Effects of Egg Yolk from Hens Fed with Acanthopanax senticosus Extract on Physical Endurance and Reproductive Parameters in Rats

  • Acanthopanax senticosus, a member of the Araliaceae used as an invigorant in traditional Korean medicine, is known to relieve physical fatigue. The principal objective of this study was to evaluate the effects of eggs laid by hens fed on Acanthopanax senticosus extracts on physical endurance and reproductive parameters. Male Sprague-Dawley rats at 4 wk of age were divided into 3 groups of 15 rats each: group A (14% egg yolk powder from hens fed with Acanthopanax senticosus extracts+86% AIN-93G), group B (14% plain egg yolk powder + 86% AIN-93G) and group C (7% fat source + 93% AIN-93G), and studied for 5 wk. The rats' physical endurance was measured via forced swimming tests. According to the results, 53.3% rats of group A swam for longer than 20 min, in group B and C, only 48.9% and 46.7% of the rats achieved this. In terms of reproductive parameters, sperm motility was significantly higher in group A than in groups B and C (p<0.05). However, no differences in sperm count were detected among the groups. Additionally, the serum testosterone levels of groups A and B were higher than that of group C. Our findings suggest that rats fed with egg yolk powder from hens fed on Acanthopanax senticosus extracts may improve athletic endurance and reproductive parameters in rats.

Effects of Onion Extract and Onion-Acanthopanax Senticosus Mixture Extracts on Obese Rats (양파와 가시오가피 혼합 추출액이 비만 흰쥐에 미치는 영향)

  • The aim of this study is to investigate the effects of onion and acanthopanax senticosus on body weight change, serum total cholesterol, HDL cholesterol, LDL cholesterol, triglyceride, free fatty acid, total lipid, phospholipid level, renal and liver function test, and CBC in obese rats and mice. Obese rats induced by high-fat fed are medicated for 7 weeks. Rats are divided into four groups depending on the medication; normal group (general-fat fed and no-medication), control group (high-fat fed and no medication), sample A group (high-fat fed and onion 100% extracted medication), sample B group (high-fat fed and onion 50% & acanthopanax senticosus 50% extracted medication), sample C group (high-fat fed and red onion 50% & acanthopanax senticosus 50% extracted medication). After medication, obesity related index, renal and liver function test, and CBC are analysed. There are significant statistical differences among control group and all experimental groups for the body weight change. There are significant statistical differences among control groups and all experimental groups for the total cholesterol, HDL cholesterol, LDL cholesterol, triglyceride level, free fatty acid, and phospholipid level. These results suggest that medications of onion and acanthopanax senticosus extracted products are effective for the treatment of obesity. Especially, onion 100% extracted product is more effective than the others.

Comarison of Major Constituents in Acanthopanax Taxa and Variety Cheongsong in Korea by GC-MS (GC-MS에 의한 오갈피나무 분류군과 청송 변종의 주요 성분 비교)

  • Species of genus Acanthopanax(Araliaceae) are long-lived trees primarily distributed throughout East Asia. These species are regarded as medically and ecologically important in Korea. A variety of Cheongsong in Korea is one of these cultivated varieties, however this variety is much longer(>100 years) than those of other cultivated groups. The components of variety of Acanthopanax in Cheongsong were analyzed for the first time and were compared to those of all Acanthopanax taxa in Korea. Nineteen components were specific to variety in Cheongsong. The main components of this variety were $\beta$-caryophyllene, hexadecanoic acid and ethyl stearate. Although some components are differ from each other, variety Cheongsong was similar to A. senticosus at phonetic topology with content of the chemicals, In addition, six species of genus Acanthopanax were investigated to compare the major chemical components by GC-MS.

A study on the effect comparative of acanthopanax stem bark (ASB) and acanthopanax root bark (ARB) on the monosodium iodoacetate (MIA)-induced osteoarthritis rats (오가피(五加皮) 수피(樹皮)와 근피(根皮)의 MIA 유도 골관절염 흰쥐에 미치는 영향 비교)

