a biological active substance. The partial crystallized substance had a strong inhibitory activity against hemorrhagic action of snake venom of Colubridae. For the inhibitory action of the sample on the lethality of venom, the substance prevented completely the lethal action of the hemorrhagic factor when they were treated with enough amount of the substance. The edema factor of whole venom of Agristrodon bromohoffi brevicaudus was completely inhibited, but those of HR-I and HR-II of Trimeresurus flavoviridis venom were inhibited about 50%, when they were treated with the substance of half amount of venom. On the other hand, from the result of subcutaneous hemorrhage in a rabbit, it was concluded that two kinds of antihemorrhagic substance might be produced by the strain used in this work.
A biological active substance was isolated from the cultured medium of Streptomyces sp. and its biochemical characteristics were investigated. Isolation process of the substance was as follows; the pH of filterate of the cultured medium was adjusted to 3.0 with N-hydrochloric acid and saturated with sodium chloride, then chloroform was added to this filterate in one fifth portions and stirred vigorously. After extracting the active substance with chloroform in 3 stages, the chloroform layer combined and evaporatea after dehydrating with sodium sulfate. The substance was found to be to be toxic to various fresh water fishes; the lethal dose for an average size Pseudorasbora parva T. et. S. was 50ug per ml. In the acidic condition, the toxicity of the substance remained fora long time, while in the alkaline state, the toxicity was decreased very fast. This substance was found to be stable to organic solvents, but labile to heat treatment. The maximal revival time of Pseudorasbora parva T. et. S. was about 20 minutes in 25 ug/ml of the substance solution.

Fermentation was carried out in the synthetic medium at 28$\CIRC$C for 48 hours. The fermentation yield reached about 12 mg per liter of the broth. The YS-943 substance was obtained as white powder, mp 194$\CIRC$C and has the molecular formular of C$_{4}$H$_{8}$N$_{2}$O$_{4}$. Its structure was determined to be o-carbamyl-D-serine by spectroscopic data. It is active against some Gram-positive bacteria and reversed by L-methionine and L-histidine.
Antipyretic substance is isolated from roots of Scrophularia Oldhami which is used as antipyretic and anti-inflammatory drug. This substance is identified as p-methoxycinnamic acid which rarely occurs in plants.
To develop an environment-friendly fungicide for controlling tomato wilt diseases, antifungal active substance was isolated Rheum australe D. Don roots against Fusarium oxysporum f. sp. lycopersici, a pathogen of tomato wilt, in this study. Methanol extract obtained from Rheum australe roots was successively fractionated with hexane, chloroform, ethyl acetate, butanol and water. The ethyl acetate fraction, which showed the highest antifungal activity, was separated by column chromatography, and 60 subfractions were obtained. The 60 subfractions were anlayzed for antifungal activities by bioassay. The active compound was identified as 5-[(E)-2- (3-hydroxy-4-methoxyphenyl)ethenyl]benzene-1,3-diol (rhapontigenin) by NMR and GC-MS analysis. As a result of testing antifungal activity of rhapontigenin against Fusarium oxysporum, EC50 of rhapontigenin was showed strong antifungal activity at 7.48 mg/L. Therefore, this study showed that the Rheum australe roots extract can be a potential candidate which is a environment-friendly fungicide against Fusarium oxysporum.
An amino acid antimetabolite named YS-460 was isolated from the culture filtrate of a newly isolated Actinomycetes identified as Streptomyces sp. Fermentation was carried out in the synthetic medium at 30$\circ$C for 5 days. Purification was done by ion exchange resin, active carbon, silica gel column chromatography and obtained 38 mg of pure active substance per liter of the broth. YS-460, an amino acid like substance, has the molecular formula of C$_{7}$H$_{11}$NO$_{3}$- Its structure determined to be furanomycin by spectral analysis. It is active against some bacteria on a chemically defined medium and reversed competitively by L-isoleucine and non-competitively by L-leucine and L-valine.
