Polymeric Acetate-Selective Electrodes Based on meso-(α,α,α,α)-Tetrakis-[(2-arylphenylurea)phenyl]porphyrins: Electormic and pH Effects

  • Polymeric membrane electrodes for acetate anion based on meso-(${\alpha}$,${\alpha}$,${\alpha}$,${\alpha}$)-5,10,15,20-tetrakis[2-(penta-fluorophenylurea) phenyl]porphyrin I and similar urea-functionalized porphyrins Ⅱ-Ⅳ as neutral ionophores were prepared. The membrane based on porphyrin I exhibits the best potentiometric properties in pH 6.0 rather than pH 7.0: linear stable response over a wide concentration range (6.0 ${\times}$$10^{-5}$-1.0 ${\times}$$10^{-2}$) with a slope of -59.6 mV/decade and a detection limit of log[CH3CO$O^-$] = -5.32. Selectivity coefficients obtained from the matched potential method (MPM) in pH 6.0 indicate that interferences of hydrophobic anions are very small for the membranes of porphyrins I and II having the strong withdrawing group. The electronic effect of urea-functionalized porphyrins and pH effect of buffer solutions are discussed on the potentiometric response.

Improvement of Radiosynthesis Yield of [11C]acetate ([11C]아세트산의 방사화학적 수율 증가를 위한 연구)

  • Purpose $[^{11}C]$acetate has been proved useful in detecting the myocardial oxygen metabolism and various malignancies including prostate cancer, hepatocellular carcinoma, renal cell carcinoma and brain tumors. The purpose of study was to improve the radiosynthesis yield of $[^{11}C]$acetate on a automated radiosynthesis module. Materials and Methods $[^{11}C]$acetate was prepared by carboxylation of grignard reagent, methylmagnesium chloride, with $[^{11}C]$$CO_2$ gas, followed by hydrolysis with 1 mM acetic acid and purification using solid phase extraction cartridges. The effect of the reaction temperature ($0^{\circ}C$, $10^{\circ}C$, $-55^{\circ}C$) and cyclotron beam time (10 min, 15 min, 20 min, 25 min) on the radiosynthesis yield were investigated in the $[^{11}C]$acetate labeling reaction. Results The maximum radiosynthesis yield was obtained at $-10^{\circ}C$ of reaction temperature. The radioactivities of $[^{11}C]$acetate acquired at $-10^{\circ}C$ reaction temperature was 2.4 times higher than those of $[^{11}C]$acetate acquired at $-55^{\circ}C$. Radiosynthesis yield of $[^{11}C]$acetate increased with increasing cyclotron beam time. Conclusion This study shows that radiosynthesis yield of $[^{11}C]$acetate highly dependent on reaction temperature. The best radiosynthesis yield was obtained in reaction of grignard reagent with $[^{11}C]$$CO_2$ at $-10^{\circ}C$. This radiolabeling conditions will be ideal for routine clinical application.

Rumen fermentation, methane production, and microbial composition following in vitro evaluation of red ginseng byproduct as a protein source

  • The main objective of this in vitro study was to evaluate red ginseng byproduct (RGP) as a protein resource and its effects on rumen fermentation characteristics, microflora, CO2, and CH4 production in ruminants. Four treatments for in vitro fermentation using buffered rumen fluid over a 48 h incubation period were used: 1, RGP; 2, corn gluten feed (CGF); 3, wheat gluten (WG); and 4, corn germ meal. In vitro dry matter digestibility (IVDMD), in vitro neutral detergent fiber digestibility (IVNDFD), in vitro crude protein digestibility (IVCPD), volatile fatty acids, pH, and ammonia nitrogen (NH3-N) were estimated after 48 h incubation. Gas production was investigated after 3, 6, 12, 24, 36 and 48 h. The CO2 and CH4 were evaluated after 12, 24, 36, and 48 h. A significant difference in total gas production and CO2 emissions was observed (p < 0.01) at all incubation times. CH4 production in RGP were higher (p < 0.05) than that in other treatments but a higher CH4 portion in the total gas production was observed in WG (p < 0.05) at 48 h incubation. The IVDMD, IVNDFD, and IVCPD of RGP was lower than those of other conventional ingredients (p < 0.01). The RGP had the lowest NH3-N value among the treatments (p < 0.01). The RGP also had the lowest total VFA concentration (p < 0.01), but presented the highest acetate proportion and acetate to propionate ratio among the treatments (both, p < 0.01). The abundance of Prevotella ruminicola was higher in RGP than in WG (p < 0.01), whereas RGP has lower methanogenic archaea (p < 0.01). In conclusion, based on the nutritive value, IVDMD, low NH3-N, and decreased methanogenic archaea, RGP inclusion as a protein source in ruminant diets can be an option in replacing conventional feed sources.

