치구계획의 자동화시스템 구성 및 데이텀 체계의 결정 (System Architecture and Datum Reference Frame for Computer Aided Fixture Planning System)

  • This paper deals with the development of a computer aided fixture planning system that automatically selects set-ups, set-up sequence and fixture design for prismatic parts. This study presents the hierarchical data structure for feature-based part model and the preprocessing procedure for the proposed system. The preprocessing procedure generates tools such as DDR(Degree of Dimensional Relationship), AMV(Admissible Misalignment Value) and the datum reference frame of each feature according to the proposed decision table. The proposed system is called AFIX(Automated FIXture planning system) which is implemented by using C language on the workstation. A case study for a cavity plate is presented to show the performance of the AFM.

  • KMA successfully began to receive and utilize the GOES-9 GVAR data since May 22nd 2003 when GOES-9 replaced the long-lived GMS-5 for Western Pacific and East Asian region until operation of MTSAT-1R in 2004. To take advantage of improvements of the GOES-9 data over the GMS-5 data, such as the increase of the temporal and spat ial resolution and addition of 3.9${\mu}$m channel, we have improved several algorithms to derive the meteorological products. Here we show two examples of algorithms, sea surface temperature and atmospheric motion vector, and preliminary results of validation of the improved algorithm.

내병성 관련유전자의 운반체 재조합 및 인삼(Panax ginseng C.A. Meyer)의 형질전환 (Vector Construction and Transformation of Ginseng (Panax ginseng C.A. Meyer) Using Disease Resistant Genes)

  • 인삼의 형질전환 시스템의 개발과 내병성관련 유전자의 도입에 관한 연구의 일환으로 soybean에서 cloning한 chitinase 유전자와 내병성 관련유전자(DR gene)가 식물에서 발현 및 형질전환될 수 있도록 운반체를 재조합하여 Agrobacterium을 이용해서 인삼에 형질전환시키고자 수행하였다. 식물세포에서 발현될 수 있는 promoter(35S-35S-AMV)에 내병성유전자인 DR-49 gene을 부착한 후 다시 식물형질전환용 binary vector에 도입하여 인삼에 도입되어 발현될 수 있도록 재조합하였으며, Disarmed Ti-plasmid 함유 Agrobacterium에 내병성유전자인 chitinase(pCH18)와 disease resistant gene(pDR)가 도입되었다. 인삼 callus 배양 과정을 거치지 않고 바로 multi-shoots를 생산하여 형질전환체 획득에 사용하고자 인삼 embryo와 petiole를 이용하여 direct shoots 생산을 유도한 결과 2,4-D 1 mg/ι와 kinetin 0.5 mg/ι 복합처리구에서 multi-shoot가 형성되었다. 또한 인삼 자엽을 이용한 형질전환에서도 MS 기본배지에 2,4-D 1 mg/ι와 kinetin 0.5 mg/ι를 첨가한 복합처리구에서 가장 효과적이었으며, dieases resistant와 chitinase 유전자에 의한 형질전환율은 각각 14%,그리고 18%를 나타내었다. 자엽으로부터 유기된 형질전환된 조직은 pre-embryoid 상태이기 때문에 성숙배의 과정을 거쳐 정상적인 shoot의 형성이 필요하였다.

국내 지황에 발생하는 바이러스병 발생 현황 (The Incidence of Virus Diseases in Rehmannia glutinosa in Korea)

  • 최근 국내 지황에서는 Rehmannia mosaic virus (ReMV), Youcai mosaic virus (YoMV), Broad bean wilt virus 2 (BBWV2), Plantago asiatica mosaic virus (PlAMV), Rehamnnia virus 1 (ReV1) 등 5종의 바이러스 감염이 확인되었다. 국내 주요 지황 재배지역의 포장에서 지황 바이러스의 발병률을 조사하기 위해 2017년부터 2018년까지 여섯 지역의 포장에서 바이러스병 조사를 실시하였다. 지황에 나타나는 주요 바이러스 증상은 잎에 모자이크와 노란 반점 및 변색이었으며 생육이 저조해지는 증상이 관찰되었다. 총 145개의 시료를 채집하여 5종 바이러스에 대한 유전자 진단을 실시한 결과, 전 시료에서 2종 이상의 바이러스가 복합 감염되어 있음을 확인하였다. 지황은 뿌리로 영양번식하는 재배 특성 상 품종별 종근에서도 바이러스 감염여부를 조사하였으며, 종근에서도 5종의 바이러스가 검정됨에 따라 지황에서의 바이러스병 관리를 위해서는 무병 종근의 생산 및 보급이 매우 중요할 것으로 판단된다.

