Highly purified chitosan reduce blood alcohol concentration, aspartate aminotransferase, and alanine aminotransferase levels in human

  • The purpose of this study was to examine the effect of supplementary highly purified chitosan (HPC) on blood alcohol concentration in healthy human. The human study was performed with two sections. Each section of the study was conducted by two-phase cross-over design with a week wash-out period. All volunteers took HPC in one phase, and took a placebo in the next phase. Blood alcohol concentrations were different between in those taking HPC and in those taking the placebo in the human. And the concentration of serum aspartate aminotransferase (AST, GOT) and alanine aminotransferase (ALT, GPT), the indicator of liver cell damage, was lowered in those taking HPC, compared to those taking the placebo. In conclusion, taking HPC prior to drinking alcohol can somewhat reduce alcohol concentration in human blood and liver cell damage.

적출간 관류법을 이용한 카드뮴과 니켈의 간독성 비교 (Comparative Hepatotoxicity Assessment of Cadmium and Nickel with Isolated Perfused Rat Liver(IPRL))

  • 목적 : 이 연구의 목적은 적출간 관류법을 통하여 염화니켈과 염화카드뮴의 간 독성을 비교하는 데에 두었다. 방법 : AST(aspartate aminotransferase ferase), ALT(alanine aminotransferase), LDH(lactate dehydrogenase) 등의 간기능을 나타내는 생화학적 지표와 관류속도를 간독성 지표로 이용하였으며 산소 소모율은 생존력을 나타내는 지표로 이용하였다. $300({\pm}50)g$ 정도의 흰쥐를 대조군과 각 노출군($50{\mu}M$$200{\mu}M$의 염화니켈과 염화카드뮴)에 5마리씩 총 25마리를 무작위로 할당하였다. Krebs-Ringer bicarbonate buffer solution을 관류액 용도로 제조하여 간문맥에 투입하고 간세포를 통과하여 vena cava로 배출되는 관류액을 일정한 시간 간격을 두고 계속적으로 채취하였으며 채취한 관류액은 간독성 지표를 측정하는 데에 이용하였다. 결과 : AST, ALT, LDH 모두 염하니켈 보다는 염화카드뮴 투여군에서 시간의 경과에 따라 큰 폭으로 증가하였으며 반복 측정된 2요인 분산분석을 실시한 결과, 통계적으로 유의한 차이를 나타냈다. 생존력은 두 물질 모두에서 시간의 경과에 따라 감소하는 경향을 나타냈다. 결론 : 결론적으로, 이 실험상에서는 염화니켈 보다 염화카드뮴의 간독성이 더욱 강한 것으로 드러났다. 적출간 더욱 강한 것으로 드러났다. 적출간 관류법의 특성상 급성 간독성 평가에서 적절한 기법으로 보이며 국내 연구에서도 이 방법이 널리 이용되기를 기대한다.

누에와 누에번데기의 혈청지질에 미친 영향에 대한 메타분석 (Meta-Analysis on the Effect of Serum Lipids Levels in Silkworm and Silkworm Pupae)

  • 본 연구는 경제적, 사회적, 환경적으로 가치가 있는 미래의 식량자원인 곤충식품의 산업적 활용도를 높이고자 최근 10년간(2009년~2019년) 발표된 곤충식품들 중 누에와 누에번데기의 생리활성 연구자료를 수집하여 혈청 지질 수준 개선에 대한 메타분석을 실시하였다. 누에와 누에번데기 연구물의 총 편수는 20편이었으며, 총 사례 수는 261개로 산출되었다. 연구단위로 분석한 연구의 수는 총 20개였다. 전체 연구결과의 효과크기는 0.852로 큰 효과크기였으며, 95% 신뢰구간의 범위는 0.662~0.999였다. Q 값은 23.264(p<0.001), Higgin's I2 값은 92.217로 각각의 연구가 이질성이 있다고 확인되었다. 관측값과 추정치를 보정한 관측값이 0.654로 동일하게 나타나 출판편향은 없었다. 누에 연구는 혈당과 혈중지질이 각각 0.801, 0.948로 큰 효과크기였으며, 그중 LDL-콜레스테롤은 1.371로 매우 큰 효과크기였다. 간 세포 손상(AST/GOT)은 0.768, 만성 간 세포 손상(ALT/GPT)은 0.788로 중간 효과크기였다. 실험기간은 5~8주가 1.170, 섭취량은 1~50 mg/kg이 1.020으로 매우 큰 효과크기였다. 누에번데기는 혈중지질과 총 콜레스테롤이 각각 0.991, 0.951로 큰 효과크기였다. 실험기간은 9주 이상이 1.103으로 매우 큰 효과크기였으며, 섭취량은 5001 mg/kg 이상이 0.855로 큰 효과크기를 보였다.

