In Vitro Propagation by Shoot-tip and Node-bud Culture of Rehmannia glutinosa (정단 및 마디조직 배양을 통한 지황의 기내 증식)

  • Multiple shoots obtained in MS medium suppler with 5.0 mg/L BA though shoot-tip culture. The frequency of vitrified shoot was lower on Bacto-agar medium than on Gelrite as gelling agent. Addition of activated charcoal at concentrations of 0.1~0.3% reduced vitrification and markedly increased shoot growth, and formation and growth of roots, but significantly reduced the number of shoots formed. The ratio of fresh weight to dry weight was decreased by increasing light intensity and agar concentration. Eight-tenths times of macroelement of MS medium was observed to be effective for shoot formation. Addition of IAA effectively promoted shoot formation in both shoot tip and node-bud explants. Effects of Activated Charcoal on in vitro Ruminal Fermentation Characteristics and Nutrient Disappearances (사료에 대한 활성탄의 첨가가 in vitro 시험시의 발효성상 및 영양소 소실율에 미치는 영향)

  • An in vitro study was conducted to examine the effects of the addition of activated charcoal (AC) on the ruminal fermentation characteristics, nutrient disappearance, and ruminal gas production. AC was added at the levels of 0.00, 0.25, and 0.50 % to each of the four types of diets (roughage/concentrate ratio : 8/2, 6/4, 4/6 and 2/8), respectively. Although not significant, ruminal pH tended to increase by adding AC, and as the concentrate level increased, ruminal pH decreased (P<0.05). Acetate concentration and acetate/propionate molar ratio tended to decrease in AC diets. but molar % of propionate tended to increase by the addition of AC. Ruminal degradation of dry matter, crude protein, NDF, ADF, and hemicellulose in AC diets tended to increase than in non-AC diet, however, no tendency in ruminal degradation of crude fat was observed. As the concentrate level increased, rumunal degradation of dry matter and nutrients in AC diets increased significantly(P<0.05). Ruminal gas production tended to decrease in the 0.50 % AC diets, however, it tended to increase in high roughage diets. Although there appeared some beneficial effects in adding AC to ruminant diets in this study, more works should be done with AC before we can make clear conclusion on the use of AC in the ruminant diets.

Effect of Seaweeds and Adsorbents on Volatile Flavor Components of Onion Juice (해조류 및 흡착제의 첨가가 양파즙의 냄새에 미치는 영향)

  • In order to retain or remove the flavor components of onions, this study was performed to investigate the effect of seaweeds and adsorbents on volatile flavor components of onion juice by testing a sensory evaluation, measuring the amount of pyruvate and thiosulfinate, and investigating the changes of volatile components by SPME/GC. The main flavor compounds in raw onion juice were dipropyl tetrasulfide, 1-propenyl propyl trisulfide, methyl propyl trisulfide, dipropyl trisulfide. Volatile flavor compounds in onion juice treated with tangleweed, brown-seaweed, laver and celite showed a relatively decreasing tendency compared to untreated onion juice, but most of volatile flavor compounds in onion juice treated with activated charcoal and ${\beta}-cyclodextrin$ were removed. The result of the sensory evaluation with various materials for masking onion flavor showed a significant difference at the p<0.05 level. The amount of pyruvate and thiosulfinate treated with tangleweed, brown-seaweed, laver and celite showed little difference compared to untreated onion juice, but the amount of pyruvate and thiosulfinate treated with activated charcoal and ${\beta}-cyclodextrin$ decreased largely compared to untreated onion juice. As a result of the study, onion juice treated with tangleweed, brown-seaweed, laver and celite showed the effect of preserving the pungent taste and masking the onion flavor. Activated charcoal and ${\beta}-cyclodextrin$added to onion juice might have a role in removing the onion flavor by adsorbing volatile flavor compounds in onion.

