Mixed organic acids improve nutrients digestibility, volatile fatty acids composition and intestinal microbiota in growing-finishing pigs fed high-fiber diet

  • Objective: The objective of this study was to investigate effects of mixed organic acids (MOA) on nutrient digestibility, volatile fatty acids composition and intestinal microbiota in growing-finishing pigs fed high wheat bran diet. Methods: Six crossbred barrows ($Duroc{\times}Landrace{\times}Yorkshire$), with an average body weight $78.8{\pm}4.21kg$, fitted with T-cannulas at the distal ileum, were allotted to a double $3{\times}3$ Latin square design with 3 periods and 3 diets. Each period consisted of a 5-d adjustment period followed by a 2-d total collection of feces and then a 2-d collection of ileal digesta. The dietary treatments included a corn-soybean-wheat bran basal diet (CTR), mixed organic acid 1 diet (MOA1; CTR+3,000 mg/kg OA1), mixed organic acid 2 diet (MOA2; CTR+2,000 mg/kg OA2). Results: Pigs fed MOA (MOA1 or MOA2) showed improved (p<0.05) apparent total tract digestibility (ATTD) of gross energy, dry matter and organic matter, and pigs fed MOA2 had increased (p<0.05) ATTD of neutral detergent fiber compared to CTR. Dietary MOA supplementation decreased (p<0.05) pH value, and improved (p<0.01) concentrations of lactic acid and total volatile fatty acids (TVFA) in ileum compared to CTR. Pigs fed MOA showed higher (p<0.05) concentration of acetic acid, and lower (p<0.05) content of formic acid in feces compared to CTR. Pigs fed MOA1 had increased (p<0.05) concentration of TVFA and butyric acid in feces. Pigs fed MOA1 showed higher concentration of Lactobacillus and lower concentration of Escherichia in feces compared to CTR. Conclusion: Dietary supplementation of MOA 1 or 2 could improve nutrients digestibility, TVFA concentration and intestinal flora in growing-finishing pigs fed high fiber diet.

Identification and Characterization of Microbial Community in the Coelomic Fluid of Earthworm (Aporrectodea molleri)

  • Earthworms play an important role in soil fertilization, interacting continually with microorganisms. This study aims to demonstrate the existence of beneficial microorganisms living in the earthworm's immune system, the coelomic fluid. To achieve this goal, a molecular identification technique was performed, using cytochrome c oxidase I (COI) barcoding to identify abundant endogenic earthworms inhabiting the temperate zone of Rabat, Morocco. Then, 16S rDNA and ITS sequencing techniques were adopted for bacteria and fungi, respectively. Biochemical analysis, showed the ability of bacteria to produce characteristic enzymes and utilize substrates. Qualitative screening of plant growth-promoting traits, including nitrogen fixation, phosphate and potassium solubilization, and indole acetic acid (IAA) production, was also performed. The result of mitochondrial COI barcoding allowed the identification of the earthworm species Aporrectodea molleri. Phenotypic and genotypic studies of the sixteen isolated bacteria and the two isolated fungi showed that they belong to the Pseudomonas, Aeromonas, Bacillus, Buttiauxella, Enterobacter, Pantoea, and Raoultella, and the Penicillium genera, respectively. Most of the isolated bacteria in the coelomic fluid showed the ability to produce β-glucosidase, β-glucosaminidase, Glutamyl-β-naphthylamidase, and aminopeptidase enzymes, utilizing substrates like aliphatic thiol, sorbitol, and fatty acid ester. Furthermore, three bacteria were able to fix nitrogen, solubilize phosphate and potassium, and produce IAA. This initial study demonstrated that despite the immune property of earthworms' coelomic fluid, it harbors beneficial microorganisms. Thus, the presence of resistant microorganisms in the earthworm's immune system highlights a possible selection process at the coelomic fluid level.

Changes in fermentation pattern and quality of Italian ryegrass (Lolium multiflorum Lam.) silage by wilting and inoculant treatments