Study on the production of porous CuO/MnO2 using the mix proportioning method and their properties (반응몰비에 따른 다공성 CuO/MnO2의 제조 및 특성 연구)

  • In this study, the porous CuO/MnO2 catalyst was prepared through the co-precipitation process from an aqueous solution of potassium permanganate (KMnO4), manganese(II) acetate (Mn(CH3COO)2·4H2O) and copper(II) acetate (Cu(CH3COO)2·H2O). The phase change in MnO2 was analyzed according to the reaction molar ratio of KMnO4 to Mn(CH3COO)2. The reaction mole ratio of KMnO4 to Mn(CH3COO)2·4H2O was varied at 0.3:1, 0.6:1, and 1:1. The aqueous solution of Cu(CH3COO)2 was injected into a mixed solution of KMnO4 and Mn(CH3COO)2 to 10~75 wt% relative to MnO2. The Cu ion co-precipitates as CuO with MnO2 in a highly dispersed state on MnO2. The physicochemical property of the prepared CuO/MnO2 was analyzed by using the TGA, DSC, XRD, SEM, and BET. The different phase types of MnO2 were prepared according to the reaction mole ratio of KMnO4 to Mn(CH3COO)2·4H2O. The results confirmed that the porous CuO/MnO2 catalyst with γ-phase MnO2 was produced in the reaction mole ratio of KMnO4 to Mn(CH3COO)2 as 0.6:1 at room temperature.

The Effect of Vegetable Sources Supplementation on In vitro Ruminal Methane Gas Production (식물원료 첨가가 In vitro 반추위 메탄가스 발생에 미치는 영향)

  • The researchers have tried to reduce ruminal methane gas ($CH_4$) and to convert it into beneficial nutrient for several decades. This study was conducted to screen the methane-reducing vegetables among lettuce, hot pepper, spring onion, onion, turmeric, sesame leaf, garlic, radish sprout, leek and ginger nutritiously on the in vitro ruminal fermentation. The heat-treated vegetables at the 10% of substrate (timothy) were used to reduce methane production on the in vitro anaerobic experiment of 0, 6, 12, 24 and 48 h incubation time. Total gas production, pH, ammonia, $H_2$, $CO_2$, $CH_4$, and volatile fatty acid (VFA) were measured as indicators of in vitro fermentation product containing methane gas. All treatments except garlic showed a tendency to increase in total gas production. The result of ammonia showed that garlic and hot pepper affected rumen bacteria concerned protein metabolism and that lettuce and spring onion increased ammonia production. Garlic decreased $CH_4$ production in inverse proportion to $H_2$. Lettuce, spring onion, onion, garlic, radish sprout, leek and ginger increased propionate of VFA. Garlic balanced the ruminal fermentation in the pH, $H_2$, $CH_4$, acetate and propionate. This results showed that methane production at in vitro study was inhibited by heat-treated garlic supplementation. In conclusion, this study suggests that ruminal fermentation covering methane production might be controled by proper vegetables.

The Microstructure of Magnetite Coated on Honeycomb and Characteristics of CO2 Decomposition (허니컴에 코팅한 마그네타이트의 미세구조 및 CO2 분해특성)

  • In this study, we fabricated magnetite coated on a cordierite honeycomb which has complex shape by ultrasound-enhanced ferrite plating. The effects of the plating condition on the formation of the magnetite and its microstructure were investigated. The magnetite coated on the honeycomb became an oxygen-deficient ferrite by H$_2$ gas reduction, then the effects of the molar concentrations of ammonium acetate for $CO_2$ gas decomposition have been studied. As the molar concentration of a pH buffer($CH_3$COONH$_4$, 0.1946∼0.3892 M) solution increased, the average particle size increased about 200∼250 nm. The magnetite coated on the honeycomb was reduced by H$_2$ gas for 2 h at 30$0^{\circ}C$. The inner pressure change in the cell began to occur at 315∼34$0^{\circ}C$. The H$_2$-Reduced magnetite coated on the honeycomb at 35$0^{\circ}C$ contained an oxygen deficient magnetite and $\alpha$-Fe phase. The thermogravimetric analysis with H$_2$ reduction and $CO_2$ decomposition were carried out with the magnetite coated on the honeycomb. A weight loss in process of H$_2$ reduction occurred between 32$0^{\circ}C$ and 34$0^{\circ}C$, while a weight gain was observed during the $CO_2$ decomposition.

  • The crude calcium acetate (CCA) was made from egg-shell and acetic acid to use the CCA as a coagulant for Tofu (soy bean curd). Amount of CCA. $CO_2$ and residue made from 5.6g egg-shell (average weight of a egg-shell) and 10% acetic acid(50m1) was 3.8g, $300{\pm}10ml(at\;25^{\circ}$ and 2.1g. respectively. Tofu formable minimum concentration of the CCA was about 0.14% similar to that of $CaCl_2$, $MgCl_2$. Volume, hardness, taste and content f protein and lipid of Tofu by addition of CCA were not significantly different in those of Tofu that magnesium chloride and calcium chloride were used.

