  • Serum glutamic oxaloacetic transaminase (GOT), latic dehydrgenase (LDH), and alkaline phosphatase (ALP) activities were determined in 5 Korean native cattle to obtain the base line of blood enzyme activities in relation to the stage of gestation. Serum GOT activities showed an increasing tendency with the course of gestation period, serum LDH levels were within normal range through all the stage of pregnancy, and the slight increase in serum ALP activities was acknowledged at the 2nd and 3rd stage of gestation.

Effect of Testoeterone and Cyclic AMP-theophylline on Ultrastructure and Several Enzymes' Activities in Rat Epididymis (Testosterone과 Cyclic AMP-theophyllin이 흰쥐 부정소의 미세구조 및 몇 가지 효소활성에 미치는 영향)

  • This study was made to investigate the effect of testosterone and cyclic-AMP (cAMP) on rat epididymis. Peritoneai injections of testosterone and cAMP to rats were earned out The activities of acid phosphatase (ACP), alkaline phosphatase (ALP) and lactate dehydrogenase (LDH) were measured and ultrastructural changes of luminal epithelial cells were observed. As a result, the activity of ACP was significanily decreased on the third day, that of ALP on the fifth day and that of LDH on the seventh day respectively in castrated group. In addition, the activities of ACP and ALP were significanily increased when treated with testosterone for 5 days, that of LDH when treated with testosterone for 7 days. In case of cAMP and cAMP - theophylline injection, the activities of ACP and LDH were increased but the range of increase was of no significance. However a significant increase in the activity of ALP was seen on both cases. On electron microscopic examination, gradually deformed Golgi complex, destructed mitochondria and disrupted stereociha were observed in castrated group. In case of testosterone injection, disrupted Golgi complex, mitochondria and stereocilia showed recovery. When cAMP and cAMP-theophylline were injected as an alternative, various cytoplasmic organelles as well as Golgi com- plex were recovered but stereocilia remained unrecovered.

  • Effects of restraint stress and its modulations by ursodeoxycholic acid(UDCA) were evaluated on some biochemical and biophysical parameters in rats. Restraint stress induced elevations in blood alkaline phosphatase (ALP). cholesterol (CHOL), aspartate transaminase (GOT), alanine transaminase (GPT), lactate dehydrogenase (LDH) levels. It was also caused adrenal hypertrophy, decrease in weight of spleen and contents of ascorbic acid in stressed rats. As a results, stress indicators such as spleen, ascorbic acid, GOT, GPT, LDH were fastly changed after imposing stress, but those such as ALP, CHOL, adrenal were induced relatively later. UDCA was tested if it has an inhibitory effect against 18-hr restraint induced stress. UDCA lowered ALP, CHOL, LDH level and also effectively elevated the ascorbic acid contents in 25 mg/kg dosage of UDCA. In organ weights. the restraint stress induced increases in spleen and adrenal were attenuated by UDCA in 50 mg/kg dosage. However. stress-induced GOT and GPT levels were unaffected by UDCA.

Changes of Serum Chemistry Values on Size and Cultured Method in Korean Rockfish, Sebastes schlegeli (사육방법 및 크기에 따른 조피볼락의 혈청 화학성분 변화)

  • The purpose of this study was to obtain reference values of serum chemistry in Korean rockfish (Sebastes schlegeli), 5 juvenile (n=25) and 3 broodstock (n=15) subpopulations which were differentiated by cultured method. Serum samples were analyzed for the following commercial clinical kits: total protein, albumin, glucose, AST (aspartate aminotransferase), ALT (alanine aminotransferase), total cholesterol, free cholesterol, LDH (lactate dehydrogenase) and ALP (alkaline phosphatase). Significant differences were observed in the values of total protein, albumin, total cholesterol, AST, ALT, LDH and ALP between juvenile and broodstock subpopulations. In the comparison of total protein, albumin, total cholesterol and ALP, broodstock subpopulations showed higher levels. On the other hand, increased AST, ALT and LDH were observed in juvenile subpopulations. These results indicate that cultured method are major contributors to health-related physiology of Korean rockfish, juveniles and broodstocks.

