Inhibiting Effect of Nicotinic Acid Hydrazide on Corrosion of Aluminum and Mild Steel in Acidic Medium

  • The corrosion behavior of aluminum and mild steel in hydrochloric acid medium was studied using a nicotinic acid hydrazide as inhibitor by potentiodynamic polarization, electrochemical impedance spectroscopy technique and gravimetric methods. The effects of inhibitor concentration and temperature were investigated. The experimental results suggested, nicotinic acid hydrazide is a good corrosion inhibitor for both aluminum and mild steel in hydrochloric acid medium and the inhibition efficiency increased with increase in the inhibitor concentration. The polarization studies revealed that nicotinic acid hydrazide exhibits mixed type of inhibition. The inhibition was assumed to occur via adsorption of the inhibitor molecules on the aluminum and mild steel surface and inhibits corrosion by blocking the reaction sites on the surface of aluminum.

  • Precipitation samples were collected at sixteen sites in Northeast Asia from June 1995 to February 1997, and were analysed for the anions $SO_4^{2-}$, $NO_3^-$and $Cl^-$ and for the cartons $Na^+, NH_4^+, K^+, Mg^{2+} and $Ca^{2+}$in addition to pH and conductivity measurements. The quality assurance of chemical composition data was checked by considering the ion balance evaluating by 1 h 1 value and the conductilfty balance. The sum of cation concentrations were slightty greater than the sum of anion concentraions. For the anions, $SO_4^{2-}$ clearly dominates in most of sampling states whereas $Cl^-$ is more abundant in coast and rural sites In. Tapan. For the cations. $Ca^{2+}$ and $NH_4^+$ are generally the more abundant tons except when sources of Na+ exist. The contribution rate of nss-$SO_4^2-and NO_3^-$ to acidity are about 70% and 10-30%. respectively. The neutralizing capacity by a major neutralizing cations such as $NH_4^+$ and nuts -$SO_4^2$. are above 98%(heavy polluted and urban sites in China), above 70%(urban sites in Japi,n and in Korea, coast sites In Chinas and above 60%(rural sites in Japan and in Korea), respectively.

Binding Characteristics of Molecularly Imprinted Polymers for Ibuprofen Enantiomers (아이뷰프로펜 이성질체에 대한 molecularly imprinted polymers의 binding 특성)

  • The molecularly imprinted polymers(MIPs) synthesized at various polymerization conditions were examined as ibuprofen receptors in terms of binding characteristics. The 4-vinylpyridine polymers had 1.2 times higher adsorption capability for (S)-(+)-ibuprofen than the methacrylic acid polymers. The methacrylic acid polymers synthesized by UV radiation had 1.9 times higher selectivity for (S)-(+)-ibuprofen compared to those by thermal initiation. Effects of various solvents for binding were also examined in this research. According to the Scatchard analysis, the (S)-(+)-ibuprofen artificial receptors had two different kinds of binding sites for (S)-(+)-ibuprofen while having only single kind of binding site for ketoprofen. The binding sites of (S)-(+)-ibuprofen, n were calculated as 4.3~4.9 $\mu$mol/g and the dissociation constants, $K_D$ were 0.68 mM for the specific binding.

Cloning and mRNA Expression Analysis of the Gene Encoding Phenylalanine Ammonia-Lyase of the Ectomycorrhizal Fungus Tricholoma matsutake

  • The ectomycorrhizal fungus Tricholoma matsutake grows symbiotically with Pinus densiflora. Phenylalanine ammonia-lyase (E.C. 4.3.1.24) catalyzes the conversion of L-phenylalanine to trans-cinnamic acid. The role of fungal phenylalanine ammonia-lyase, however, has not been clear until now. In this study, the gene encoding phenylalanine ammonia-lyase (PAL), which was isolated from T. matsutake, was cloned and characterized. The PAL gene (tmpal) consists of 2,160 nucleotides, coding for a polypeptide containing 719 amino acid residues. The deduced amino acid sequence of tmpal from T. matsutake shows high identity (70%) with that from Laccaria bicolor. Comparative analysis of the PAL genes among T. matsutake and other species of the class Agaricomycetes showed that both active sites and binding sites were significantly conserved among these genes. The transcriptional analysis of the PAL gene revealed a differential gene expression pattern depending on the developmental stages (mycelium, primordium, stipe, pileus, and gills) of T. matsutake. These results suggest that the PAL gene in T. matsutake plays an important role in multiple physiological functions.

