Effect of Drenching Soil with Benomyl, Propiconazole and Fluazinam on Incidence of Disappearing Root Rot of Ginseng

  • Three fungicides, Orbit (propiconazole), Benlate (benomyl) and ASC-66835 (fluazinam), were tested as soil drenches to control disappearing root rot (DRR) of ginseng (Panax quinquefolius) in gardens artificially infested with Cylindrocarpon dsstrutans. The incidence of DRR was low (0∼3.5%) in uninfected plots and significantly higher in infested plots (2.6∼19.9%). Significant reductions in the incidence of DRR were observed in 1-year-old and 2-year-old gardens that were treated and assessed for disease in the same year Significant control was not obtained in 3-year-old gardens treated and as secede in the same year, or in 1-year-old or 2-year-old gardens assessed in the year following information. Disease incidence was significantly reduced by 49-77% by low and high rates of benomyl (45 and 1,250 mg a.1./L) and propiconazole (10 and 40 mg a.i./L) and by fluazinam at 150 mg a.i./L. These fungicides seem to be worthy of further investigation as soil drenches to control DRR of ginseng.

Preventable effect of L-threonate, an ascorbate metabolite, on androgen-driven balding via repression of dihydrotestosteroneinduced dickkopf-1 expression in human hair dermal papilla cells

  • In a previous study, we recently claimed that dihydrotestosterone (DHT)-inducible dickkopf-1 (DKK-1) expression is one of the key factors involved in androgen-potentiated balding. We also demonstrated that L-ascorbic acid 2-phosphate (Asc 2-P) represses DHT-induced DKK-1 expression in cultured dermal papilla cells (DPCs). Here, we investigated whether or not L-threonate could attenuate DHT-induced DKK-1 expression. We observed via RT-PCR analysis and enzyme-linked immunosorbent assay that DHT-induced DKK-1 expression was attenuated in the presence of L-threonate. We also found that DHT-induced activation of DKK-1 promoter activity was significantly repressed by L-threonate. Moreover, a co-culture system featuring outer root sheath (ORS) keratinocytes and DPCs showed that DHT inhibited the growth of ORS cells, which was then significantly reversed by L-threonate. Collectively, these results indicate that L-threonate inhibited DKK-1 expression in DPCs and therefore is a good treatment for the prevention of androgen-driven balding.

스터드 기반의 레이저 레이다 삼차원 영상센서기술

  • 본 발표에서는 최근 무인전투기 및 무인자율주행차량 등의 어플리케이션에 응용되고 있는 삼차원 영상 센서 시스템 기술에 관한 소개와 더불어 한국전자통신연구원에서 독자적인 구도를 기반으로 최근 시연에 성공한 스터드(STUD) 레이저 레이다의 동작원리 및 구현결과를 소개하고자 한다. 최근 전세계의 삼차원 레이저 영상 센서 시스템은 1) 한 관측점을 회전을 통해 스캔하는 2D 스캔 방식에서 벗어나, 2) 일차원 배열 형태로 구현된 관측점을 센서가 회전하면서 얻어지는 3D 스캔 방식과 3) 이차원 어레이 형태의 검출기로 삼차원 영상을 검출하는 방식으로 삼차원 영상을 확보하는 노력으로 구분되어 진행되어 왔다. 이번 시연에 성공한 제안된 방법은, 기존의구도와는 다른 독자적인 방식으로, 대면적 검출기를 기반으로 센서부가 회전하지 않으면서, 하나로 통합된 검출기 출력을 이용할 수 있는 구도인 스터드(STUD: STatic and Unitary Detector)기반의 삼차원 레이저 레이다 시스템이다. 최근 구현 결과는 수평해상도 320, 수직해상도 240인 QVGA 영상 수준으로, 이는 지금까지 세계 최고해상도인 ASC사의 128x128 해상도를 뛰어 넘는 우수한 결과이다. 제안된독자적인 구도의 삼차원 레이저 영상 시스템은 다양한 관련 기술들과의 접목하여 향후 군수용 뿐만아니라 민수용 시장의 기술발전에 큰 영향을 미칠 것으로 예상되고 있다.

  • In recent years, the occurrence of nitrosamine, which are produced by the interaction of nitrite and secondary amine, in foods has been the subject of considerable interest and controversy. In this experiment, changes in nitrate, nitrite, TMAO, TMA and DMA nitrogen of salted and dried corvenia, which were incorporated with sodium benzoate, ascorbic acid, cysteine and tetracycline in salt, during processing and storage were analyzed. Levels of nitrate nitrogen was decreased while those of nitrite was increased in salted and dried products during processing and storage, but sample was incorporated with ascorbic acid, cysteine and sodium benzoate in salt inhibited the reduction of nitrate to nitrite while with tetracycline in salt accelerated this reduction in salted and dried products during processing and storage. Contents of TMAO nitrogen in all salted and dried products as well as in the control was decreased, but was increased during storage, while TMA nitrogen was increased in salted and dried products during processing and storage. Contents of DMA nitrogen was increased in all salted and dried products during processing and storage, DMA contents of raw in the control was increased to more than 11.6 times after storage for 30 days, but sample was incorporated with ascorbic acid, cysteine and sodium benzoate in salt inhibited the production of DMA nitrogen while with tetracycline in salt accelerated in salted and dried products during processing and storage.