Expression of gus and gfp Genes in Ggrlic (Allium sativum L.) Cells Following Particle Bombardment Transformation

  • The activity of promoter sequences was evaluated in garlic cells using the $\beta$-glucuronidase (GUS) gene as a reporter. Histochemical GUS assay indicated transient GUS activity in leaf, callus and root cells 48 hours after particle bombardment transformation. Quantitative fluorometric assays in extracts of transformed leaves demonstrated that the CsVMV promoter induced the highest level of gene expression, which was, on average, ten fold the level induced by CaMV35S and by the Arabidopsis Act2 promoters and two fold the level expression observed with a construct containing a double CaMV35S plus the untranslated leader sequence from AMV. No activity or very low levels were observed when cells were transformed with plasmids rontaining the typical monocot promoters, Actl, from rice or the Ubi-1, from maize. The green fluorescent protein (GFP) was also tested as a marker gene for garlic transformation. Intense fluorescence was observed in leaf, callus and root cells transformed with a construct containing the gfp gene under control of the CaMV35 Promoter. No fluorescence was detected when the gfp was under control of the Ubi-1 promoter.

Status and Prospects of Marine Wind Observations from Geostationary and Polar-Orbiting Satellites for Tropical Cyclone Studies

  • Satellite-derived sea surface winds (SSWs) and atmospheric motion vectors (AMVs) over the global ocean, particularly including the areas in and around tropical cyclones (TCs), have been provided in a real-time and continuous manner. More and better information is now derived from technologically improved multiple satellite missions and wind retrieving techniques. The status and prospects of key SSW products retrieved from scatterometers, passive microwave radiometers, synthetic aperture radar, and altimeters as well as AMVs derived by tracking features from multiple geostationary satellites are reviewed here. The quality and error characteristics, limitations, and challenges of satellite wind observations described in the literature, which need to be carefully considered to apply the observations for both operational and scientific uses, i.e., assimilation in numerical weather forecasting, are also described. Additionally, on-going efforts toward merging them, particularly for monitoring three-dimensional TC wind fields in a real-time and continuous manner and for providing global profiles of high-quality wind observations with the new mission are introduced. Future research is recommended to develop plans for providing more and better SSW and AMV products in a real-time and continuous manner from existing and new missions.

장애함수법에 의한 신뢰성기반 최적설계 (Barrier Function Method in Reliability Based Design Optimization)

  • The need to increase the reliability of a structural system has been significantly brought in the procedure of real designs to consider, for instance, the material properties or geometric dimensions that reveal a random or incompletely known nature. Reliability based design optimization of a real system now becomes an emerging technique to achieve reliability, robustness and safety of these problems. Finite element analysis program and the reliability analysis program are necessary to evaluate the responses and the probabilities of failure of the system, respectively. Moreover, integration of these programs is required during the procedure of reliability based design optimization. It is well known that reliability based design optimization can often have so many local minima that it cannot converge to the specified probability of failure. To overcome this problem, barrier function method in reliability based design optimization is suggested. To illustrate the proposed formulation, reliability based design optimization of a bracket is performed. AMV and FORM are employed for reliability analysis and their optimization results are compared based on the accuracy and efficiency.

