Ethanol Extracts of Achillea millefolium and Hypericum perforatum Low Anti-Toxoplasma Activity

  • Objectives: This study was performed to determine the lethal and the inhibitory effects of ethanol extracts of Achillea millefolium (A. millefolium) and Hypericum perforatum (H. perforatum) on Toxoplasma gondii (T. gondii) RH strain tachyzoites in vitro. Methods: The tachyzoites were treated with concentrations of 10, 50, and 100 mg/mL of A. millefolium and H. perforatum extracts within 10, 30, and 45 minutes in the wells. The mortality rates of tachyzoites treated with extracts were determined by using alkaline methylene blue staining. Also, the tachyzoites in cell cultures were treated with concentrations of 50, 100, and 200 mg/mL of these extracts. The cell viability, inhibition concentration ($IC_{50}$), and selectivity were determined from MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assays. Results: In the cell-free in vitro study, all of tachyzoites were killed at concentrations of 100 mg/mL of both extracts while at concentration 10 mg/mL, the mortality was 4.53% - 5.31%. In the cell culture study, the values of the effective concentration ($EC_{50}$) were 215 and $153{\mu}g/mL$ and the selectivities were 0.73 and 0.69 for the A. millefolium and the H. perforatum extracts, respectively. Conclusion: We conclude that neither extracts has any significant effect on the tachyzoites of T. gondii in cell cultures.

Insecticidal Activities of 10 Plant Essential Oils against Plodia interpunctella and Tribolium castaneum (화랑곡나방과 거짓쌀도둑거저리의 유충과 성충에 대한 10종 식물 정유의 살충효과)

  • The insecticidal activities of the essential oils of Artemisia vulgaris, Achillea millefolium, Anethum graveolens, Cimicifuga heracleifolia, Citrus paradisi, Chrysanthemum indicum, Eucalyptus dives, Neroli bigard, Nigella sativa, and Ruta graveolens against stored-grain insects, Plodia interpunctella and Tribolium castaneum larvae and adults were evaluated using fumigant and contact bioassays. Based on the $LD_{50}$ values in fumigant bioassay, A. millefolium ($0.12{\mu}g/cm^3$) and A. graveolens ($93.14{\mu}g\;cm^{-3}$) oil were most effective against P. interpunctella and T. castaneum adults. In addition, N. bigard ($139.11{\mu}g\;cm^{-3}$) and A. graveolens ($52.87{\mu}g\;cm^{-3}$) oil exhibit strong insecticidal activities against P. interpunctella and Tribolium castaneum larvae in contact bioassay. These results indicate that A. millefolium, A. graveolens, N. bigard, and A. graveolens oils could be effective natural acaricides for managing stored-grain insects, P. interpunctella and T. castaneum.

Screening the extracts of the seeds of Achillea millefolium, Angelica sylvestris and Phleum pratense for antibacterial, antioxidant activities and general toxicity

  • Various extracts of higher plants have been used in traditional medicine systems for centuries. While tropical and sub-tropical plants have received considerable attention from the researchers for evaluation of their bioactivity, temeperate plants have always been neglected somewhat. Similarly, seeds of the plants have not been considered seriously compared to other plant parts, e.g. leaves, stems, roots, flowers, etc. as a potential source for biologically active compounds. As part of our on-going evaluation of the extracts of the seeds of temperate plants, especially from Scotland, for biological activity, Achillea millefolium, Angelica sylvestris and Phleum pratense have been chosen for the present study. Both A. millefolium and A. sylvestris are well known for their traditional medicinal uses in Europe and also in the orient, but there is no report on any medicinal properties of P. pratense available to date. Extracts of the seeds of these plants have been assessed for their antioxidant and antibacterial potential and also for general toxicity. Both DCM and MeOH extracts of A. millefolium showed the most significant broad spectrum antibacterial activity among the three plants and inhibited the growth of almost all test strains of bacteria. The DCM extracts of all three species were active against methicillin resistant Staphylococcus aureus (MRSA) and Citrobacter freundii $(MIC=6.25{\times}10^{-1}\;mg/mL)$. While the MeOH extracts of A. millefolium and P. pratense were active against C. freundii, that of P. pratense was also active against MRSA. The MeOH extract of A. sylvestris did not show any antibacterial activity against any of the eight bacterial strains at test concentrations. The MeOH extract of P. pratense showed the most prominent antioxidant activity $(IC_{50}=145\;{\mu}g/ml)$ and there was no antioxidant activity observed with the DCM extract of A. millefolium. The DCM extract of P. pratense was the most toxic $(LC_{50}=20\;{\mu}g/ml)$ among the extracts.

