GAPCON-THERMAL-2 Revision2 코드를 이용한 핵분열 생성물 방출 모델 비교 연구

  • 핵분열 생성물 방출량을 계산하는 모델들에 대한 비교 분석을 위해 GAPCON-THERMAL-2 Revision 2 (GT2R2) 코드를 이용하여 Beyer-Hann , Beyer-Hann with NRC High Burnup Correction, ANS5.4와 Modified ANS5.4 핵분열 생성물 방출 모델들을, RISO-M2-2C 핵연료봉의 실험결과와 비교하였다. Beyer-Hann 모델은 실험결과보다 낮게 예측한반면 ANS5.4 모델은 실험결과 보다 높게 예측하였다. 한편 NRC High Burnup Correction을 한 Beyer-Hann 모텔과Modified ANS5.4 모델은 실험 결과와 비슷한 방출비를 예측하였다. 이러한 결과를 확인하기 위해 국부적인 핵연료 온도와 연소도를 검토한 결과 ANS5.4 모델이 .Modified ANS5.4 모델보다 온도와 연소도에 따라 더 민감한 반응을 보이고 있으며, Beyer-Hann 모텔은 연소도 영향이 없이 각 온도 영역에서 일정하였고, Beyer-Hann with NRC High Burnup Correction 모델은 20,000MWd/MTU 연소도 이상영역에서 연소도 영향을 보이고 있다.

Interaction of ${\alpha}$-Ketoglutarate Dehydrogenase Complex with Allosteric Regulators Detected by a Fluorescence Probe, 1,1'-bi(4-aniline)naphthalene-5,5'-disulfonic acid, an Inhibitor of Catalytic Activity

  • The interaction of ${\alpha}-ketoglutarate$ dehydrogenase complex (${\alpha}-KGDC$) with a hydrophobic fluorescent probe [1,1'-bi(4-aniline)naphthalene-5,5'-disulfonic acid] (bis-ANS) was studied. The punfied ${\alpha}-KGDC$ was potently inhibited by bis-ANS with an apparent half maximal inhibitory concentration ($IC_{50}$) of 9.8 ${\mu}m$ at pH 8.0. The catalytic activities of both the E1o and E2o subunits were predominantly inhibited while that of the E3 component was hardly affected. The binding of bis-ANS to the enzyme caused a marked enhancement and blue shift from 523 nm to 482 nm in the fluorescence emission spectrum. The dissociation constant ($K_d$) and the number of binding sites (n) were calculated to be 0.87 mM and 158, respectively. Allosteric regulators such as purine nucleotides and divalent cations further increased the fluorescence intensity of the $bis-ANS-{\alpha}-KGDC$ binary complex. These data suggest that the binding of these allosteric regulators to ${\alpha}-KGDC$ may cause the conformational changes in the enzyme and that bis-ANS could be used as a valuable probe to study the interaction of the multi-enzyme complex and its allosteric regulators.

수용성 폴리파라시클로판류와 약물과의 상호작용(제 3보)-수용액 중 수용성 폴리파라시클로판류와 형광 소수 나프탈렌 유도체류와의 복합체 형성- (Interactions between Water-Soluble Polyparacyclophanes and Drugs (III) -Complex Formation of Water-Soluble Polyparacyclophanes with Fluorescent Hydrophobic Naphthalene Derivatives in Aqueous Solution-)

  • Complex formation of water-soluble polyparacyclophanes bearing two diphenylmethane or two diphenyl ether skeletons with l-anilinonaphthalene-8-sulfonate (ANS) and 2-p-toluidinylnaphthalene-6-sulfonate (TNS) was investigated quantitatively to develop useful host compounds comparing with ${\alpha}\;-\;and\;{\beta}-cyc1odextrins$$({\alpha}-\;and\;{\beta}-CyDs$) in aqueous solution. Benesi-Hildebrand type analysis of the fluorescent intensity showed that the dissociation constants (Kd) of paracyclophane-ANS complexes were $1.55\;{\times}\;10^{-4}M$ for 1,6,20,25-tetraaza[6.1.6.1]paracyclophane(CPM 44) and $1.23\;{\times}\;10^{-4}M$ for 1,7,21,27-tetraaza[7.1.7.1]paracyclophane (CPM 55), and those of paracyclophane-TNS complexes were $6.99\;{\times}\;10^{-6}M$ for CPM 44 and $6.23\;{\times}\;10^{-5}M$ for CPM 55, in 1:1 molar ratio. On the other hand, the Kd values of 1,7,21,27-tetraaza-14,34-dioxa[7.1.7.1]paracyclophane (CPE 55)-ANS, 1,8,22,29-tetraaza-15,36-dioxa[8.1.8.1]paracyclophane (CPE 66)-ANS, CPE 55-TNS, CPE 66-TNS complexes were $1.75\;{\times}\;10^{-3}M$, $3.07\;{\times}\;10^{-3}M$, $3.75\;{\times}\;10^{-3}M$ and $2.15\;{\times}\;10^{-3}M$, respectively. On the contrary, the Kd values of ${\alpha}-CyD-ANS$, ${\beta}-CyD-ANS$, ${\alpha}-CyD-TNS$ and ${\beta}-CyD-TNS$ complexes were found to be $3.98\;{\times}\;10^{-2}M$, $1.05\;{\times}\;10^{-2}M$, $1.38\;{\times}\;10^{-2}M$ and $3.52\;{\times}\;10^{-4}M$, respectively. These results mean that the complexation of CPMs with ANS or TNS is by 5.6-1,975 fold stronger than that for ${\alpha}-or\;{\beta}-CyDs$, and the complex formation of CPEs with ANS or TNS is nearly same as or somewhat stronger than that for ${\alpha}-or\;{\beta}-CyDs$. From the Kd values determined at different temperatures, thermodynamic parameters were calculated and the complexation was found to be a spontaneous exothermic reaction. The effects of pH on Kd values of CPM 44-ANS, and CPM 55-ANS complexes were negligible in the range of pH 1.2-1.8. However, the Kd values of these complexes increased significantly with increasing ionic strength.

