Molecular Identification and Chemical Analysis of Aconiti Kusnezoffii Tuber on the Domestic Markets (국내 시장에서 유통되는 초오의 DNA 감별과 화학적 분석)

  • Aconiti Kusnezoffii Tuber has been traditionally used to treat the symptoms of rheumatoid arthritis and joint pain. The main constituents are diterpenoid alkaloids such as benzoylmesaconine, benzoylaconine, mesaconitine, aconitine, and hypaconitine. In Korea, Aconiti Kusnezoffii Tuber is officially defined as the tubers of Aconitum kusnezoffii Reichb., A. ciliare Decasisne, and A. triphyllum Nakai. On the other hand, only the tuber of A. kusnezoffii is to be used in China. In order to identify the botanical origin of Aconiti Kusnezoffii Tuber circulated in Korea, we analyzed 24 samples of Aconiti Kusnezoffii Tuber obtained from local markets for comparative DNA analysis. The sequence analysis of nrRNA ITS 1 was useful to distinguish Aconitum species and revealed that the roots of A. karakolicum were circulated in Korean markets without discretion. HPLC quantitative analysis showed that aconitine was detected at the highest amount in A. karakolicum. Authentic diterpenoid alkaloids were coinjected for quantification of aconitine-type ingredients. All data were statistically grouped by Principal Component Analysis (PCA). This study suggests that both molecular and chemical analyses should be utilized for the standardization and the quality control for Aconiti Kusnezoffii Tuber.

The Study of Dosages of Herbal Medicine Including Aconiti Lateralis Preparata Radix and Aconiti Tuber on Aspartate Aminotransferase, Alanine Aminotransferase (부자(附子) 및 천오(川烏)를 포함한 처방이 혈중 Aspartate Aminotransferase, Alanine Aminotransferase 수치에 미치는 영향)

  • Objectives : Aconiti Lateralis Preparata Radix (附子) and Aconiti Tuber (川烏) are not commonly prescribed, but are necessary for some clinical conditions, despite of the fact that some negative effects have been known to occur with these medicines. This study shows the consequences for aspartate aminotransferase (AST), alanine aminotransferase (ALT) due to herbal medicines, including Aconiti Lateralis Preparata Radix (附子) and Aconiti Tuber (川烏). Methods : From 1st June 2007 to 10th May 2009, the results were analyzed for 64 patients belonging to the OO Oriental Medical Hospital who took herbal medicine, including Aconiti Lateralis Preparata Radix (附子) and Aconiti Tuber (川烏), more than 20 days. This is the study about the comparison and the investigation of AST, ALT. And the results were taken upon their hospitalization and to their departure. The Criteria of Diagnosis in Liver Injury of CIOMS was used as a standard for their examinations. Results : AST, ALT (total 64). The number of patients who were within normal reference value : 49. The number of patients who exceed the normal reference value after dosage, without satisfying the Criteria of Diagnosis in Liver Injury : 5. The number of patients who were recovered in normal reference value after dosage : 9. The number of patients who satisfying the Criteria of Diagnosis in Liver Injury after dosage : 1. Conclusions : This results suggest that these herbal medicines, including Aconiti Lateralis Preparata Radix (附子) and Aconiti Tuber (川烏), didn't cause adverse side-effects on AST, ALT of the patients who are taking them.

  • A cardiac principle of Aconiti tuber, higenamine was analyzed with a differential pluse voltammetry employing a glassy carbon electrode. The peak potentials for the oxidation of higenamine were varied depending on the pH of the media. Higenamine at the concentration of $5.4{\times}10^{-7}M$ (0.146ppm) was easily determined using a pH 7.0 phosphate buffer. The amounts of higenamine in the crude Aconiti tuber and tablets containing A. tuber were determined by a standard addition method after extracting with methanol, partitioning with a mixture of $H_2O$ and $CHCl_3$, and freeze-drying the aqueous layer.

