Dedifferentiation State Specific Increase of Trypsin- and Chymotrypsin-like Protease Activities during Urodele Limb Regeneration and Their Enhancement by Retinoic Acid Treatment (유미양서류 다리 재생 기간중 탈분화 시기 특이적 트립신, 키모트립신 유사 단백질 효소의 활성도 증가)

  • Treatment of regenerating amphibian limbs with retinoic acid (RA) is known to induce paftern duplication, which is closely related to the extent of dedifferentiation. In the present study, the activities of trypsin- and chymotrypsin-like proteases are examined to delineate a possible role in the process of dedifferentiation in the regenerating limbs of urodeles, the Korean salamander (Hynobius leechii) and the Mexican axolod (Ambystoma mexicanum). Specifically, we were interested to know if there is any correlation between trypsin- and chymotrypsin-like protease activities and the state of dedifferentiation which is augmented by RA treatment. We were also interested in expoloring if there is any species-specific difference in the profile of enzyme activities during limb regeneration. The results showed that the activities of these two enzymes reached a peak level at dedifferentiation stage, and RA treatment caused elevation of their activities, especially in the case of trypsin-like protease. The increase of trypsin-like protease activity after RA treatment was pronounced in the Korean salamander, which might reflect a species-specific responsiveness to RA. The present results imply that trypsin and chymotrypsin or similar proteases may play an active role in the process of dedifferentiation in regenerating limbs, and that trypsin or trypsin-like eryrymes might be involved in the RA-evoked enhancement of dedifferentiation which precedes overt pattern duplication.

Changes in Free Sugars, Organic Acids and Fatty Acid Composition of Kanjang Prepared with Different Cooking Conditions of Whole Black Bean (원료콩의 증자조건을 달리한 검정콩 간장의 유리당, 유기산 및 지방산의 변화)

  • pH values in all samples were in the range of 5.04 to 5.32. pH of kanjang prepared with normal pressure(NPK) was higher than that of kanjang prepared with high pressure(HPK) and kanjang prepared with steam(SPK). Titratable acidity(TA) in all samples were found to be 4.26~7.72 mL. TA in HPK(5.99~6.96 mL) was lower than that in SPK(4.26~5.61 mL) and NPK(7.32 mL). Reducing sugar(RS) contents in all samples were in the range of 25.18 to 194.81 mg%. RS contents of HPK and SPK were a similar pattern and were higher than that of NPK. Major free sugars(FS) were glucose, fructose and xylose. In total FS, SPK was higher than that of NPK and HPK. Major organic acids(OA) were succinic acid, oxalic acid, tartaric acid, malic acid and citric acid. In total OA contents, SPK(40.38~76.96 mg%) was higher than that of NPK(55.85 mg%)and HPK(27.78~34.21mg%). OA contents in HPK and SPK were decreased slightly according to the heating time of whole black bean. The highest percentage of fatty acid (FA) in all samples was palmitic acid(33.28~43.51 % as area % of GC chromatogram), followed in a decreasing order by stearic acid(14.15~19.16%) and myristic acid(5.93~19.14%). The composition ratios of three FA to total FA ranged from 55.97 to 72.85&. In all samples, there was myristic acid that was not contained in raw soybean.

Biological activities in Aronia melanocarpa depending on drying methods (건조방법에 따른 아로니아의 생리활성)