  • Objectives : This study was designed to compare the effects of acanthopanax stem bark (ASB) and acanthopanax root bark (ARB) on the monosodium iodoacetate (MIA)-induced osteoarthritis rats. Methods : The antioxidant activities were evaluated through radical scavenging assays using 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radicals and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals. Also, we examined total poly phenol and flavonoids contents. Osteoarthritis was caused by injection MIA($50{\mu}{\ell}$ with $80mg/m{\ell}$) into the knee joint cavity of rats. Rats were divided by 4 groups (normal group, control group, ASB treated group, ARB treated group, each n=6). The changes in the levels of reactive oxygen species (ROS), alanine aminotransferase (ALT) and aspartate aminotransferase (AST) in serum were analyzed after experiment. Also, the anti-oxidant, inflammatory protein levels were investigated western blot analysis. Knee joint tissue, histopathological observation hematoxylin & eosin staining and safranin-O staining were measured. Results : In the present study, ARB treated group showed superior inhibitory effects on the inflammatory parameters than the ASB treated group. ARB aqueous extract was effective in antioxidant measurements. The administration of ARB showed a significant reduction of changes in relative hind paw weight distribution. Morever, it decreased ROS, ALT and AST levels in serum, compared with those of the control rats. The ARB administration inhibited the biomarkers of inflammatory in tissues. Conclusions : ASB aqueous extract and ARB aqueous extract have a great effect on osteoarthritis, and ARB aqueous extract has excellent effect on osteoarthritis through antioxidant and anti-inflammation.

Effects of Extruded Acanthopanax Folium Extracts on Complete Freund's Adjuvant Induced Arthritis of Rats (오가엽(五加葉) 압출성형(壓出成形) 추출물의 Complete Freund's Adjuvant로 유발한 흰쥐의 관절염에 대한 효과)

  • Objectives : This study was designed to investigate the effects of extruded Acanthopanax Folium extracts on Complete Freund's Adjuvant (CFA) induced arthritis of rats. Methods: To induce arthritis in the ankle joint of rats, CFA was injected in the proximal part of the tail subcutaneously. After CFA injection, arthritic conditions were examined with macrography. The volume of paw edema and thickness of the ankle joints were checked regularly within 20 days. At 20 days, histopathological examination was performed on the ankle joint. Inflammation levels were determined by total WBC counts and differential WBC counts using a blood analyzer. Tumor necrosis factor-$\alpha$ and interleukin-1$\beta$ concentration in paw exudate were measured by ELISA method. Results: Several arthritic conditions induced by CFA were alleviated by Acanthopanax Folium treatment. Morphologically, reduction of arthritic conditions were observed and the volume of paw edema and thickness of the ankle joints were significantly decreased. Additionally, cytokines in paw exudate were diminished and histopathological improvement was observed. Conclusions: This study showed that the extruded Acanthopanax Folium extracts have the beneficial effects on the CFA induced arthritis of rats and might be used for chronic arthritis patients.

Effect of Acanthopanax Ethanol Extract on Experimental Renal Hypertension in Rats (오가피(五加皮) Ethanol Extract의 실험적(實驗的) 신성고혈압(腎性高血壓)에 미치는 영향(影響))

  • This study was carried out for the purpose of observing the effect of Korean Acanthopanax Radicis Cortex on renal hypertension and to clarify the mechanism of this effect, making use of its ethanol extract. Adult male or female rats, weighing 180-250g, were divided into 3 groups; the first for normotensive control, the second for hypertensive control and the third for hypertensive Acantopanax-treatment. Rats in the normotensive and hypertensive control group were administered 0.9% saline subcutaneously only, whereas those in the Acanthopanax-treated hypertensive group were administered 50mg/kg Acanthopanax ethanol extract subcutanously once a day. Changes of original blood pressure, and responses of blood pressure to various 4gents(norepinephrine, angiotensin, acetylcholine, serotonin and histamine) were recorded for each group on the initial, 18th, 32nd and 46th days of the experiment. The results obtained in this experiment are as follows: 1) The initial blood pressure was $102.6{\pm}7.6mmHg$ on the average. The blood pressures of the normotensive control group were not observed to alter significantly at any period in the course of the experiment. 2) The mean blood pressures in the hypertensive control group were recorded at $120.3{\pm}10.4mmHg$ on the 18th day, at $134.5{\pm}9.2$ on the 32nd day and at $138.8{\pm}8.3$ on the 46th day, thus revealing significant elevation in comparison with the corresponding normotensive control group blood pressures. On the other hand, the mean blood pressures in Acanthopanax-treated hypertensive group on the 18th, 32nd and 46th days were $118.3{\pm}9.7,\;129.9{\pm}8.3\;and\;120.2{\pm}8.3mmHg$ respectively. The blood pressures of the hypertensive-Acanthopanax group recorded. on the 46th day revealed a significant difference as compared with those of the corresponding hypertensive control group. 3) On the 46th day of this experiment, the responses of blood pressure to acetylcholine in the hypertensive-Acanthopanax group were suppressed significantly as compared with those of the hypertensive control group, and in the latter group, angiotensin was decreased markedly as compared with the corresponding normotensive control group. In contrast, pressor action of norepinephrine and depressor action of serotonin and histamine did not differ significantly among the three groups. These results suggest that Acanthopanax ethanol extract suppresses the induction of renal hypertension by means of a cholinergic action such as that caused by acetylcholine.

