Verlamelin, an Antifungal Compound Produced by a Mycoparasite, Acremonium strictum

  • A strain of Acremonium strictum, the mycoparasite of Botrytis cinerea, showed strong antifungal activities both in vitro and in vivo against several phytopathogenic fungi. An antifungal substance was purified from the liquid cultures of A. strictum and identified as verlamelin by instrumental analyses. Verlamelin exhibited in vitro antifungal activity against some phytopathogenic fungi such as Magnaporthe grisea, Bipolaris maydis, and Botrytis cinerea, while it was net active against all the bacteria tested. In viva, verlamelin exhibited strong protective and curative activities, particularly against barley powdery mildew. At 100 μg/ml, it inhibited the development of barley powdery mildew with control values of more than 90% in 7-day protective and 2-day curative applications. This is the first report on the production of verlamelin by Acremonium species.

  • The Purpose of this study was to isolate and identify fungi found in excavated clothes dating on the 16th century. As a result of microscoping with LPCB staining, seven isolated strains were classified-Acremonium strictum, Acremonium kiliense, Fonsecaea sp., Candida sp., Trichoderma sp., Cladosporium spp., Penicillium spp.. In the process of isolation and identification of fungi by the sort of textile, more fungi were found in cotton, cotton batt and silk than in ramie and hemp, suggesting that hemp has antibacterial characteristics due to the presence of lignin. In the Comparison washed samples with unwashed ones, there were more kinds of fungi in washed samples. In the cellulase activity test, Acremonium kiliense had the highest level cellulase activity, suggesting that Acremonium killiense has an important Influence on damage of textiles.

  • Cephalosporium acremonium MAR-80, a strain of methionine analogue-resistant mutant, showed good activity of sulfate utilization as only sulfur source. The effect of methionine on the sulfate uptake system was investigated by using $Na_{2}^{35}SO_{4}$ as a tracer in the resting cell system. From this result, it was revealed that sulfate permease of this strain was less repressed and/or less inhibited by methionine than parent type. This deregulation was due to low actibity of methionine uptake, which was operated by somewhat simple diffusion. From these studies, it could by anticipated that the improved productivity of cephalosporin C and lower dependence of cephalosporin C production on methionine were related to increased uptake rate of sulfate.

  • YIp5를 사용하여 Cephalosporium acremonium ATCC 2033으로부터 Saccharomyces cerevisiae SHY3에서 발현되는 자율복제 기점 (ARS)를 분리하였다. 자율복제 기점을 갖는 40종의 vector 가운데 가장 안정성 (stability)이 높은 plasmid를 pCY-2라 명명하였으며 pCY-2는 C.acremonium에서 유래하는 3.7kb의 단편을 갖고 있었다. 또 Southern hybridization과 재형질전환을 통해 pCY-2가 효모내에서 자율적으로 복제되고 있다는 것을 확인하였다. 또한 pCY-2는 형질전환율과 안정성에 있어 효모의 ARS를 갖는 YRp7 vector보다 우수하였다.

Plant Growth-promoting Activity of Acremonium strictum MJN1 Isolated from Roots of Panax ginseng

  • The plant growth-promoting activity of Acremonium strictum MJN1 isolated from roots of Panox ginseng was explored. The myceliaI extract of A. strictum MJN1 enhanced the rice seedling growth by 14.5 and 9.0% in the dried weight of shoots and roots, and the growth of red pepper by 54 and 85% in the top length and the dried weight in pot experiments, respectively. The plant growth-promoting substances in the myceliaI extract of Acremonium strictum MJN1 were identified as D-adenosine and glycerol. Both commercial D-adenosine and glycerol also promoted significantly the rice seedling growth but, unlike the mycelial extract of A. strictum MJN1, hardly affected the yields of plants grown in pots or field. Therefore, it is possible that other plant growth-promoting substances are produced by A. strictum MJN1. However, this study shows that A. strictum MJN1 has a great potential as a biofertilizer.

The Improvement of Cephalosporin C Production by Fed-batch Culture of Cephalosporium acremonium M25 Using Rice Oil

  • The objective of this study is to improve cephalosporin C (CPC) production byoptimization of medium and culture conditions. A statistical method was introduced to optimize the main culture medium. The main medium for CPC production was optimized using a statistical method. Glucose and corn steep liquor (CSL) were found to be the most effective factors for CPC production. Glucose and CSL were optimized to 2.84 and $6.68\%$, respectively. CPC produc­tion was improved $50\%$ by feeding of $5\%$ rice oil at day 3rd and 5th day during the shake flask culture of C acremonium M25. The effect of agitation speeds on CPC production in a 2.5-L bio­reactor was also investigated with fed-batch mode. The maximum cell mass (54.5 g/L) was obtained at 600 rpm. However, the maximum CPC production (0.98 g/L) was obtained at 500 rpm. At this condition, the maximum CPC production was improved about $132\%$ compared to the re­sult with batch flask culture.

Identification of Acremonium acutatum and Trichothecium roseum isolated from Grape with White Stain Symptom in Korea

  • During 2010 and 2012 grape harvest seasons in Gyeonggi-do, Korea, a white stain symptom was observed on the harvested grape fruits in 'Campbell-Early' and 'Kyoho' varieties. In samples collected from the infected vine, two different strains of pathogenic fungi have been found and identified as Acremonium acutatum and Trichothecium roseum based on fungal morphology and nucleotide sequence of internal transcribed spacer (ITS) and supported by the phylogenetic analysis of the rDNA-ITS region. The DNA homologies of the isolated strains were 99.8% and 99.6% identical with T. roseum (IFB-22133) and A. acutatum (CBS682.71), respectively. In the pathogenicity test, the spores of A. acutatum and T. roseum sprayed on the grapes caused white stain symptoms on the fruits in two weeks after the artificial inoculation, which is similar to observations in the field. To our knowledge, this is the first report of white stain symptoms caused by A. acutatum and T. roseum on the grapes in Korea.
