사염화탄소 투여로 인한 랫드의 급성 간 손상에 대한 루틴의 예방 효과 (Protective Effect of Rutin on Carbon Tetrachloride-Induced Acute Hepatotoxicity in Rats)

  • This study was performed to investigate hepatoprotective effect of rutin on acute hepatic damage induced by carbon tetrachloride in rats. Twenty-four male Sprague-Dawley rats were randomly divided into four groups; normal control group, $CCl_4$ control group, two rutin treatment groups (rutin 200+$CCl_4$ and rutin 400+$CCl_4$). Dissolving vehicles were applied to the rats in the normal control group. The rutin was administrated to the rats in rutin 200+$CCl_4$ and rutin 400+$CCl_4$ groups at the levels of 200 mg/kg and 400 mg/kg, 3 consecutive days orally, with 24 hours interval before inoculating $CCl_4$. $CCl_4$ was intraperitoneally administered an hour after the last treatment of rutin to the rats in every group except the normal control group. The body weight of rats in $CCl_4$ control group were significantly lower than other groups (p < 0.05), but the liver weight and relative liver weight were higher than normal control group (p < 0.05). The activities of ALT, AST, ALP, GTP, LDH and the level of total bilirubin in sera of rats in $CCl_4$ control group were higher and the levels of total protein, albumin and globulin were lower than the normal group (p < 0.05). The activities of ALT, AST, ALP, GTP, LDH and the level of total bilirubin in rutin 200+$CCl_4$ and rutin 400+$CCl_4$ groups were lower than $CCl_4$ control group (p < 0.05). Therefore the pre-treatment of rutin before $CCl_4$ inoculation in rats effectively inhibited the elevation of serum ALT, AST and total bilirubin which are the parameters of hepatic damage.

발효강화쑥의 간장해 보호효과 (Hepatoprotective Effect of Fermented Artemisia princeps PAMPANINI by Lactic Acid Bacteria)

  • Artemisia princeps PAMPANINI has been used in traditional medicine of the treatment of inflammatory, liver dysfunction and order disorder in the far east countries including Korea. The present study was carried out to investigate the hepatoprotective effects of ethanol extract of Artemisia princeps (AP) and its fermented agents (AP-F) by lactic acid bacteria derived from human intestinal bacteria on liver injured rat induced by $CCl_4$ and d-galactosamine. Hepatoprotective activity was monitored by estimating serum ALT, AST, ALP, LDH, superoxide dismutase (SOD), glutathione redeuctase (GR) and glutathione peroxidase (Gpx) activities in the liver injured by hepatotoxin. Pretreating rats with AP or AP-F at the same dosage regimen significantly suppressed the acute elevation of serum transaminase, ALP, LDH and GR activities, and significantly increased the lowering of blood SOD and GR activites induced by hepatoxin. Based on these findings, it is presumed that AP and APF may have the hepatoprotective effect on $CCl_4$ and d-galactosamine-induced hepatotoxicity rat.

A Study on the Mechanism of Oxidative Stress, Screening of Protective Agents and Signal Transduction of Cell Differentiation in Cultured Osteoblast and Osteoclast Damaged by Reactive Oxygen Species

  • It is well known that oxidative stress of reactive oxygen species (ROS) may be a causative factor in the pathenogenesis of bone disorder on osteoblast or osteoclast. The purpose of this study was to evaluate the cytotoxicity of oxidative stress, protective effect of glutamate receptor antagoinst against ROS-induced osteotoxicity, secretion of tumor necrosis factor $(TNF)-\alpha$ and the expression of c-fos gene in the cultured rat osteoblasts and osteoclasts. Cell viability by MTS assay or !NT assay, activity of glutathione peroxidase (GPx), lipid peroxidation (LPO) activity, protein synthesis by sulforhodamine B (SRB) assay, alkaline phosphatase (ALP) activity, lactate dehydrogenase (LDH) activity, MTS assay for NMDA (N-methyl-D-aspartate) receptor antagonist or AMPA/kainate receptor antagonist, measurement for $TNF-\alpha$, and c-fos gene expression were performed after these cells were treated with or without various cocentrations of xanthine oxidase (XO), hypoxanthine (HX), D-2-amino-5-phosphonovaleric acid (APV), 7-chlorokynurenic acid (CKA), 6-cyano-7-nitroquinoxaline-2,3-dione (CNQX) and 6,7-dinitroquinoxaline-2,3-dione (DNQX), respectively. In this study, XO/HX showed decreased cell viability and glutathione peroxidase (GPx) activity, but it showed increased LPO activity, $TNF-\alpha$ secretion and c-fos expression. APV and CKA incresed protein sythesis and ALP activity. While, CNQX or DNQX did not show any protective effect in LDH activity or cell viability. From these results, XO/HX showed cytotoxic effect in cultured rat osteoblast or osteoclast, and also NMDA receptor antagonist such as APV or CKA was effective in blocking XO/HX-induced osteotoxicity in these cultures.

