육계.부자의 약침과 경구투여가 2형 당뇨 모델 생쥐의 혈당강하에 미치는 영향 (Effects of Cinnamomum Cassia and Aconitum Carmichaeli's Pharmacopuncture and Oral Administration on Blood Sugar in Type II Diabetic Mice)

  • Objectives : This study was designed to evaluate the antidiabetes effects of Cinnamomum cassia and Aconitum carmichaeli in type II diabetes mellitus model mice. Methods : The experimental animals were divided into 3 groups and treated for 50 days as follows:control group, pharmacopuncture group and extract administration group. The effects of Cinnamomum cassia and Aconitum carmichaeli were observed by measuring body weight, blood glucose, insulin, insulin immunoreactivity of the pancreas and subpopulation of splenocytes. Results : Cinnamomum cassia sample groups were reduced a little body weight than control group. Glucose level of extract administration group lowered than control and pharmacopuncture group. Insulin immunoreactivity of Aconitum carmichaeli extract group appeared more effective than control and pharmacopuncture group. Conclusions : This study shows that Cinnamomum cassia and Aconitum carmichaeli have a distinct antidiabetes effect in type II diabetes mellitus model.

부자류 및 Aconitine이 흰쥐 간 내 유전자발현에 미치는 영향 (Effects on Gene Expressions in the Rat Liver of Aconitine and Aconitum Species.)

  • Objectives : Aconitine is one of the toxic components of aconitum species. This study was carried out to evaluate gene expressions of herbal prescriptions containg aconitum species and oriental medicinal plants of aconitum species. Methods : We have measured gene expressions in the liver of aconitine, Aconitum carmichaeli DEBX., Aconitum ciliare DC. Yong Ho Whan using Sprague-Dawley rat. Gene expression in rat liver has been analyzed using codelink 10k microarray. Results : 1. Genes up-regulated over than 4 fold were 118 and down-regulated less than 4 fold were 91 in aconitine 50 ${\mu}g/kg/day$ over control. 2. Genes up-regulated of over than 4 fold were 124 and down-regulated less than 4 fold were 98 in Aconitum ciliare DC. 4g/kg/day and 169 of over than 4 fold, and 110 of less than 4 fold for Aconitum carmichaeli DEBX. 4g/kg/day, respectively. 3. Regulated genes in treatment group of Aconitum carmichaeli DEBX, Aconitum ciliare DC. and aconitine was only 2 different genes, Sulfotransferase family 4A, member 1 and Lin-7 homolog b (C. elegans). Conclusions : Gene expression profiles in liver were different among aconitine, Aconitum carmichaeli DEBX., Aconitum ciliare DC. and herbal prescription YongHo-whan. Furthermore, we can find many new genes related with effects of aconitum species.

수치(修治) 한약재가 사람의 간세포 WRL68와 원숭이의 신장세포 Vero에 미치는 영향 (Cytotoxic activity of processing the traditional drug on monkey kidney cell (Vero) and human liver cell (WRL68))

  • The cytotoxic activities of Pinellia ternate, Aconitum carmichaeli, Arisaema amurense, Aconitum kusnezoffii and Scutellaria baicalensis on monkey kidney cell (Vero) and human liver cell (WRL68) were evaluated by Sulforhodamine B Protein (SRB) and Tetrazolium-based (MTT) colorimetric assay methods The results were as fellows : 1 The Pinellia ternate and Arisaema amurense did not show the cytotoxic activities at any concentration without processing or natural drugs. 2 The extracts of Aconitum carmichaeli, Aconitum kusnezoffii and Scutellaria baicalensis showed cytotoxic activities. However, the cytotoxic activities of processing drugs were less effective than the natural drugs. 3. The cytotoxic activities on monkey kidney cell (Vero) and human liver cell (WRL68) of Aconitum carmichaeli was determined by MTT assay. The Kyungpo(京?) of Aconitum carmichaeli showed less concentrate than that of the Dangpo(唐?). The $IC_{50}$ value on monkey kidney cell (Vero) and human liver cell (WRL68) of Kyungpo was $937{\pm}29\;and\;731{\pm}31{\mu}g/ml$, respectively 4. The cytotoxicity on monkey kidney cell (Vero) and human liver cell (WRL68) of Scutellaria baicalensis showed strong activities without processing or natural drugs.

