한국약용식물의 최종당화산물 생성저해활성 검색 (II) (Screening of Korean Herbal Medicines with Inhibitory Activity on Advanced Glycation End Products (AGEs) Formation (II))

  • Advanced glycation end products (AGEs) contribute to the progression of micro and macrovsacular complication of diabetes and therefore present a promising target for therapeutic agents. In this study, 40 Korean herbal medicines have been investigated with an in vitro evaluation system using AGEs inhibitory activity. Of these, 21 herbal medicines $(IC_{50}<50{\mu}g/ml)$ exhibited an inhibitory activity against AGEs formation compared with anminoguanidine $(IC_{50}=72.12{\mu}g/ml)$. Particularly, 7 herbal medicines, Actinidia arguta (root and stem), Crataegus pinnatifida (twig), Camellia japonica (whole), Kalopanax pictus (bark), Lagerstroemia indica (leaf-stem), Reynoutria sachalinensis (root) showed more potent inhibitory activity (approximately 3-10 fold) than the positive control aminoguanidine.

Ecological Characteristics and Distribution of Plant Resources of Pyrus and Malus sp. in Jindong Valley, Gangwon Province

  • Four major naturally populated areas around the Jindong valley in Inje-Gun, Gangwon province for wild Pyrus ussuriensis, Malus sieboldii, and Malus baccata are mostly located on the southeast side of the mountains at 630-745m in altitude at sea level and are in very good sunny areas. The total of 77 taxa dividing into 32 families, 58 genera, 65 species, and 12 varieties has been inhabited in these areas. Most of these plants are heliophytes, which love sunshine, but some of rhizo-plants such as Sasa borealis, climbing plants such as Actinidia arguta and Celastrus orbiculatus, or naturalized plants like Bidens frondosa are also shown. These areas have been continually destroyed, so it is possibly thought that the second transition has been progressed. The index of species diversity of plant groups in these areas is 0.672∼1.465 based on the Shannon-Wiener's method, but its index for the area that rhizo-plants like Sasa borealis are growing as an under planting decreases relatively. In this study, the oldest plant of Pyrus ussuriensis in Korea, which is 120cm in diameter at breast height (DBH), 19m in tree height and 25m in width, has been found.

'Goldone', a Yellow - fleshed Kiwifruit Cultivar with Large Fruit Size

  • In Korea, kiwifruit is grown within a limited region on the southern coast where the climate is warm. Since the yellow - fleshed kiwifruit variety, 'Hort16A', first became commercially available, we have focused on breeding additional yellow - fleshed kiwifruits. Here, we describe the cultivar 'Goldone', which originated from 'Red Princess' and is characterized by red coloration around the fruit core. Conventional field crosses were performed in 2003, and permission for final release of 'Goldone' was obtained in 2011. This cultivar is very productive, with an average fruit weight of 129 g, which is 39% heavier than that of the cultivar 'Hayward'. 'Goldone' is harvested in late October, approximately 165 - 170 days after anthesis. In general, 'Goldone' has approximately eight flowers per fruiting shoot; these flowers must be thinned before blooming for commercial production. 'Goldone' was registered at the Korean Seed & Variety Service in 2014 for plant variety protection rights (grant no. 4835).

Characterization of Antibacterial Strains against Kiwifruit Bacterial Canker Pathogen

  • Kiwifruit (Actinidia spp.) is an economically important crop and a bacterial canker disease, caused by Pseudomonas syringae pv. actinidiae (Psa), is the most destructive disease in kiwifruit production. Therefore, prevent and control of the disease is a critical issue in kiwifruit industry worldwide. Unfortunately, there is no reliable control methods have been developed. Recently, interest in disease control using microbial agents is growing. However, kiwifruit microbiota and their roles in the disease control is mainly remaining unknown. In this study, we secured bacterial libraries from kiwifruit ecospheres (rhizosphere, endospere, and phyllosphere) and screened reliable biocontrol strains against Psa. As the results, Streptomyces racemochromogenes W1SF4, Streptomyces sp. W3SF9 and S. parvulus KPB2 were selected as anti-Psa agents from the libraries. The strains showed forcible antibacterial activity as well as exceptional colonization ability on rhizosphere or phyllosphere of kiwifruit. Genome analyses of the strains suggested that the strains may produce several anti-Psa secondary metabolites. Our results will contribute to develop biocontrol strains against the kiwifruit canker pathogen and the disease management strategies.