  • Objective: This study was conducted to investigate the effects of wilting and microbial inoculant treatment on the fermentation pattern and quality of Italian ryegrass silage. Methods: Italian ryegrass was harvested at heading stage and ensiled into vinyl bags (20 cm×30 cm) for 60d. Italian ryegrass was ensiled with 4 treatments (NWNA, no-wilting noadditive; NWA, no-wilting with additive; WNA, wilting no-additive; WA, wilting with additive) in 3 replications, wilting time was 5 hours and additives were treated with 106 cfu/g of Lactobacillus plantarum. The silages samples were collected at 1, 2, 3, 5, 10, 20, 30, 45, and 60 days after ensiling and analyzed for the ensiling quality and characteristics of fermentation patterns. Results: Wilting treatment resulted in lower crude protein and in vitro dry matter digestibility and there were no significant differences in acid detergent fiber (ADF), total digestible nutrient (TDN), water-soluble carbohydrate (WSC), ammonia content, and pH (p>0.05). However, wilting treatment resulted in higher ADF and neutral detergent fiber content of Italian ryegrass silage (p<0.05), and the WNA treatment showed the lowest TDN and in vitro dry matter digestibility. The pH of the silage was higher in the wilting group (WNA and WA) and lower in the additive treatment group. Meanwhile, the decrease in pH occurred sharply between the 3-5th day of storage. The ammonia nitrogen content was significantly lower in the additive treatment (p<0.05), and wilting had no effect. As fermentation progressed, the lactic and acetic acid contents were increased and showed the highest content at 30 days of storage. Conclusion: The wilting treatment did not significantly improve the silage fermentation, but the inoculant treatment improved the fermentation patterns and quality of the silage. So, inoculation before ensiling is recommended when preparing high quality of Italian ryegrass silage, and when wilting, it is recommended to combine inoculation for making high quality silage.

액아배양을 통한 야생 백하수오(Cynanchum wilfordii)의 기내증식 (In Vitro Propagation of Wild Cynanchum wilfordii Through Axillary Bud Culture)

  • 본 연구는 야생 백하수오를 대상으로 기내증식의 최적 조건을 구명하기 위해 수행되었다. 기내 종자발아를 위하여 야생 백하수오 종자를 $GA_{3}$ 용액(100 ppm)에 24시간 침지처리 후 1.0 mg/L BA가 첨가된 WPM 배지에 치상하였을 때 91.6%의 발아율을 보였다. 신초 증식은 1.0 mg/L $GA_{3}$가 첨가된 배지에서 최대 5.2 cm까지 신초가 신장하였으나 비정상적으로 생장하여 결국 고사하였고, 신초를 0.1 mg/L BA가 첨가된 배지에 치상 4 주 후 2.4개의 신초를 획득하였다. 발근은 식물생장조절제(Plant Growth Regulators, PGRs)를 첨가하지 않은 환기처리 배지에 치상하였을 때 3개의 뿌리가 분화되었고, 0.1 mg/L NAA가 첨가된 환기처리 배지에 치상하였을 때 2 cm로 가장 높게 유도되었다. 기외이식 후 생존율은 기내에서 환기 전처리한 개체군이 70%로 가장 높게 나타났다.

Effects of formic acid and lactic acid bacteria inoculant on main summer crop silages in Korea

  • To improve the fermentation quality of silage and reduce the nutrients loss of raw materials during the ensiling process, silage additives are widely used. The effect of additives on silage is also affected by the species of crop. Therefore, this study was designed to explore the effects of formic acid (FA) and lactic acid bacterial inoculant on the quality of main summer crop silage. The experiment was consisted on split-plot design with three replications. The experiment used the main summer forage crops of proso millet ("Geumsilchal"), silage corn ("Gwangpyeongok"), and a sorghum-sudangrass hybrid ("Turbo-gold"). Treatments included silage with Lactic acid bacterial Inoculant (Lactobacillus plantarum [LP], 1.0 × 106 CFU/g fresh matter), with FA (98%, 5 mL/kg), and a control (C, without additive). All silages were stored for 60 days after preparation. All additives significantly increased the crude protein content and in vitro dry matter digestibility (IVDMD) of the silages and also reduced the content of ammonia nitrogen (NH3-N) and pH. Corn had the highest content of IVDMD, total digestible nutrients and relative feed value among silages. Compared with the control, irrespective of whether FA or LP was added, the water soluble carbohydrate (WSC) of three crops was largely preserved and the WSC content in the proso millet treated with FA was the highest. The treatment of LP significantly increased the lactic acid content of the all silage, while the use of FA significantly increased the content of acetic acid (p < 0.05). The highest count of lactic acid bacteria (LAB) was detected in the LP treatment of corn. In all FA treatment groups, the total microorganism and mold numbers were significantly lower than those of the control and LP groups (p < 0.05). In conclusion, both additives improved the fermentation quality and nutritional composition of the main summer forage crops. The application of FA effectively inhibited the fermentation of the three crops, whereas LAB promoted fermentation. So, both FA and LP can improve the quality of various species of silage.

