  • Microbial characteristics of bacterial population were investigated in human remains and soil inside the bones in excavated grave no.4 and no.5 at Keundokgol site, Osu-ri, Buyeo. Phylogenetic characteristics of bacterial populations were analyzed by direct extracting of ancient DNA. In this study, based on the 16S rDNA sequences, in case of grave no.4, 319s from human remain were classified into 11 phyla, and 462s from soil were classified into 16 phyla. In case of grave no.5, 271s from human remain were classified into 10 phyla, and 497s from soil were classified into 11 phyla. Especially, Actinobacteria phylogenetic group are dominant group of bacterial populations in grave no.4 and no.5. Also, most of these were analyzed uncultured group. Thus, the discovery of a diversely microbial community and uncultured group was thought to be due to the specificity of the sample. Conclusively the general excavated human bones were contaminated with soil bacteria species their near around. This results contribute to preservation and management of ancient human bone from archaeological sites.

  • Hydrochemical characteristics and microbial communities of spring (Samtong), surface water, and groundwater in Cheorwon, Korea, were analyzed. Field surveys and water quality analyses were undertaken at 10 sampling points for five spring, two surface, and three groundwater samples on 15 December 2022. Hydrochemical analysis revealed that most water samples were Ca-HCO3 type and that water-rock interactions were the predominant mineral source. Radon concentrations were <1 kBq m-3 for surface water, 1~10 kBq m-3 for spring water, and 1~1,000 kq m-3 for groundwater. Microbial cluster analysis showed that the main phyla were Proteobacteria, Planctomyceta, Verrucomicrobia, Acidobacteria, and Actinomycetota.Non-metric multidimensional scaling (NMDS) analysis indicated that water temperature, pH, and Si content were closely related to microorganism content. NMDS and canonical correspondence analysis results revealed that environmental factors affecting spring water were temperature, and Mg and Si concentrations, particularly for Acidobacteria and Proteobacteria, and Pseudomonas brenneri. Both hydrochemical and microbial community analyses yielded similar results at some spring and groundwater sampling points, likely due to the effects of a basalt aquifer.

  • A novel phytase of Acidobacteria was identified from a soil metagenome, cloned, overexpressed, and purified. It has low sequence similarity (<44%) to all the known phytases. At the optimum pH (2.5), the phytase shows an activity level of 1,792 μmol/min/mg at physiological temperature (37℃) and could retain 92% residual activity after 30 min, indicating the phytase is acidophilic and acidostable. However the phytase shows poor stability at high temperatures. To improve its thermal resistance, the enzyme was redesigned using Disulfide by Design 2.0, introducing four additional disulfide bridges. The half-life time of the engineered phytase at 60℃ and 80℃, respectively, is 3.0× and 2.8× longer than the wild-type, and its activity and acidostability are not significantly affected.

  • In this study, the structure and diversity of bacterial community were investigated in the surface and subsurface marine sediments using a NGS method (i.e. illumina sequencing technology). The bacterial community in the surface was distinct from that in the subsurface of marine sediment; with the exception of the phylum Proteobacteria, the relative abundance of Bacteroides phylum were higher in the surface than subsurface, whereas the sequences affiliated to the phyla Chloroflexi and Acidobacteria were relatively more copious in the subsurface than surface sediment. Moreover, interestingly, we observed that the phyla Nitrospinae and Nitrospirae contribute to nitrogen cycle in the marine sediment. This study may present the possibility for the presence of novel microorganisms as unexplored sources and provide basic information on the microbial community structure.

  • Marine beach sands with bacterial pathogens may cause increased outcomes of illness among beachgoers in summer. In this study, pyrosequencing of 16S ribosomal DNAs extracted from 12 beach sands was performed to understand how the environmental factors of wastewaters or human wastes affected the distribution of bacterial communities at the beach of North Gyeongsang province (Yeongdeok and Pohang counties) in the middle of October. It was found that Acidobacteria were dominantly distributed in the sands near the clean seawaters, Proteobacteria in the sands near the river waters, Cyanobacteria in the sands near the wastewaters, and Bacteroidetes in the sands near the beach park. Other phyla groups such as Actinobacteria, Chlorobi, Deferribacteres, Deinococcus-thermus, Firmicutes, Gemmatimonadetes, Nitrospirae, and/or Verrucomicrobia were distributed at low relative abundance (1-5%).

