백운산 지역에서 계류수의 이화학적 성질에 미치는 양어장 개발의 영향 (Influences of Fish Farm Development on the Physicochemistry of Stream Water Quality in (Mt.) Paekun Area)

  • Quantifications of fish farm influences on stream water quality may provide basic informations on watershed management to reduce environmental impact due to fish farm development and to conserve stream water quality in forested watershed area. In this research stream water qualities around (Mt.) Paekun area were monitored by seasons and the following results were obtained. Due to the increase of pH in effluent water from the fish farm it was believed that alkalization of stream water can be accelerated by large scale development of fish farms in the forested watershed area. Negative effects on stream water quality was also observed by indications of increase in electrical conductivity and temperature of effluent water from the fish farm. Decreases of physicochemical indexes such as the amount of dissolved oxygen, percentage of dissolved oxygen and total amount of ion in effluent water from the fish farm were also negative aspects in downstream ecology. It is recommended that water purification system as well as eco-friendly fish farm design be incorporated to large scale fish farm development plan in forested watershed area.

Improving Solubility through Carboxymethylation of Different-sized Endosperm, Bran, and Husk Rice Powders

  • The surfaces of different-sized endosperm, bran, and husk rice powders were modified using carboxymethylation. Carboxymethylation was carried out using aqueous alkalization and neutralization. After the carboxymethylation process, the centrifuged products were milled and classified by size: particles passed through sieves of 45, 106, and $300-{\mu}m$ width. The effect of carboxymethylation on physical properties such as solubility and dispersibility of endosperm, bran, and husk particles were studied. Overall, carboxymethylation increased solubility of the particles, while size reduction increased dispersibility. In particular, carboxymethylation created good aqueous suspensions by minimizing interparticle agglomeration. Our results show that the combination of size reduction and carboxymethylation improves solubility and dispersibility, resulting in better stability of the suspension. This study may be helpful for expanding the use of rice and its byproducts as ingredients in a variety of food and beverage applications.

Effect of Natural Fiber Surface Treatments on the Interfacial and Mechanical Properties of Henequen/Polypropylene Biocomposites

  • The surfaces of henequen fibers, which can be obtained from the leaves of agave plants, were treated with two different media, tap water and sodium hydroxide, that underwent both soaking and ultrasonic methods for the fiber surface treatment. Various biocomposites were fabricated with untreated and treated, chopped henequen fibers and polypropylene using a compression molding method. The result is discussed in terms of interfacial shear strength, flexural properties, dynamic mechanical properties, and fracture surface observations of the biocomposites. The soaking (static method) and ultrasonic (dynamic method) treatments with tap water and sodium hydroxide at different concentrations and treatment times significantly influenced the interfacial, flexural and dynamic mechanical properties of henequen/polypropylene biocomposites. The alkali treatment was more effective than the water treatment in improving the interfacial and mechanical properties of randomly oriented, chopped henequen/PP bio-composites. In addition, the application of the ultrasonic method to each treatment was relatively more effective in increasing the properties than the soaking method, depending on the treatment medium and condition. The greatest improvement in the properties studied was achieved by ultrasonic alkalization of natural fibers, which was in agreement with the other results of interfacial shear strength, flexural strength and modulus, storage modulus, and fracture surfaces.

External pH Effects on Delayed Rectifier $K^+$ Currents of Small Dorsal Root Ganglion Neuron of Rat

  • Under certain pathophysiological conditions, such as inflammation and ischemia, the concentration of H^+$ ion in the tissue surrounding neurons is changed. Variations in H^+$ concentration are known to alter the conduction and/of the gating properties of several types of ion channels. Several types of K^+$ channels are modulated by pH. In this study, the whole cell configuration of the patch clamp technique has been applied to the recording of the responses of change of external pH on the delayed rectifier K^+$ current of cultured DRG neurons of rat. Outward K^+$ currents were examined in DRG cells, and the Charybdotoxin and Mn^{2+}$ could eliminate Ca^{2+}-dependent$ K^+$ currents from outward K^+$ currents. This outward K^+$ current was activated around -60 mV by step depolarizing pulses from holding potential -70 mV. Outward K^+$ currents were decreased by low external pH. Activation and steady-state inactivation curve were shifted to the right by acidification, while there was small change by alkalization. These results suggest that H^+$ could be alter the sensory modality by changing and modifying voltage-dependent K^+$ currents, which participated in repolarization.