대두 펩타이드의 표면소수도가 흰쥐의 혈청 콜레스테롤 농도 및 분변 스테로이드의 배설량에 미치는 영향 (Effect of Surface Hydrophobicity of Soybean Peptides on the Concentration of Serum Cholesterol and Fecal Steroid Excretion in Rats)

  • 대두 펩타이드의 표면소수도가 혈청 콜레스테롤의 농도에 미치는 영향을 조사하기 위하여, 대두 단백질(ISP), 카세인(CNP), 이들 단백질을 펩신으로 가수분해하여 pH에 따른 펩타이드 침전 획분들(SHT, SH8, SH6, SH4, CHT, CH6, CH5, CH4)을 흰쥐에 섭취시키고 혈청 콜레스테롤 농도 및 분변 스테로이드이 배설량을 측정하였다. 그리고 각 펩타이드의 표면소수도를 ANS 형광법 및 SDS 결합법으로 측정하여, 이들의 상관관계를 분석한 결과, 펩타이드의 ANS 표면소수도가 높아질수록 분변으로 배설된 스테로이드량은 증가하였으며(r=0.81), 혈청 콜레스테롤 농도는 낮아졌다.(r=-0.868). 그러나 SDS 표면소수도는 그들과 상관관계가 없었다. 또한 대두 단백질은 가수분해에 의하여 ANS 표면소수도가 증가하였다. 이상의 결과는 흰쥐의 담즙염이, 소화중 생성된 높은 표면소수도의 펩타이드와 결합하여 체외로 배출되므로서 대두단백질의 섭취에 의한 혈청 콜레스테롤 농도가 낮아짐을 시사하였다.

  • The purpose of this study is to obtain the enshortening and elongation rate of image in orthopantomograph. The subjects were consisted of 15 dry skulls attached with radiopaque materials on each anatomical points. The length measurements were performed between two points on dry skull, and between two points on film. The results are as follows: 1. The enshortening and elongation rate between two anatomical points (1) ANS↔infraorbital foramen (left:+1.3%, right:+0.7%) (2) ANS↔maxillary tuberosity(left:-11.7%, right:-14.3%) (3) Zygomatic arch length(left:-4.8%, right:-8.9%) (4) first molar↔infraorbital foramen (left:+19.8%, right:+24%) (5) inter-infraorbital foramen length(-21.4%) (6) inter-first molar length (-31.8%) (7) inter-mental foramen length(+1.4%) (8) mental foramen↔mandible angle (left:+3.3%, right:+3.3%) (9) mental foramen↔lingula(left:+8.2%, right:+3.3%) (10) mental foramen↔condyle head(left:+5.2%, right:+4.5%) (11) mandible↔condyle head (left:+15.4%, right:+16.4%) 2. The closer the object is to the occlusal plane and the median line, the smaller it appears.

RSA분석과 자율신경기능을 평가하는 호흡주기 설정에 관한 연구 (A Study on an Optimal Respiration Rate for the ANS Assessment based on RSA Analysis)