부자(附子)의 우울증 억제효과에 관한 연구 (Anti-depressant Effect of the Extracts of Aconitum carmichaeli)

  • Objectives : The water and methanol extracts of Aconitum carmichaeli(Aconiti Tuber Preparat) were investigated for their anti-depressant effects. Methods : In this study, reserpine-induced hypothermia test, tail suspension test and hot plate test. Additionally, the brain monoamine oxidase activity was determined in vivo. Results: In the reserpine-induced hypothermia test, both extracts suppressed the fall of body temperature compared to the control group in a dose-dependent manner, suggesting the inhibition on hypothermia. In the tail suspension test, the methanol extract dose-dependently reduced the duration of immobility by 28.4% at a dose of 1 g/kg compared to control group, which is more effective than the water extract. In the hot plate test, the water extract and methanol extract increased the jump latency time compared to the control group, showing the inhibition rate of 198% and 182%, respectively, at a dose of 1 g/kg. Methanol extracts potently inhibited the brain monoamine oxidase activity in an in vivo assay compared to the control group, showing 84.6% inhibition, but the water extract revealed very weak activity. Conclusions : Above results suggested that the extract of Aconitum carmichaeli can be useful for the prevention and treatment of depression.

고지방식이로 유도된 비만형 동물모델에서 부자 물추출물의 비만 및 당대사 개선 효능 평가 (Evaluation of Efficacy of Aconitum carmichaeli Debx Extract on Obesity and Glucose Tolerance in Diet Induced Obese Mice)

  • Objectives: This study investigated the effects of water extract of Aconitum carmichaeli Debx (ACD) on obesity and glucose tolerance in high fat diet induced obese mice. Methods: Five-week-old C5BL/6 mice were fed a high fat diet (HFD) containing or not containing ACD (100 or 300 mg/kg) for 16 weeks. Body weight, food intake, fasting blood glucose, and body temperature were checked every week and then organs, blood serums were collected after 16-week treatment. Furthermore oral glucose tolerance test (OGTT) was carried out after treatment. Results: ACD treated mice showed no significant decreases in body weight and adipose tissue weight as compared with HFD mice. Lipid accumulations in liver and serum lipid levels were not different between ACD treated and HFD mice. However, ACD extract administration significantly and dose-dependently reduced fasted plasma glucose and glucose tolerance as determined by OGTT. Conclusions: The present study demonstrates that ACD might prevent diet-induced glucose tolerance in mouse models of obesity.

  • Objectives: The prevalence of obesity and metabolic syndrome is increasing worldwide. Regulation of cellular energy metabolis has the potential to be manipulated therapeutically to serve as a target for obesity and insulin resistance. Skeletal muscle is regarded as a target for regulation of energy metabolism and insulin resistance. In this study, the authors investigated the regulatory effect of (Cinnamomum cassia Blume, CCB), Aconitum carmichaeli Debx (ACD), and Benth (Pueraria lobata Benth, PLB) on energy and glucose metabolism in C2C12 myotubes. Methods: The water extracts of CCB, ACD, and PLB (0.5 mg/ml) were treated in differentiated C2C12 myotubes. The expressions of adenosine monophosphate-activated protein kinase (AMPK) and phosphorylation AMPK were detected with western blotting. Glucose metabolism was investigated with glucose uptake assay and glucose consumption assay, total adenosine triphosphate (ATP) content was also analyzed. Results: CCB, ACD, and PLB activated the phophorylation of AMPK, they also increased the glucose metabolism and total ATP contents in C2C12 myotubes. Conclusions: This study suggests that CCB, ACD, and PLB have the potential to increase energy and glucose metabolism in skeletal muscle.

부자추출물의 골격근 세포에서의 에너지 조절 작용 (Effect of Aconitum carmichaeli Debx on Energy Metabolism in C2C12 Skeletal Muscle Cells)

  • Objectives: The all anti-obesity drugs currently approved by the US Food and Drug Administration work by reducing energy intake. In fact, no approved drug targets energy expenditure. In Korean medicine, it is known to Qi or Yang invigorating therapy could increase energy metabolism. Aconitum carmichaeli Debx (ACD) is a Yang invigorating herb, often used for treat obesity in Korean medicine. In the present study, the authors investigated the regulatory effects of ACD in energy metabolism and mitochondrial biogenesis in C2C12 skeletal muscle cells. Methods: The water extract of ACD (0.2, 0.5 and 1.0 mg/ml) were treated in differentiated C2C12 cells. The protein or mRNA levels of target genes were analyzed and mitochondrial mass were investigated. Results: ACD activated the expressions of peroxisome proliferator-activated receptor gamma coactivator 1-alpha ($PGC-1{\alpha}$), nuclear respiratory factor 1 and TFAM and increased mitochondrial mass. ACD also increased adenosin monophosphate-activated protein kinase (AMPK), and acetyl-CoA carboxylase. Conclusions: This study suggests that ACD has the potential to increase energy metabolism and mitochondrial biogenesis by activating AMPK and $PGC1{\alpha}$.

