Liquid Chromatography-Tandem Mass Spectrometric Determination of Geniposide in Rat Plasma and its Pharmacokinetic Application

  • Geniposide is a biologically active ingredient of gardenia fruit. A liquid chromatography-tandem mass spectrometric method was developed and validated for the determination of geniposide in rat plasma. The plasma samples were pretreated by solid-phase extraction and introduced into a BDS Hypersil $C_{18}$ column ($50{\times}2.1mm$, $5{\mu}m$) for chromatographic separation. The mobile phase consisted of 0.1% formic acid and 0.1% formic acid in acetonitrile, and gradient elution was performed at a flow rate of 0.25 mL/min. For mass spectrometric detection, multiple reaction monitoring was performed via an electrospray ionization source in positive mode. The calibration curve for geniposide was linear ($r^2=0.997$) in the concentration range of $0.005-1{\mu}g/mL$. The intra- and inter-day accuracies and precisions fulfilled the required criteria (${\pm}15%$). The developed method was subsequently used for pharmacokinetic analysis of geniposide after oral administration to rats at a dose of 50 mg/kg. The mean maximum plasma concentration of geniposide was $0.68{\pm}0.29{\mu}g/mL$ at $0.44{\pm}0.13h$, and the mean area under the plasma concentration versus time curve was $1.46{\mu}g{\cdot}h/mL$.

Screening of Korean Traditional Prescriptions with Inhibitory Activity against Protein Tyrosine Phosphatase 1B and Analysis of Jakgamhwangsinbu-tang (芍甘黃辛附湯) Prescription (전통 처방의 Protein Tyrosine Phosphatase 1B 저해 활성 검색 및 작감황신부탕(芍甘黃辛附湯) 처방 분석)

  • In order to search for protein tyrosine phosphatase 1B (PTP1B) inhibitors as therapy of type 2 diabetes and obesity from Korean traditional prescriptions, we selected 58 traditional prescriptions based on a review of the Korean traditional medicine books. The hot water extracts of Korean traditional prescriptions were screened for the inhibitory activity against PTP1B. Among the tested extracts, water extracts of Jakgamhwangsinbu-tang, Seonbanghwalmyung-eum, and Takreeonjoong-tang showed relatively good inhibitory activity against PTP1B at the concentration of $30{\mu}g/ml$. Additionally, we evaluated PTP1B inhibitory effect for each herbal ingredient and composition in Jakgamhwangsinbu-tang (芍甘黃辛附湯). Of the tested ingredients from this herbal medicine, water extracts of Paeoniae Radix rubra and Rhei Rhizoma, and ethanol extracts of Paeoniae Radix alba, Rhei Rhizoma, Asiasari Radix, and Aconiti Tuber showed good PTP1B inhibitory effect. Herbal compositions composed of these active herbal ingredients exhibited significant activity for PTP1B inhibition over 70% at $7.5{\mu}g/ml$.

Investigation of Ingredients and Hazardous Substances in Disinfectants Used against COVID-19 and Some Livestock Diseases (코로나바이러스감염증-19와 일부 가축전염병 방역소독제품의 함유성분 및 유해물질 조사)

  • Objectives: The Coronavirus Disease 2019 (COVID-19) pandemic has caused the death of 740,000 people around the world as of August 12, 2020. Foot-and-Mouth Disease, Avian Influenza, and African Swine Fever are serious livestock diseases. Government agencies in Korea have provided ingredient information and usage instructions for disinfectants used to counter those infectious diseases. The purpose of this study was to provide information on the chemical ingredients in disinfectant products used against COVID-19 and certain livestock diseases. Methods: We collected information from the Korean government. The Central Disaster Management Headquarters and Central Disease Control Headquarters provided information on disinfectant products used against COVID-19. The Animal and Plant Quarantine Agency of Korea provided information on efficacy-certified disinfectant products for use against selected livestock diseases. Health hazard and environmental hazard information on the ingredients in the disinfectants was collected from the Korea Occupational Safety & Health Agency's Material Safety Data Sheets, and toxicity value information was collected from United States Environmental Protection Agency's CompTox Chemicals Dashboard. Results: There were 76 COVID-19 disinfectant products in use, and the most common ingredients were benzalkonium chloride (51%), alkylbenzyl dimethyl ammonium (30%), and ethanol (3%). There were 216 livestock disease disinfectant products comprised of 89 acidic, 88 oxidic, 30 aldehydic, three alkaline, and six other products. Among the 49 active ingredients used in the disinfectants that were investigated, health and environmental hazard information was provided for many of them, but only 20 chemicals had official toxicological information. Conclusion: Since the disinfectants included numerous chemicals, an understanding of their chemical characteristics could be critical to prevent unintended human or environmental exposure.