Suppression of Ripe Rot on 'Zesy002' Kiwifruit with Commercial Agrochemicals

  • Ripe rot caused by Botryosphaeria dothidea is one of the serious diseases of postharvest kiwifruit. In order to control ripe rot on Actinidia chinensis cultivar 'Zesy002', several commercial agrofungicides were selected by an antifungal test on an artificial medium. Furthermore, disease suppression by the selected fungicides was evaluated on the kiwifruit by inoculation with a conidial suspension of B. dothidea. On the artificial media containing boscalid + fludioxonil was shown to be the most effective antifungal activity. However, in the bio-test pyraclostrobin + boscalid and iminoctadinetris were the most effective agrochemicals on the fruit. On the other hand, the infection structures of B. dothidea on kiwifruit treated with pyraclostrobin + boscalid were observed with a fluorescent microscope. Most of the fungal conidia had not germinated on the kiwifruit treated with the agrochemicals whereas on the untreated fruit the fungal conidia had mostly germinated. Electron microscopy of the fine structures showed morphological changes to the conidia and branch of hyphae on the kiwifruit pre-treated with pyraclostrobin + boscalid, indicating its suppression effect on fungal growth. Based on this observation, it is suggested that ripe rot by B. dothidea may be suppressed through the inhibition of conidial germination on the kiwifruit treated with the agrochemicals.

Causal Fungus of Side Rot on 'Zesy002' Kiwifruit in Jeju Island, South Korea

  • Since the past few years, dimple rot has become one of the most serious diseases affecting Actinidia chinensis var. chinensis 'Zesy002' kiwifruit in Korea, leading to a low quality and subsequent losses of income. In the current study, the causal pathogen of side rot on 'Zesy002' kiwifruit was identified to be Botryosphaeria dothidea, a widespread pathogen that could penetrate the fruit directly without wounding. Diaporthe sp., another pathogen causing dimple rot, was isolated from infected kiwifruit. It was not able to infect fruits without wounding. Dimple-rot symptoms are known to be associated with other pathogens from the Pseudocercospora genus. In this study, B. dothidea was ratified as the causal disease agent in Jeju Island. To illustrate the pathogenicity of this fungus on 'Zesy002', further studies such as observation of infection structures of fungus may be needed.

Evaluation of control methods for the best practicing conditions for the control of bacterial blossom blight of kiwifruit

  • Control of bacterial blossom blight of kiwifruit (Actinidia deliciosa) has been mainly depended on chemical control. Recently cultural practices such as trunk girdling of kiwifruit trees and rainproof installation over kiwifruit trees also were conducted as the alternative control practices. Each of the control methods was evaluated for the best practicing conditions for the control of bacterial blossom blight of kiwifruit. Among the various combinations of spray times and spray periods, optimum spray program of antibiotics was turned out to be 3 times with intervals of 10 days from early May during the flowering season of kiwifruits. Optimum periods of trunk girdling of kiwifruit trees were from late March to late April. Trunk girdling with 20-30 mm wide showed best control efficacies on bacterial blossom blight, irrespective of the heights of girdling on trunks of kiwifruit trees. Optimum period of rainproof installation over kiwifruit trees was from March till late April, irrespective of installation methods.