Establishment of proliferation and regeneration system of PLBs in Phalaenopsis by treatments of a variety of types of medium, sucrose concentrations and anti-browning agents (다양한 배지종류, sucrose 농도 및 갈변억제물질 처리에 의한 팔레놉시스 PLB 증식 및 재분화 체계확립)

  • To establish an efficient proliferation and regeneration of PLBs (protocorm-like bodies) of Phalaenopsis plants, a variety of propagation medium types, various concentraions of sucrose as well as liquid and solid type were tested in this study. Further, activated charcoal, citric acid and ascorbic acid were compared whether these agents are suppose to reduce the browning in culture process using PLBs of Phalanopsis plants. With regard to the proper propagation medium, VW medium showed 1.3 ~ 2 times highr than those of other medium in an index of increasing for fresh weight and 50% higher than those of other medium in the frequency of shoot regeneration. However, regarding liquid and solid types of culture, there were no significant differences in the proliferation of PLBs and regeneration of shoots from PLBs. In the experiment for a variety of sucrose concentrations (0 ~ 50 g/l), 10 g of sucrose showed 30 ~ 50% higher than other concentrations in increasing index and 10 ~ 50% higher in the regeneration of shoots from PLBs. Regarding the reduction of browning in tissue culture via PLBs of Phalaenopsis plants, 1 g of activated charcoal showed only 1.5% browning of PLBs cultured. Whereas, other treatments including citric acid and ascorbic acid showed 6 ~ 16% of browning of PLBs. Therefore, activated charcoal was selected as an efficient anti-browning agents for the culture of PLBs in Phalaenopsis plants. Using above-described results can be contibuted to the establishment of mass propagation system using PLBs of Phalaenopsis plants in the future.

Culture condition for gametophyte and sporophyte masspropagation of bamboo fern (Coniogramme japonica) using tissue culture (조직배양을 이용한 가지고비고사리의 전엽체와 포자체 대량번식을 위한 배양조건)

  • This study was conducted to investigate the optimal culture method for gametophyte and sporophyte propagation in Coniogramme japonica (Thunb.) Diels, which can be used in various fields. The propagation of prothallus were cultured in 1/4 - 1 Murashige and Skoog medium and Knop medium for 10 weeks. The results indicated that the fresh weight of prothallus was the highest (14.5 g) in 1MS medium. Subsequently, various concentrations of sucrose, activated charcoal and nitrogen source were also added to 1MS medium and cultured for 8 weeks. The results provided that the sucrose concentration was 3% and the fresh weight of prothallus was the highest 10.8 g. According to the concentration in the range of 8.8 ~ 10.8 g, in the case of activated charcoal, the four treatments showed no significant difference. The nitrogen source was added at a concentration of 30, 60 and 120 mM with the ratio of ${NH_4}^+:{NO_3}^-$ being 1 : 2. As a result, the fresh weight of all treatments increased to similar level but there was no significant difference. We investigated sporophyte formation according to soil type and the highest number of sporophytes at 228.0 was formed in soils mixed with horticultural substrate and decomposed granite at 2 : 1 (v : v). On the other hand, sporophyte was not formed in soils containing peatmoss except for the one with peatmoss and decomposed granite at 2 : 1 (v : v).

Study on the Growth Factors for Rapidly Cultivating Mycobacterium spp. (마이코박테리움을 신속하게 배양할 수 있는 성장 인자에 관한 연구)

  • Mycobacteria grow slowly. Therefore, a solid medium should be used for eight weeks and a liquid medium for six weeks. The purpose of this study was to find the growth factors that can grow Mycobacterium rapidly and to help develop a solid medium for rapid identification. Three types of Mycobacterium growth factors were evaluated with 10 Mycobacteria by adding activated charcoal, defibrinated sheep blood, and L-ascorbic acid to $Difco^{TM}$ Mycobacteria 7H11 agar (Becton, Dickinson and Company, Sparks, MD, USA). The time to detection and the distinguishability of a colony were compared with that of the current method. In the rapidly growing Mycobacterium, the difference in detection time between the new media and conventional media confirmed that the new media was faster. M. kansasii and M. intracelluare grew faster in 7H11 C than in 7H11 medium. MTB grew faster than the other media in 7H11 C. This study confirmed that the two growth factors affect fast-growing Mycobacteria and slow-growing Mycobacteria. 7H11 C showed better distinguishability than the conventional media in all 10 Mycobacterium due to the color contrast. In particular, when the MTB was grown, the size of the colonies was larger than with other media, so visualization was easy.