형방패독산(荊防敗毒散)과 독활지황탕(獨活地黃湯)이 Wistar rat의 노화(老化)에 미치는 영향(影響) (Effects of Hyeongbangpaedok-san and Dokhwaljihwang-tang that Get Weight, Hematology, Biochemistry Change by Wistar Rat's Aging)

  • 1. Objectives The purpose of this study is to find out effects of Hyeongbangpaedok-san(HBPDS) and Dokhwaljihwang(DHJH) against decline of physical function as aging. 2. Methods Administrating HBPDS and DHJH to 40-week-old Wistar rat for 10 weeks so, I researched weight change, weight change of internal organs, and hematological and serological changes. 3. Results & Conclusions 1. Both examining groups, which were taken HBPDS and DHJH, got more weight than control group. But that was regardless. 2. Both examining groups got more weight on internal organs than control group. But that was regardless, too. 3. Both examining groups decreased in amount of MDA in serum, as contrasted with control group. But it was regardless. 4. Both examining groups improved on hematological condition. WBC, RBC, Hgb, monocytes and eosinophil rates were decreasing and HCT and PLT were increasing. Especially monocytes(p<0.001) and eosinophil(p<0.05) rate of DHJH taken group was decreased remarkably. 5-1. Both examining groups show decline in each item of functional examination of liver, such as ALT, AST, T-bilirubin, T-protein, ALB, A/G. T-chol, TG, etc. HBPDS taken group showed meaningful decline in Albumin(p<0.01) and A/G(p<0.01) and DHJH taken group showed meaningful decline in T-bilirubin(p<0.01). 5-2. Both examining groups showed decline in items of functional examination of kidneys. Specially HBPDS taken group showed meaningful decline in CRN rate(p<0.05) and DHJH taken group showed meaningful decline in BUN rate(p<0.05). As those results, HBPDS and DHJH are effective against decline of physical function as aging.

자색고구마가 사염화탄소 투여에 의한 흰쥐의 간손상 보호에 미치는 영향 (Pretective Effect of Purple Sweet Potato (Ipomoea batatas) on Hepatotoxicity Rats Induced by Carbon Tetrachlolide)

  • The purpose of this study was to investigate the effects of dietary purple sweet potato(Ipomoea batatas) powder on serum lipid levels and antioxidative enzymes in normal and pretective effect on hepatotoxicity rats induced by carbon tetrachlolide. Four groups of rats (3-week-old inbred Sprague-Dawley male rats) were normal rats fed control diet(C), induced hepatotoxicity rats fed control diet(EC), normal rats fed purple sweet potato diet(P), and induced hepatotoxicity rats fed purple potato sweet diet(EP). Rats were induced by single injection of 50% carbon tetrachlolide(0.1 mL/100 g B.W., i.p.). The rats were fed ad libitum each of the experimental diet for 5 weeks. After 5 weeks the rats were sacrificed and activities of antioxidant enzymes and lipid peroxidation products were determined in their liver homogenates. But serum concentrations of lipid was not significant in all groups. Serum alanine aminotransferase(ALT/GPT) and aspartate aminotransferase(AST/GOT) of the EC and EP groups were heigher than the C and P groups. The hepatic glucose 6-phosphatase(G6Pase) activity of the group fed purple potato diet(P) was lower than the other groups(p<0.05). However, The glutathione peroxidase(GPx) activities was not statistically different between the groups. Renal glutathione S-transferase(GST) activity of the EC and EP groups were lower than the C and P groups(p<0.05). In conclusion, these results suggest that purple sweet potato is believed to be possible protective effect on hepatotoxicity rats induced by carbon tetrachlolide.

소자호탕이 $CCl_4$로 유발된 Rat의 간 장해 전후에 미치는 영향 (The Pro and Post Effects of Soshiho-tang on Rat's Liver Damage induced by $CCl_4$)

  • In studying the specific effects of some drugs, animals under experiments get some stress through laboratory environments, drug injection, and adaptation period. These stimuli do harms on liver function. Nowadays studies on liver intoxication and its protection are under research, but the function of dissolution is rarely under studies. It is widely accepted that Soshiho-tang has function of clearing away low spirits, and that it enables liver bloods to move stronger, and to have calm mind. So I injured rats liver by injectioning CCI₄. And the rats took in Soshiho-tang solution. I made a comparison between the functions before and after rat's liver damage. There are many representative serums used to note an index on liver damage. I used total protein, albumin, ALP, GOT, GPT activity, P450, SOD, Catalase, GST, GR, and GPx. I got the following results. When Soshiho-tang was injected after CCI4 intoxication, total protein and albumin decreased. When Soshiho-tang was injected, ALP decreased, compared with control group. When Soshiho-tang was injected after CCI₄ intoxication, AST and ALT decreased. When Soshiho-tang was injected before CCI₄ intoxication, P450 was restrained. When Soshiho-tang was injected, LPO was all restrained. When Soshiho-tang was injected, SOD, Catalase, GST, GR, and, GPx increased. These results show that blood test reveals that it is good to inject Soshiho-tang after CCI₄ intoxication, but that it is good to inject Soshiho-tang before CCI₄ intoxication in case of P450, LPO, SOD, Catalase, GST, GR, and GPx. It is estimated that the medication period and time of liver damage by CCI₄ have counter results, and that it needs more modified study.

