Study on the distribution of marine bacteria and the consumption of oxygen in Wonmun bay (원문만의 해양세균분포와 산소소모량에 관한 연구)

  • Hypoxic bottom $(\leq2.0ml/l),\;40\%\;oxygen\;saturation)$ is formed in the semi-closed Wonmun bay during summer and autumn early. This study was carried out to know seasonal distribution of marine bacteria and the role of marine bacteria for forming the hypoxic bottom at Wonmun bay during summer and autumn early, 1990. During the study periods, 170 bacterial strains were isolated from sea water and sediment. Viable cell counts were ranged between $10^5-10^7\;cells/ml$. The dominant species were Acinetobacter spp. in spring, Flavobacerium spp. in summer, Pseudomonas spp. in autumn, Serratia spp. in winter. Because ETSA(Electron Transport System Activity) reveals potential consumption of oxygen in the aquatic microorganisms, the ETSA was used as potential consumption of oxygen in this study. The potential consumption of oxygen was in the range of $232.4-637.5{\mu}l/O_2/l/day$ by marine organism and $142.6-432.4{\mu}l/O_2/l/day$ by marine bacteria during the study periods. The ratio of potential oxygen consumption of marine bacteria to total marine microorganism was 0.54. The potential consumption of oxygen by marine bacteria closely related with the number of viable cells. Consequently, bacteria play an important role to form Hypoxic bottom at marine environment.

Isolation and Characterization of Aniline Degrading Bacteria (아닐린 생분해 균주의 분리 및 특성)

  • Twenty-nine bacterial isolates capable of growing on aniline as n sole source of carbon and nitrogen were obtained. Ten of these isolates were identified. Nine isolates were Identified as Pseudomonas spp. and one was Acinetobacter sp.. Five strains among 29 isolates had one or several plasmids. Four of these five strains utilized aniline through meta pathway and one through ortho pathway. Pseudomonas acidovorans 4A1 which utilized aniline through meta pathway harbored a plasmid of ca. 230 kilobases shown to be involved in aniline metabolism.

Biodegradation of and comparison of adaptability to dectergents (미생물에 의한 계면활성제의 분해능과 적응력의 비교)

  • Microorgansims utilizing anionic detergent as their carbon and sulfur sources were isolated from soils and sewages. Alkyl benzene sulfonate (Hiti) and sodium dodecyl sulfonate (SDS) were the detergent compound tested. Three of these isolated microorganisms were identified as Pseudomonas spp. and the others asKlbsiella, Enterobacter and Acinetobacter. Biodegradation rate of the detergents and growth rate of Acinetobacter Strain II-8, Pseudomonas strain H-3-1 and 554 among six isolated microorganisms were investigated with colorimetric, warburg manometric, and ultraviolet absorption analyses. By performance of 4 serial successive tranfer to new culture broth for the purpose of adaptation method, ABS and SDS could be degraded to far more than 40%-60% and 70%-75%, respectively. However the employment of nonadaptation method, ABS and SDS were degraded to 30%-45% and 45%-65%, respectively. In another words, detergents degradation ability was increased to a certain extent by successive transfer to the new minimal media. We would conclude that the development of adaptation was effective in the removal of recalcitrant compounds.

Changes in the Viable Counts and Microflora of Oyster and Weakfish during Cold Storage (굴과 Weakfish의 저온저장중 생균수 및 Microflora의 변화)

  • Oyster (Crassostrea virginica) and Weakfish (Cynoscion regalis) were stored at 6, 0, -4 and -20$^{\circ}C$ for up to 45 days and examined for changes in microflora. Aerobic plate counts (incubated at 21$^{\circ}C$) were performed at selected times during storage and 495 isolates (255 isolates from oyster and 240 isolates from Weakfish) were randomly selected from the plates during the storage. Before the storage of the fishes, viable counts of oyster were 4.9${\times}$10$\^$5/ CFU/g of meat and those of Weakfish were 1.5${\times}$10$^4$ CFU/cm$^2$of skin. Microflora of oyster before storage, the major isolates identified as Pseudomonas spp. (67%) and Vibrio spp. (20%). Pseudomonas ll1/1V-H and Flavobacterium/Cytophaga were predominant genus in the microflora of oyster during cold storage at 6, 0, -4 and -20$^{\circ}C$. The composition of the microflora of Weakfish before storage, Acinetobacter (40%) and Moraxella (33%) were the major species, with Pseudomonas and Vibrio constituting a small percentage of the total isolates. The microflora shifted to predominantly Pseudomonas spp. during storage at 6. 0 and -4$^{\circ}C$, making up from 60 to 100% of isolated strains. During frozen storage, the percentage of isolates identified as Mnraxella increased to 40-60% of the total isolates. During cold storage, halophilic bacteria (Pseudomonas lII/IV-H and Vibrio) were predominant in oyster while nonhalophilic bacteria (Pseudomonas III/IV-NH and Moraxella) were predominant in Weakfish. Vibrio spp. were higher in oyster than in Weak fish. Listeria spp. were not isolated but unidentified ${\beta}$-hemolytic bacteria were islolated from both of the fishes during cold storage.

