주상(酒傷)에 응용되는 성주청간탕(醒酒淸肝湯)이 알코올대사 및 간장애에 미치는 영향 (Effects of the Sungjuchunggan-tang(Xingjiuqinggan-tang) on alcoholic metabolism and alcoholic liver damages)

  • Objectives : The aim of this study was to investigate effects of the Sungjuchunggan-tang(Xingjiuqinggan-tang), which has been used for alcoholic diseases in Oriental medicine, on alcoholic metabolism and alcoholic liver damage. Methods : We evaluated the activities of alcohol dehydrogenase(ADH)and aldehyde dehydrogenase(ALDH), and measured the levels of AST, ALT, glucose, triglyceride, HDL-cholesterol, LDL-cholesterol and phospholipid in serum of guinea pigs. Results and Conclusions : In this experiment, Sungjuchunggan-tang, Pharbitidis Semen and Puerariae Radix significantly suppressed the activity of ADH which is a precursor enzyme of acetaldehyde. Sungjuchunggan-tang and Pharbitidis Semen significantly suppressed the activity of ALDH, which is a catabolic enzyme, and increased its level. Due to alcohol consumption, level of ALT and AST were significantly reduced. These experimental results suggest that Sungjuchunggan-tang and Pharbitidis Semen inhibit the formation of acetaldehyde in the metabolism of alcohol, and affect the recovery of weakened liver functions caused by alcohol.

청간해주탕이 알코올대사 및 손상간에 미치는 영향 (Effects of Chungganhaeju-tang(Qingganjiejiu-tang) on Alcohol Metabolism and Alcoholic Liver Damages)

  • Objectives: This experiment was conducted to verify the effects of Chungganhaeju-tang(Qingganjiejiu-tang) on the alcohol metabolism and liver functions, by measuring the activity levels of ADH and ALDH, as well as glucose, triglyceride, and BUN. Damage of the liver cells caused by alcohol was determined through the examination of serum levels of AST, ALT, ALP, LDH, and uric acid. Methods: Sprague-Dawley rats were used in this experiment and the rats were divided into control and experiment groups. Chungganhaeju-tang(Qingganjiejiu-tang) extract was orally administered in the experiment group for three weeks. Each group was further classified into two sub-groups, and control group's blood was taken without oral ingestion of alcohol, while the experiment group' s blood was withdrawn after ingestion of alcohol. Evaluation of damage level was done considering the presence of extract and alcohol. Results: In this experiment, Chungganhaeju-tang(Qingganjiejiu-tang) significantly suppressed the activity of ADH which is a precursor enzyme of acetaldehyde, but didn't cause significant changes in the activity of ALDH which is a catabolic enzyme. Decreased glucose level due to alcohol consumption was recovered back to the normal level and increased levels of triglyceride, BUN, AST, ALT, ALP, LDH, and uric acid were significantly reduced. Conclusions: These experiment results suggest that Chungganhaeju-tang(Qingganjiejiu-tang) inhibits the formation of acetaldehyde in the metabolism of alcohol, and affects the recovery of weakened liver functions due to alcohol.

알코올 의존 환자에서의 Aldehyde Dehydrogenase II와 CYP2E1 유전자 다형성과 임상적 특성간의 연관성 (Association of Genetic Polymorphisms of Aldehyde Dehydrogenase II and CYP2E1 and Clinical Characteristics of Patients with Alcohol Dependence)

