Genetics and Breeding for Modified Fatty Acid Profile in Soybean Seed Oil

  • Soybean [Glycine max(L.) Merr.] oil is versatile and used in many products. Modifying the fatty acid profile would make soy oil more functional in food and other products. The ideal oil with the most end uses would have saturates(palmitic + stearic acids) reduced from 15 to < 7%, oleic acid increased from 23 to > 55%, and linolenic acid reduced from 8 to < 3%. Reduced palmitic acid(16:0) is conditioned by three or more recessive alleles at the Fap locus. QTLs for reduced palmitic acid have mapped to linkage groups(LGs) A1, A2, B2, H, J, and L. Genes at the Fad locus control oleic acid content(18:1). Six QTLs($R^2$=4-25%) for increased 18:1 in N00-3350(50 to 60% 18:1) explained four to 25% of the phenotypic variation. M23, a Japanese mutant line with 40 to 50% 18:1 is controlled by a single recessive gene, ol. A candidate gene for FAD2-1A can be used in marker-assisted breeding for high 18:1 from M23. Low linolenic acid(18:3) is desirable in soy oil to reduce hydrogenation and trans-fat accumulation. Three independent recessive genes affecting omega-3 fatty acid desaturase enzyme activity are responsible for the lower 18:3 content in soybeans. Linolenic acid can be reduced from 8 to about 4, 2, and 1% from copies of one, two, or three genes, respectively. Using a candidate gene approach perfect markers for three microsomal omega-3 desaturase genes have been characterized and can readily be used in for marker assisted selection in breeding for low 18:3.

Effect of DHA-Rich Fish Oil on Brain Development and Learing Ability in Rats (DHA가 풍부한 어유가 새끼쥐의 뇌발달과 학습능력에 미치는 영향)

  • Effect of DHA-rich fish oil on brain development and learning ability has been studied in Sprague Dawley rats. Female rats were fed experimental diets containing either corn oil fish oil at 10%(w/w) level throughout the gestation and lactation. Corn oil was added in fish oil diet to supply essential fatty acid at 2.3% of the calories. All male pups were weaned to the same diets of dams at 21-days after birth. Plasma fatty acid composition was analyzed for dams and pups at 21-days, 28-days and 22-weeks after birth. The analysis of DNA and fatty acid profile in the brain were undertaken at birth, 3, 7, 14, 21, 28 days and 22 weeks after birth and learning ability was tested at 18-20 weeks of age. Regardless of dietary fats, arachidonic acid(AA) and docosahexaenoic acid(DHA) were the principal polyunsaturated fatty acids in the brain. Rats fed CO diet showed a continouus increase of AA content in the brain from 10.9%(at birth) to maximum 15.3% level (14-days old), while the rars fed FO diet showed 78-79% of CO group throughout the period. Rats fed FO diet showed higher incorparation of DHA from 15.2% at birth to a maximum level of 18.5% at 140days, while the rats fed CO diet showed only 7.0% incorporation of DHA at birth and a maximum level of 11.1% at 21-days. Compared to CO group, FO group showed lower ratio of chol/PL and higher content of DHA in brain microsomal membrane, resulting in better membrane fluidity. Total amount of DNA per gram of brain was reached maximum level at 21 days in both groups. This would be a period of the cell proliferation during brain development. Overall, the rats fed fish oil diet showed a higher incorporation of DHA and membrane fluidity in the brain and better learning performances (p<0.05).

Quality Evaluation on Use of Camellia Oil as an Alternative Method in Dried Seaweed Preparation

  • The fatty acid and volatile compound compositions of camellia oil were analyzed in this study. The impacts of the replacement of conventional vegetable oil with camellia oil on the sensory attributes of dried seaweed were also determined. C18:1 (83.59%), followed by C16:0 and C18:2, were the most abundant fatty acids in camellia oil. A total of 11 and 32 volatile compounds were identified in camellia oil and sesame oil, respectively. In the preference test, the camellia oil samples received a higher, although insignificant, liking rating in overall acceptability of appearance. Overall, there were no differences between the sensory attributes of camellia oil and sesame oil. This finding, combined with the unique fatty acid composition, thermal stability, and health benefits of camellia oil indicate that further study into the use of camellia oil in foods is warranted.

Differential Effect of n6 and n3 Polyunsaturated Fatty Acids on Plasma Lipids in Rats Fed Low and High Fat Diets (식이지방 수준에 따라 n6 와 n3 계 불포화지방산이 혈장 지질수준에 미치는 영향에 관한 비교연구)

  • To compare the hypolipidemic effects of n6 and n3 PUFA at different fat levels, male Sprague Dawley rats were fed either low fat (LF, 10% Cal) or high fat (HF, 40% Cal) diet which was different only in fatty acid composition for 6 weeks. Dietary fats were beef tallow, corn oil, perilla oil, and fish oil concentrate as a source of saturated fatty acid, n6 linoleic acid(LA). n3 ${\alpha}-linolenic$ acid(LL) and n3 eicosapentaenoic acid(EPA)+docosahexaenoic acid(DHA), respectively. VLDL fraction was separated by ultracentrifugation and chemical composition was determined by thin layer chromatography. Plasma cholesterol level was increased by n6 LA but decreased by n3 LL and n3 EPA in LF and HF diets, and the hypocholesterolemic effect of n3 EPA was most significant in HF diet. HDL-Chol level was raised by n6 LA in LF and HF diets, but significantly reduced by n3 EPA in HF. Plasma TG level was reduced by n6 LA n3 LL and EPA in LF and HF with the reduction of lipogenic enzyme activity only by n3 PUFAs. The proportion of TG in VLDL fraction was significantly lowered by n3 EPA in LF and HF. The proportion of apo-B in VLDL fraction was not changed in LF, but was significantly decreased in HF by n3 EPA. Therefore, the hypotriglyceridemic effect of n3 PUFA could be from the reduced lipogenesis in liver and resulted in the depressed secretion of TG as VLDL in LF and HF with significant lower production of apoB in HF diet.