  • The objective of this study was to analyze the phylogenetic composition of bacterial community in the soil of an earth-cave (Niu Cave) using a culture-independent molecular approach. 16S rRNA genes were amplified directly from soil DNA with universally conserved and Bacteria-specific rRNA gene primers and cloned. The clone library was screened by restriction fragment length polymorphism (RFLP), and representative rRNA gene sequences were determined. A total of 115 bacterial sequence types were found in 190 analyzed clones. Phylogenetic sequence analyses revealed novel 16S rRNA gene sequence types and a high diversity of putative bacterial community. Members of these bacteria included Proteobacteria (42.6%), Acidobacteria (18.6%), Planctomycetes (9.0 %), Chloroflexi (Green nonsulfur bacteria, 7.5%), Bacteroidetes (2.1%), Gemmatimonadetes (2.7%), Nitrospirae (8.0%), Actinobacteria (High G+C Gram-positive bacteria, 6.4%) and candidate divisions (including the OP3, GN08, and SBR1093, 3.2%). Thirty-five clones were affiliated with bacteria that were related to nitrogen, sulfur, iron or manganese cycles. The comparison of the present data with the data obtained previously from caves based on 16S rRNA gene analysis revealed similarities in the bacterial community components, especially in the high abundance of Proteobacteria and Acidobacteria. Furthermore, this study provided the novel evidence for presence of Gemmatimonadetes, Nitrosomonadales, Oceanospirillales, and Rubrobacterales in a karstic hypogean environment.

  • The bacterial community structures of the marine sponge, Dactylospongia metachromia, collected from Chuuk of Micronesia on February 2012, were analyzed by denaturing gradient gel electrophoresis (DGGE). The DGGE fingerprints of two individuals of D. metachromia, CH607 and CH840 showed the same band patterns. The sequences derived from DGGE bands revealed 93~100% similarities with known bacterial species in the public database and high similarity with uncultured bacterial clones. The bacterial community structures of both D. metachromia sponges (CH607, CH840) were composed of 6 phyla, 8 classes: Alphaproteobacteria, Betaproteobacteria, Gammaproteobacteria, Acidobacteria, Actinobacteria, Chloroflexi, Cyanobacteria, Spirochaetes. DGGE fingerprint - based phylogenetic analysis revealed that the bacterial community profiles were identical in two individuals of the same sponge species collected from the same geographical location.

  • In this study, bacterial diversity in the living room and bathroom of a residential environment was analyzed using the pyrosequencing method. There was no difference in the diversity index of bacteria between the 2 rooms; however, differences were noted in the composition of bacteria. The classes ${\beta}$-Proteobacteria and ${\delta}$-Proteobacteria were found in the bathroom at higher abundances than in the living room. The phyla Acidobacteria, Chlorobi, Chloroflexi, Fusobacteria, Nitrospirae, and Planctomycetes were found in the bathroom, but not in the living room, indicating a broader range of bacteria. However, the living room showed a more diverse range of bacterial genera than the bathroom did. In both the living room and the bathroom, the genus Methylobacterium was dominant.

  • The diversity of epiphytic bacterial communities on deciduous oak tree (Quercus dentate Thunb.) leaves was examined both in the natural forest area with a clean air and in the industrial estate to assess effects of acidic deposition to the phyllosphere using 16S rDNA sequence data. In addition, acid-tolerant epiphytic bacterial communities were compared. A total of 78 epiphytic and 444 acid-tolerant clones were obtained from clone libraries, resulting in 20 and 17 phylotypes by analysis of restriction fragment length polymorphism (RFLP) for PCR-amplified 16S rDNA products. A low bacterial diversity in both areas was found. As tree leaves grow older, bacterial diversities were slightly increased in the level of subphylum. The community structure of epiphytic bacteria in both areas in April consisted of only two subphyla, $\beta-and\;\gamma-Proteobacteria$. In August two additional subphyla in both areas were found, but the composition was a little different, Acidobacteria and Cytophaga-Flexibacter-Bacteroids (CFB) group in the industrial estate and a -Proteobacteria and CFB group in the natural area, respectively. Acidobacteria could be an indicator of epiphytic bacteria for acidic deposition on plant leaves, whereas a -Proteobacteria be one of epiphytic bacteria that naturally survive on leaves that are not affected by acidic deposition. The acid-tolerant bacterial communities in April were composed of two subphyla, $\gamma-Proteobacteria$ and Low G+C gram-positive bacteria in both areas, and in August a-Proteobacteria was added to the community just in the natural forest area. The direct influence of acidic deposition on the acid-tolerant bacterial phylogenetic composition could not be detected in higher taxonomic levels such as subphylum, but at narrower or finer levels it could be observed by a detection of Xanthomonadales group of $\gamma-Proteobacteria$ just in the industrial estate.

  • In this study, variations of the organic and nitrogenous compounds in wastewater were investigated in a single reactor with intermittent aeration. Over 90% of organic and nitrogen removals are accomplished with C/N ratio of 3 : 1 and 20/20 min of aeration/non-aeration period. Longer non-aeration period on the aeration/non-aeration cycle showed more stable nitrogen removal, showing various microbial community in the reactor. From PCR-DGGE analysis, it is conclusive that Dysgonomonas mossii strain Melo40, Eubacterium sp. oral clone JN088, Uncultured bacterium clone SPESB2_718, and Bacterium enrichment culture clone LE are related with the organics and nitrogen oxidation. Uncultured Acidobacteria bacterium clone AKYG487, Lactobacillus harbinensis strain FQ003, Erythrobacter litoralis strain Gi-3, Phytobacter diazotrophicus strain Ls8, and Mycobacterium sp. enrichment culture clone GE10037biofNNA are distinctly appeared under denitrification condition.