A Case Report of Severe Melosalgia with Aconiti Tuber Toxicity (하지 통증을 주요 증상으로 한 부자 중독 환자에 대한 증례 보고)

  • Objective: We report a case of non-specific Aconiti Tuber poison complaining only of severe peripheral neurotoxicity without cardiac dysfunction. Methods: The authors evaluated the symptom changes of a patient who was hospitalized in an Oriental hospital for fourteen days. The patient received acupuncture, herbal medicine, moxibustion and analgesics. Result: No abnormality in examination for cardiac function or biochemical parameters was present. The severity of pain and dysesthesia in lower extremities gradually receded during the period of treatment with herbal and western medicines. Conclusion: This study provides helpful information for treatment of Aconiti Tuber toxicity.

The effect of Aconiti tuber butanol fraction on the rabbit heart microsomal $Na^+-K^+$-activated ATPase activity (부자 Butanol Fraction이 가토 심장근 Microsomal $Na^+-K^+$-activated ATPase 활성도에 미치는 영향)

  • Aconiti tuber butanol fraction shows positive inotropic effect on the isolated atrium of rabbit heart. To investigate the mechanism, the effect on microsomal ATPase activity of rabbit heart is observed. The microsomal fraction which contains the $Na^+$- and $K^+$-activated ATPase in the presence of $Mg^{++}$ is isolated from the left ventricle of rabbit heart. The microsomal ATPase activity is maximally stimulated at $Na^+$ and $K^+$ concentration of 100 mM and 10 mM respectively. Microsomal $Na^+-K^+$-activated ATPase is inhibited by ouabain and Aconiti tuber butanol fraction. Ouabain and Aconiti tuber butanol fraction depress $Na^+$-stimulation on microsomal ATPase activity, and the inhibitory effects are not completely reversed at $Na^+$ concentration of 300 mM. Also, $K^+$-stimulation on microsomal ATPase activity is inhibited by ouabin and Aconiti tuber butanol fraction and the inhibitions are not compeletely reversed at $K^+$ concentration of 30 mM. It is, therefore, suggested that the inhibitory effect of Aconiti tuber butanol fraction on the microsomal ATPase activity may contribute to leading to the positive inotropic effect.

Studies on the Positive Inotropic Mechanism of Aconiti Tuber (부자 강심성분의 작용기전에 관한 연구)

  • Aconiti tuber butanol fraction, which is isolated from the chloroform insoluble and water soluble extract of Aconitum volubile, has been recently known to have a potent positive inotropic effect in the isolated cardiac muscle preparations of various animals. The positive inotropic mechanism of Aconiti tuber butanol fraction, in relation with the external calcium, was studied using the isolated cat papillary muscle. The positive inotropic effect was dependent on the calcium concentration in the nutrient medium, and a synergistic relation could be demonstrated between Aconiti tuber butanol fraction and the external calcium. The inotropic effect of $10^{-4}g/ml$ of Aconiti tuber butanol fraction was equivalent to that of 0.06mM of calcium in the medium. After the treatment with a calcium influx inhibitor, Verapamil$(2{\pm}10^{-7}-10^{-6}M)$, the contractile force of the papillary muscle was markedly inhibited. In these preparations, Aconiti tuber butanol fraction restored the decreased contractility in a dose-dependent manner. It was suggested that the positive inotropic effect of Aconiti tuber butanol fraction might be related with the stimulating action on the calcium influx through the slow inward calcium channels in the cardiac cell membrane. In contrast with digitalis cardiac glycoside, Aconiti tuber butanol fraction infused intravenously into the anesthetized rabbit decreased the systemic arterial blood pressure and increased the carotid blood flow, but produced no prominent changes in the heart rate.

