알코올 투여한 흰쥐 간세포내 알코올 탈수소효소의 활성과 항산화에 미치는 식물추출물들의 영향 (Effect of Plant Extracts on the Activity of Alcohol Dehydrogenase and the Antioxidation in Alcohol-treated Rat Hepatocyte)

  • 본 실험은 알코올 투여한 흰쥐를 대상으로 콩나물, 솔잎, 표고버섯과 오가피 추출물의 알코올 탈수소효소 활성과 항산화 효과를 비교하기 위해 수행되었다. 약 200g 정도의 Sprague-Dawley계 쥐들을 정상군, 알코올 단독투여군, 4가지 식물추출물 투여군으로 나우어서, 각 식물추출물을 알코올을 주입하기 전 8일 동안 200mg/kg b.w. 을 하루에 한번 경구투여하였다. 모든 동물은 알코올을 주입하고 90분 후에 도살시켰다. 콩나물과 솔잎추출물 투여군의 혈중 알코올 농도는 알코올 단독투여군, 표고버섯, 오가피군 보다 유의적으로 낮게 나타났다. 또한 콩나물과 솔잎 추출물 투여군의 알코올 탈수소효소활성은 알코올 단독투여군과 표고버섯, 오가파군보다 유의적인 증가를 보여주었다. Catalase 활성은 식물추출물 투여군이 알코올 단독투여군보다 다소 높게 나타났지만, 유의성 있는 차이는 보이지 않았다. 이러한 결과는 흰쥐에게 일회성으로 알코올을 다량 투여했을 때 알코올 탈수소효소가 catalase보다 우선적으로 알코올 대사에 반응함을 보여준다. 모든 식물추출물 투여군들의 지질과산화와 glutathione peroxidase 활성은 알코올 단독투여군보다 유의적으로 낮게 나타났다. 이 결과는 본 실험에 사용된 식물추출물들이 알코올의 산화에 대한 항산화 효과를 가진다는 것을 보여주며, 특히 콩나물과 솔잎 추출물 투여군이 알코올 탈수소효소의 활성증가오 항산화에 대한 효과가 높음을 보여주었다.

생약복합제 GCSB-5의 품질 표준화를 위한 구척의 지표성분 탐색 및 HPLC 분석 (HPLC Analysis and Screening of Standard Compound on Cibotii Rhizoma for Standardization of GCSB-5 Preparation)

  • GCSB-5 preparation is a purified extract from a mixture of 6 medicinal plants(Acanthopanacis Cortex, Achyranthis Radix, Saposhnikoviae Radix, Cibotii Rhizoma, Glycine Semen Nigra, Eucommiae Cortex) that have been widely used for the treatment of various bone disorders. The aim of this study was to investigate HPLC analysis method and screening of standard compound on Cibotii Rhizoma for quality standardization of a medicinal crude drug GCSB-5. Onitin-4-O-$\beta$-D-glucopyranoside was isolated from Cibotii Rhizoma as the standard compound and identified on the basis of spectroscopic data such as NMR. HPLC analysis method for the determination of onitin-4-O-$\beta$-D-glucopyranoside was established for the quality control of the medicinal plants of Cibotii Rhizoma species, GCSB-5 raw material and GCSB-5 preparation. And validation of HPLC analysis methods were conformed for verification of HPLC methods by check to specificity, linearity, intra-day precision, inter-day precision and accuracy following ICH guideline.

  • Nuclear-kappa B(NF-$\kappa$B) plays a role in the regulation of genes responsible for inflammatory and immune responses as well as growth control of cells. A cell-based assay system for guiding NF-KB activity was developed to determine the influence of activated NF-KB in human keratinocytes. It suggested that this system could be used to determine the quantitative measurement of NF-$\kappa$B activity in the human skin and allow the monitoring of anti-inflammatory agent for dermatological means from various environmental stimuli. 수종의 전통약제가 일차 배양 간세포에서 $CCl_4$ 유발 세포독성에 미치는 영향 (Effects of Traditional Drugs on $CCl_4-induced$ Cytotoxicity in Primary Cultured Rat Hepatocytes)

  • 80% Methanol extracts of 44 traditional drugs used for the treatment of liver diseases or tonic effects were screened for anti-hepatotoxic activity by in vitro assay using $CCl_4-induced$ cytotoxicity in primary cultured rat hepatocytes. $CCl_4-induced$ cytotoxicity was evaluated by determination of LDH, GOT or GPT activity in the medium. Rehmaniae Radix Preparata and Gelantina nigra inhibited the release of LDH, GOT or GPT from $CCl_4-treated$ hepatocytes. Gibotii Rhizoma and Eucommiae Cortex showed inhibitory effect on release of LDH from normal hepatocytes as well as $CCl_4-treated$ hepatocytes. Eucommiae Cortex and Lili Bulbus decreased release of GOT and LDH from normal hepatocytes, respectively. Astragali Radix inhibited release of GPT in $CCl_4-treated$ hepatocytes. Phlomidis Radix, Imperatae Rhizoma, Cistanchis Herba, Broussonetiae Fructus, Asparagi Tuber, Trigonellae Semen and Polgonati Rhizoma inhibited release of LDH from $CCl_4-treated$ hepatocytes. Among 44 traditional drugs, most of them released LDH, GOT or GPT at the dose of 1 mg/ml in normal hepatocytes, and Drynariae Rhizoma, Acanthopanacis Cortex, Longanae Arillus, Atratylodis Rhizoma and Ecliptae Herba increased $CCl_4-induced$ cytotoxicity.