  • To investigate biological activities in Aronia melanocarpa various drying methods were employed such as vacuum freeze drying, hot air drying and cold air drying. DPPH radical scavenging activity and ABTS radical scavenging activity of vacuum freeze dried Aronia melanocarpa was higher than hot and cold air dried Aronia melanocarpa. Vacuum freeze drying method showed the greatest contents of total phenol (15.34 g GAE/100 g), flavonoid (3.10 g GE/100 g) and tannin (2.46 g TE/100 g). Total anthocyanin content decreased to 163.52 mg C3G/100 g and 50.15 mg C3G/100 g for hot and cold air drying, respectively. Vacuum freeze-dried method increased the total anthocyanin content (743.09 mg C3G/100 g) when compared with fresh Aronia melanocarpa (163.52 mg C3G/100 g). Total proanthocyanidin content of vacuum freeze dried Aronia melanocarpa has increased to 6.21 g CE/100 g more than eight times compared with fresh Aronia melanocarpa (0.71 g CE/100 g). Chlorogenic acid and neochlorogenic acid content of vacuum freeze dried Aronia melanocarpa were higher than hot air dried and cold air dried Aronia melanocarpa, increasing about three times compared with fresh Aronia melanocarpa. These results suggested that vacuum freeze drying is optimal drying method to enhance biological activities in Aronia melanocarpa.

Effect of Packaging on Quality of Stored Gangjung (포장방법이 강정의 저장 중 품질특성에 미치는 영향)

  • This study was conducted to improve the packaging technology used for Gangjung and the product's quality characteristics during storage. Packaging materials consisting of (1) IP: OPP (oriented polypropylene)/PE (polyethylene) + Paper, (2) LP1: OPP/PE + Paper, (3) LP2: OPP/PE + Can, (4) LP3: OPP/PE + Plastic, (5) BP1: OPP/PE, (6) BP2: OPP/DL/CPP (cast polypropylene), (7) BP3: PET/PE/CPP, and (8) BP4: OPP/PVDC (polyvinylidene chloride)/PE/CPP were used. The major fatty acids including 54.2% linoleic acid, 24.1% oleic acid, 11.2% palmitic acid and 6.0% linoleic acid were assessed. The packages effective in maintaining moisture were in the order of LP2, BP4 > IP > LP3, BP1, BP2 > LP1 > BP3. There were no significant differences in water content and the products in IP, BP3, and BP showed the highest level in the increase in hardness on day 60 of storage. Acid values of IP, LP1, LP2, LP3, BP1, and BP2 were maintained at <2.0 during 40 days of storage. IP, LP1, LP2 and BP1.

  • This study was designed to examine effects of the various levels of sardine and safflower oil mix on lipid contents of serveral tissues in dietary hyperlipidemic rats. Experimental oils were 16% butter(control group), 8% butter+8% olive oil(group 2), 8% butter+8% sardine oil(group 3), 8% butter+6% sardine oil+2% safflower oil(group 4), 8% butter+4% sardine oil+4% safflower oil(group 5), 8% butter+2% sardine oil+6% safflower oil(group 6) and 8% butter+8% safflower oil(group 7). The diet administered to the male rats of Sprague-Dawley were fed for 4 weeks. In the fatty aicd composition of brain phospholipid, n-3 EPA and DHA contents were increased, and DHA content was remarkably high in the phosphatidylethanolamine(PE) component. Arachidonic acid content were high in the cardiolipin component. In the fatty acid composition of heart phospholipid, PUFA contents were highest in the group 5 and DHA content was higher in the groups 4 and 5, particularly. Fatty acid composition of spleen lipid showed that n-3 EPA and DHA contents were higher in the group 3 than in the other groups.

A STUDY ON THE SHEAR BOND STRENGTH OF LUTING GLASS IONOMER AND DENTIN TREATED WITH CALCIUM SOLUTION (칼슘수용액으로 처리한 상아질과 합착용 글래스아이오노머의 전단결합강도에 관한 연구)