Identification and Distribution of the Pathogenic Microorganisms Isolated from Edible Ice in North Area of Daegu, Korea (대구시 북구지역의 식용얼음에서 세균 분포 및 동정)

  • The definition of edible ice is frozen water for the use of food manufacturing, processing, or cooking, as well as for the direct eating. It has been reported that in the process of ice manufacturing and its selling, edible ice is contaminated with some microorganisms, which causes food poisoning and gastroenteritis. It was shown that besides in the edible ice, germ growth caused by various reasons occurred in the mineral water, tap water, water filtering system, and water purifier. With public awareness, in order to examine the sanitary conditions of edible ice in the Northern area of Daegu metropolitan city, 15 places were randomly selected. As a result, 14 places were found to be contaminated with microorganisms. After incubating on the Brain Heart Infusion (BHI) agar plate, 80% of Gram-negative bacilli, 17% of Gram-positive cocci, and 3% of Gram-negative cocci were cultured. Enterobacter cloacae, Chryseomonas luteola, Pantoea spp., Klebsiella pneumoniae, Acinetobacter baumannii, Acinetobacter calcoaceticus or Providencia rettgeri were detected. Gram-positive cocci cultured in BHI agar plate from 5 specimens were identified as Staphylococcus aureus or Staphylococcus xylosus, which is well known bacteria causing strong food poisoning. This present paper raises questions on the importance and awareness of sanitary conditions of edible ice and the identification of pathogenic microorganisms living in the edible ice in relation to their distribution. The examination of sanitary conditions of edible ice in other areas in Daegu seems to be also needed to find out if there are similar cases.

Distribution of Marine Bacteria and Celluar Fatty Acid Composition of Dominated Genus in Suyeong Bay (수영만의 해양세균의 분포와 우점속의 균체 지방산 조성)

  • This study was performed to examine the distribution patterns, ecological characteristics and celluar fatty acid compositions of marine bacteria in Suyeong Bay. During study periods, total cell count (TC) and viable cell count (VC) were $10^7-10^8/m\ell\;and\;10^4-10^6\;cfu/m\ell$, respectively. The temporal variations of TC showed similar patterns between surface and bottom layer, but the VC at bottom decreased gradually from winter to summer. Among the 303 bacterial strains isolated in the study area, which belong to 10 genus types, Pseudomonas spp., $(32.3\%)$, Acinetobader sup. $(19.1\%)$, Vibrio spp. $(11.2\%)$, Flovobacterium spp. $(10.6\%)$ and Bacillus spp. $(7.9\%)$ were dominant. Thirty-one fatty acids were detected from Pseudomonas spp. and Bacillus spp., which were the most predominated among Gram positive and Gram negative bacteria. Celluar fatty acid compositions of Bacillus spp. were relatively simple compared to those of Pseudomonas spp.. Relatively high ratio of monounsaturated forms were detected in Pseudomonas spp. while branched types were dominant in those of Bacillus spp.. Hydroxy and cyclopropane fatty acid were detected only in the cellular fatty acid of Pseudomonas spp.. Cellular fatty acid compositions of Pseudomonas spp. revealed relatively high percentage of $C_{16:1},\;C_{17:1},\;C_{18:1}\;and\;C_{16:0}$, While Bacillus spp. predominated $C_{16:0}\;iso\;C_{16:0}\;anteiso,\;C_{17:0}\;anteiso\;and\;C_{16:1}$.

Population of Pesticide Resistant Strains in Cultivated Soils of Honam Area and Degradation of Chlorothalonil in soil (호남지역 농경지 토양에 분포된 농약내성균의 밀도와 살균제 Chlorothalonil의 분해)

  • To obtain the basic information for degradation of remaining pesticide accumulated in cultivated soil of Honam area, the resistant bacterial strains were investigated in Chlorothalonil(TPN). Mancozeb, Bentazone, and Butachlor levels of 100, $500{\mu}g\;ml^{-1}$, and degradation of TPN by TPN-resistant bacteria in sterilized soil was studied under TPN levels 0, 10, 50 and $100{\mu}g\;g^{-1}$. A number of resistance strains were decreased with higher at concentration level of pesticide, and were higher in greenhouse than upland or paddy soil. The resistance of bacteria was strong in other of Bentazone> Butachlor> TPN> Mancozeb. The percentage of bacterial strains of resistance for pesticides isolated from the cultivated soil were the highest in Acinetobacter spp. and Corynebacterium spp., and the lowest in Moraxella spp. A number of TPN-resistant strains were the highest at the TPN level of $10{\mu}g\;g^{-1}$, and 5 days after strains inoculation, and were higher in Pseudomonas spp. TD-25 than TC-23 or strains in non-sterilized soil. The degradation of TPN was fast in order of strain TD-25>strain in non-sterilized soil >TC-23.