Removal of Cesium and Cobalt within Soil around TRIGA Reactor by Electrokinetic method (동전기적방법을 이용한 TRIGA 연구로 주변 토양내의 세습과 코발트 제거)

  • The characteristics of cesium and cobalt removal from soil around the TRIGA reactor using the electrokinetic method were analyzed and a device to restrain the pH increase in the soil column was suggested. When a NaCl solution was used as the electrolyte to raise the electric field strength, a precipitate was formed in the cathode in the soil column, resulting in a low removal efficiency. Thus, an acetate buffer solution (compound solution of $CH_3COONa$ and $CH_3COOH$) was injected into the soil column and acetic acid was periodically infected into the cathode reservoir to restrain any pH increase. Many $Cs^{2+}$ and $Co^{2+}$ ions were transferred by electromigration rather than electroosmosis during the initial remediation period, and no precipitate was formed in the soil column. 96% of the total amount of cesium in the soil column was removed after 5.9 days, while 94% of the total amount of cobalt was removed. Furthermore, the residual concentrations predicted by the developed model were similar to those obtained by experiment.

Effects of Defaunation on Fermentation Characteristics, Degradation of Ryegrass Hay and Methane Production by Rumen Microbes In Vitro When Incubated with Plant Oils

  • This study was conducted to examine the effects of defaunation (removal of live protozoa) on fermentation characteristics, degradation of ryegrass hay and $CH_4$ (methane) production by rumen microbes when incubated with plant oils (SO, sunflower oil and LO, linseed oil) in vitro. Sodium lauryl sulfate (0.000375 g/ml) as a defaunation reagent was added into the culture solution and incubated anaerobically up to 24 h at $39^{\circ}C$. pH from defaunation was increased for all treatments from 6 h incubation times (p<0.01-0.001) compared with those from fauantion. Concentration of ammonia-N from defaunation is higher than that from faunation at 3 h (p<0.001), 12 h (p<0.05) and 24 h (p<0.001) incubation times. Defaunation decreased (p<0.01-0.001) total volatile fatty acid concentration at all incubation times. Molar proportions of $C_2$ (acetate, p<0.05-0.001) and butyrate (p<0.01-0.001) were also decreased by defaunation at all incubation times. Molar proportion of $C_3$ (propionate), however, was increased by defaunation at all incubation times (p<0.001). Thus the rate of $C_2$ to $C_3$ was decreased by defaunation at all incubation times (p<0.001). Defaunation decreased ED (effective degradability) of dry matter (p<0.001) and ED of neutral detergent fiber (p<0.001) of ryegrass hay. Defaunation decreased total gas, $CH_4$ production, $CH_4$ % in total gas and $CH_4/CO_2$ at all incubation times (p<0.001). Oil supplementation decreased total gas (p<0.05-0.001), $CH_4$ production (p<0.001) and $CH_4$ % in total gas (p<0.001) compared with control at all incubation times. The result of this study showed that defaunation combined with oil supplementation may cause an alteration of microbial communities and further medicate the fermentation pattern, resulting in both reduction of degradation of ryegrass hay and $CH_4$ production. No difference, however, was observed in all the examinations between SO and LO.

Acidification of Pig Slurry with Sugar for Reducing Methane Emission during Storage (메탄 배출 저감을 위한 설탕을 이용한 돈 슬러리의 산성화)

  • The major problem encountered during the storage of pig slurry (PS) is the release of huge amounts of greenhouse gases (GHGs), which are dominated by methane ($CH_4$). To reduce this, concentrated sulfuric acid has been used as an additive to control the pH of pig slurry to 5.0-6.0. However, other low-risk substitutes have been developed due to some limitations to its use, such as corrosiveness, and hazards to animal and human health. In this study, sugar addition was proposed as an eco-friendly approach for limiting $CH_4$ emission from PS during storage. The pH of PS has been reduced from $7.1{\pm}0.1$ (control) to $5.8{\pm}0.1$, $4.6{\pm}0.1$, $4.4{\pm}0.1$, $4.1{\pm}0.1$, and $4.0{\pm}0.1$, by the addition of 10, 20, 30, 40, and 50 g sugar/L, respectively. Lactate, acetate, and propionate were detected as the dominant organic acids and at sugar concentration above 20 g/L, lactate concentration represented 42-72% (COD basis) of total organic acids. For 40 d of storage, $20.6{\pm}2.3kg\;CO_2\;eq./ton\;PS$ was emitted in the control. Such emission, however, was found to be reduced to $8.7{\pm}0.4$ and $0.4{\pm}0.1kg\;CO_2\;eq./ton\;PS$ at 10 and 20 g/L, respectively. Small amount of $CH_4$ from PS at 10 g/L was emitted until 30 d of storage, while for rest of storage period, it has increased to $8.7{\pm}0.4kg\;CO_2\;eq./ton\;PS$ ( 40% of the control) when methanogens have recovered by increasing pH to 7.0. By the end of storage, VS and COD removal in the control reached 24% and 27%, while their ranges reached 15-4% and 12-17% in the sugar added experiments, respectively. It was found that more than 90% of COD removal was done by aerobic biological process.