  • Objective:This study was to explore the relation of genetic polymorphisms of ALDH2 and CYP2E1 to clinical characteristics of alcoholic patients and alcohol induced liver damage. Methods:The genotype and allele frequencies of 128 male hospitalized patients who met DSM-IV criteria for alcohol dependence were compared with 128 healthy male control subjects. The genetic informations of ALDH2 and CYP2E1 were identified with the technique of polymerase chain reaction and restriction fragment length polymorphism. The clinical characteristics of the alcoholic patients were assessed and analyzed in relation to the family history of alcoholism. For the relation of CYP2E1 genetic polymorphism to the liver damage, the blood levels of various liver function indicators such as ALT, AST, and protein were checked out. Results:1) The alcoholic patients with the family history of alcoholism had the earlier onset of age (p=0.001), the longer duration of illness(p=0.045), and higher NCA scores(p=0.018) than those without the family history of alcoholism. 2) Most alcoholic patients were homozygous for $ALDH2^*1$, compared to control subjects.(p=0.000) 3) There was no difference of CYP2E1 distribution between alcoholic patients and control subjects. However, alcoholic patients having mutant c2 allele showed higher alcoholism severity scores(p=0.004) and more hospitalizations(p=0.014) than those having c1 allele. 4) There was no relationship between CYP2E1 genotype and the functional abnormalities of the liver. Conclusion:This study suggests that $ALDH2^*1$ is highly related with alcohol dependence. Also mutant c2 allele of CYP2E1 is correlated with the severity of alcoholism and the number of hospitalization. But genetic polymorphim of CYP2E1 seems to have no relation to liver damages.

Lactobacillilus brevis를 이용한 다시마 발효물의 알코올 분해 활성 (Alcohol Metabolizing Activity of Fermented Sea Tangle Juice)

  • Alcohol metabolizing activity of fermented sea tangle juice (FSYJ) using Lactobacillilus brevis BJ20 were evaluated by measuring relative alcohol dehydrogenease (ADH) and aldehyde dehydrogenase (ALDH) activities. According to the results of MTI assay. the fermented sea tangle juice by Lactobacillilus brevis BJ20 appeared safe in the cytotocxity. The relative ADH activity of FSTJ showed 124% at 10 mg/mL, which increased with increasing concentration. The relative ALDH activity showed, however, insignificant difference (P>0.05) between concentrations of FSTJ up to 50 mg/mL. These results suggested that fermented sea tangle juice by L. brevis BJ20 could be used as a potential material for metabolizing alcohol.

Two novel mutations in ALDH18A1 and SPG11 genes found by whole-exome sequencing in spastic paraplegia disease patients in Iran

  • Hereditary spastic paraplegia is a not common inherited neurological disorder with heterogeneous clinical expressions. ALDH18A1 (located on 10q24.1) gene-related spastic paraplegias (SPG9A and SPG9B) are rare metabolic disorders caused by dominant and recessive mutations that have been found recently. Autosomal recessive hereditary spastic paraplegia is a common and clinical type of familial spastic paraplegia linked to the SPG11 locus (locates on 15q21.1). There are different symptoms of spastic paraplegia, such as muscle atrophy, moderate mental retardation, short stature, balance problem, and lower limb weakness. Our first proband involves a 45 years old man and our second proband involves a 20 years old woman both are affected by spastic paraplegia disease. Genomic DNA was extracted from the peripheral blood of the patients, their parents, and their siblings using a filter-based methodology and quantified and used for molecular analysis and sequencing. Sequencing libraries were generated using Agilent SureSelect Human All ExonV7 kit, and the qualified libraries are fed into NovaSeq 6000 Illumina sequencers. Sanger sequencing was performed by an ABI prism 3730 sequencer. Here, for the first time, we report two cases, the first one which contains likely pathogenic NM_002860: c.475C>T: p.R159X mutation of the ALDH18A1 and the second one has likely pathogenic NM_001160227.2: c.5454dupA: p.Glu1819Argfs Ter11 mutation of the SPG11 gene and also was identified by the whole-exome sequencing and confirmed by Sanger sequencing. Our aim with this study was to confirm that these two novel variants are direct causes of spastic paraplegia.