식물에 의한 납, 카드뮴 흡수 기작에 미치는 자유이온 및 유기산-중금속 복합체의 영향 (Effects of free metal ions and organo-metal complexes on the absorption of lead and cadmium by plants)

  • 토양 중 중금속은 자유이온, 유기산-중금속 복합체를 포함한 다양한 화학종으로 존재한다. 이 중 중금속 자유이온 비율은 식물체의 중금속 흡수와 밀접한 관련이 있는 것으로 알려져 있다. 본 연구는 중금속 자유이온과 유기산-중금속 복합체 존재 비율이 식물의 납, 카드뮴 흡수에 어떤 영향을 미치는지 알아보기 위하여 수행되었다. 이를 위해 순환식 양액재배 장치에서 28일간 키운 상추를 48시간 동안 납과 카드뮴 용액에 침지시키는 실험을 진행하였다. 침지 전 시트르산, 옥살산, 아세트산, 휴믹산을 이용해 납과 카드뮴 용액의 중금속 자유이온과 유기산-중금속 복합체의 비율이 100:0, 90:10, 70:30, 60:40이 되도록 조절하였다. 침지 후 상추 뿌리와 잎의 납, 카드뮴 농도를 분석한 결과, 용액 중 중금속 자유이온의 비율과 식물이 흡수한 납, 카드뮴 농도는 상관관계를 보이지 않았다. 반면에 처리한 유기산의 종류와 중금속 종류에 따른 차이가 더 큰 것으로 나타났다. 시트르산은 납 흡수는 증가시켰지만 카드뮴 흡수는 감소시켰다. 시트르산은 다른 유기산보다 상추의 납 흡수율을 높였는데 이는 시트르산이 분자량이 크기 때문에 중금속 이온과 복합체를 형성할 수 있는 용량이 더 크기 때문인 것으로 보인다. 그러므로 본 연구 결과는 식물에 의한 중금속 흡수가 단순히 자유이온의 양에 따라 달라지기 보다는 존재하는 유기산의 분자량에 의한 결합용량 차이가 중금속 종류에 따라 달리 작용하여 결정된다는 것을 보여준다.

산처리에 의한 적양파 분말의 quercetin 배당체와 aglycone의 농도변화 (Acid treatment effects on the contents of quercetin glycosides and aglycone in red onion powder)

  • 열풍건조로 제조된 껍질을 포함한 적양파 분말은 강력한 항산화제인 플라보노이드 함량이 생양파즙에 비해 약 22배로 고농도로 농축되어 있으며, 60-70% ethanol 농도에서 70℃, 2시간 추출시 가장 높은 수율을 나타내었다. 이때 추출된 플라보노이드의 함량은 DPPH radical 소거능과 상관계수 0.877의 높은 상관관계를 나타내었다. 적양파 분말의 플라보이드는 주로 quercetin 배당체인 Q3,4'G, Q4'G와 QA로 이루어져 있으며, 15:39:47의 비율로 구성되어 있었다. 염산, 젖산, 초산 등 다양한 종류의 산처리에 의해 적양파 분말 속 quercetin 배당체는 QA로 전환될 수 있었으며 저농도 초산용액에서 QA로 전환되는 비율이 높았다. 이는 적양파 분말에 포함된 glucosidase가 저농도 산용액에서 활성화되면서 일어나는 반응으로 사료되었다. 초산 처리초기에는 diglycoside인 Q3,4'G의 de-glycosylation이 급격히 일어났으며, 초산처리 6시간 이후에는 Q4'G의 분해와 함께 QA가 증가하여 24시간 경과 후에 최대 QA의 함량에 도달하였으며, QA의 증가는 DPPH radical 소거능과 유의적인 상관성(r=0.90)을 나타내어 생리활성의 증가를 나타내었다. 본 연구결과, 양파의 주요 플라보노이드인 quercetin 배당체는 저농도 초산 처리로 빠르고 간편하게 QA로 전환이 가능하였으며, 이와 더불어 생리활성의 증가도 도모할 수 있었다. 적양파 분말을 이용하여 저농도 산처리를 통하여 QA로의 전환률을 높이면, 고농도의 quercetin을 함유한 양파 소재의 개발이 가능할 것이며, 이를 통해 기능성 양파 가공품의 개발로 이어질 수 있을 것이다.