  • During storage of $45^{\circ}Bx$ cloudy, $50^{\circ}Bx\;and\;72^{\circ}Bx$ clear apple juice concentrates for 18 months, the changes in microbial counts and physicochemical properties were investigated. Little viable microbial counts were detected in $50^{\circ}Bx\;and\;72^{\circ}Bx$ clear concentrates during all storage time and a few in $45^{\circ}Bx$ cloudy concentrates during the only first storage stage. In all three concentrates, any changes were not found in pH and titrable acidity. But Hunter L values were decreased and a, b values were increased. Alcohol soluble color(ASC) were increased and vitamin C contents rapidly decreased, especially during the first three months. The contents and compositions of free sugars were showed a little change. It seemed that the change in ASC was more closely related with that of vitamin C contents than that of reducing sugar contents.

Neural-based Blind Modeling of Mini-mill ASC Crown

  • Neural network can be trained to approximate an arbitrary nonlinear function of multivariate data like the mini-mill crown values in Automatic Shape Control. The trained weights of neural network can evaluate or generalize the process data outside the training vectors. Sometimes, the blind modeling of the process data is necessary to compare with the scattered analytical model of mini-mill process in isolated electro-mechanical forms. To come up with a viable model, we propose the blind neural-based range-division domain-clustering piecewise-linear modeling scheme. The basic ideas are: 1) dividing the range of target data, 2) clustering the corresponding input space vectors, 3)training the neural network with clustered prototypes to smooth out the convergence and 4) solving the resulting matrix equations with a pseudo-inverse to alleviate the ill-conditioning problem. The simulation results support the effectiveness of the proposed scheme and it opens a new way to the data analysis technique. By the comparison with the statistical regression, it is evident that the proposed scheme obtains better modeling error uniformity and reduces the magnitudes of errors considerably. Approximatly 10-fold better performance results.

High-Level Expression of Pseudomonas sp. LBC505 Endoglucanase Gene in Escherichia coli

  • Endoglucanase gene of Pseudomonas sp. LBC505 was previously cloned in pUC19 to yield plasmid pLCl. The Pseudomonas sp. LBC505 endoglucanase gene was subcloned in a temperature-regulated Es-cherichia coli expression vector, pAS1, containing the leftward promoter $P_L$ of bacteriophage lambda. The level of gene expression was controlled by the thermal inactivation of the heat-sensitive lambda cI857 repressor. Best yield of endoglucanase was obtained by lowering the incubation temperature to $37^{\circ}C$ after induction at $42^{\circ}C$ for 1h. Under these conditions enzyme production continued for about 5h at a gradually decreasing rate. Ecoli harboring recombinant plasmid pASC10 expressed 4.3 times as much CMCase activity as E.coli containing pLCl. To enhance the expression level of endogl, ucanase gene, we have also changed the presumptive Shine-Dalgamo sequence (AGAGGT) of the gene to consensus sequence (AGGAGGT) by site-directed mutagenesis. The genes mutated were subcloned in pASl resulting in the formation of recombinant plasmid pASS50. E.coli harboring the plasmid pASS50 expressed 6.2-fold higher levels of CMCase activity than that of E.coli harboring pLC1.

miR-30a-5p Augments the Anti-inflammatory Effects of Dexmedetomidine in LPS-induced BV2 Cells (LPS로 유도된 BV2 세포에서 Dexmetomidine이 갖는 항염증효과에 대한 miR-30a-5p의 시너지 효과)