홍합과 여러 식물의 열수 추출물의 알코올분해효소에 미치는 영향과 항산화 및 ACE 저해 활성 (Effect of Hot Water Extracts of Blue Mussel and Several Plants on Alcohol Metabolizing Enzymatic, Antioxidant, and Angiotensin Converting Enzyme Inhibitory Activities)

  • 홍합을 함유한 숙취 해소용 식품 개발의 근거를 확보하기 위하여 12종류의 열수 추출물을 제조하였다 - A(겨우살이); B(냉이); C(칡); D(콩나물); E(헛개나무); F(홍합); G(홍합과 겨우살이); H(홍합과 냉이); I(홍합과 칡); J(홍합과 콩나물); K(홍합과 헛개나무). C 추출물이 alcohol dehydrogenase(ADH)와 acetaldehyde dehydrogenase(ALDH)의 활성을 가장 크게 증가시켰으나, 홍합과 혼합한 경우에는 상승 효과를 보이지 않았다. 칡을 함유한 경우를 제외하고는 H 추출물이 높은 ADH($237.4{\pm}1.7%$)와 ALDH($136.5{\pm}2.1%$) 활성을 보였다. 또한, H 추출물은 가장 높은 DPPH 라디칼 소거능($93.9{\pm}0.1%$)과 안지오텐신 전환효소(ACE) 저해능($42.7{\pm}1.6%$)을 나타내었다. 홍합과 냉이를 혼합한 경우에는 alcohol dehydrogenase 활성과 ACE 저해능이 상승되는 시너지 효과를 확인하였다.

  • In the course of evaluation of hepatoprotective components against alcohol-induced toxicity from Aloe spp., the methanol extract was found to cause a significant inhibition of rat liver cytosolic alcohol dehydrogenase activity. Systematic fractionation of active tractions monitored by bioassay led to isolation of four compounds; aloe-emodin, aloenin, ethylidene-aloenin and ${\beta}-sitosterol$, which were estimated as active principles for inhibition of c-ADH and c-ALDH activities in vitro.

발효한약추출물 HP-1이 알코올을 투여한 쥐의 알코올 대사에 미치는 영향 (Effect of Fermented Herbal Extracts, HP-1 on Enzyme Activities and Gene Expressions Related to Alcohol Metabolism in Ethanol-loaded Rats)

  • Recently, much attention has been paid to developing various kinds of fermented herbal extracts, a new type of traditional herbal medicine, in the field of Korean traditional medicine. The fermentation of medicinal herbs is intended to exert a favorable influence on digestability, bioavailavility and pharmacological activity of herbal extract in the gastrointestinal tract. It also produces a number of fermentation products that intensify the nutritional and pharmacological aspects of the medicinal herbs. In order to develop a functional beverage of alleviating the aftereffects of the previous drinks, the extracts (HP-1) of fermented herbal mixture, including Artemisia capillaris Thunb., Lonicera japonica Thunberg, and Hovenia dulcis Thunb., were prepared and the medicinal effect as a hangover cure was evaluated in ethanol-loaded rats. The enzyme activities of alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase-2 (ALDH2) were analyzed by measuring the concentration of blood ethanol and acetaldehyde. The mRNA expression of ADH and ALDH2 was also investigated through RT-PCR analysis. In the HP-1-treated group, the concentration of blood ethanol was significantly reduced at one hour after loading of ethanol, as compared to that in the saline-treated group. The reduced ethanol was converted to acetaldehyde, which resulted in rapid increase in acetaldehyde concentration in an hour. Acetaldehyde was started to decrease at 5 hours after ethanol loading. It implies that HP-1 is highly effective to stimulate the activities of ADH and ALDH2. The HP-1 treatment also activated the mRNA expression of ADH and ALDH. This study suggests that fermented herbal extract, HP-1 can be used as a functional beverage of alleviating the alcohol-induced hangover symptoms by stimulating the activities and gene expression of hepatic alcohol metabolizing enzymes.

막걸리 열수 추출물의 아질산염 소거능, 알코올 분해능 및 angiotensin converting enzyme 저해 효과 (Nitrite Scavenging and Alcohol Metabolizing Activities of Hot Water Extract from Makgeoly and Its Angiotensin Converting Enzyme Inhibitory Effect)