Influences of Sodium Fluoride Contents on Hepatic Functional Enzyme Activities in Rats (Sodium Fluoride 함량이 흰쥐의 간 기능 효소활성에 미치는 영향)

  • The purpose of this study was to probe the influences of NaF oral administration on a dose-effect relationship between fluoride levels of serum enzyme activity such as alkaline phosphatase (ALP), aspartate aminotransferase (AST), alanine aminotransferase (ALT), and lactate dehydrogenase (LDH) in rats fed experimental diets for 5 weeks. All groups increased the activity of serum ALP, AST, ALT, and LDH levels with increasing NaF. In addition the fluoride levels of serum and organ tissues (liver, brain, heart, lung, kidney) in oral NaF groups (NF3~NF50) were significantly increased by adding sodium fluoride in comparison with normal diet group (ND) (p<0.05). These results, a high concentration of sodium fluoride was determined that the toxicity to various organ tissues.

  • The purpose of this study was to obtain reference data of parameters for hematological health diagnosis in cultured freshwater fish and also evaluate application of automatic dry-type chemistry analyzer (FUJI DRI-CHEM 3000) used to those blood tests. A blood profile of total 200 fish for rainbow trout (Onchorhynchus mykiss), israel carp (Cyprinus carpio), tilapia (Oreochromis niloticus) and eel (Anguilla japonica) cultured in Inland Fisheries Research Institute of NFRDI was determined by hemoglobin (Hb) and plasma chemistry tests: total protein (TP), albumin (ALB), alkaline phosphatase (ALP), blood urea nitrogen (BUN), lactate dehydrogenase (LDH), triglyceride (TG), total cholesterol (TCHO), creatinine (CRE), aspartate aminotransferase (AST), alanine aminotransferase (ALT), glucose (GLU). The values of ALT, TG, LDH, ALB, TCHO, AST and ALP were outside from the minimum and/or maximum of the established detectable range of the analyzer. ALT and TG were not detectable in the range of 67%~61.5%. LDH, ALB and TCHO were not detectable in the range of 36~17%. AST and ALP were not detectable in the range of 5.5~0.5%. However, the values of BUN, CRE, GLU, Hb and TP were below the detectable limits of the analyzer.

Radiomodifying Potential of Panax ginseng in Liver of Swiss Albino Mice against Gamma Radiation

  • Panax ginseng occupies an important role in the folk medicine of China, Korea and Japan. The present study was undertaken to determine the radioprotective efficacy of ginseng root extract in the liver of Swiss albino mice. The animals were divided into 4 groups. Group I-Only vehicle was administered. Group II-The animals received 10 mg/kg body weight ginseng root extract i.p. for 4 consecutive days. Group III-Animals were irradiated with 8Gy gamma radiation at the dose rate of 1.69 Gy/min at the distance of 80 ems. Group IV-Animals were given by ginseng root extract (10 mg/kg body weight) continuously for 4 days and on 4th day they were irradiated with 8 Gy gamma radiation after 30 min. The animals from above groups were autopsied on 1,3,7,14 and 30 days. Biochemical estimations of phosphatases (acid & alkaline), LDH (lactate dehydrogenase), LPO (lipid peroxidation) and GSH (reduced glutathione) in liver and SGOT (serum glutamate oxaloacetate transaminase), SGPT (serum glutamate pyruvate transaminase) and alkaline phosphatase in serum were done. In ginseng treated group acid phosphatase (ACP), alkaline phosphatase (ALP), LPO and LDH in liver and SGOT, SGPT and alkaline phosphatase in serum did not show any significant alteration. However, a significant increase in GSH content in liver was recorded. In irradiated group there was a significant increase in ACP, ALP and LPO content in liver and SGOT & SGPT in serum was noted. Whereas, a significant decrease was recorded in GSH and LDH activity in liver and alkaline phosphatase activity in serum. Pretreatment of ginseng with radiation significantly alters the biochemical parameters in liver and serum. A significant decline in ACP, ALP activity and LPO content in liver and SGOT and SGPT activity in serum was observed. However, a significant increase in GSH content and LDH activity in liver and ALP activity in serum was estimated. The present study suggests that pretreatment of ginseng before irradiation significantly protects the liver and maintains the enzyme activity.

Effect of Ginseng Extract on Enzyme activities of Rats Fed Lard and Alcohol (Lard와 Alcohol을 섭취시킨 흰쥐 혈청중의 효소활성에 인삼추출물이 미치는 영향)

  • The present study has been undertaken to investigate the effects of ginseng extract on the activities of several enzymes in serum of rats fed lard and alcohol. Thirty-five males of Sprague-Dawley strains weighed about 130 g were divided into 7 groups, each group receiving a different diet for 10 weeks; i.e. basal diet plus 15% lard, basal diet plus 5% alcohol, basal diet plus 5% ginseng extract, basal diet plus 15% lard and 5% ginseng extract. Determinations were carried out on the net weight gain, food efficiency ratio, weight of organs, and AST, ALT, lactate dehydrogenase, alkaline phosphatase activities in serum of rats. The results obtained were as follows:Rats given feed containing lard and alcohol showed significant decrease in net weight gain, but ginseng extract caused an increase in food efficiency ratio. Lard supplementation caused an increase in the weight of liver, kidney, spleen, but another groups did not. AST, ALT, ALP, LDH of serum were significantly increased in lard and alcohol containing group but ginseng extract feeding decreased enzyme activities compared to lard and alcohol containing group. The above results suggest that ginseng extract would prevent the metabolic disease of liver by preventing hyperlipemia caused by high fat diet.

