A potential role for fatty acid biosynthesis genes during molting and cuticle formation in Caenorhabditis elegans

  • Caenorhabditis elegans undergoes a developmental molting process that involves a coordinated interplay among diverse intracellular pathways. Here, we investigated the functions of two fatty acid biosynthesis genes; pod-2, encoding acetyl-CoA carboxylase, and fasn-1, encoding fatty acid synthase, in the C. elegans molting process. Although both the pod-2 and fasn-1 genes were expressed at constant levels throughout C. elegans development, knockdown of the proteins encoded by these genes using RNA interference produced severe defects in triglyceride production, molting, and reproduction that were coupled to suppression of NAS-37, a metalloprotease. An assessment of the structure and integrity of the cuticle using a COL-19::GFP marker and Hoechst 33258 staining showed that downregulation of either pod-2 or fasn-1 impaired cuticle formation and disrupted the integrity of the cuticle and the hypodermal membrane.

한약재 물 추출물이 간세포 Glucokinase, Pyruvate Dehydrogenase, Acetyl-CoA Carboxylase mRNA 발현에 미치는 영향 (Effects of medicinal herb water extracts on expression of hepatic glucokinase, pyruvate dehydrogenase and acetyl-CoA carboxylase mRNA)

  • 본 연구에서는 산수유, 목단피, 산약, 숙지황, 지골피, 야생배의 한약재 물 추출물이 당대사 관련 효소인 GCK, PDH, ACC mRNA 발현정도에 미치는 영향을 측정하였다. HepG2 세포에 대한 세포독성을 측정한 결과, GCK, PDH, ACC mRNA 발현량을 측정하기 위한 물 추출물의 농도 범위는 세포 생존율에 영향을 주지 않는 100, 250, 500 ppm로 결정하였다. GCK mRNA 발현은 100 ppm에서 숙지황 물 추출물이 165%로 가장 높게 나타났고, 250 ppm에서는 숙지황과 지골피 물 추출물이 각각 180%, 154%로 높았으며, 500 ppm에서는 산수유(195%), 목단피 (157%), 야생배 (139%), 산약 (122%), 지골피 (117%), 숙지황 (113%)의 순으로 GCK mRNA 발현이 증가되었다. PDH mRNA 발현량은 250 ppm 농도에서 지골피, 숙지황 물추출물에서 각각 141%, 118% 증가되었고, 500 ppm에서는 지골피, 숙지황 물 추출물에서 각각 191%, 124% 증가되었다. ACC mRNA 발현량은 500 ppm에서 지골피 (188%), 숙지황 (126%)로 가장 높게 나타났다. 결과적으로 GCK, PDH, ACC mRNA 발현량을 증가시킬 수 있는 소재로 산수유, 숙지황, 지골피 등을 꼽을 수 있겠으며 이들 소재들은 식후 혈당상승을 억제할 수 있는 항당뇨 천연소재로 이용될 수 있음을 제시하였다.

Biotinoyl Domain of Human Acetyl-CoA Carboxylase;Structural Insights into the Carboxyl Transfer Mechanism

  • Acetyl-CoA carboxylase (ACC) catalyzes the first step in fatty acid biosynthesis: the synthesis of malonyl-CoA from acetyl-CoA. As essential regulators of fatty acid biosynthesis and metabolism, ACCs are regarded as therapeutic targets for the treatment of metabolic diseases such as obesity, In ACC, the biotinoyl domain performs a critical function by transferring an activated carboxyl group from the biotin carboxylase domain to the carboxyl transferase domain, followed by carboxyl transfer to malonyl-CoA. Despite the intensive research on this enzyme, only the bacterial and yeast ACC structures are currently available, To explore the mechanism of ACC holoenzyme function, we determined the structure of the biotinoyl domain of human ACC2 and analyze its characteristics using NMR spectroscopy. The 3D structure of the hACC2 biotinoyl domain has a similar folding topology to the previously determined domains from E. coli and P. Shermanii, however, the 'thumb' structure is absent in the hACC2 biotinoyl domain. Observations of the NMR signals upon the biotinylation indicate that the biotin group of hACC2 does not affect the structure of the biotinoyl domain, while the biotin group for E. coli ACC interacts directly with the thumb residues that are not present in the hACC2 structure. These results imply that, in the E. coli ACC reaction, the biotin moiety carrying the carboxyl group from BC to CT can pause at the thumb of the BCCP domain. The human biotinoyl domain, however, lacks the thumb structure and does not have additional non-covalent interactions with the biotin moiety; thus, the flexible motion of the biotinylated lysine residue must underlie the "swinging arm" motion. This study provides insight into the mechanism of ACC holoenzyme function and supports the "swinging arm" model in human ACCs.

Anti-obesity Effect of Black Vinegar Fermented with Herbal Extracts

  • Vinegar has been widely produced for a variety of industrial and domestic use as well as medicinal use. For sale of the commercial vinegar with herbal extracts, we produced an experimental black vinegar through sequential fermentation of alcohol, followed by acetic acid according to the manufacturer's procedure. To investigate the effect of anti-obesity of black vinegar on biochemical values, we evaluated enzyme activities via acetyl-CoA carboxylase (ACC), which plays a critical role in the lipid metabolism. We found that increased phosphorylated adenosine monophosphate (AMP) activated protein kinase (AMPK) and ACC in L6 mouse muscle cells treated with the manufactured vinegar. Based on the results, supplementation of experimental herbal black vinegar inactivates ACC, enhancing the phosphorylation of AMPK. Thus, the lipid oxidation and inhibitory effect of fatty acid synthesis by the black vinegar expects to facilitate the anti-obesity activity.