Effect of Scutellaria Baicalensis Georgi Extraction (SbGE) on H2O2-induced Inhibition of Phosphate Transport in Renal Epithelial Cells (황금약침액(黃芩藥鍼液)이 신장상피세포(腎臟上皮細胞)에서의 H2O2에 의한 인산염(燐酸鹽) 운반(運搬)의 억제(抑制)에 미치는 영향(影響))

  • Objective : This study was performed to determine if Scutellaria balicalensis Georgi extract (SbGE) prevents oxidant-induced membrane transport dysfunction in renal tubular cells. Methods : Membrane transport function was estimated by measuring $Na^+$-dependent inorganic phosphate transport in opossum kidney (OK) cells. $H_2O_2$ inhibited phosphate transport in a dose-dependent manner. Results : The inhibitory effect of $H_2O_2$ was significantly prevented SbGE over concentration range of 0.005-0.05%. $H_2O_2$ caused ATP depletion, which was prevented by SbGE. $H_2O_2$ induced the loss of mitochondrial function as evidenced by decreased MTT reduction and its effect was prevented by SbGE. The $H_2O_2$-induced inhibition of phosphate transport was not affected by a potent antioxidant DPPD, but the inhibition was prevented by an iron chelator deferoxamine, suggesting that $H_2O_2$ inhibits $Na^+$-dependent phosphate transport via an iron-dependent nonperoxidative mechanism in renal tubular cells. Conclusion : These data suggest that SbGE may exert the protective effect against oxidant-induced membrane transport dysfunction by a mechanism similar to iron chelators in renal epithelial cells. However, furher studies should be carried out to find the active ingredient(s) of SbGE that exerts the protective effect.

A Study on Nobel SUNBLOCK Materials Containing an Active Ingredient Extracted from Symbiodinium (심바이오디니움으로부터 추출된 유효성분을 함유하는 신규 SUN BLOCK 물질에 관한 연구)

  • Symbiodinium is a dinoflagellate genus that coexists with coral reefs and is known to provide ultraviolet (UV) protection in nature through the synthesis of mycosporin-like amino acids (MAA). In order to develop a natural and ecofriendly sunblock for use in summer resorts, the possibility of using a Symbiodinium microbiome extract or an MAA was investigated. Two sunblocks, one containing 7% Symbiodinium extract and the other containing MAA were prepared to be tested on hairless mice and human skin. In this clinical study, the sun protection factor (SPF) and Protection factor of UV A (PFA) values of the sunblock containing either Symbiodinium extract or MAA were determined. The SPF values of the sunblock containing Symbiodinium extract and MAA were 10.43 and 10.83, respectively, and the PFA values were 3.42 and 3.39, respectively. Based on their PFA values of ≥2, the UV A protection can be graded as PA+ (low) for both the sunblocks. In addition, the UV-blocking extract of Symbiodinium has a low phototoxicity and cytotoxicity, reducing the possibility of a heavy feeling to the skin or a foreign-body sensation caused by residue build-up. The low toxicity feature of the major sunblock component will also prevent side effects, such as allergic reactions. Although Symbiodinium extract or MAA alone do not provide effectiveprotection against UV irradiation, their protection capacity can be complemented by the addition of inorganic UV-protecting compounds.

Effects of Functional Ingredients Addition on Quality Characteristics of Kochujang (기능성 소재의 첨가가 고추장의 품질특성에 미치는 영향)

  • To produce bio-active Kochujang, 1.2% of pomegranate powder, 1.0% of hawthorn berry extract powder, 0.01% of ginko biloba extract, 1.2% of red yeast rice powder, 1.2% of silk powder, 0.2% of garlic oil, 0.2% of chitosan, 1.2% of wax gourd extract powder, and 1.0% of pine mushroom were added individually with raw materials that are needed for Kochujang process. These Kochujangs were fermented for 45 days at $30^{\circ}C$, and examined for changes in their physicochemical properties. There were no significant differences in pH, acidity, formol-N content, color and total cell count by plate count agar between Kochujangs fortified individually with functional ingredients and the control Kochujang. When 0.2% of garlic oil was added, antioxidant activity of its Kochujang product increased twice as much as that of the control Kochujang. Improved ACE inhibiting activities were observed in 1.2% of red yeast rice powder. When 1.2% of silk powder or 0.01% of ginko biloba extract were added, antithrombotic abilities were remarkably enhanced up to two times of the control Kochujang.