Sequence Analysis and Expression of Xylanase Gene (xynY) from Alkalophilic Bacillus sp. YC-335

  • The nucleotide sequence of the xylanase gene (xynY) from alkalophilic Bacillus sp. YC-335 was determined and analyzed. An open reading frame of 1, 062 base pairs for xynY gene was observed and encoded for a protein of 354 amino acids with a molecular weight of 38, 915. S1 nuclease mapping showed that the transcription initiation sites of the xynY gene were different in Bacillus sp. YC-335 and Escherichia coli HB101 (pYS55). S1 mapping also showed that -10 region of the xynY gene recognized by RNA polymerases of E. coli and Bacillus sp. YC-335 were TACAGT and TATGAT , respectively. A ribosome binding site sequence with the free energy of -17.0 Kcal/mol was observed 9 base pairs upstream from the unusual initiation codon, TTG. The proposed signal sequence consisted of 27 amino acids, 2 of which were basic amino acid residues and 21 were hydrophobic amino acid residues. When the amino acid sequences of xylanases were compared, Bacillus sp. YC-335 xylanase showed more than 50% homology with xylanases from B. pumilus, B. subtilis, and B. circulans.

  • Glycerol-3-phosphate cytidylyltransferase from Bacillus subtilis was modified with various chemical modifiers to determine the active sites of the enzyme. Treatment of the enzyme with group-specific reagents diethylpyrocarbonate, N-bromosuccinimide, or carbodiimide resulted in complete loss of enzyme activity, which shows histidine, tryptophan, and glutamic acid or aspartic acid residues are at or near the active site. In each case, inactivation followed pseudo first-order kinetics. Inclusion of glycerol-3-phosphate and/or CTP prevented the inactivation, indicating the presence of tryptophan and glutamic acid or aspartic acid residues at the substrate binding site. Analysis of kinetics of inactivation showed that the loss of enzyme activity was due to modification of a two histidine residues, single tryptophan residue, and two glutamic acid or aspartic acid residues.

KDICical Characteristics and Microbial Activity of Streams Contaminated by The Abandoned Coal Mine Drainage (폐탄광 배수에 의해 오염된 하천의 화학적 특성과 미생물 활성)

  • A survey was carried out to investigate the contamination of streams by the acid mine drainage originated from the abandoned coal mines and coal refuse piles. The physico-KDICical characteristics such as pH, sulfate and elements concentrations in the water and sediment in streams were analyzed. Microbial activity in the sediment was evaluated by measuring dehydrogenase activities. At sites contaminated by acid mine drainage, the pH of the water and sediment declined to acidic range from neutral due to the accumulation of sulfate. The dehydrogenase activity ranged from 12 to $170{\mu}g-TPF{\cdot}g-dry\;soil^{-1}{\cdot}24h^{-1}$ at the contaminated sites, whereas uncontaminated sites had activities of 1,176~4,259 ${\mu}g-TPF{\cdot}g-dry\;soil^{-1}{\cdot}24h^{-1}$. The dehydrogenase activity was significantly affected by low pH of the sediment, indicating that high concentration of sulfate inhibited microbial activity. The concentrations of heavy metals such as Pb and Fe in contaminated sdeiment (37~46 ppm Pb; 46,000~464,000 ppm Fe) were much higher than those in the uncontaminated sediment. The concentration of Al in the contaminated water acidfied by coal mine drainage was in the range of 11 to 42 ppm. Compared with those in the uncontaminated sediment, the concentrations of Mn, Mg and Ca in contaminated sediment were low because of the leaching from soil to water by the acidfied stream water.

Effects of atmospheric environmental changes on annual ring growth of Cryptomeria japonica in Southern Korea

  • Annual ring formation is considered a source of information to investigate the effects of environmental changes caused by temperature, air pollution, and acid rain on tree growth. A comparative investigation of annual ring growth of Cryptomeria japonica in relation to environmental changes was conducted at two sites in southern Korea (Haenam and Jangseong). Three wood disks from each site were collected from stems at breast height and annual ring growth was analyzed. Annual ring area at two sites increased over time (p > 0.05). Tree ring growth rate in Jangseong was higher than that in Haenam. Annual ring area increment in Jangseong was more strongly correlated with environmental variables than that in Haenam; annual ring growth increased with increasing temperature (p < 0.01) and a positive effect of $NO_2$ concentration on annual ring area (p < 0.05) could be attributed to nitrogen deposition in Jangseong. The correlation of annual ring growth increased with decreasing $SO_2$ and $CO_2$ concentrations (p < 0.01) in Jangseong. Variation in annual growth rings in Jangseong could be associated with temperature changes and N deposition. In Haenam, annual ring growth was correlated with $SO_2$ concentration (p < 0.01), and there was a negative relationship between precipitation pH and annual ring area (p < 0.01) which may reflect changes in nutrient cycles due to the acid rain. Therefore, the combined effects of increased $CO_2$, N deposition, and temperature on tree ring growth in Jangseong may be linked to soil acidification in this forest ecosystem. The interactions between air pollution ($SO_2$) and precipitation pH in Haenam may affect tree growth and may change nutrient cycles in this site. These results suggested that annual tree ring growth in Jangseong was more correlated with environmental variables than that in Haenam. However, the further growth of C. japonica forest at two sites is at risk from the long-term effects of acid deposition from fossil fuel combustion.