Inhibition of Cell Proliferation and Induction of Apoptosis by Methanolic Extract of Aconiti Tuber in Human Renal Cell Carcinoma Cells (부자의 메탄올 추출물에 의한 신세포암의 증식억제 및 세포사멸 유도)

  • Aconiti Tuber is a traditional medicinal plant generally used in Oriental medicine therapy. In this study, we investigated the biochemical mechanisms of anti-proliferative effects by the methanol extract of Aconiti tuber (MEBJ) in Caki-1 human renal cell carcinoma cells. It was found that MEBJ could inhibit, in a dose-dependent manner, cell growth which was associated with apoptotic cell death such as formation of apoptotic bodies, DNA fragmentation and increased populations of apoptotic-sub G1 phase. Apoptosis of Caki-1 cells by MEBJ was associated with an up-regulation of pro-apoptotic Bax expression, and a down-regulation of anti-apoptotic Bcl-2 in a dose-dependent manner; however, the levels of IAP family were not affected. MEBJ treatment also induced the proteolytic activation of caspase-3 and -8, and a inhibition of poly(ADP-ribose) polymerase (PARP) and $PLC{\gamma}1$ protein. Furthermore, MEBJ treatment caused a dose-dependent inhibition of iNOS and cyclooxygenase-2 (Cox-2). Though further studies will be needed to identify the active compounds that confer the anti-cancer activity of MEBJ, the present findings provide important new insights into the possible molecular mechanisms of the apoptotic activity of MEBJ in cancer cells.

Isolation of Flavonoids from Processed Aconiti Tuber

  • A processed Aconiti tuber (Korean name, Kyeong-Po Buja) was extracted with 80% aqueous EtOH, and the concentrated extract was partitioned with EtOAc and water ($H_2O$). From the EtOAc fraction, two flavonoids were isolated through repeated silica gel column chromatographies. From the result of physico-chemical data including NMR, mass spectrometry and IR, the chemical structures of the compounds were determined to be liquiritin (1) and liquiritigenin (2). This is the first study to isolate flavonoids (1) and (2) from the processed Aconiti tuber.

  • Aconiti Tuber (Aconitum spp. tuber, Ranunculaceae) which contains bioactive but toxic alkaloids has been used as analgesic. cardiotonic, diuretic. and stimulant. We have previously reported two new C-19 norditerpenoid alkaloids and five known norditerpenoid alkaloids. Relationship of Bogi, Boyang, Onri herbs activated ion currents and excitatory neurotransmitter, glutamate, receptor (보기, 보양, 온리약에 의해 유발된 이온전류와 흥분성 신경전달물질인 glutamate 수용체와의 관계)

  • The effects of Bogi, Boyang and Onri herbs on glutamate receptor using nystatin perforated patch clamp under voltage clamp were investigated and the following results was obtained. Ginseng radix and Astragali radix were chosen as Bogi herbs. Cervi cornu and Boshniakiae herba were used as Boyang herbs. Aconiti tuber and Zingiberis rhizoma were selected as Onri herbs. 1. $10^{-6}M$ of glycine brought about the increase of the ion currents by Astragali radix. Cervi cornu. Boshniakiae herba and Aconiti tuber to a minute extent that has no experimental meaning. 2. It was assumed that Bogi herb had no effect on glutamate receptor since the ion currents of Ginseng radix and Astragali radix that are Bogi herbs had not been inhibited to a great extent by D-AP5 and CNQX. 3. Boyang herbs were seem to have meaningful relationship with non-NMDA receptor since the ion currents by Cervi cornu and Boshniakiae that are Boyang herbs were inhibited by CNQX but not inhibited to a meaningful extent by D-AP5. 4. D-AP5 had not a significant effect on the ion currents activated by Onri herbs such as Aconiti tuber and Zingiberis rhizoma. CNQX had a significant effect on the ion currents of Aconiti tuber but no effect on the ion currents of Zingiberis rhizoma. Therefore, it seemed that Aconiti tuber had a relationship with non-NMDA receptor while Zingiberis rhizoma had no relationship with glutamate receptor. In summary, Bogi, Boyang and Onri herbs had different effects through different mechanisms on glutamate receptor. Hence it was believed that. through experiments on acetylcholine receptor new classification could be made.