Effects of Bee Venom on Glioma Cells (봉독(峰毒)이 Glioma Cell에 미치는 효과(效果))

  • Objective: Bee venom (BV) has been used for the treatment of inflammatory diseases such as rheumatoid arthritis and relief of pain in Oriental medicine. The two main components of BV are melittin and phospholipase A2 (PLA2). Of these, melittin, the major active ingredient of BV, has been reported to induce apoptosis and to possess anti tumor effects. Several studies have established that the agents inducing apoptosis in target organs suppress tumorigenesis. As the other component, PLA2 has been reported to induce neurite outgrowth in PC12 cells. However, there was no report about proliferative effect of BV in neuronal cells. In order to examine the effect of BV on glioma cell, human glioma cell line, U87 was used. Methods: Analysis of proliferation was confirmed by MTT assay. BV increased cell number through dose and duration dependent manner and these effects are apparent at a concentration of 10 ug/ml. To observe which signaling molecules will be activated by BV, phosphorylation of Akt, MAPK, PYK2 or CREB were examined by Western blot analysis. To study the long term effect of BV in U87 cells, the image of cells treated with BV for 4 days were obtained. Results: The phosphorylation levels of PYK2 and Akt were increased at 5 min after addition of 10 ug/ml of BV and sustained to 2 hours. On the other hand, phosphorylation of MAPK and CREB were increased at 5 min, maximum at 10 min, and returned to 30 min. These imply that BV may activate two different signaling pathways, PYK2/Akt and MAPK/CREB. BV treated cells showed increased neurite number and length. Conclusion: These results propose that BV may induce differentiation as well as proliferation of U87 cells through the activation of PYK2/ Akt and MAPK/ CREB.

  • Glyphosate is the active ingredient in widely used herbicides. It acts through inhibition of the shikimate metabolic pathway in plants. This pathway does not exist in mammals, however, so glyphosate is presumably less toxic to humans. Nevertheless, fatal cases of glyphosate poisoning in humans have still occurred. Cases of glyphosate poisoning reported in the previous literature were almost always caused by intentional ingestion. Therefore, intramuscular injection of glyphosate with suicidal intent has not been reported. We report a case of 43-year-old man with poisoning due to intramuscular injection of glyphosate herbicide. He was admitted to the emergency department with a chilling sensation, local hotness, swelling, and tenderness at the site of glyphosate injection. He was treated with intravenous antibiotics and analgesics for 10 days and was discharged without any other complication.

Evaluation of Acute Toxicity about Leakage Waters of Antifouling Paints on Sebastes shlegeli and Artemia (방오도료 용출수의 조피볼락과 알테미아에 대한 급성독성 평가)

  • The use of antifouling(AF) paints is the effective method for the protection of underwater structures from the development of marine fouling organisms. The ban on harmful substances in antifouling paints requires the development of new antifouling strategies although Tributyitin (TBT) compound had been used extensively as an active ingredient Alternatives should be as effective as conventional paints but have lower toxicity. In the present study, a TBT-free self-polishing (Cu SPC) AF paint containing $Cu_2O$, a Cu free SPC AF paint, and a Foul-release silicone AF paint, which were commercially available, were examined to investigate environmental erects of leakage waters employing Sebastes shlegeli and Artemia. Survival rates were inversely proportional to the concentration of leakage waters from AF paints and the acute toxicity of SPC AF paints was relatively higher than that of foul release AF paints.

The Improving Method of Facial Recognition Using the Genetic Algorithm (유전자 알고리즘에 의한 얼굴인식성능의 향상 방안)

  • As the security system using facial recognition, the recognition performance depends on the environments (e. g. face expression, hair style, age and make-up etc.) For the revision of easily changeable environment, it's generally used to set up the threshold, replace the face image which covers the threshold into images already registered, and update the face images additionally. However, this usage has the weakness of inaccuracy matching results or can easily active by analogous face images. So, we propose the genetic algorithm which absorbs greatly the facial similarity degree and the recognition target variety, and has excellence studying capacity to avoid registering inaccuracy. We experimented variable and similar face images (each 30 face images per one, total 300 images) and performed inherent face images based on ingredient analysis as face recognition technique. The proposed method resulted in not only the recognition improvement of a dominant gene but also decreasing the reaction rate to a recessive gene.