Synthesis and base pairing properties of DNA-RNA heteroduplex containing 5-hydroxyuridine

  • 5-Hydroxyuridine (5-OHU) is a major lesion of uridine and cytosine produced in RNA by various chemical oxidants. To elucidate its biochemical and biophysical effects on RNA replication, the site-specifically modified oligoribonucleotides containing 5-OHU were synthesized with C5-hydroxy-5'-ODMTr-2'-TBDMS-uridine phosphoramidite using automated solid phase synthesis. The base-pairing properties of nucleotides opposite 5-OHU in 24 mer oligoribonulcleotides with dNTP were studied using three reverse transcriptases (Super-$Script^{TM}II$-, AMV-, MMLV-RT) in cDNA synthesis. Adenine as well as guanine was incorporated preferentially by all reverse transcriptases. In the UV-melting temperature experiment, the results from the relative stabilities of the base pairs were A : 5-OHU > G : 5-OHU > T : 5-OHU $\approx$ C : 5-OHU. Circular Dichroism (CD) studies showed that DNA-RNA containing 5- OHU heteroduplexes exhibit a similar conformation between the A-type RNA and B-type DNA. These results suggest that 5- OHU from oxidative damage was mainly influenced by adenine mismatch.

A study on the patterns of expression of the DAZ and HSP genes in the testicular tissue of men with azoospermia

  • Spermatogenesis is known to be regulated by a number of genes and several factors such as hormones, growth factors, cytokines and others. This study was done to evaluate the relationship between HSPs and DAZ genes in human spermatogenesis; we observed the expression pattern of HSP gene in azoospermia men with DAZ gene that regulated the gene expression related with human spermatogenesis. RT-PCR method was used to detect DAZ, HSP70A, and HSP70B transcripts in all RNA samples. Total RNA was extracted from 21 testis tissues using TRIZOL reagent. cDNAs were synthesized with reverse transcriptase, AMV. All PCR reaction were performed on a PCR themocycler with DAZ, HSP70A, and HSP70B-specific primers. Semen analysis, karyotyping and testis histology were performed. DAZ gene, known as a candidate gene of azoospermia factor(AZF), was deleted in 2 of 21 patients. To evaluate the only effects of HSPs in this patients, 2 DAZ deleted cases were removed. We observed the mRNA of HSP70B in 5 whereas none could be seen with regard to HSP70A. Furthermore, the sperm of these 5 men were discovered to be immature. In conclusion, HSP70B as well ad DAZ gene seem to be involved causing spermatogenic failure. We suggest that HSP70B plays an important role in spermatogenesis and it is one of factors induced sperm maturation in human.

Occurrence and Distribution of Viruses Infecting Pepper in Korea

  • We conducted a survey on pepper virus diseases in 31 regions in Korea from November 2001 to December 2004. Using electron microscopy, test plant reaction, rapid immuno-filter paper assay (RIPA), reverse transcription-polymerase chain reaction (RT-PCR) and/or analysis of viral nucleotide sequences, we found a number of viruses from 1,056 samples that we collected. These included Cucumber mosaic virus (CMV), Pepper mottle virus (PepMoV), Pepper mild mottle virus (PMMoV), Broad bean wilt virus 2 (BBWV2), Tobacco mild green mosaic virus (TMGMV), and Tomato spotted wilt virus (TSWV). Of the samples analyzed, $343(32.5\%)$ were infected with CMV, $209(19.8\%)$ with PepMoV, $141(13.4\%)$ with PMMoV, $12(1.1\%)$ with BBWV2, $40(3.8\%)$ with TMGMV, $5(0.5\%)$ with TSWV, $153(14.5\%)$ with CMV and PepMoV, $54 (5.1\%)$ with CMV and PMMoV, $31(2.9\%)$ with PepMoV and PMMoV, $3(0.3\%)$ with CMV and BBWV2, $1(0.1\%)$ with CMV, PepMoV and BBWV2, $8(0.8\%)$ with CMV, PepMoV and PMMoV, and $30 (2.8\%)$ samples were infected with viruses which were not identified. CMV was the most predominant virus in all inspected fields and the number of the samples infected with PMMoV was relatively low as compared PepMoV infection level in pepper. TMGMV was only found in the southern part of Korea, while TSWV was isolated in Anyang and Yesan. However, we did not encounter in this survey the Alfalfa mosaic virus (AMV), Potato virus Y (PVY), Tobacco mosaic virus (TMV), and Pepper vein chlorosis virus (PVCV).