Study on the Growth Characteristics, Botanical Composition and Fauna Distribution of Wildflower Pasture (야생화 도입 초지의 생육특성, 식생비율 및 동물상에 관한 연구)

  • The purpose of this study was to suggest the possibility of utilizing the wildflower pasture for promoting public interest. Growth characteristics, botanical composition, fauna distribution on wildflower pastures were observed. The experimental design included two treatments: native wildflower pasture(NWP, turf grasses 6 species + native wildflower 11 species) and introduced wildflower pasture(lWP, turf grasses 6 species + introduced wildflower 9 species). The field trials were carried out on the experimental pasture plots at Chungnam National University from 1997 to 2000. The results obtained are as follows: 1. The flowering of wildflower was maintained continuously from May to September, and the colors of wildflowers; varied seasonally during this period. With native flowers, however, Hemerocallis fulva, Belamcanda chinenis and Aster koraiensisi showed problems in lately germination and early establishment. Meanwhile, Introduced wildflower showed not only excellent germination and early establishment compared to native flowers species but also maintained brighter colors. But Coreopsis tinctoria, Achillea mi/lefolium and Rudbeckia bicolor had colonized at a higher height or possessed stronger rhizome. 2. The appropriate species of turf grass which maintained continuous seasonal distribution are thought to be tall fescue, perennial ryegrass. Kentucky bluegrass in NWP and IWP. 3. Botanical composition of wild flower in NWP was arranged in the order of Achillea sibirica > Lotus corniculatus var. Japonicus > Dianthus chinensis > Plantago asiatica > Taraxacum pla~ycarpum > Viola mandshurica > Aster koraiensis > Vicia tetasperma > Lespedeza stipulacea > Hemerocallis fulva, respectively. The highest seasonal distribution of native wildflower, Achillea sibirica was in spring and summer, Lotus corniculatus var. Japonicus was in autumn. Botanical composition of wild flower in IWP was arranged in the order of Achillea millefolium Coreopsis tinctoria > Silene armeria > Coreopsis lanceolata > Rudbeckia bicolor > Sanguisorba oficinalis > Centaurea cyanus > Chrysanthemum leucanthemum > Dianthus petraeus, respectively. The highest seasonal distribution of introduced wildflower, Silene armeria was in spring, Achillea millefolium was in summer, and Coreopsis tinctoria was in autumn.

Essential Oil Isolated from Iranian Yarrow as a Bio-rational Agent to the Management of Saw-toothed Grain Beetle, Oryzaephilus surinamensis (L.) (머리대장가는납작벌레의 합리적 방제 물질로 이란 서양가새풀 정유의 살충효과 평가)

  • Overuse of synthetic pesticides caused negative side-effects such as environmental contamination, development of insect pests' resistance, and effects on non-target organisms. Plant origin substances without/or with low mammalian toxicity have been considered as promising alternatives to the synthetic pesticides. Fumigant toxicity of the essential oil of Iranian Yarrow, Achillea millefolium L., was investigated against a cosmopolitan stored-product insect pest: saw-toothed grain beetle (Oryzaephilus surinamensis L.). Chemical profile of this essential oil was studied by Gas Chromatography-Mass Spectrometry. Tested concentrations were significantly effective to the mortality of insect pest. A positive correlation between essential oil concentrations and pest mortality were realized. LC50 value (lethal concentration needed to 50% mortality) was achieved as $17.977(16.195{\pm}20.433){\mu}l/l$ air. The main components were 1,8-Cineole (13.17%), nerolidol (12.87%), ${\alpha}$-cubebene (12.35%), artemisia ketone (6.69%), ${\alpha}$-terpineol (5.27%), alloaromadendrene oxide (4.71%) and borneol (3.99%). Terpenic compounds including monoterpene hydrocarbons (8.19%), monoterpenoids (44.23%), sesquiterpene hydrocarbons (21.69%) and sesquiterpenoids (22.24%) were 96.35% of the total identified compounds. Results indicated that the terpene-rich A. millefolium essential oil may be considered as a safe bio-agent in the O. surinamensis management.

Effect of Ethephon, Paclobutrazol, and Uniconazole on Growth and Flowering of Potted Yarrow, Achillea millefolium 'Cerise Queen' (Ethephon, Paclobutrazol과 Uniconazole 처리가 분화 서양톱풀의 생장과 개화에 미치는 영향)

  • The purpose of this study was to investigate the effects of ethephon, paclobutrazol, and uniconazole on the flowering of potted Yarrow (Archillea millefolium). Each plant growth regulator was sprayed two times before stem elongation period of yarrow. The tallest was 45.7 cm at $1000mg{\cdot}L^{-1}$ ethephon and the shortest was 11.1 cm at 20 uniconazole. Uniconazole delayed flowering and decreased the number of flowers. Both uniconazole at 20 ppm and paclobutrazol at 200 ppm decreased the number of florets to 90.0% and 43.6% of control, respectively. In ethephon, number of florets was increased 34.1% compared with control. In addition, ethephon and paclobutrazol increased the number of mean auxillaly flowers to 28.6% and 23.5% of control, respectively. On the other hand, uniconazole decrease the number of auxillary flowers to 55.3%. Thus, paclobutrazol was recommended as a suitable plant growth regulator for yarrow pot plant production.