  • Heart rate variability(HRV) is the clinical consequence of various influences of the autonomic nervous system(ANS) on heart beat. HRV can estimate the potential physiologic rhythm from the interval between consecutive beats(RR interval or HRV data), but cardiovascular system governed by ANS is in relation to respiration and autonomic regulation. It is known as RSA representing respiration-related HR rhythmic oscillation. Because the mechanism linking the variability of HR to respiration is complex, it has so far been unknown well. In this paper, we tried to evaluate 5-min RR interval segments under control of respiration in order to find out a proper respiration rate that can estimate the ANS function. 10 healthy volunteers were included to evaluate 5-min HRV data under 4 different respiration-controlled environments; 0.03Hz, 0.1Hz, 0.2Hz, and 0.4Hz respiration. HRV data were analyzed both in the frequency and the time domain, with cross-correlation coefficient(cross-coeff.) for HRV and respiration signal. The results showed maximum cross-coeff. of 0.84 at 0.1 Hz and minimum that of 0.16 at 0.4Hz respiration. Cross-coeff was decreased at a faster rate from 0.1Hz respiration. All mean SDNN, RMSSD, and pNN50 of time domain measures were 108.7ms, 71.85ms, and 28.47%, respectively, and LF, HF, and TP of frequency domain measures were $12,722ms^2,\;658.8ms^2$, and $7,836.64ms^2$ at 0.1Hz respiration, respectively. In conclusion, 0.1Hz respiration was observed to be very meaningful from time domain and frequency domain analysis in relation to respiration and autonomic regulation of the heart.

우리나라 미승인 유전자변형 장미의 duplex PCR검출법 (Detection Method for Unapproved Genetically Modified Rose Plants in Korea Using Duplex Polymerase Chain Reaction)

  • 유전자변형 장미 개발에 이용된 형질전환 벡터 pSPB130을 검출할 있는 duplex PCR이 개발되었다. 유전자변형 장미를 검출하기 위해 anthocyanin synthase($ANS$)가 장미 내 재유전자로 PCR 검사법에 이용하였다. 107bp의 PCR 산물을 얻을 수 있는 RHANS-KF/KR primer가 ANS 유전자를 증폭시키는데 이용되었고, 이것을 이용하여 9개의 다른 식물에 대해서 PCR한 결과 장미에서만 특이적인 증폭산물이 확인되었다. GMRH-KF/KR primer가 형질전환 벡터 pSPB130에 삽입된 35S promoter와 flavonoid 3',5'-hydroxylase($F3^{\prime}5^{\prime}H$) 사이의 염기서열을 확인할 수 있도록 제작되었다. 본 연구에서 개발된 duplex PCR의 검정한계치는 약 0.5%이며, 이러한 duplex PCR이 우리나라 미승인 유전자변형 장미를 모니터링 하는데 유용하게 사용될 수 있을 것이다.

전치부 반대교합 아동의 비강통기도에 관한 연구 (A STUDY ON NASAL RESPIRATORY PATENCY IN THE GROWING CHILDREN WITH ANTERIOR CROSSBITE)

  • This study was designed to analyze nasal respiratory patency and its correlation with skeletal components in growing children with anterior crossbite. The subjects consisted of 40 control patients, 24 nose breathers with anterior crossbite and 18 mouth breathers with anterior crossbite. The mean age was 11.4 years in the control group, 10.1 years in nose breathing group and 9.5 years in mouth breathing group. The results were as follows, 1. In anterior cross bite group, and nasal respiratory airflow rates (N.R.A.R.) was significantly lower than that of control group regardless of nasal decongestants application. 2. The N.R.A.R. of mouth breathers with anterior crossbite in male group was significantly lower than that of mouth breathers, but increased to the level of control group after nasal decongestants application. But in female group, the N.R.A.R. was significantly lower in mouth breathing group at both conditions. 3. Mouth breathing group showed smaller anterior vertical nasal cavity height (ANS-ANS'), lower upper anterior facial height ratios (N-sp'/N-Me) and higher maxillary occlusal plane ratios (OL-ML/ML-NL) than those of nose breathing group with anterior crossibte. 4. Items showing nasal height (ANS-ANS', PNS-PNS'), anterior upper facial height (N-sp') was were strongly correlated with N.R.A.R. at 150 pascal in inspiration. But item showing maxillary occlusal plane ratios (OL-ML/ML-NS) was negatively correlated with N.R.A.R. at 150 pascal in inspiration. 5. There were forward tongue position in mouth breathing group, but it was not significantly correlated with N.R.A.R. at 150 pascal in inspiration.

영광 3,4호기 시뮬레이터 실시간 NSSS모델 개발 및 검증

  • 운전원 훈련용 시뮬레이터의 국산화 개발계획에 따라 영광 원자력 3,4호기 발전소 운전원 훈련용 시뮬레이터가 국내 최초로 개발되었다. STK(space and time kinetics)와 RETACT (Real Time Advanced Core and Thermohydraulics)코드를 이용하여 영광 3,4호기 시뮬레이터의 실시간 NSSS 모델을 생성하였다. 생성된 모델의 검증(Verification Validation)을 위해 정상상태(Steady State)에서의 주요인자들이 ANS3.5의 오차범위이내임을 확인하였다. 과도상태(Transient)의 검증을 위해 터빈정지 과도상태와 주증기 관파열(Main Steam Line Rupture)사고를 실제 발전소 시험 데이터 및 성능해석 코드(NPA)를 이용하여 분석한 결과와 비교하였다. 비교 결과 기준발전소의 반응과 큰 차이 없이 운전원 훈련용 시뮬레이터의 규격인 ANS 3.5를 잘 만족함을 확인 할 수 있었다.