Characterization of the Biodiversity of the Spoilage Microbiota in Chicken Meat Using Next Generation Sequencing and Culture Dependent Approach

  • This study investigated the psychrotrophic bacteria isolated from chicken meat to characterize their microbial composition during refrigerated storage. The bacterial community was identified by the Illumina MiSeq method based on bacterial DNA extracted from spoiled chicken meat. Molecular identification of the isolated psychrotrophic bacteria was carried out using 16S rDNA sequencing and their putrefactive potential was investigated by the growth at low temperature as well as their proteolytic activities in chicken meat. From the Illumina sequencing, a total of 187,671 reads were obtained from 12 chicken samples. Regardless of the type of chicken meat (i.e., whole meat and chicken breast) and storage temperatures ($4^{\circ}C$ and $10^{\circ}C$), Pseudomonas weihenstephanensis and Pseudomonas congelans were the most prominent bacterial species. Serratia spp. and Acinetobacter spp. were prominent in chicken breast and whole chicken meat, respectively. The 118 isolated strains of psychrotrophic bacteria comprised Pseudomonas spp. (58.48%), Serratia spp. (10.17%), and Morganella spp. (6.78%). All isolates grew well at $10^{\circ}C$ and they induced different proteolytic activities depending on the species and strains. Parallel analysis of the next generation sequencing and culture dependent approach provides in-depth information on the biodiversity of the spoilage microbiota in chicken meat. Further study is needed to develop better preservation methods against these spoilage bacteria.

In vitro and In vivo Antibacterial Activity of a New Fluoroquinolone Containing C7-bicyclic Structure (C7-이환체 구조를 갖는 새로운 플루오르퀴놀론계 항생제의 in vitro와 in vivo 항균작용)

  • The in vitro and in vivo antibacterial activities of a new fluoroquinolone, DWP20364(1-cyclopropyl-5-amino-6,8-difluoro-7-(2,7-diazabicyclo[3.3.0]oto-4-ene-7-yl)-1 ,4-di-hydro-4-oxoquinoline-3-carboxylic acid) were evaluated in comparison with those of ciprofloxacin(CPFX), sparfloxacin(SPFX) and ofloxacin(OFLX). DWP20364 was more potent than CPFX and OFLX against Staphylococcus spp., Streptococcus spp. and Enterococcus faecium MD8b and it was similarly or slightly less active than CPFX against Escherichia spp. and Pseudomonas spp.. For MRSA and OFLX resistant strains (Staphylococcus spp.(14),Enterococcus spp.(4), Acinetobacter spp.(2), Pseudomonas spp.(9), Klebsiella spp.(2) and Serratia spp.(6)),DWP20364(MICs for 90% of strains,0.025 and 12.5${\mu}$g/ml, respectively) was 4 to 32 folds more potent than SPFX and CPFX. The activity of DWP20364 decreased moderately in the presence of 5mM $Mg^{2+}$. However, various pHs and the concentrations of various serum had no effect on the activity of DWP20364. DWP20364 possessed a bacteriocidal effect at the 1MIC against gram positive and gram negative strains. The protective effect of DWP20364 against systemic infections in mice caused by S. aureus Smith or S. aureus L2379 was superior to that of CPFX and SPFX but it was less active than that of CPFX against infection by P. aeruginosa E-2.

Evaluation of Characteristics of Microorganisms Isolated from Public Drinking Water Facilities in Gwangju City (광주지역 먹는물 공동시설의 미생물 특성 및 분포조사)

  • Objective: This study was performed to detect indicator bacteria in drinking spring water samples in Gwangju City and to identify their genus using the VITEK-II system. Methods: The subjects were ten drinking spring water sites in Gwangju. Samples of spring water were taken every month from September 2019 to August 2020. We analyzed for the indicator bacteria Yersinia and microorganisms isolated from the spring water. Result: According to the research results on indicator bacteria, general bacteria in st1-st7 with sterilization facilities in the spring and summer were investigated in the range of 0-2 CFU/mL and 0-12 CFU/mL. In st9, where a sterilization facility was not installed, the most general bacteria were detected (160 CFU/mL). Total coliform and fecal coliform showed unsatisfied rates of 16.7 and 11.1% in spring and 14.7 and 11.8% in summer, respectively. The unsatisfied rates of total coliform for the designated and non-designated spring water facilities were 3.8 and 47.1%, respectively, and for the fecal coliform group they were 2.5 and 35.3%. The difference was confirmed according to the presence of a sterilization facility. Yersinia spp. was not detected in all drinking spring water. Forty-one strains in 25 species were isolated from ten sites. The results classified as major dominant species are Pseudomonas spp. 14.6%, Pantoea spp. 9.8%, Serratia spp. 9.8%, Acinetobacter spp. 9.8%, Citrobacter spp. 7.3%, Bordetella spp. 7.3%, Delftia spp. 4.9%, and Enterobacter spp. 4.9%. Conclusions: Based on the result that various species derived from fecal pollution and artificial pollutants were detected in the non-specified public spring water facilities that many people use, the facilities need institutional complements such as continuous management or complete shutdowns.