  • This experiment was carried out to study the effect of pure nitrogen fertilization on botanical composition of grassland in 1987~ 1988 under practical conditions at the " Federal Institute for Agriculture in the Alps" in Austria. The application rates were 0, 30, 60, 90 and 120kg/ha/cut, the cutting regimes 3-, 4 , 5- and 6-cuts/year. The results were as follows: 1. Only PK fertilization resulted in higher botanical composition of Trifolium repens among legumes in grassland, which was increased by cutting frequency .2. Nitrogen fertilization on three-cut areas resulted in higher existence of generally valuable tall grasses such as Arrhenotherum elatius Trisetum flavescens and Dactylis glomerata . On the other hand, nitrogen fertilization on four-, five- and six-cut areas showed mainly Doctylis glomerato and Poa pratensis appearance. 3.For some cases, appearance of less valuable grasses, herbs and weeds such as Agropyron repens, Poa trivialis, Poa annua, Setoria viridis, Aegopodium podagra rio, Men landrium rubrum, Taraxacum officlnale, Achillea millefolium, Rorippa sylvestris and Polygonum ocleulare was *Bundesanstalt fur alpenlandische Landwirtschaft Gumpenstein(A-8952 Irdning, 6sterreich) increased on medium and high rates of N fertilized areas. 4.Reduction of sward density may also diminish the advantages of nitrogen fertilization and may be threatened by mainly high dressings of nitrogen combined with too late utilization of the sward. 5.Location altered the efficiency of nitrogen fertilization on botanical composition.mposition.

Evaluation of Chemical Composition and In vitro Digestibility of Appennine Pasture Plants Using Yak (Bos grunniens) Rumen Fluid or Faecal Extract as Inoculum Source

  • Pastures of the Apennines of Central Italy contribute to feed resources of high altitude (above 1,300 m sea level) grazing systems. The objective of this study was to evaluate the effectiveness of faecal extract from the yak (Bos grunnienes) as an alternative microbial inoculum to rumen fluid for estimation of digestibility of several forage species. Forage samples produced at high altitude were tested in this study: four legumes (Lathyrus sativus L., Lotus corniculatus L., Onobrychis viciaefolia L. and Trifolium pratense L.), three forbs (Achillea millefolium L., Potentilla reptans L. and Teucrium flavum L.) and one grass (Brachipodyum pinnatum L.) were incubated with yak rumen fluid or faecal extract. A large variability in chemical composition was observed among the species collected. Rumen liquor and faecal samples were collected from adult healthy yak. The $Daisy^{II}$ incubator was used to evaluate the nutrient digestibility of forages using rumen liquor as control and faecal extract as alternative microbial inoculum sources. Filter bags containing samples of browse species were added to the four digestion vessels along with their respective inoculum and then incubated for 48 h and dry matter (DM), organic matter (OM), crude protein (CP), neutral and detergent fiber (NDF) digestibility was determined. There was a significant relationship between estimates, indicating that faecal liquor has the potential to be used instead of rumen fluid for estimation of in vitro digestibility of plants. It is concluded that the $Daisy^{II}$ incubator results are appropriate for the determination of in vitro digestibility of nutrients using faecal liquor to define the potential for adaptation of yak to new pastures.

The Flora of Vascular Plants in Eung-bong (Hwacheon-gun, Gangwon-do) (응봉 일대(강원도 화천군)의 관속식물상)

  • 본 연구에서는 강원도 화천군 응봉 일대의 중심으로 식물상을 조사하였다. 관속식물은 64과 162속 222종 3아종 15변종 2품종으로 총 242분류군으로 조사되었다. 분류군별로는 양치식물이 6과 10분류군, 나자식물 1과 2분류군, 쌍자엽식물이 52과 200분류군, 단자엽식물이 5과 30분류군이 확인되었다. 주요 식물로는 특산식물은 진범(Aconitum pseudolaeve), 무늬족도리풀(Asarum versicolor), 각시서덜취(Saussurea macrolepis) 등 10과 13분류군, 희귀식물은 관심대상종(Least Concern)인 홀아비바람꽃(Anemone koraiensis), 세잎종덩굴(Clematis koreana), 도깨비부채(Rodgersia podophylla) 등 14분류군, 취약종(Vuinerable)인 백작약(Paeonia japonica), 멱쇠채(Scorzonera austriaca subsp. glabra) 등 3분류군, 멸종위기종(Critically Endangered)인 닻꽃(Halenia corniculata) 등 2분류군으로 총 20분류군이 확인되었다. 귀화식물은 서양톱풀(Achillea millefolium), 미국가막사리(Bidens frondosa), 주걱개망초(Erigeron strigosus) 등 4과 11분류군이 조사되었다. 산림청에서 지정한 특별산림보호대상종으로는 왕벚나무(Prunus yedoensis)와 금강초롱꽃(Hanabusaya asiatica)이 조사되었다. 유용식물로는 용도를 모르는 52분류군(21.5%)을 제외하면 190분류군(78.5%)이 자원식물로 이용가치가 있는 것으로 나타났다. 이들을 용도에 따라 구분해 보면 식용 93분류군(38.4%), 섬유용 3분류군(1.2%), 약용 67분류군(27.7%), 관상용 45분류군(18.6%), 사료용 77분류군(31.8%), 산업용 1분류군(0.4%), 염료용 3분류군(1.2%), 목재용 12분류군(5.0%)으로 조사되었다.