Enhancing Effect of Acanthopanax senticosus Extracts on Mouse Spleen and Macrophage Cells Activation (가시오가피 물 추출물에 의한 마우스 비장세포 및 대식세포 활성의 항진효과)

  • Acanthopanax senticosus is an herb that has been used as a traditional remedy and medicine source. Its anti-inflammatory and, anti-oxidative effects have been reported in previous studies. This study aimed to investigate the effect of Acanthopanax senticosus water extracts on mouse macrophage cell in vitro. Mouse splenocyte proliferation increased after application of Acanthopanax senticosus water extract supplement of 5, 10, 50, 100, 250, 500, $1,000{\mu}g/mL$ after 48 h pre-treatment with a mitogen (ConA or LPS). The production of cytokines secreted by LPS and non LPS stimulated macrophages was detected by ELISA assay using a cytokine kit. Cytokine production (IL-2, IFN-${\gamma}$, and TNF-${\alpha}$) increased after water extract supplementation. The result of this in vitro study, showed that splenocyte proliferation and cytokine production by activated peritoneal macrophages were increased after Acanthopanax senticosus water extract in the range of $500{\sim}1,000{\mu}L/mL$. Thus, it is suggested that supplementation with Acanthopanax senticosus water extracts may enhance immune function by regulating splenocyte proliferation and enhancing cytokine production by activated macrophage.

Quantitative Analysis of Lignans in the Fruits of Acanthopanax Species by HPLC

  • High performance liquid chromatography (HPLC) was used for the analysis of the lignans eleutheroside B and eleutheroside E in the fruits of Acanthopanax species. A reverse-phase system using a gradient of $H_2O$ and acetonitrile as the mobile phase was developed and detection was at 210 nm. The analysis was successfully carried out within 20 min. The content of eleutheroside B and eleutheroside E in Acanthopanax species was measured in the fruits of A. senticosus (0.58 and $1.66\;{\mu}g/mg$, respectively), A. sessiliflorus (1.15 and $8.49\;{\mu}g/mg$, respectively), A. koreanum (2.16 and $1.80\;{\mu}g/mg$, respectively), and A. divaricatus (1.06 and $7.08\;{\mu}g/mg$, respectively).

Modulation of IL-12 and IFN-γ Secretions by Eleutheroside E, Tortoside A, and Syringaresinol from Acanthopanax koreanum Nakai

  • Acanthopanax koreanum Nakai (Araliaceae) is a medicinal plant indigenous to Korea. The root and stem barks of Acanthopanax species have been used as a tonic and sedative as well as in the treatment of rheumatism and diabetes. In our study, three lignans, eleutheroside E (EE), tortoside A (TA), and syringaresinol (SY), were isolated from the stem and root of A. koreanum in an effort to study the immunomodulating effect. We treated natural killer cells and dendritic cells with lignans (EE, TA, or SY), and analyzed their cytokine (IL-12 and IFN-${\gamma}$) secretion. EE, TA, or SY markedly enhanced IL-12 secretion in mouse lymphoid (DC1) and myeloid type (DC2.4) dendritic cells after 48 hr of treatment. There were no significant differences in the cytokine stimulatory effects between EE, TA, or SY. Moreover, treatment of EE, TA, or SY significantly induced IFN-${\gamma}$ secretion by human NK cells (NK92MI) confirmed by ELISA assay. This study suggests that lignans from A. koreanum modulate cytokines, and that such modulation may provide the mechanism of action for many of their therapeutic effects.

Analysis of Lignans in Acanthopanax sessiliflorus Fruits and Their Fermented Wine by HPLC

The lignans were quantified by a reversed-phase system using a gradient of $H_2O$ and acetonitrile as a mobile phase within 20 min. The analysis was successfully carried out within 20 min. The contents of eleutherosides Band E as main active principles of Acanthopanax species were measured in A. sessiliflorus fruits (1.15 and $8.49\;{\mu}g/mg$, respectively), their fermented wine (0.45 and $1.33\;{\mu}g/mg$, respectively) and wine residues (no detection).
