The effect of AcNPV infection conditions such as serum concentration, pH, CaCl2, lysosomotropic agent, cell density at infection, agitation, aeration and nutritional supplementattion on recombinant protein production in Spodoptera frugiperda 21 cells was investigated using tissue culture flask, bottle and spinner flask. It was shown that serum, CaCl2, pH and cell density at infection affected recombinant production. The lysosomotropic agent did not significantly influence recombinant protein production.
A novel method is developed to yield virus titers in 10 h, is easy to .perform using 96-well plates, and applicable to both any Autographa californica nucleopolyhyderovirus (AcNPV) and Bombyx mori nucleopolyhedrovirus (BmNPV)-based recombinant baculovirus. This assay uses an antibody to a DNA-binding protein to detect the infected cells via immune-staining. The titer is determined by counting foci produced due to infection of virus under a fluorescent microscopy. The required incubation period was shortened considerably because infected cells expressed viral antigens at the post infection time of 4 h. Therefore, 10 hours were enough to estimate the virus titer including virus infection time, insect cell culture, and estimation of virus titer.

Transformed Spodoptera frugiperda (Sf9) cells expressing baculovirus very late factor (VLF-1) were constructed by using Autograha nuclear polyhedrosis virus (AcNPV) immediate earthy gene (ie1). Neomycin-resistance gene as a selectable marker was introduced under the control of AcNPV ie1 promoter, and Bombyx mori nuclear polyhedrosis (BmNPV-K1) vlf-1 gene was introduced under the control of the Drosophila heat shock protein gene (hspr70) promoter to yield dual expression plasmid with two independent transcription units. It was transfected into Sf9 cells and cell clones expressing vlf-1 were selected by G4l8 treatment. Genomic DNA from transformed cells was isolated and integration of AcNPV iel harboring vlf-1 was confirmed by PCR using AcNPV iel-specific primers and Southern blot analysis. The transformed cells expressing VLF-1 in an infection-independent manner expressed foreign gene product of recombinant baculovirus in the earlier stage of infection compared with control Sf9 cells. These results suggest the possible to develop highly efficient transformed insect cells for baculovirus expression vector system.

Baculoviruses base vectors come to be regarded as methods for in vivo gene delivery and transient expression to the silkworm. In the case of silkworm, B. mori, two types of baculoviruses, AcNPV (Autographa californica nuclear polyhedrosis virus) and BmNPV (Bombyx mori nuclear polyhedrosis virus), are potentially applicable as vectors. Recently, AcNPV showed promising results with some silkworm strains despite different host-specificities. We searched for a highly-permissive silkworm strain in the B. mori stocks of Kyungpook National University that could produce high levels of recombinant protein. Seventy strains were screened using the recombinant AcNPV/BmA3-Luc virus. Based on the measured luciferase activity, the strains could be divided into three groups, high-, middle-, and low-permissive strains, according to their relative recombinant protein expression levels. At 48 hours post-injection, the luciferase activity in the high-permissive strains was 500-fold greater than that of the low-permissive strains. At 72 hours post-injection, a significant elevation in luciferase activity was observed in the hemocytes of all strains. Then, based on the above results, the High Permissive Strain (HPS) S10 and the Low Permissive Strain (LPS) S39 were pick up and was carried out Dot blotting, RT-PCR and Real time PCR.
유용물질을 생산할 수 있는 곤충 baculovirus expression vector system의 국내 개발을 위하여, Bm-NPV의 다각체 단백질 유전자를 탐색, 클로닝 하고 그 구조를 분석한 결과는 다음과 같다. 1. AcNPV의 다각체 단백질 유전자를 포함하고 있는 EcoRI-Ⅰ fragment내의 0.93kb를 probe로 하여 southern 분석한 결과, BmNPV의 다각체 단백질 유전자는 PstⅠ-F fragment(7.4Kb)에 위치하였다. 2. BmNPV의 다각체 단백질 유전자를 포함하는 PstⅠ-F fragment를 E. coli에 클로닝시켜서 pBmP-F라 명명하고, 다시 southern분석을 통해 subcloning하여 pBmP-H라 명명하였으며 pBmP-H의 제한효소 지도를 작성하였다. 3. pBmP-H의 염기서열분석결과 구조유전자를 포함한 1,259 bp의 염기서열이 결정되었다. Iatrou 등이 보고한 BmNPV 다각체 단백질 유전자의 염기서열과 비교한 결과 10개의 염기서열에서 차이를 보였으며, 74 Val이 Ile로, 76 Asn이 Ser으로 155 Met이 Val로 아미노산 변경된 결과를 보였다.
베큘로바이러스는 일부 나비목 해충을 대상으로 방제하는 데 사용되고 있다. 그러나 화학농약에 비해 느린 살충효과 및 좁은 적용 해충으로 응용 범위에 한계를 갖고 있다. 본 연구는 이러한 한계를 극복하고자 곤충의 면역억제을 통해 바이러스 병원력을 제고시킬 수 있는 기술을 소개한다. 폴리드나바이러스는 일부 맵시벌 및 고치벌에 공생하는 곤충 DNA 바이러스 분류군이다. 프루텔고치벌(Cotesia plutellae) 유래 CpBV(Cotesia plutellae bracovirus)는 브라코바이러스에 속한 폴리드나바이러스로서 면역어제를 발휘하는 여러 유전자를 함유하고 있다. 이 가운데 7개의 CpBV유전자를 선발하고 이를 야생형Autographa California multiple nucleopolyhedrovirus(AcNPV)에 재조합하였다. 이들 재조합 베큘로바이러스를 이용하여 파밤나방(Spodoptera exigua)과 배추좀나방(Plutella xylostella)을 대상으로 생물 검정한 결과, 이들 대부분은 야생형의 바이러스와 유사하거나 우수한 살충력을 나타냈다. 특히 CpBV-ELP를 포함한 재조합 베큘로바이러스가 대조바이러스에 비해 살충시간을 약 2 일 이상단축시킴으로 가장 우수하였다. 이 재조합 베큘로바이러스는 농도에 따른 살충력증가와 배추를 가해하는 파밤나방을 대상으로 한 바이러스 살포 처리가 뚜렷한 방제효과를 나타내어 현장 적용 가능성을 제시하였다. 또한 본 연구는 이 재조합 바이러스의 살충력 제고 현상을 CpBV-ELP의 항바이러스 기작 억제라는 측면에서 고찰했다.
Autographa californica 핵다각체병 바이러스(AcNPV)의 다각체 단백질과 초록색 형광 단백질의 융합단백질의 특성을 분석하였다. 초록색 형광 단백질 유전자는 AcNPV의 완전한 다각체 단백질 유전자의 앞쪽과 뒤쪽에 융합하여 다각체 단백질 유전자의 프로모터 조절하에 도입하였다. 이렇게 작성된 재조합 바이러스를 각각 Ac-GFPPOL 또는 Ac-POLGFP이라고 명명하였다. 이들 재조합 바이러스에 의해 감염된 곤충세포주에서는 56kDa의 융합단백질이 발현되었다. 한편, 흥미롭게도 재조합 바이러스 Ac-POLGFP에 의해 감염된 세포주에서는 초록색 형광이 핵내에서만 다각체 유사 granular particle 형태로 관찰되었다. 반면에 Ac-GFPPOP에 의해 감염된 세포도주에서는 대부분 핵내에 존재하였지만, 세포질과 핵 모두에서 초록색 형광을 관찰할 수 있었다. 그러나 발현된 융합단백질은 분명히 다각체단백질을 포함하고 있음에도 다각체는 형성하지 않았다. 이러한 결과들은 융합단백질에서 다각체단백질의 위치와 관련이 있는 것으로 보여진다.