Synthesis and Characterization of Calcium Derivative Combined with High-Surface-Area Activated Carbon Composites for Fine Toxic Gas Removal

  • Highly toxic gases such as hydrogen sulfide ($H_2S$), carbon dioxide ($CO_2$), and ammonia ($NH_3$) are generated by both nature and human activities and affect human health. In this research, activated carbon combined with $Ca(OH)_2$ and $CaCO_3$ (AC-CO and AC-CC, respectively) were fabricated and applied in absorbing toxic gases from air pollutants. Activated charcoal powder was compressed in the form of pellets and used in the designated conditions. The optimum operating conditions and material properties, such as adsorption capacity, effect of weight ratio of the mixture, and hardness, have been investigated after combining with the calcium derivative. The good performance exhibited in this study suggests that this material is expected to be an effective and economically viable adsorbent for $NH_3$, $CO_2$, and $H_2S$ removal from the air system.

Effects of Browning Products and Charcoal on the Degradation of Aflatoxin B, in Korean Soy Sauce (Kanjang) and its Model System (간장 및 모델시스템에서 간장 갈색물질과 숯이 Aflatoxin $B_1$의 파괴에 미치는 영향)

  • The effects of browning Products (BP) from Kanjang(soy sauce) and charcoal on the degradation of aflatoxin $B_1(AFB_1)$ in Kanjang and its model system were studied. Approximately 60% of $AFB_1$ was degraded in the presence of 0.05% BP at pH 7 of phosphate buffer after 2 days of incubation at $30^{\circ}C$. The mutagenicity of the $AFB_1$ which reacted with the BP was decreased to about 50% and 70% in Salmonella typhimurium TA98 and TA100 strains, respectively (p<0.05). When a few pieces of charcoal were added to home made Kanjang, $AFB_1$ was quite stable for 5days at $30^{\circ}C$, however, about 80% of $AFB_1$ was removed when the charcoal was either in distilled water or in 20% of NaCl solution after 2 days of incubation. Activated carbon instead of the charcoal removed $AFB_1$ completely in the all samples under the same conditions.

Adsorption Removal of Odor Compounds (IBMP, IPMP) (이.취미물질(IBMP, IPMP)의 흡착제거)

  • The purpose of this study was carried out to estimate removal possibility of IBMP and IPMP causing odor in raw water. As a result of Freundlich isotherm. IBMP was superior to IPMP in adsorptive capacity. Adsorptive capacities of activated carbon were found to be in order of Lignite, Coconut shell, and Charcoal. These were well fitted with Freundlich isotherm. According to adsorption breakthrough tests for Lignite GAC, breakthrough time of IPMP and IBMP were 5.7hr and 5.5hr, respectively. Because adsorptive capacities of target material were very different with pore size distribution, it seemed that Lignite and Coconut shell based activated carbons were recommended in order to remove door compounds.

Development of Adsorbent for Heavy Metals by Activation of the Bark (활성화 수피를 이용한 중금속 흡착제 개발)

  • The objective of this research was to develop the adsorbent far heavy metals by activating the bark sample. Barks from pine tree with diameters of $2{\sim}4\;mm$ were activated in the muffle furnace under a high relative humidity condition at temperatures of $600{\sim}900^{\circ}C$. The removal efficiency of the activated bark (ACTBARK) for Cu and Cd was temperature dependent showing the order of $900^{\circ}C$ > $800^{\circ}C$ > $700^{\circ}C$ > $600^{\circ}C$. The critical temperature was considered to be $900^{\circ}C$ to become an efficient adsorbent for Cu and Cd. The bark samples activated at temperatures lower than $700^{\circ}C$ showed a less removal efficiency than the crude bark. The ACTBARK activated at $900^{\circ}C$ removed more Cu and Cd from solution than the commercial activated carbon and charcoal. The ACTBARK (activated at $900^{\circ}C$) adsorbed all of the Cu and Cd in solution with concentrations less than 150 mg/L. The selectivity of the ACTBARK was in the order of Cu > Zn > Ni > Pb > Fe > Cd > Mn.