The Changes of Physico-chemical Properties of the Frying Oils during Potato and Chicken Frying (감자 및 닭튀김 중의 튀김유의 물리화학적 특성의 변화)

  • The changes of the physico-chemical characteristics of potato and chicken frying oil with frying number were examined. The changes in temperatures of frying oil during the potato flying were greater than that during the chicken frying. The weight loss of potato during deep frying was about 21.9% and about 12.3% for chicken. The acid values and conjugated dienoic acid values of the potato and chicken frying oils increased with increased frying number. Their values were greater in the chicken frying oil than in the potato frying oil. The peroxide values of frying oil did not change regularly as frying number increased. But iodine values of the frying oils decreased with increased frying number. Linoleic acid content of the frying oil decreased, whereas oleic acid content increased with frying number.

Effect of Seeding Time on Fatty Acid Composition, Oil Contents and Seeds Yield in Flax

  • Influences of different seeding dates on growth, seed yield, fatty acid composition and oil content were investigated in flax plants for two years. The results indicated that plant height in early seeding date was higher than that of delayed seeding dates during first season. Furthermore, seeding date also significantly affected the ripened seed rate and the rate increased with the delay in seeding date in first season. Seed yield in the first crop season was significantly higher than the second crop season. Palmitic acid showed variation in different seeding dates. Contrarily, stearic acid was stable and did not changed by different seeding dates. Linolenic acid was found in highest amount in all seeding dates consecutively in two cropping years. Highest oil content was recovered from the seeds of flax sown at 29 Apr. and May 9 in first and second cropping year respectively.

Comparative Study of Fatty Acid Composition and Characterization of Fixed Oil of Four Peanut Varieties Available in Pakistan

  • four varieties of ground nut (Arechis hypogea) were collected from different areas of Pakistan (Tillagang, Gujar khan, Hanoi and Pipplan) from Punjab province. The oils were extracted for the evaluation of lipid. Physico-chemical values of oils were determined like refractive index, peroxide value, unsaponifiable matter, acid value, iodine value, free fatty acid and ester value. The lipid profile as indicated by GLC showed that Palmitic acid ranged from 8.2-8.8%, proportions of oleic acid and linoleic acid varied from 57.8-59.87% and 22.5-24.1% respectively. Gujar khan variety of peanut has higher % age of oil (56.79). The higher yield of oil in this variety is not at the cost of any nutritional quality of peanut oil.

Synergistic Effect of Tocopherol, Citric Acid and Sodium Polyphosphate on the Oxidative Stability of Heated Frying Oil (가열유지(加熱油脂)의 산화안정성(酸化安定性)에 대(對)한 Tocopheol, 구연산 및 인산염(燐酸鹽)의 상승효과(相乘效果))

  • The oxidative stability of palm oil with moisture addition during heationg was investigated and the antioxidant effects of tocopherol, citric acid and sodium polyphosphate were evaluated. The addition of natural tocopherol up to 200 ppm level did not improve the oxidative stability of palm oil with moisture (1m1 $H_2O$ additive to 200g of oil at $180^{\circ}C$). The maximum antioxidant effect was observed at the Concentration of 400 ppm. Addition of citirc acid did not influence the effective concentration of tocopherol. Whereas addition of sodium polyphosphate and citric acid increased the antioxidant effect of tocopherol. The maximum synergistice effect was observed at the concentrations of 50 ppm citric acid and 50 ppm of sodium polyphosphate. Addition of 50 ppm citric acid and 50 ppm sodium polyphosphate to 100 ppm of tocopherol exceeded the antioxidant effect of 400 ppm tocopherol alone.

  • This study was designed to examine the effect of ${\omega}-3$ fatty acid, linlenic acid, EPA, DHA on serum lipid and cytokines of male rats(Sprague-Dawley). Animals of 3 groups were administrated perilla oil, salmoon oil, and tuna oil of 0.4 $m{\ell}/day$ for 8 weeks respectively. These oils were used for a source of linolenic acid, EPA and DHA. ${\omega}-3$ polyunsaturated fatty acid decreases significantly body weight, serum $PGE_2$ content and serum cytokines content of the rat, and increases internal organs weight, specially liver weight and serum HDL-cholesterol level of the rat. In the results, authors propose to use perilla oil for source of effective ${\omega}-3$ poly-unsaturated fatty acid(linolenic acid) to Prevent cardiovascular and immune diseases.

  • Structured lipids (SLs) are defined as triacylglycerols to change the fatty acid composition in the glycerol backbone and lipases are known as a powerful tool for the syntheses of SLs. Structured lipid from corn oil, capric acid, and conjugated linoleic acid (CLA) by transesterification reaction and using several amounts of immobilized lipase RM IM (from Rhizomucor miehei) was studied, and 4% of lipase amount was selected for further study as the optimal amount. Comparison the chemical properties (free fatty acid value, iodine value, saponification value, tocopherols, and color analysis), solidification behavior, and volatile fractions (from headspace SPME GC-MS) between com oil and SL com oil was obtained.