Experimental Study of the Effect of Injinsaryungsan and Sosihotang on cholestatic liver injury induced by $ANIT({\alpha}-naphtylisothiocyanate)$ (인진사령산(茵陳四岺散)과 소시호탕(小柴胡湯)이 ANIT 로 유발(誘發)된 담즙울체성(膽汁鬱滯性) 간장애(肝障碍)에 미치는 영향(影響))

  • In an attempt to evaluate the effect of high and low concentration of Injinsaryungsan and high and low concentration of Sosihotang on cholestatic liver injery induced by $ANIT({\alpha}-naphthylisothiocyanate)$, biochemical changes in serum transaminase(GOT, GPT), alkaline phosphate, lactate dehydrogenase, total cholesterol, triglyceride, total-bilirubine were studied and the following results were obtained. 1. High concentration of Injinsaryungsan(2.2g/Kg) inhibited significantly the activity increases of GOT, GPT, ALP, LDH, TC, TG, T-Bilirubine induced by $ANIT({\alpha}-naphthylisothiocyanate)$. 2. Low concentration of Injinsaryungsan(1.1g/Kg) inhibited the activity increases of ALP, LDH, TC, TG with statistical significance, while inhibited the activity increase of GOT ,but with no statistical significance. 3. High concentration of Sosihotang(2.4g/Kg) inhibited the activity increases of LDH, TG, TC with statistical significance while inhibited the activity increases of GOT, GPT, ALP, T-bilirubine with no significance. 4. Low concentration of Sosihotang(1.2g/Kg) inhibited the activity increase of TG, while inhibited the activity increase of ALP, TC with no statistical sig-nificance, but didn't inhibite the activity increases of GOT, GPT, LDH, T-Bil. These results suggest that Injinsaryungsan has more significant effect on the liver injury induced by $ANIT({\alpha}-naphthylisothiocyanate)$ compared with Sosihotang and so can be applicable clinically to virus hepatitis and cholestatic liver injury. Further study will be required to evaluate the effect of Sosibotang on cholangitis and cholecystitis.

Effect of Puerariae Radix Methanol Extract on Benzo(a)pyrenc -in - duced Hepatotoxicity in Rats (갈근 메탄올 엑기스가 흰쥐에 있어서 Benzo(a)pyrene에 의해 유도된 간장해에 미치 는 영향)

  • The present study was conducted to evaluate the hepatoprotective effect of puerariae Radix methanol extract on benzo(a) pyrene(B(a)P) - induced liver injuries in rats. In vitro experiment, primary cultured hepatocytes (5X105 cells/$m\ell$) were cultured for 20~24 hours after adding puerariae Radix mehtanol extract(32$\mu\textrm{g}$/$m\ell$) and B(a)P(50 uM). In vivo experiment, Puerariae Radix methanol extract(0.25 g/kg/day, per os) was administered for 7 days and B(a)P(0.1 mg/kg/day, intraperitoneally) was given after the last administration of extract. And then the hepatoprotective effect of Puerariae Radix methanol extract was investigated biochemically through in vitro and in vivo experiments. Namely, activities of enzymes (GOT, GPT and LDH) were measured and 3-(4,5-dimethylthiazol-2-yl)-2, 5-diphenyl tetrazolium bromide(MTT) assay were carried out in vitro cell culture study and GOT, GPT, LDH and ALP activities and HDL-cholesterol, total cholesterol and triglyceride contents were performed in vivo study. In vitro experiment, as a result of enzyme activity measurement(GOT, GPT and LDH) and MTT assay, GOT,GPT and LDH activities changed by B(a)P were recovered to normal levels and hepatocytes impaired by B(a)P were recovered to normal. In vivo experiment, Puerariae Radix methanol extract significantly decreased the enzyme activities(GOT, GPT, ALP and LDH in serum and GPT and ALP in tissue) and lipid contents in comparison to B(a)P-treated group.