신규 cyclohexanedione계 화합물 EK3143과 EK3150의 제초작용 발현특성과 Acetyl CoA Carboxylase 활성에 미치는 효과 (Action Characteristics of EK3143 and EK3150, New Cyclohexanedione Herbicides, and Their Effects on Acetyl CoA Carboxylase Activity)

  • 신규로 합성한 cyclohexanedione계 화합물 EK3143과 EK3150의 제초작용 발현의 특성을 기존약제와 비교조사하고, 이들이 ACCase에 대해 어떠한 저해활성을 가지는지를 알아보고자 제반실험을 수행하였다. Sethoxydim은 화본과식물에만 제초효과를 가지며 생육정지와 함께 조직의 황화, 괴사가 나타난다, 반면에 Ek3143과 EK3150은 화본과식물에 대한 활성은 상대적으로 감소되었으나 까마중, 어저귀 등의 광엽잡초에 대해서는 보다 강한 활성을 가졌다. EK3143은 저농도의 보다 폭넓은 범위에서는 백화증상이, 고농도에서는 sethoxydim과 동일한 증상을 나타내었고 EK3150은 모든 농도에서 식물체의 백화를 유기시켰으며 광엽잡초의 경우 분열조직에서 그 증상이 뚜렷하였다. 이들 화합물의 화본과식물에 대한 생육저해 활성과 옥수수 ACCase에 대한 저해정도간에는 일치된 경향을 보였으나 광엽잡초인 어저귀와 까마중에 대한 ACCase 활성과 전식물체(intact plant)에서의 제초활성간에는 일치된 경향을 보이지 않았다. 즉 EK3143과 EK3150은 어저귀와 까마중의 ACCase에 대한 저해활성이 sethoxydim보다 전반적으로 높은 특징을 보이지만, 광엽잡초에 대한 제초활성이 상대적으로 높았던 EK3150이 ACCase에 대해서는 EK3143보다 오히려 낮은 저해활성을 나타내었다. 한편 색소감소 정도가 신장저해보다 빨리 나타나는 norflurazon 및 EK3140 처리에서와는 달리, EK3143과 EK3150 처리에서는 색소감소 보다 신장저해가 보다 빠르게 나타나 다른 저해양상을 보였다. 이상의 결과로 보아 EK3143 EK3150은 ACCase 이외에 다른 작용점을 추가로 저해하는 것으로 생각되며, 약간의 구조변경에따라 제초작용점간의 결합정도에 차이가 생김으로써 생육저해 또는 백화증상이 서로 비중을 달리하여 나타나는 것으로 추정된다.

Effects of Dietary Gum Phospholipid on Lipid Metabolism in Broiler Chicks

  • This experiment was to determine the usefulness of gum safflower phospholipid as a feed ingredient. Forty female broiler chicks were divided into four groups and fed experimental diets containing following fats and oils; beef tallow (Tallow), the blend of safflower oil and palm oil (SP-oil), gum rapeseed phospholipid (Rap-PL), or gum safflower phospholipid (Saf-PL) for 21days. There were no differences in growth performances among the treatments. Abdominal fat weight tended to be reduced in the chicks fed. phospholipids. The activity of hepatic acetyl-CoA carboxylase was significantly reduced in the Rap-PL and Saf-PL as compared to that of Tallow. Feeding dietary phospholipids resulted in a slight reduction in total fat and triglyceride contents in the breast and thigh muscles. In addition, total fat and triglyceride contents in the thigh muscle were significantly decreased by dietary Saf-PL as compared to those of Tallow. These results suggested that dietary gum phospholipids, either from rapeseed or safflower, had desirable effects of lowing abdominal and muscle fats, and could be used as a feed ingredient for broiler diets.

Effect of Single Nucleotide Polymorphisms of Acetyl-CoA Carboxylase α(ACACA) Gene on Carcass Traits in Hanwoo (Korean Cattle)

  • Meat production and quality traits in beef cattle are largely affected by genetic factors. Acetyl-Coenzyme A carboxylase-${\alpha}$ (ACACA) plays a key role in the regulation and metabolism of fatty acid biosynthesis in mammalian animals. The gene encoding ACACA enzyme was chosen as a candidate gene for carcass and meat traits. In this study, we investigated effects of single nucleotide polymorphisms (SNPs) in the ACACA gene on beef carcass and meat traits in Hanwoo (Korean cattle) populations. We have sequenced a fragment of intron I region of the Hanwoo ACACA gene and identified two SNPs. Genotyping of the two SNP markers (g.2344T>C and g.2447C>A) was carried out using PCR-SSCP analysis in 309 Hanwoo steers to evaluate their association with carcass and meat production traits. The g.2344C SNP marker showed a significant increasing effect on LW (p = 0.009) and CW (p = 0.017). Animals with the CC genotype had higher CW and LW compared with TT and TC genotypes (p<0.05). The g.2447A SNP marker was associated with higher MC (p = 0.019). Animals with the AA genotype had higher MC than animals with CC and CA genotypes (p<0.05). Although the degree of linkage disequilibrium (LD) was not strong between g.2344T>C and g.2447C>A in the LD analysis, four major haplotype classes were formed with two SNP information within the ACACA gene. We constructed haplotypes using the HaploView software package program and analyzed association between haplotypes and carcass traits. The haplotype of ACACA gene significantly affected the LW (p = 0.027), CW (p = 0.041) and MC (p = 0.036). The effect of h1 haplotype on LW and CW was larger than that of h3 haplotype. Animals with the h1 haplotype also had greater MC than did animals with h2 haplotype. Consequently, the ACACA gene could be useful as a DNA marker for meat production traits such as carcass yield and meat contents in Hanwoo.