생약복합제 GCSB-5의 급성통증 모델에서의 진통작용 (Evaluation of the Antinociceptive Properties of GCSB-5, a Herbal Formulation)

  • This study examined the antinociceptive properties of GCSB-5, a herbal formulation consisting of 6 Oriental herbs (Ledebouriellae Radix, Achyranthis Radix, Acanthopanacis Cortex, Cibotii Rhizoma, Glycine Semen, and Eucommiae Cortex) that are used in traditional medicine to treat various bone disorders, mainly of which involve analgesic processes. Peripheral and central analgesic models were established in experimental animals in order to evaluate the antinociceptive effects of the agent. GCSB-5 significantly inhibited the number of acetic-induced writhing (33.3%-34.3% inhibition at 100-600 mg/kg) but increased the pain threshold (38.0% increase at 300 mg/kg) in the Randall-Selitto test. However, GCSB-5 had no effect on the hot plate-induced nociception and hyperalgesia from the tail-pinch method. These results suggest that the antinociceptive effect of GCSB-5 may be mediated via peripheral mechanisms.

  • The water extracts of 82 Korean medicinal herbs were prepared and were examined for the binding affinity on the glycine binding site of NMDA (N-methyl-D-aspartate) receptor prepared by the synaptic membranes from the forebrains of male Sprague-Dawley rats. Among the tested, the extracts of Dioscoreae Rhizoma, Hoveniae Semen cum Fructus, Astragali Radix, Armeniacae Semen, Huttuynia cordata Herba, Acanthopanacis Cortex, Aurantii nobilis Pericarpium, Phellinus linteus, Amomi Fructus, Artemisiae capillaris Herba, Polyporus, Agastachis Herba and of Galli Stomachichum Corium were found to exhibit significant competitions with $[^3H]-MDL$ 105,519 for the glycine specific binding site of NMDA receptor in a dose dependent manner, respectively.

  • GCSB-5 preparation is a purified extract from a mixture six herbal medicines (Acanthopanacis Cortex, Achyranthis Radix, Saposhnikoviae Radix, Cibotii Rhizoma, Glycine Semen Nigra, Eucommiae Cortex) that have been widely used in traditional medicine to treat various bone disorders. This study was carried out to obtain the HPLC analysis method that can be used to establish quantitative analysis of Glycine Semen Nigra and Eucommiae Cortex for standardization of GCSB-5 preparation. HPLC analysis methods for the simultaneous determination of genistin (Glycine Semen Nigra) and geniposide (Eucommiae Cortex) were established for the quality control of herbal medicinal raw material and preparation. And validation of HPLC analysis methods were conformed for verification of HPLC methods by check to specificity, linearity, intra-day precision, inter-day precision and accuracy following ICH guideline. As the result of quantitative analysis, the contents of genistin and geniposide in the raw material of GCSB-5 preparation were 0.0426-0.0427 mg/g and 0.431-0.432 mg/g. And GCSB-5 preparation contained genistin of 0.0202-0.0203 mg/capsule and geniposide of 0.211-0.212 mg/capsule, respectively.

충북 지역 학교 급식 영양사들의 약선에 대한 인식 조사 (A Survey on the Perception of Yaksun(Medicated Diet) among School Foodservice Dietitians in the Chungbuk Area)

  • In this study, dietitians working in the school foodservice industry in the Chungbuk region were surveyed to determine their perception of medicinal herbs and medicinal foods in order to obtain basic data for developing medicinal food menus. The findings of this study are as follows. Many of the dietitians had low confidence in medicinal herbs used in medicinal foods. However the more the subjects ate such food, the higher their perception was. The higher the perception and reliability were, the more positive the answer was in terms of taste, nutrition and efficacy. Regarding the familiarity with medicinal herbs, the subjects, regardless of their age, showed a high perception of easily accessible medicinal herbs; younger subjects preferred Schisandrae fructus, Rubi fructus and Acanthopanacis cortex. The higher the perception and the eating frequency were, the higher the familiarity was. Regarding the development of medicinal food menus, the older the subjects were, the more positive they viewed the development of menus. Also, dietitians with 15 years of experience showed a high interest in the development of new menus.

General Pharmacological Study of GCSB-5, a Herbal Formulation

  • The general pharmacological properties of GCSB-5, a herbal formulation consisting of 6 Oriental herbs(Ledebouriellae Radix, Achyranthis Radix, Acanthopanacis Cortex, Cibotii Rhizoma, Glycine Semen and Eucommiae Cortex), were investigated in mice, rats, guinea pigs and rabbits. The administration of GCSB-5 had no effect on general behavior, and did not influence the central nervous system. Mean blood pressure, heat1 and respiratory rate and contractile response of the isolated guinea pig atrium were unaffected by the treatment of GCSB-5. Addition of GCSB-5 did not cause spontaneous relaxation and contraction of the isolated guinea pig ileum and rat uterus. And also, GCSB-5 had no effect on the gastrointestinal system and the blood system of the animals examined in this study. GCSB-5, at higher doses(1,000 and 3,000 mg/kg), increased the urinary excretion of electrolytes, however, the urine volume and pH in rats were unaffected. Taken together, these results indicate that GCSB-5 does not induce any adverse effects in experimental animals and is expected to have no significant general pharmacological activities.
