Isolation and Characterization of Lactobacillus buchneri Strains with High ${\gamma}$-Aminobutyric Acid Producing Capacity from Naturally Aged Cheese

  • Two lactic acid bacteria (LAB) with high ${\gamma}$-aminobutyric acid (GABA)-producing capacity were isolated from naturally aged cheese. Examination of the biochemical features using an API kit indicated that the two strains belonged to Lactobacillus. They were gram positive, rod-type bacteria, and fermented arabinose, melezitose, melibiose and xylose, but did not utilize cellobiose or trehalose. 16S rDNA sequencing analysis confirmed that they were Lactobacillus buchneri and Lactobacillus sp. They were accordingly named as Lactobacillus buchneri OPM-1 and Lactobacillus sp. OPM-2, and could produce GABA from MRS broth supplemented with 10 g/L of monosodium glutamate (MSG) at a productivity of 91.7 and 116.7 mg/L/hr, respectively. Cell extracts of L. buchneri OPM-1 and Lactobacillus sp. OPM-2 showed glutamate decarboxylase (GAD) activity, for which the optimum pH and temperature were 5.5 and $30^{\circ}C$, respectively.

  • For investigation of the food safety and influence of biofilm on the bacterial quality of tap water, it was detected by selective media, and identified by API kit and MIDI system. Biofilm and effluent showed much more colonies than influent. Although there were many presumptive positive colonies, no pathogenic bacterium detected. However opportunistic pathogens, such as Pseudomonas aeruginosa and Aeromonas species, and members of family Enterobacteriaceae were detected. Compared to influent, biofilm and effluent showed more diverse bacteria. This study implies that there would be the negative influence of biofilm on the bacterial qualities of tap water.

  • 본 논문에서는 3차원 게임 물리 엔진의 개발을 지원하는 물리 시뮬레이터 및 개발도구에 대해 설계하였다. 3차원 게임 물리 엔진을 개발하는 과정에서 다양한 물리 현상을 실시간으로 테스트하여 인터렉티브한 게임에서의 물리효과와 사용자가 물리 시뮬레이터를 이용하여 테스트한 환경을 시뮬레이터 개발도구를 통해 API를 생성하여 라이브러리 형태로 제공함으로써 좀더 정확한 물리 현상 구현과 개발기간의 단축 및 개발 공정을 효율적으로 처리할 수 있도록 하였다.

  • The purpose of this research is to develop co-simulation methodology of codes developed in different modeling and simulation environment. We develop aerodynamic FMU(Functional Mock-up Unit) meeting FMI(Functional Mock-up Interface) specification version2 utilizing Legacy FORTRAN aerodynamic code based on unsteady vortex lattice method. It is concluded that making FMU is possible utilizing Legacy code made in any language which can be compiled and linked with object in FMI API coded in C language. This paper explains QTronic's method of using FMU SDK(Software Development Kit) and suggestion for using FORTRAN properly. Finally, we make articulated rotor/aerodynamics co-simulation by integrating aerodynamics FMU and rotor FMU developed by Modelica.

  • In this study we isolate lactic acid bacteria from kimchi, which were identified as pediococcus pentosaceus and Lactobacillus bevis by using API 50 CHL Kit, some morphological and physiological tests, The water extract of green tea(50, 100, 500, 1000ppm) inhibieted the growth of pediococcus pentosaceus and Lactobacillus brevis in broth system. Furthermore, their time were prolonged to about 4hours at 500 to 1000ppm. The leakage amount of intracellular materials and Mg++ in tested bacteria cells increased with increasing concentration of green tea water extract.

  • A bacteriocin-producing lactic acid bacteria was isolated from contaminated milk products, which was identified by using the API50 CH kit as Lactococcus lactis subsp. lactis with reliability of 98%. Fatty and analysia of the cell membrane showed that this strain contained same fatty acids profiles as type strain, Lactococcus lactis subsp. lactis ATCC 19435. The bacteriocin of Lactococcus sp. HY 449 showed relatively wide range of inhibition spectrum against gram positive and some gram negative bacteria such as Escherichia coli and maintained the inhibitory activity between pH2.0 and pH9.0 The thermostability of this bacteriocin was higher in acidic solution than in distilled water and was stable at 60$\circ $C for 1 hour.

Description of unrecorded wild yeasts from soil in Republic of Korea under cold conditions

  • The purpose of this study was to isolate and identify wild yeasts from soil collected in Daegu City and Cheongyang County, Republic of Korea. Among 11 strains isolated in this study, nine strains were previously reported and two strains were unreported in Republic of Korea. To identify wild yeast strains, pairwise sequence comparisons of the D1/D2 region of the 26S rRNA gene sequence were done using Basic Local Alignment Search Tool (BLAST). The cell morphologies were observed by phase contrast microscope and assimilation test are done using API 20C AUX kit. All strains were assigned to the phylum Basidiomycota. Of the two unrecorded yeast strains, CY-9-10C belongs to the genus Mrakia (family Mrakiaceae, order Cystofilobasidiales, class Tremellomycetes) and PG3-4-10C belongs to the genus Slooffia (family Chrysozymaceae, order Microbotryomycetes incertae sedis, class Microbotryomycetes). Both strains had oval-shaped and polar budding cells. This research described the morphological and biochemical properties of the two unreported yeast species that had not officially reported in Korea.

  • The purpose of this study is to isolate and identify wild yeasts from the soil samples collected in Daegu and Daejeon City, Republic of Korea. Among 15 strains isolated in this study, 13 strains were previously reported and two strains had not been reported in Republic of Korea. To identify wild yeast strains, pairwise sequence comparisons of D1/D2 region of the 26S rRNA gene sequence were done using Basic Local Alignment Search Tool (BLAST). The cell morphologies were observed by phase contrast microscope and assimilation tests were done using API 20C AUX kit. All strains were assigned to the phylum Basidiomycota. The two unrecorded yeast strains, PG2-2-10C and DJ2-14-10C, belong to the genus Holtermanniella (family Holtermanniaceae, order Holtermanniales, class Tremellomycetes) and Goffeauzyma (family Filobasidiaceae, order Filobasidiales, class Tremellomycetes), respectively. The two unrecorded yeast strains had oval shape and polar budding cells. This research describers the morphological and biochemical properties of the two unreported yeast species that had not officially reported in Korea.

  • The purpose of this study was to isolate and identify wild yeasts from soil samples collected in Seoul and Daejeon, Republic of Korea. To identify wild yeast strains, pairwise sequence comparisons of D1/D2 region of the 26S rRNA gene sequence were done using Basic Local Alignment Search Tool (BLAST). The cell morphologies were observed by phase contrast microscope and carbon source assimilation test were done using API 20C AUX kit. Among the 13 isolated strains, 11 strains were previously reported, but two strains have never been reported from Republic of Korea. The 13 strains were assigned to the phylum Basidiomycota. The two unrecorded yeast strains B2UV-201 and DJ1-5-B-10C belong to the genera Rhodotorula and Rhodosporidiobolus, respectively. The two unrecorded yeast strains are oval shaped and polar budding cells. This research focuses on the morphological and biochemical properties of the two unreported yeast species that have not officially been reported in Korea.