Effects of polyphenols of Cocos nucifera husk fibreon selected indices of cardiovascular diseases in mice

  • Cocos nucifera (C. nucifera) oil is indigenously used to treat cardiovascular diseases. However, coconut husk fibre (which is rich in polyphenols) has not been screened for this property. Based on the ethnomedicinal use of polyphenols in treating cardiovascular diseases, this study was carried out to evaluate the effects of polyphenols of C. nucifera husk fibre on selected cardiovascular disease indices in mice. Fifty adult male Swiss albino mice were assigned randomly into five groups (A-E). Mice in groups B, C, D and E were administered 31.25, 62.5, 125, and 250 mg/kg body weight polyphenols of ethyl acetate extract of C. nucifera husk fibre respectively while the control group (A) mice received 5% DMSO for seven days. The mice were sacrificed twenty four hours after the last administration of polyphenols. Heart and plasma lactate dehydrogenase (LDH), alanine aminotransferase (ALT), aspartate aminotransferase (AST) and alkaline phosphatase (ALP) activities and plasma lipid profile were determined. Results revealed significant reduction (*p< 0.05) in plasma levels of total cholesterol and LDL-cholesterol with no significant change (*p> 0.05) in HDL-cholesterol, triglyceride and VLDL levels in the plasma at all doses of polyphenols administered compared to controls. There was significant reduction (*p< 0.05) in the activities of heart AST and LDH while plasma ALT, AST, and ALP activities were not significantly altered (*p> 0.05) at all doses of polyphenols administered compared to controls. These results suggest that the polyphenols of C. nucifera husk fibre possess cardio-protective properties and also indicate their possible use in the treatment of cardiovascular diseases.

인진(茵蔯)의 생식.발생 독성 시험 -배자(胚子)및 태자(胎子)발생을 중심으로- (Terato Genicity Test of Oral Administration of Artemisiae capillaris herba Extracts on New Zealand White Rabbit)

  • Objectives : The objective of this study was to access the effect of Artemisiae capillaris herba Extracts on the organogenetic reproductive toxicity of New Zealand White rabbits when administered by oral gavage. Methods : Gestational New Zealand White rabbits were dosed from 6 days of gestation to 18 days of gestation. It was conducted in accordance with the recommendations of the KFDA Guideline for Detection of Toxicity to Reproduction for Medicinal Products. In addition, serum AST, ALT, LDH and ALP levels were detected with their necropsy and histopathological observation Results: No significant changes of body weights, gains and food consumption were demonstrated in all dosed groups compared to those of vehicle control group except for 2,000mg/kg-dosing group, which showed significantly increase of body weight compared to that of vehicle control group. Normal gross findings were demonstrated in all tested groups. No significant changes of number of corpora lutea, implantation, implantation rate, number of fetal death, loss rate of post-implantation, number of live youngs at C-section, survival rate of post-implantation, number of male live youngs at C-section, number of female live youngs, sex ratio of live young, external anomalies of live youngs and body weight of live youngs were demonstrated in all dosed groups compared to those of vehicle control group. Although some visceral variations such as thymic reminant in the neck, dilation of renal pelvis, dilated ureter and malpositioned left common carotid artery, no significant changes were demonstrated in all dosed groups compared to that of vehicle control group. Some malformations and variation were demonstrated with retardations. However, no significant changes were demonstrated in all dosed groups compared to that of vehicle control group. In addition, similar ossification numbers were detected in sternebrae, metacarpals, metatarsals, phalanges, cervical vertebrae, and sacral and caudal vertebrae. No Artemisiae capillaris herba Extracts treatment-related changes of serum AST, ALT, LDH and ALP levels were demonstrated in all dosed levels in this study. Conclusions : We could conclude that oral administration of Artemisiae capillaris herba Extracts hasn't an influence on the organogenetic reproductive toxicity.

