Studies on the Constituents of Seeds of Acanthopanax sessiliflorum Seemann (I)-On Fatty Acids and Sterols- (오갈피나무 종자(種子)의 성분(成分)에 관한 연구(硏究)(제1보)(第1報)-지방산(脂肪酸)과 Sterol 성분(成分)에 관하여-)

  • Fatty acids and sterols were isolated from petroleum ether extract of seeds of Acanthopanax sessiliflorum Seeman. Fatty acids were obtained from the saponified fraction and sterols from nonsaponified fraction of petroleum ether extract. Fatty acids were identified by gas liquid chromatography, and its composition was myristic acid, palmitic acid, stearic acid, oleic acid, linoleic acid and linolenic acid. The composition of sterols were determined by preparative TLC and gas liquid chromatography. It was confirmed to be a mixture of stigmasterol and ${\beta}-sitosterol$. These fatty acids and sterols were not previously reported from seeds of Acanthopanax sessiliflorum Seemann.

Determination of Chiisanoside in Acanthopanax Species by High Performance Liquid Chromatography

  • The content of chiisanoside in the Acanthopanax Species was determined by reversed-phase high performance liquid chromatographic method. Chiisanoside was separated from the other components in the plant extracts using Zorbax 300 SB $C_{18}$ column with gradient elution of acetonitrile. Identification of chiisanoside was carried out by comparison in the LC/MS spectrum of separated peak from extract with that of standard. By HPLC analysis in this experiment, Acanthopanax species could be classified into two groups based upon the content of chiisanoside-one with low concentration of chiisanoside, such as A. senticosus and A. koreanum, and another with high concentration of chiisanoside, such as A. senticosus f. inermis, A. Divaricatus var. albeofructus, and A. chiisanensis.

  • "Ga Si O Gal Pi" is one of the Korean crude drugs used mainly to cure arthritis, acute gastritis, and beriberi. With regard to the botanical origin of "Ga Si O Gal Pi", it has been considered to be Acanthopanax species of Araliaceae, but there was no pharmacognostical confirmation on it. To clarify the botanical origin of "Ga Si O Gal Pi", the anatomical characteristics of the branch of Acanthopanax species growing wild in Korea and Japan, A. chiisanensis, A. divaricatus, A. koreanum, A. rufinerve, A. senticosus, A. seoulense, A. sessiliflorus, A. sieboldiaum were studied. As a result, it was clarified that "Ga Si O Gal Pi" was the branch and rarely the bark of branch of Acanthopanax senticosus.

  • Acanthopanax cortex (Acanthopanax sessiliflorum, Araliaceae, KP VIII), an important herbal drug, has been used as tonic, antistress and immuno-enhancing drugs in Korea. And Peiplocae cortex(Periploca sepium, Asclepiadaceae, CP 2000) has been used as cardiotonic, anti-inflammatory, and sedative effect in china. These are called “Namogapi” of Acanthopanax cortex and “Bukogapi” of Periplocae cortex in Chinese herbal market. These herbal medicines are sometimes circulated as same herbal medicine “Ogapi”. Antioxidant activity of medicinal plant extracts cultivated in Jecheon (제천산 약용식물 추출물의 항산화 특성)

  • Antioxidant activity of Astragalus membranaceus, Polygonatum stenophyllum, Angelica gigas, Acanthopanax sessiliflorus and Angelica pubescens extracts cultivated in Jecheon prepared with different solvents were determined by 1,1-diphenyl-2-picrylhydrzyl(DPPH) radical scarvenging ability and thiocyanate method . The highest hydroxy radical scarvenging activity was shown in Acanthopanax sessiliflorus, while the lowest was in Polygonatum stenophyllum. Angelica gigas showed strong antioxidant activity by thiocyanate method. Methanol extracts and water extracts showed higher antioxidant activities than ethanol extracts. Angelica pubescens showed the highest polyphenol contents and Acanthopanax sessiliflorus showed the highest flavonoid contents. Direct correlation between the hydroxy radical scavenging activity and polyphenol contents or flavonoid contents was established by simple regression(r>0.8) in each solvent extracts. Among medicinal plants cultivated in Jecheon, Acanthopanax sessiliflorus and Angelica pubescens showed strong antioxidant activities. These results suggest that methanol or water extracts of several medicinal plant cultivated in Jecheon could be used as natural antioxidants.