  • 전통발효주 막걸리의 기능성을 증명하기 위하여 S사막걸리로부터 구입한 막걸리 침전물의 여러 가지 생리활성에 대하여 조사하였다. 우선 막걸리 침전물 열수 추출물의 항산화 효과를 측정하기 위해 DPPH radical 소거능과 SOD 유사활성을 측정하였다. 그 결과 DPPH법을 통해 측정한 막걸리 침전물 열수 추출물의 radical 소거능은 10 mg/ml에서 48.0%으로 나타났으며, 농도가 증가함에 따라 유의적으로 증가하는 경향을 나타내었다. 또한 SOD 유사활성은 10 mg/ml 농도에서 98.7%로 비교적 높은 SOD 유사활성을 보였다. 항고혈압 활성 측정 실험에서는 현재 시판되고 있는 항고혈압제인 captopril은 0.1 mg/ml에서 93.4%의 ACE 억제효과가 나타났고, 막걸리 침전물 열수 추출물 10 mg/ml에서는 74.0%의 높은 저해 활성을 나타내었다. 따라서, 막걸리 침전물 열수 추출물은 인체에 부작용이 적은 천연 항고혈압소재로서 이용가능성이 높은 것으로 사료된다. 아질산염 소거능 측정 실험에서는 positive control인 Vit. C1 mg/ml의 경우 pH 1.2와 3.0에서는 74~64%, pH 6.0에서는 45%의 소거능을 보인반면, 막걸리 침전물 열수 추출물의 경우 pH 1.2와 3.0에서는 51~42%, pH 6.0에서는 28%의 소거능을 나타내었다. 막걸리 침전물 열수 추출물의 숙취해소 효능은 ADH와 ALDH 활성증진에 막걸리 침전물 열수 추출물이 미치는 영향을 조사함으로써 증명하고자 하였다. 그 결과, 알콜과 acetaldehyde 분해능은 높게 나타났다. 이상의 결과들은 막걸리 침전물의 우수한 기능성으로서의 이용 가능성에 대한 기초자료로 그 가치가 기대된다.

흰쥐에서 황칠나무 열수 추출물을 포함한 혼합물의 혈중 알코올 농도와 숙취 해소 효과 (Eliminatory Effect of Mixture including Hot Water Extract of Dendropanax morbifera Lev. on Alcohol-induced Blood Alcohol Concentration and Hangover in Rat)

  • 본 연구에서는 숙취해소에 좋은 것으로 알려진 식품 소재와 황칠나무 추출물을 복합하여 in vitro 및 in vivo에서 알코올 유도된 간 손상에 대한 보호효과를 검토하였다. HepG2세포에서 300 mM 알코올과 SBJ 혼합물을 처리하였을 때 농도 의존적으로 LDH 수치가 감소하였으나 황칠나무 추출물을 제외한 복합물인 SBJ-복합물에서는 그 효과가 유의적으로 낮아지는 것을 관찰하였다. In vivo에서 SBJ 혼합물의 보호효과를 확인하고자 흰쥐에 알코올과 SBJ 혼합물을 투여한 결과, 알코올 투여 후 1시간까지 급격하게 혈중 알코올 수치가 EtOH군에서 증가하는 것이 관찰되었으며, SBJ 혼합물 투여군은 유의적인 차이로 감소되었음이 관찰되었으며 또한 농도의존적인 경향을 확인하였다. ADH 및 ALDH 활성의 증가는 SBJ 혼합물의 알코올 분해 및 대사산물의 제거 활성에 기여할 것으로 예상된다. 뿐만 아니라, 알코올에 의하여 증가한 LDH의 농도가 대조군과 유사한 수준으로 유지하는 것을 확인하였으며, GST, SOD, GPx 및 reduced glutathione와 같은 항산화 인자 및 효소의 활성은 대조군보다 EtOH군이 유의적으로 감소했으며, SBJ 혼합물에 의해 개선되는 경향을 관찰하였다. 이를 통해 SBJ 혼합물은 ADH, ALDH 활성 증가 및 항산화 방어계를 향상시킴으로써 산화적 스트레스 감소를 통한 간보호 효과가 있는 것으로 생각되며, 이러한 간보호에 미치는 주요한 추출물은 황칠나무 추출물임을 알 수 있었다. 이러한 결과를 토대로 향후 숙취해소 작용을 갖는 신규 식품소재로 활용될 수 있을 것으로 기대된다.