하고초산(夏枯草散)이 Thioacetamide에 의(依)한 백서(白鼠) 간손상(肝損傷)에 미치는 영향(影響) (Studies on the Effect of 'Hagocho-San Against Liver Damage in Thioacetamide Induced Rats)

  • To experimentally research out the effect of 'Hagocho-San' used for liver disease on the clinical treatment of oriental medicine, I induced liver damage rats by thioacetamide and dosed the extracts orally and measured the activity rate of GOT, GPT, ALP and LDH. The following results were obtained: 1) GOT activity was more decreased in the experimental group than in the control group. Extracts from mixed decoction was more efficacious than those mixed form of the extracts after individual decoction. Extracts from decoction showed continuous significance after 48 hours on, but mixed form of extracts after individual decoction was significant only after 48 hours and 96 hours. 2) GPT activity was more decreased in the experimental group than in the control group. Extracts from mixed decoction was effective after 48 hours on but mixed form of extracts after individual decoction was significant after 48 hours and 72 hours. 3) ALP activity was more decreased in the experimental group than in the control group. Extracts from mixed decoction was more efficacious than the mixed form of extracts after individual decoction and significance was verified after 72 hours on. 4) The significance of LDH activity in the extracts from mixed decoction was revealed after 72 hours and 96 hours but mixed form of extracts after individual decoction showed significance 72 hours later. 5) From the above results, it was showed that 'Hagocho-San' extract was effective on liver disease and extracts from mixed decoction was more effective than mixed form of extracts after individual decoction.

간수, 기문혈의 당귀 약침자극이 acetaminophen으로 유발된 흰쥐의 손상간에 미치는 영향 (Effects of Angelicae gigantis Radix Aqua-acupuncture at Ganshu(BL18) and Qimen(LR14) on Liver Damage induced by Acetaminophen in Rats)

  • This study was done in order to investigate the protective effects of A.G.R.(Angelicae gigantis Radix) aqua-acupuncture on acetaminophen induced liver damage in rats. The liver damage was induced by acetaminophen (500mg/kg) injection into the peritoneum. The A.G.R. aqua-acupuncture solution was injected into the corresponding loci to Ganshu($BL_{18}$) and Qimen($LR_{14}$) of human body and a blank locus of the root of tail on four consecutive times at 0, 3, 6, and 12 hours after acetaminophen injection. And the serum GOT, GPT, LDH, ALP activities, total bilirubin, direct bilirubin levels were measured in the rats. The serum GOT, GPT, LDH ALP activities and bilirubin level were decreased comparing with that of a control group in case of A.G.R. aqua-acupuncture treated group, specially Ganshu and Qimen aqua-acupuncture treated groups showed an obvious significant decrease.

D-Galactosamine으로 유발된 흰쥐의 간손상에 대한 치자두시탕 추출액이 미치는 영향 (Effect of the Chizadochi-Tang on Hepatotoxicity of D-Galactosamine in Rats)