Growth-Promoting Effects of Vegetable Extracts on Selected Human Lactic Acid Bacteria

  • Ethanol extracts from 36 vegetable samples were assayed for their growth-promoting effects on Bifidobacterium bifidum, B. longum, and Lactobacillus casei The growth-promoting effects varied according to bacterial strain and vegetable species. In modified Gy rgy broth, extracts of Lactuca sativa, Lycopersicon esculentum and L escutentum var. cerasiforme exhibited strong growth-promoting responses toward B. longum, and significant and strong growth- promoting response toward B. bifidum was observed in extracts of Actinidia arguta, Allium cepa, A. sativum, Brassica campestris subsp. napus vats. pekinensis, Capsicum frutescens, Daucus carota var. sativa, L sativa, 1. esculentum and L. esculentum var. cerasforme, Nelumbo nucifera, Cucurbita moschata, Lackca sativa var. capitata, and Rubus coreanus. For L casei, extracts of A. fshlosum, A. hberosum, Cichorium intbus, Cucurbita moschat\ulcorner Ipomoea batatas, 1. sativa var. capitata, L. esculentum, P. brachycarpa, Raphanus sativus, R. coreanus, and S. melongena strongly enhanced the growth of this bacteria. In modified Gy rgy broth, the promoting effect was most pronounced with B. bifidum and L. casei among lactic acid bacteria used. In MRS broth, A. arguta, A. cepa, A. sativum, B. campestris subsp. napus var. pekinensis, C. frutescens, and D. carota var. sativa L. satiw var. capitata, and R. coreanus strongly enhanced the growth of B. bifidum, Growth of B. longum was strongly affected by the addition of extracts from L. sativa var. capitata. For L casei, moderate growth-promoting responses were observed in 9 vegetable extracts. The promoting effect in MRS broth was most pronounced with B. bifidum among lactic acid bacteria used.

건조 다래순의 조리 중 라디칼 소거 활성과 알파글루코시데이스 억제 활성의 변화 (Changes in Radical Scavenging Activity and α-Glucosidase Inhibitory Activity of Dried Daraesoon (Shoot of Hardy Kiwi, Actinidia arguta) during Cooking)

  • 다래순 묵나물을 찬물에 16시간 불리고, 30분 삶은 후 1시간 찬물에 불리는 재수화 과정을 거친 후 다래순의 라디칼 소거 활성과 알파글루코시데이스 억제활성은 유의하게 감소하였으며 재수화된 다래순을 $180^{\circ}C$에서 10분 또는 20분 동안 가열 조리한 경우 들기름 첨가와 관계없이 라디칼 소거활성은 증가하였으나 알파글루코시데이스 억제 활성은 들기름을 첨가하고 가열 조리한 경우에만 증가하였다. 다래순의 라디칼 소거 활성과 알파글루코시데이스 억제 활성 변화는 색소보다는 산화방지제, 플라보노이드 또는 토코페롤보다는 폴리페놀 화합물 함량 변화와 더 유사한 경향을 보였다. 따라서 본 결과는 다래순 묵나물의 재수화 또는 가열 조리 시 산화 방지, 항당뇨 등 건강기능성 개선을 위해 폴리페놀 화합물의 손실을 줄이는 것이 바람직함을 시사하였다.

오대산 월정사지역 전나무림의 생태학적 연구 (Ecological Research of Abies holophylla Forest at Wol-jong Temple(Mt. Odae, Kangwon-do))

  • 본 연구는 오대산 국립공원내 월정사지역에 분포하고 있는 전나무림의 식생구조를 분석하여 이 지역 전나무림의 관리에 대한 기초자료를 제공하기 위하여 수행되었다. 그 결과 전나무림의 층위구조는 교목층(2종류), 아교목층(20종류), 관목층(46종류) 그리고 초본층(87종류)으로 구성되어있었다. 조사지역에 출현한 전체식물의 중요치는 전나무가 34.09%로 가장 높게 나타났고, 당단풍(23.07%), 청시닥나무(14.44%), 고광나무(10.72%), 오리방풀(8.49%), 복자기 (7.80%), 고추나무(7.65%), 산거울(6.79%), 방아풀(5.76%), 고로쇠나무 (5.47%), 개다래(5.4%)의 순으로 나타났다. 특히, 전나무의 치수가 전혀 없어 앞으로 아교목층과 관목층을 형성하고 있는 단풍나무류, 참나무류, 피나무류 등의 경쟁에 의하여 천이가 있을 것으로 사료된다.