Norditerpenoid Alkaloids and Other Components from the Processed Tubers of Aconitum carmichaeli

  • A new norditerpenoid and a known alkaloid were isolated from the alkaloidal fraction of the processed tubers of Aconitum carmichaeli. The structure of the new norditerpenoid alkaloid was elucidated as lipoforesaconitine (1) on the basis of spectroscopic analysis. The known norditerpenoid alkaloid was characterized as lipoyunanaconitine (2). In addition, a new flavonoid, 6'-O-acetylliquiritin (7), along with a known ceramide, (2S,3S,4R,8E)-2-[(2'R)-2'­hydroxylignoceroylamino]-8(E)-octadecene-1,3,4-triol (3), as well as a known steroid saponin, gracillin (8), and three known flavonoids, Iiquiritigenin (4), isoliquiritigenin (5), and liquiritin (6), were also isolated and characterized. All known compounds were isolated from this plant for the first time. The structures of the isolates were established by spectroscopic and chemical methods.

마황천오약침이 비만에 미치는 영향 (The Effect of Mahuang(Ephedra sinica Stapf)-Chuanwu(Aconitum carmichaeli Debx) Pharmacopuncture Treatment in Obese Patients)

  • Objectives : We investigated the effects of Mahuang-Chuanwu Pharmacopuncture Treatment in Obese Patients by analysing Body mass index(BMI), waist measurement and weight. Methods : We investigated on 52 volunteers consisted of 24 subjects in experiment(Mahuang-Chuanwu Pharmacopuncture) group and 28 subjects in control(Normal Saline) group. The Study form was a single-blind and placebo-controlled clinical trial. 24 subjects in experiment group were injected Mahuang-Chuanwu Pharmacopuncture with additional common treatment and 28 subjects in control group were injected Normal Saline with additional common treatment with Dietic Theraphy and Exercise. We measured BMI, waist measurement and weight on 3 times : before 1st session, after first 5 sessions, and after 10 sessions. The SPSS 13.0 for Windows was used to analyze the data and the wilcoxon signed ranks test (in group), mann Whitney u t-test(between two groups) and ${\chi}^2$-test(description of number) were used to verify the result. Results : In experiment group. BMI, waist measurement and weight were significantly low at the period of after first 5 sessions and after 10 sessions. In control group, BMI, waist measurement and weight were significantly low at the period of after 10 sessions but weren't significantly low at the period of after first 5 sessions. The changes of BMI, waist measurement and weight in experiment group were significantly more than that in control group. Conclusions : The Results suggest that Mahuang(Ephedra sinica Stapf)-Chuanwu(Aconitum carmichaeli Debx) Pharmacopuncture is effective on the treatment of obesity, compared to Normal Saline.

바꽃(烏頭)의 포제(抱製)에 관한 연구 (Study on The Drug Processing of of the Roots of Aconitum carmichaeli)

  • From the tuberous root of Aconitum carmichaeli Debx.(Ranunculaceae), the main root is called as common monkhood mother root and the later root is called as the prepared aconite root. From the prepared aconite root. Looking at the processing method of the prepared aconite root, it is divided into Yeombuja (prepared aconite root processed in salt) and heuksoonpyeon (baekbupyeon) following the processing method after removing the soil and this is a way of processing the prepared aconite root without damage it. The recently produced raw prepared aconite root is easily damaged, thus it shall be preserved in salt to have the crystal shape on the surface of the prepared aconite root and store and transport in firmly solidified yeombuja condition. Therefore, yeombuja shall remove the salt before use and requires processing for use but heuksoonpyeon or baekbupyeon may use immediately. For the succession of the unique processing techniques of our ancestors, there has to be studies on the techniques. Prepared aconite root is generally used as holy medicines to cure the yang depletion syndrome, kidney-yang deficiency syndrome, and obstruction of qi in the chest syndrome. However, they are the substances with toxicity. It is contemplated that the contents of processing are broadly understood through the document on the processing method, and based on such foundation, the systematic set and proof on the documents are made along with the addition of the contemporary scientific theory and technology to develop the traditional processing technology to maximize the treatment effect and safety of prepared aconite root. In this study, the historic data and records on the processing method of latteral root of aconitum carmichaeli Debx will be rearranged to contribute to the standardization of medicinal herbs, maximization of efficacy and minimization of the side effects.