복숭아 전통발효식초 원료로서의 복숭아즙 유용성 연구 (A study on the Usefulness of Peach-juice as a Raw Material for Traditional Fermented Peach Vinegar)

  • 복숭아의 부가가치를 높이고 과잉 생산된 복숭아를 활용하기 위해서는 다양한 형태로 복숭아 원료를 개발할 필요가 있다. 본 연구에서는 4종류 즙(생과원액, 냉장생즙, 중탕즙, 냉동퓨레)으로 제조한 4종류 전통발효식초의 특성을 비교·분석하여 식초 원료로서 즙의 활용가치를 알아보았다. 본 연구에서 제조한 4종류 발효식초의 당도(6.20~6.50°Brix), 총산도(5.25~5.61%), 산도pH(3.42~3.74) 및 초산 함량(48.81~54.29 mg/mL)은 기존 전통발효식초들과 유사하였다. 그러나 총페놀 및 플라보노이드 함량은 냉장생즙 및 중탕즙으로 제조한 식초가 월등히 높은 것으로 나타났다. 따라서 복숭아 전통발효식초를 제조하기 위한 원료로 중탕즙 및 냉장생즙이 매우 적합하며, 특히 페놀 및 플라보노이드 함량이 높은 중탕즙은 고부가가치 기능성 식품 소재로도 활용 가능함을 확인하였다. 본 연구결과는 고부가가치 복숭아 원료를 개발하는데 중요한 지침이 될 것으로 사료된다.

Optimization of an Industrial Medium and Culture Conditions for Probiotic Weissella cibaria JW15 Biomass Using the Plackett-Burman Design and Response Surface Methodology

  • The objective of this study was to optimize industrial-grade media for improving the biomass production of Weissella cibaria JW15 (JW15) using a statistical approach. Eleven variables comprising three carbon sources (glucose, fructose, and sucrose), three nitrogen sources (protease peptone, yeast extract, and soy peptone), and five mineral sources (K2HPO4, potassium citrate, ⳑ-cysteine phosphate, MgSO4, and MnSO4) were screened by using the Plackett-Burman design. Consequently, glucose, sucrose, and soy peptone were used as significant variables in response surface methodology (RSM). The composition of the optimal medium (OM) was 22.35 g/l glucose, 15.57 g/l sucrose, and 10.05 g/l soy peptone, 2.0 g/l K2HPO4, 5.0 g/l sodium acetate, 0.1 g/l MgSO4·7H2O, 0.05 g/l MnSO4·H2O, and 1.0 g/l Tween 80. The OM significantly improved the biomass production of JW15 over an established commercial medium (MRS). After fermenting OM, the dry cell weight of JW15 was 4.89 g/l, which was comparable to the predicted value (4.77 g/l), and 1.67 times higher than that of the MRS medium (3.02 g/l). Correspondingly, JW15 showed a rapid and increased production of lactic and acetic acid in the OM. To perform a scale-up validation, batch fermentation was executed in a 5-l bioreactor at 37℃ with or without a pH control at 6.0 ± 0.1. The biomass production of JW15 significantly improved (1.98 times higher) under the pH control, and the cost of OM was reduced by two-thirds compared to that in the MRS medium. In conclusion, OM may be utilized for mass producing JW15 for industrial use.

Efficacy of combination of endo-xylanase and xylan-debranching enzymes in improving cereal bran utilization in piglet diet

  • Objective: This study was aimed to explore the efficacy of combination of endo-xylanase (Xyn) and xylan-debranching enzymes (arabinofuranosidase, Afd and feruloyl esterase, FE) in improving utilization of bran in piglet diet. Methods: In vitro experiments were firstly conducted to examine the enzymological properties of Xyn, Afd, and FE, concurrent with their effect on degradation of arabinoxylan (Abx) in bran. In vivo experiment was then implemented by allocating two hundred and seventy 35-d-old postweaning piglets into 3 groups (6 replicates/group), which received bran-containing diet supplemented with Xyn (1,600 U/kg) or its combination with Afd (0.8 U/kg) and FE (4 U/kg) or without enzyme. Results: Both Xyn, Afd, and FE are relatively stable against the changes in temperature and pH value. Combining Xyn with Afd and FE had a superiority (p<0.05) over Xyn alone and its combination with Afd or FE in promoting (p<0.05) degradation of Abx in different brans. Combined treatment with Xyn, Afd, and FE was more beneficial than Xyn alone to induce increasing trends (p<0.10) of average daily gain, final body weight and feed efficiency of piglets fed bran-containing diet. Moreover, combination of Xyn, Afd, and FE showed advantages (p<0.05) over Xyn alone in causing reductions (p<0.05) in diarrhea rate and cecal pH value, concurrent with increases (p<0.05) in cecal and colonic acetic acid and total volatile fatty acid concentrations, as well as cecal butyric acid concentration of piglets fed bran-containing diet. Conclusion: Combining Xyn with Afd and FE was more beneficial than Xyn alone in promoting degradation of Abx in bran, along with growth performance and intestinal volatile fatty acid profile of piglets received bran-containing diet. Thereby, combination of Xyn, Afd, and FE had a superior efficacy relative to Xyn alone in improving application of cereal bran in piglet diet.
