Isolation of an Autonomously Replicating DNA Sequence from Aspergillus nidulans

  • Using yeast, Saccharomyces cerevisiae, and the integrate vector system, we have isolated and characterized an autonomously replicating sequence (ARS) from Aspergillus nidulans. The DNA fragment, designated ANR1, is 5.0 kb in size and maintained free from the chromosome in S. cerevisiae. The YIplac211-ANR1 recombinant plasmid, which consists of sequences derived from the yeast integrative vector YIplac211 and 5.0 kb ANR1 fragment, showed a 104-fold enhancement in transformation efficiency over that found for YIplac211, and was easily recovered from the transformed yeast. Genetic analysis of transformants showed that YIplac21-ANR1 could be over 96% cured when cultured over 20 generations in complete medium and thus suggests that this sequence is mitotically unstable. In A. nidulans, recombinant plasmid PILJ16-4.5 which carries the 4.5 kb EcoRI fragment of ANR1 showed a 170-fold enhancement in transformation efficiency compared to that of the integrative vector PILJ16.

Identification of Korean Strawberry Cultivars using DNA markers (DNA 표지를 이용한 딸기 국내 육성 품종 판별)

  • This study was conducted to develop the DNA markers for identification of the strawberry cultivars in Korea and Japan. We developed fifteen cleaved amplified polymorphic sequence (CAPS) markers based on the Fragaria gene sequences. Among them six CAPS markers showed polymorphism exclusively in one cultivar. Five CAPS markers (ANR-MspI, ANR-BamHI, ACO-HinfI, DFR-AseI, FGT-MspI) provided enough polymorphism to identify eight Korean strawberry cultivars except for 'Maehyang' and 'Sunhong'. To complement the fifteen CAPS markers, we selected another fifteen sequence-related amplified polymorphism (SRAP) and one of them, me1/em5_460bp marker, made it possible to discriminate between 'Maehyang' and 'Sunhong'. Therefore, application of the five CAPS markers and one SRAP marker were sufficient to identify the nineteen Korean and Japanese strawberry cultivars. These markers could be used practically for cultivar identification of Korean and Japanese strawberry.

  • The conversion of mechanical energy into electrical energy at nanoscale using piezoelectric nanowire arrays has been in detail shown by deflection of nanowires. Recently it has performed an analytical model, both at classical and at quantum level, for describing the most important quantities concerning transport phenomena; the model predicts interesting peculiarities, as high initial charge diffusion in nanodevices constituting by nanowires and permits also in particular to deduce interesting informations about the devices sensitivity, focusing on the correlation between sensitivity and high initial diffusivity of these materials at nanometric level.

  • Forced vibration analysis of a cracked functionally graded microbeam is investigated by using modified couple stress theory with damping effect. Mechanical properties of the functionally graded beam change vary along the thickness direction. The crack is modelled with a rotational spring. The Kelvin-Voigt model is considered in the damping effect. In solution of the dynamic problem, finite element method is used within Timoshenko beam theory in the time domain. Influences of the geometry and material parameters on forced vibration responses of cracked functionally graded microbeams are presented.

  • We have obtained opal-like photonic crystals based on opals and inverted opals exhibiting a shift of the selective reflection band toward longer and shorter wavelengths with respect to the diffraction band of the initial opal consisting of $SiO_2$ spheres. The contribution of frames forming three-dimensional periodic structures and that of fillers to the spectral arrangement of the diffraction bands has been determined.

  • We study a device structure which can be used to generate pure valley current and valley polarized current using quantum adiabatic pumping. The design of the structure allows the flexibility of changing the structure from one for pure valley current generation to one for valley polarized current generation by changing the applied electric potentials through changing the symmetry of the structure. The device is useful for the development of valleytronic devices.