  • Trimorphomyces papilionaceus dikaryon 으로부터 자영적인 돌연변이에 의해 분리한 이배체를 대상으로 생장속도, U. V. 광 조사 후의 생존율, U. V. 분광광도계와 Hoechst 33258 핵염색에 의한 DNA 함량등을 monokaryon 및 dikaryon 과 비교하여 결정하였다. 이배체는 dikaryon 핵의 DNA 양과 비슷하였고 monokaryon 핵의 DNA 양의 2배 정도가 되었다. Glyoxalase II 의 band 양상에서 monokayon은 한 개의 isozyme band 만을 보인 반면, 이배체와 dikaryon은 같은 위치에서 두개의 isozyme band 가 관찰되었다. 이원전개한 전기영동실험에서 dikaryon 특이 단백질은 분자량이 66.000 보다 컸으며 분자량 24,000-29,000 사이에서 이배체 특이 단백질이 존재한다.

  • Analysis of fish farm influences on stream water quality may provide basic informations on watershed management to reduce environmental impact due to fish farm development and to conserve stream water quality in forested watershed area. In this research stream water qualities around Mt. Baegun area were monitored seasonally for three years. Due to the increase of pH in effluent water from the fish farm it was believed that alkalization of stream water can be accelerated by large scale development of fish farms in the forested watershed area. As a result of regression analyses, the linear equation of pH of influent and effluent water of fish farm was, pH of effluent water of fish farm = $0.6234{\times}pH$ of influent water of fish farm + 2.6263. Also, the linear equation of electrical conductivity of influent and effluent water of fish farm was electrical conductivity of effluent water of fish farm = $1.7275{\times}$electrical conductivity of influent water of fish farm - 14.007. Negative effects on stream water quality were observed by indications of increase in electrical conductivity and water temperature of effluent water from the fish farm. Decreases in physicochemical indices such as the amount of dissolved oxygen, total amount of cation and total amount of anion in effluent water from the fish farm were also negative aspects in downstream ecology. It is recommended that water purification system as well as environmentally-friendly fish farm design should be incorporated to large scale fish farm development plan in forested watershed area.

Dicamba 급성 중독으로 인한 사망 1례 (A Fatal Case of Dicamba Intoxication)

  • Dicamba is a benzoic acid and classified as a chemically related chlorophenoxy herbicide which is widely used for the control of broad-leaved weeds. While the chlorophenoxy herbicide poisoning is known to be uncommon, its ingestion can result in serious or sometimes fatal outcome. A 65-year-old man ingested about 300 ml of dicamba in a suicidal attempt and three hours later he was admitted hospital, complaining abdominal pain, nausea and vomiting. On admission his vital signs were normal and laboratory findings were not remarkable except metabolic acidosis in arterial blood gas analysis. Shortly after the admission endotracheal tube was inserted due to altered mental state and activated charcoal was given after performing gastric lavage. However, his vital signs became unstable 6hrs after the ingestion and mechanical ventilation was started with administration of inotropic agents. In spite of urine alkalization for rapid elimination of the absorbed dicamba, the metabolic acidosis was aggravated with concomitant rhabdomyolysis and acute renal failure, and he died 24 hrs after the ingestion.