Effects of Acanthopanax Senticosus and Onion Mixture Extract on the Collagen-induced Arthritis in Rat Model (가시오가피와 양파 혼합 추출물이 Collagen 유발 관절염에 미치는 영향)

  • This study was conducted to determine the analgesic effect of onion and acanthopanax senticosus mixture extract using the rheumatoid arthritis rat model. Rheumatoid arthritis model was made by the intradermal injection of type II collagen emulsified. Rats were divided into four groups: (1) Sham group(n=5), (2) Control group(administered DW 3 $m{\ell}$/1 day after RA induced, n=7), (3) Experimental group I(administered Onion extractor 600 mg/3 $m{\ell}$/1 day after RA induced, n=7). (4) Experimental group III(administered Onion and Acanthopanax senticosus mixture extractor 600 mg/3 $m{\ell}$/1 day after RA induced, n=7). After that, we examined the arthritic index, paw edema, pain threshold at 1st, 14th, 28th days. And also we examined histopathologic study(safranin-O green), immunohistochamical stain(COX-2) of knee joint at 28th days. Arthritic index, paw edema and pain threshold test were decrease in experimental group I, II than control group. Especially group II was most significantly inhibit effect than the other groups at 28th days. On the histopathologic view, all experimental groups were relieved and reproduced the erosion of arthritic site compared with control group. All experimental groups were COX-2 positive cells in the immunohistological stain of the knee joint were significantly decreased compared with control group. Especially group II was most significantly decreased than the other groups at 28th days. Onion and Acanthopanax senticosus mixture extractor can be used for curing rheumatoid arthritis. Anti-inflammatory effects may be somewhat better in combination of Onion and Acanthopanax senticosus.

Comparative Radical Scavenging and Nitric Oxide Production Capacity of Water Extracts of Acanthopanax divaricatus var, Albeofructus and Eleutherococcus divaricatus var. Chiisanensis (지리산오가피와 흰털오가피 열수추출물의 라디칼 제거 활성 및 대식세포에서의 산화질소 생성능 비교)

  • In the present study, water extracts of Acanthopanax divaricatus var, albeofructus and Eleutherococcus divaricatus var. chiisanensis were prepared and their antioxidant and anti-inflammatory effects were compared. The water extract of Acanthopanax divaricatus var, albeofructus showed significantly higher activity in total polyphenol and flavonoid contents, while there were no significant differences in ABTS/DPPH radical scavenging activities and ferric reducing antioxidant power. However, the water extract of Eleutherococcus divaricatus var. chiisanensis at the highest dose showed significantly lower capacity in nitric oxide production than that of Acanthopanax divaricatus var, albeofructus. Taken together, there were differences between Acanthopanax divaricatus var, albeofructus and Eleutherococcus divaricatus var. chiisanensis in total polyphenol/flavonoid contents and anti-inflammatory effect, however, the antioxidant effect was almost similar.

Effects of Cutting Date, Shading Ratio and Growth Regulator on Rooting of Acanthopanax divaricatus var. albeofructus and Acanthopanax koreanum Nakai (흰털오갈피나무와 섬오갈피나무의 삽목시기, 차광율 및 생장조절제 처리가 발근에 미치는 영향)

  • This study was conducted to enhance the propagation of Acanthopanax divaricatus and Acanthopanax koreanum through different cutting practices. For propagation by cuttings, scions were taken on September 1. This already has hard skin and showed much higher values in terms of root length, root number, rooting ratio and root weight than those taken on June 30 and August 1. Regardless of cutting date, 50% shading resulted to the highest root length, root number, root ratio and root weight. Meanwhile, 95% shading significantly reduced these parameters compared with no shading. These results suggest that over-shading may inhibit root growth. Two growth regulators, IBA (concentration of 1,000, 2,000, 3,000 ppm) and Rootone-F (0.4%) were also tested of its effect to the cuttings. Rootone-F was found to be more effective than IBA. Cutting treated with Rootone-F had slightly higher root length and root number. A. koreanum which grows well in hot climatic condition showed better rooting ability than A. divaricatus.

Effect of Shading and Pinching on Growth and Acanthoside-D content of Acanthopanax divaricatus var. albeofructus and Acanthopanax koreanum Nakai (차광과 적심높이에 따른 흰털오갈피나무와 섬오갈피나무의 생육과 Acanthoside-D 함량 변화)

  • This study was conducted to identify the effect of shading and pinching on growth and acanthoside-D content of Acanthopanax divaricatus var. albeofructus and A. koreanum Nakai. Different pinching heights showed no significant differences in terms of plant growth and acanthoside-D content but higher values showed that pinching A. divaricatus at 60 cm and A. koreanum at 30 cm favored good growth and higher fresh weight in the shoots. The content of acanthoside-D was not significantly affected by pinching heights. Also, no significant difference in acanthoside-D content was found between the lower and upper part of plant in the first year. However it was much higher in the lower part than the upper part in the second year, which indicated that the content of acanthoside-D was comparatively high in the lower part where lignification is much advanced. Shading showed benefits in terms of growth of A. divaricatus while only 50%-shading was favorable for A. koreanum to achieve superior growth. Overall, results indicated that shading had favorably affected the growth of the 2 Acanthopanax species while no-shading is better if we opt to achieve higher acathoside D content.