  • Neuroinflammation is defined as a neurological inflammation within the brain and the spinal cord. In neuroinflammation, microglia are the tissue-resident macrophages of the central nervous system, which act as the first line of defense against harmful pathogens. Dexmedetomidine (Dex) has an anti-inflammatory effect in many neurological conditions. Additionally, the microRNA-30a-5p (miR-30a-5p) mimic has been proven to be effective in macrophages in inflammatory conditions. This study aimed to investigate the synergistic anti-inflammatory effects of both miR-30a-5p and Dex in lipopolysaccharide (LPS)-induced BV2 cells. This study showed that miR-30a-5p and Dex decreased nuclear factor kappa-light-chain-enhancer of activated B cells (NF-κB) translocation in LPS-induced BV2 cells. MiR-30a-5p and Dex alleviated tumor necrosis factor-alpha (TNF-α) and interleukin-6 (IL-6), LPS-induced phosphorylation c-Jun N-terminal kinases (JNK), extracellular signal-regulated kinase (ERK) and p38. Also, the expression of the NOD-like receptor pyrin domain containing 3 inflammasome (NLRP3), cleaved caspase-1, and ASC was inhibited. Furthermore, LPS-stimulated nitric oxide (NO) production, inducible nitric oxide synthase (iNOS), and cyclooxygenase-2 (COX-2) expression were attenuated by Dex and miR-30a-5p. Our results indicate that a combination of Dex and miR-30a-5p, attenuates NF-κB activation, the mitogen-activated protein kinase (MAPK) signaling pathway, and inflammatory mediators involved in LPS-induced inflammation and inhibits the activation of the NLRP3 inflammasome in LPS-activated BV2 cells.

Renal Sympathetic Denervation Induces Acute Myocardial Inflammation through Activation of Caspase-1 and Interleukin-1β (교감신경절제술(RDN) 후 caspase-1과 interleukin-1β 활성화로 인해 유발된 염증성 급성심근손상)

  • Efferent and afferent sympathetic nerves are closely related to the development of hypertension and heart failure. Catheter-based renal sympathetic denervation (RDN) is implemented as a strategy to treat resistant hypertension. We investigated whether RDN procedure causes inflammatory damage on myocardium in the early phase of sympathetic denervation. Twenty-five female swine were divided into 3 groups: normal control (Normal, n=5), sham-operated control (Sham, n=5), and RDN groups (RDN, n=15). The RDN group was further subdivided into 3 subgroups according to the time of sacrifice: immediately (RDN-0, n=5), 1 week (RDN-1, n=5), and 2 weeks (RDN-2, n=5) after RDN. There were no significant changes in the clinical parameters between the normal control and sham-operated group using contrast-media. In the myocardium, inflammatory cytokines, $IL-1{\beta}$ and $TNF-{\alpha}$ increased at the first week, and then decreased at the second week after RDN. Anti-inflammatory cytokine, IL-10 increased immediately, and then decreased at the second week after RDN. Caspase-1 activity and apoptosis-associated speck-like protein containing a caspase recruitment domain (ASC) expression increased immediately after RDN until the second week. However, nucleotide-binding oligomerization domain, leucine rich repeat and pyrin domain containing protein 3 (NLRP3) expression did not show any significant differences among the groups. The RDN can cause acute myocardial inflammation through activation of caspase-1 and $IL-1{\beta}$. We should pay attention to protecting against early inflammatory myocardial damage after RDN.

Evaluation of Antimicrobial Activities of Sequential Spray Applications of Decontamination Treatments on Chicken Carcasses

  • The objective of this study was to evaluate the effects of sequential applications of ${\varepsilon}$-polylysine (EPL) or lauramide arginine ethyl ester (LAE) sprays followed by an acidic calcium sulfate (ACS) spray on inoculated chicken carcasses to reduce Salmonella (Salmonella enterica serovars including Salmonella typhimurium and Salmonella enteritidis) contamination during 6 days of storage ($4.4^{\circ}C$). Secondly, reductions of the resident microflora were studied on uninoculated chicken carcasses following the sequential application of the treatments, chilling and 10 days of storage at $4.4^{\circ}C$. The treatment of Salmonella inoculated carcasses with 300 mg/L EPL followed by 30% ACS (EPL300-ACS30) sprays reduced Salmonella counts initially by 1.5 log cfu/mL and then by 1.2 log cfu/mL (p<0.05) following 6 days of storage at $4.4^{\circ}C$. Likewise, 200 mg/L LAE followed by 30% ACS (LAE200-ACS30) treatment reduced initial Salmonella counts on poultry carcasses by 1.8, 1.4 and 1.8 log cfu/mL (p<0.05), respectively, after 0, 3, and 6 days storage. Immediately after the treatments, EPL300-ACS30 and LAE200-ACS30 both reduced Escherichia coli counts significantly by 2.6 and 2.9 log cfu/mL, respectively. EPL300-ACS30 and LAE200-ASC30 were effective in lowering psychrotroph counts by 1 log cfu/mL on day 10 when compared to the control and distilled water treatments. This study demonstrated that EPL300-ACS30 and LAE200-ACS30 were effective in reducing Salmonella on inoculated chicken carcasses both after treatment and during the storage at $4.4^{\circ}C$ for up to 6 days. In addition, reductions in psychrotroph counts indicated that these treatments might have the potential to increase the shelf-life of poultry carcasses.