흰쥐에서 사염화 탄소로 유도된 간독성에 미치는 더위지기 추출물의 영향- I. 사염화탄소 독성유발 심화 수준에 대한 더위지기 추출물의 회복효과 (Effects of an Extract from A. gmelini Weber on Hepatotoxicity Induced by $CCl_4$in Rats - I. Effects of an Extract from A. gmelini on Hepatotoxicity Induced by Different Levels of $CCl_4$)

  • To investigate effects of A. gmelini on the 14-day CCl$_4$induced hepatotoxicity, extracts were prepared in 3 ml saline at the dose of 5,000 mg/kg b.w. to administer orally once daily. Each concentration (5:5, 2:5 : 7.5, 1:9, CCl$_4$: olive oil v/v) of CCl$_4$was orally administered with 2.5 ml/kg b.w., During the experiment, halves of the rats were sacrificed every 7 day and hemanalysis was done. On the 7th day, hemanalysis showed following recovery values; AST 52.6∼61.4%, ALT 55.9∼86.1%, ALP 46.0∼70.9%, BUN 75.7∼100.0< %, TBIL 55.2∼96.1%, TCHO 38.0∼63.7%, and TG 55.2∼96.0%. On the 14th day, recovery values of each treatment were GOT 37.7∼43.1%, GPT 19.8∼45.9%, ALP 58.1∼95.9%, BUN 57.6∼100.0< %, TBIL 78.6∼100.0< %, TCHO 56.9∼100.0< %, and TG 10.0∼5l.2%. By histological examination of liver, hydropic degeneration, fatty change, lipid accumulation and necrosis were also recovered by administration of A. gmelini extract.

소아 지방간 환자에서 체중조절에 의한 간기능 개선 효과 (Improvement of Liver Function by Weight Control in Children with Fatty Liver)

  • Obese children have increaed in Korean in recent years as socieconomic status have got more higher than past years. Obese children have many health problems and some kinds of complications. Obese patients who have fatty liver with abnormal liver function should be managed with weight control. This study was intended to investigate the effect of weight control on hepatic abnormalities of obese children with fatty liver. We managed 6 obese children who had fatty liver with abnormal liver finction for weight control containing behavior modification, diet and exercise during 3 months. After 3 months, we compared body weight, liver function test, dietary intake and exercise. Body mass index(BMI) showed significant decrease from 28.1$\pm$2.3kg/$m^2$ to 26.2$\pm$1.7kg/$m^2$ (p<0.01). Aspartate aminotransferase(AST) showed significant improvement from 56$\pm$15IU/l(p<0.01). Alanine aminotransferase(ALT) also showed significant improvement from 108$\pm$38IU/l to 33$\pm$9IU/l(p<0.01). Therefore if we can rule out the other causes of hepatic abnormalities in obese children with fatty liver, these patients showed benefit by weight control as it can improve both obesity and liver functions.

주거용 컨테이너 화재시 발생되는 연소가스의 독성 평가에 관한 연구 - 화재용 방독면 filter의 성능평가를 중심으로 (A Study on Toxicity Evaluation of Combustion Gases Released from the Residental Container Fire - Efficiency Test for the Fire Gas Mask Filters)

  • The recent Ire incident in an elementary school of Chonan city causes the media focus on the fire safety of residential container buildings. In this study, real fire tests were conducted in this kind of buildings. Combustion products including $O_2,\; CO_2,\;CO,\;NOx,\;SOx,\;HCI,\;HCN$ were measured, and blood samples of lab rats were analyzed in terms of Co-Hb, Glucose, AST(GOT), ALT(GPT), in order to investigate the hazard-reduction effects of employing gas mask protected with filter during the fire emergency of residential container buildings. According to the test results, whether or not employing the filter showed a sheer difference in the toxicity of the fire-induced gases, and then the importance of wearing a gas mask was evidently demonstrated.

담도 폐쇄증으로 Kasai 수술 받은 환아들의 장기간의 추적관찰 (10년 이상 생존한 환아들을 대상으로) (Long-term Outcome after Surgery for Biliary Atresia (Study of 10 patients surviving more than 10 years))

  • Biliary atresia (BA) is the result of fibrosing destructive inflammatory process affecting intrahepatic and extrahepatic bile ducts, which lead to cirrhosis and portal hypertension. Kasai portoenterostomy has been the standard operative procedure in biliary atresia. Recently, there has been remarkable increase in the survival rate in cases of BA. However, long-term survivors are not clearly evaluated in Korea. To define long-term prognosis factors of patients who underwent surgery for BA, a retrospective study was undertaken of 10 (37 %) patients surviving more than 10 years among 27 patients who underwent one of Kasai procedures between 1981 and 1995. Hepatomegaly was present in 4 and splenomegaly in 7 patients. Serum bilirubin was normalized at 1 year after operation. Aspartate aminotransferase (AST, GOT), Alanine aminotransferase(ALT, GPT) were normalized at 12 years and alkaline phosphatase(ALP) was normalized at 13.5 years. Cholangitis developed mainly within 5 years after operation so close follow up is needed. Life long follow-up is needed because of progressive deterioration of liver function even after 10 years.