We have now constructed a novel recombinant baculovirus Autographa californica nuclear polyhedrosis virus (AcNPV) producing polyhedra with Bacillus thuringiensis (Bt) CryIAc crystal protein. The recombinant polyhedra produced by the recombinant baculovirus, Btrus, in insect cells was characterized. The recombinant baculovirus has two independent transcription units in opposite orientations with two promoters, p10 or polyhedrin gene promoter each initiating transcription of either native polyhedrin or fusion protein with polyhedrin and Bt Cry1Ac crystal protein. Surprisingly, this recombinant baculovirus stably produced recombinant polyhedra which were nearly similar to those of wild-type AcNPV. The immunogold staining experiment showed that the recombinant polyhedra were assembled with polyhedrin and Bt Cry1Ac crystal protein, and contained virus particles. Insecticidal toxicity of recombinant polyhedra of Btrus to the fall webworm, Hyphantria cunea, was strikingly improved in comparison with the wild-type AcNPV.

A novel recombinant baculovirus Autographa californica nuclear polyhedrosis virus (ACNPV) producing the green fluorescent polyhedra was constructed and characterized. The recombinant virus was stably produced fluorescent polyhedra in the infected cells and the morphology of the polyhedra was nearly similar to that of wild-type AcNPV. For the production of the fluorescent polyhedral the green fluorescent protein (GFP) gene was introduced under the control of polyhedrin gene promoter of AcNPV by translational fusion in the front and back of intact polyhedrin gene. The recombinant baculovirus was named as CXEP, As expected, the 93 kDa fusion protein was expressed in the CXEP-infected cells. Interestingly, however, the cells infected with CXEP also showed a 33 kDa protein band as cells infected with wild-type AcNPV. The results of Southern blot analysis and plaque assay suggested that two types of baculoviruses expressing the GFP fusion protein or only native polyhedrin were formed through homologous recombination between two polyhedrin genes in the same orientation. Thus, this system can be applied for the production of recombinant polyhedra with foreign gene product of diverse interest.

The hydrogel process is a different form of ultrafiltration and has been used to separate biological molecules. In this study, the gel pore size was predicted by pulse NMR technique and neural network using a database obtained from gel filtration chromatography and diffusion experiment. Recombinant alkaline phosphatase expressed in insect cells was concentratred 1.5 times by hydrogel ultrafiltration by swelling at 2$0^{\circ}C$ and collapsing at 35$^{\circ}C$ at 53-65% separation efficiency and 78-83% enzyme recovery. Wild and recombinant Autographa californica unclear polyhedrosis viruses (AcNPV) were also concentrated 1.4 and 1.6 times of the feed solution at 48.5 and 60.0% separation efficiency, respectively Hydrogel ultrafiltration appears to be an attractive alternative for the concentration of AcNPV and recombinant proteins from insect cells.