  • Objectives : To investigate the hepatoprotective effect of Gardeniae Fructus (Ga) and Glycine semen preparatum (Gl) aqueous extract against D-galactosamine (D-GalN, 300mg/kg body weight) was administered to the male Sprague Dawley (SD) rats. Materials and Methods : The study was carried out on male SD rats (age matched, weight $250{\pm}10$ g). Experimental groups (Exp) divided four : Normal group (Nor) was administered saline, Control (Con) group was only received D-GalN (300 mg/kg) intraperitoneally. Exp was orally administered Ga (200 mg/kg; Ga group), Gl (700 mg/kg; Gl group), and Chizadochi-Tang (200 mg/kg+700 mg/kg, GG group) after D-GalN treatment during 14 days (n=6). Results : D-GalN administration induced hepatotoxicity in rats which was manifested by increased levels of aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP) and lactate dehydrogenase (LDH) but decreased total cholesterol (HDL C) and triglyceride (TG). The serum TG concentrations were significantly increased ($^{\sharp}p$ <0.05) in the Ga group compared with Con. AST and ALP activities were significantly decreased ($^{\sharp}p$ <0.05) in the all experimental groups compared with Con. ALT activities were significantly decreased ($^{\sharp}p$ <0.05) in the Ga group compared with Con. LDH activities were significantly decreased ($^{\sharp}p$ <0.05) in the GG group compared with Con. On the light microscopic study, a number of vacuole were observed in the Con, but decreased in experimental groups. Conclusion : Ga aqueous extract and Chizadochi-Tang extract possesses hepatoprotective potential, thus validating its use in alleviating toxic effects of D-GalN.

애엽 에탄올 추출물이 사염화탄소로 유발된 흰쥐의 간 손상 보호효과 (Protective Effect of Ethanol Extract of Artemisiae vulgaris L. on hepatic injury Induced by Carbon tetrachloride In Rat.)

  • Dawley 계 숫컷 흰쥐(200~210g)를 정상군, CCl4-대조군, CCl4-실험군으로 나누어 실험군은 애엽 에탄올 추출물을 1,000mg/kg,b.w의 용량으로 1일1회7일간 경구 투여 후 사염화탄소를 0.6mg/kg,b.w의 용량으로 복강내 주사 후 다음날 개복하여 혈청내의 Alanine aminotransferase(ALT), Aspartate aminotransferase(AST),Alkaline phosphatase(ALP), Glutamyltranspeptidase(γ-GT), Lactate dehydrogenase(LDH)의 활성도와 중성지방, 콜레스테롤 함량을 측정한 결과 애엽 추출물 투여군에서 CCl4-대조군과 비교하여 유의적인 감소를 나타내었으며, HDL-콜레스테롤은 유의적인 증가를 나타내었다. 간조직중의 지질과산화 함량은 감소를 glutathione함량은 유의적인 증가를 나타내었다 이 실험 결과 애엽 에탄올 추출물은 사염화탄소 투여에 의한 간 손상을 억제하는 보호물질을 함유하고 있음을 알 수 있었다.

녹각이 Galactosamine에 의해 유도된 간장해에 미치는 영향 (Effect of Old Antler Extracts on the Galactosamine-induced Hepatotoxicity in Rats)

  • 간장해에 미치는 녹각추출물(water-ext., neutral ext. 및 ether-ext.)의 효과를 구명할 목적으로 녹각을 추출 분리하여 급여하고 매주 일정시각에 galactosa-mine (400mg/kg of body weight)으로 간장해를 유도한 후 녹각추출물이 간기능 상태의 지표인 효소활성에 미치는 영향을 생화학적 측면에서 비교.고찰하였다 체중증가량, 식이섭취량 및 식이효율은 Control군에 비해 galactosamine 투여군에서 감소를 보였고, 각각의 녹각추출물 급여로 증가하는 경향을 보였다. 체중100g당 장기무게는 간장의 경우 Balactosamine 투여로 현저한 증가를 보였으나 각각의 녹각추출물 급여가 이들의 수치를 감소시켰고, 신장, 비장 및 심장에는 별다른 차이가 없었다. 혈청 aminotransferase 활성은 galactosamine투여로 인해 증가하였으며, 각각의 녹각추출물중 water-ext.가 galactosamine투여에 의한 효소 활성도의 변화를 현저히 완화시켰다. 혈청 LDH와 ALP의 활성은 galactosamine투여로 증가하였고, 녹각추출물을 병행 투여한 군에서 Water-CaIN군의 활성이 Control군 수준에 가장 가깝게 감소하였다.