The antibacterial effect of xanthorrhizol as an endodontic irrigant on Enterococcus faecalis

  • Objectives The aim of this study was to evaluate the antibacterial effect of xanthorrhizol (XTZ) on E. faecalis, compared with 2% chlorhexidine (CHX). Materials and Methods Normal physiological state (NS), starvation state (SS), and alkalization state (AS) of E. faecalis were used. A solution containing 1% XTZ in 30% ethanol, 1% dimethyl sulfoxide (DMSO), and 100 mg/ml sodium methyl cocoyl taurate was used and is referred to as Xan in this study. To determine the minimal bactericidal concentration (MBC) of Xan and CHX, $500{\mu}l$ of E. faecalis (NS and two stress states) was added to a microtube containing $500{\mu}l$ of serial 2-fold dilutions of 1% Xan and 2% CHX (1:2-1:128). The MBC of each antimicrobial was determined by the plate count method. Results The antibacterial effect of Xan was more effective on E. faecalis in AS than in the other states (NS, SS) at 0.125% Xan and 0.03325% Xan (P<0.05). In contrast, the antibacterial effect of CHX was more effective against E. faecalis in SS than the other states (NS, AS) at 0.0625% CHX (P<0.05). In SS, the antibacterial effect of CHX was more effective than that of Xan at 0.125% and 0.0625% (P<0.05). However, in AS, the antibacterial effect of Xan was more effective than that of CHX at 0.0625% and 0.03325% (P<0.05). Conclusions In endodontic retreatment cases in which it is important to effectively remove E. faecalis from the infected root canal, Xan may be more suitable when combined with NaOCl than CHX.

Onion peel extract and its constituent, quercetin inhibits human Slo3 in a pH and calcium dependent manner

  • Sperm function and male fertility are closely related to pH dependent $K^+$ current (KSper) in human sperm, which is most likely composed of Slo3 and its auxiliary subunit leucine-rich repeat-containing protein 52 (LRRC52). Onion peel extract (OPE) and its major active ingredient quercetin are widely used as fertility enhancers; however, the effect of OPE and quercetin on Slo3 has not been elucidated. The purpose of this study is to investigate the effect of quercetin on human Slo3 channels. Human Slo3 and LRRC52 were co-transfected into HEK293 cells and pharmacological properties were studied with the whole cell patch clamp technique. We successfully expressed and measured pH sensitive and calcium insensitive Slo3 currents in HEK293 cells. We found that OPE and its key ingredient quercetin inhibit Slo3 currents. Inhibition by quercetin is dose dependent and this degree of inhibition decreases with elevating internal alkalization and internal free calcium concentrations. Functional moieties in the quercetin polyphenolic ring govern the degree of inhibition of Slo3 by quercetin, and the composition of such functional moieties are sensitive to the pH of the medium. These results suggest that quercetin inhibits Slo3 in a pH and calcium dependent manner. Therefore, we surmise that quercetin induced depolarization in spermatozoa may enhance the voltage gated proton channel (Hv1), and activate non-selective cation channels of sperm (CatSper) dependent calcium influx to trigger sperm capacitation and acrosome reaction.

Synthesis and Characterization of Magnetic Cellulose Powder from Sawdust Waste

  • Timber industry waste is being examined for cellulose manufacturing to give important benefits. The study's goal is to investigate the properties of cellulose powder derived from sawdust waste after it has been reinforced with ferrous-ferric oxide nanoparticles (Fe3O4-NPs). Sawdust cellulose powder was produced from Sengon (Albizia chinensis) wood waste in this study. The crushed sawdust waste is handled with alkalization and bleaching. Cellulose powder is then reinforced with Fe3O4-NPs at 10 wt.%, 20 wt.%, and 30 wt.%. The magnetic cellulose powder was analysed by X-ray diffraction, Fourier Transform Infrared, scanning electron microscopy morphology, magnetic vibrating sample magnetometer, Brunauer-Emmett teller, and adsorption tests for Methylene Blue and Congo Red dyes. Structure study identifies sawdust as cellulose 1β, with peaks at 14°, 16°, and 22° diffraction angles. The addition of Fe3O4-NPs reduces the crystalline index of sawdust cellulose powder from 68.50% to 63.38%, and functional group bond analysis revealed many peak shifts indicating a change in the chemical bonds of magnetic cellulose powder. Incorporating Fe3O4-NPs into sawdust cellulose powder confers magnetic and superparamagnetic properties to the sawdust cellulose. Similarly, the surface texture of magnetic cellulose seems rougher as the surface area increases. These parameters imply a 31.8% increase in Congo Red adsorption, using adsorption kinetics based on the pseudo-first-order model.