  • The objective of this paper was to evaluate the shear bond strength of luting glass ionomer cement with defferent calcium based solution treatment on dentin surface. 120 extracted human teeth were classified into 12 group based on presence of smear layer on dentin surface and type of treatment solution. Smear layer remove on dentin surface was done using 6% citric acid for 60 seconds. Five different dentin surface treatment solutions(calcium acetate, calcium carbonate, clacium chlorided, calcium hydroxide, and calcium phosphate) were evaluated in this study. After surface modification, metal ring(inner diameter : 3mm, depth : 1mm) was placed to expose the same dentin surface area and inner space was filled with luting glass ionomer cement according to the recommended procedure for stadard clinical procedure. The shear bond strength of glass ionomer cement was determined after 24 hours. SEM was used for the evaluation of the surface morphologic changes and EDAX analysis was done for determination of the change of the calcium contents of treated dentin. Follwing conclusion can be drawn : 1. In the group of the dentin surface with smear layer, the calcium carbonate solution was the most effective for the increase of the clacium content and the shear bond strength of glass ionomer cement to dentin surfaces. 2. In the group of the calcium carbonate treated dentin with msear layer, the shear bond strength was increased twice compared to the control group and cohesive failure mode was observed. 3. The shear bond strength of cement was increased significantly be the removal of smear layer using 6% citric aicd. However, additional calcium solution treatments were not effective for further bond strength increase. 4. The shear bond strength of cement was significantly improved by both of the removal of smear layer and the calcium solution treatment, and the former was more effective for bond strength improvement. 5. The smear layer removed/calcium solution treated groups showed dentinal tubule obstruction and crystal attachment in SEM evaluation. However, the shear bond strengths of these groups were not increased compared to the smear layer removed/no dentin treatment group.

A Study of Heavy Metal-Contaminated Soil Remediation with a EDTA and Boric acid Composite(II): Cd and Cr (EDTA와 붕산 혼합용출제를 이용한 중금속으로 오염된 토양의 처리에 관한 연구(II): 카드뮴 및 크롬)

  • To investigate the optimal operation factors in appling Soil Flushing Process, which use EDTA and boric acid composite as a flushing reagent, to the Cd(cadmium) and Cr(chromium) contaminated sites, Cd and Cr emediation rate was investigated at various experimental conditions; as EDTA concentration was increased from 0.001M to 0.1M, Cd remediation rate was increased from $73.2\%\;to\;98.5\%$; as boric acid concentration was increased from 0.001M to 0.1M, Cr remediation rate was increased from $59.2\%\;to\;99.0\%$. In the experiments using 0.005M of EDTA and 0.005M of boric acid composite as a flushing reagent at different pH regimes, Cd remediation rate was decreased from $92.4\%\;to;80.9\%$ as the pH was increased from 3 to 7. But Cr remediation rate was more effective ($70.4\%$) at pH 5 compared to the other pHs. Furthermore, at pH 5, Cd and Cr remediation rate was investigated at different molecular ratio of [EDTA]/[Boric aicd]; the optimized [EDTA]/[Boric acid] ratio was 0.01M/0.1M for Cd contaminated site and, as the concentration of boric acid was increased over 0.1M, Cr remediation rate was also increased. So the similar results were shown in the experiments using real contaminated soils which were sampled from a mining and a industrial area.

Role of a Phytotoxin Produced by Fusarium oxysporum f. sp. raphani on Pathogenesis of and Resistance to the Fungus (무 시들음병균이 생산하는 Phytotoxin의 병원성 및 저항성에서 역할)

  • In the course of a developing screening method for resistant radish to Fusarium oxysporum f. sp. raphani, we found that the fungus produces phytotoxic compound against Raphanus sativus. The culture filtrate of F. oxysporum f. sp. raphani KR1 represented the strongest phytotoxicity when the fungus was incubated in the malt extract broth with 150 rpm at $25^{\circ}C$ for 14 days. Under bioassay-guided purification, we isolated a substance from liquid culture of F. oxysporum f. sp. raphani KR1, with phytotoxic effect against R. sativus. The compound was identified as fusaric acid by mass and nuclear magnetic resonance spectral analyses. Phytotoxicity of the compound against cruciferous vegetable crops, including radish, cabbage, and broccoli, was investigated. Fusaric acid represented phytotoxicity on radish seedlings by concentration dependant manner. And the phytotoxin demonstrated strong phytotoxicity on the resistant cultivars as well as susceptible cultivars of radish to F. oxysporum f. sp. raphani. In addition, fusaric acid isolated from the fungus also showed a potent phytotoxic efficacy against non-host Brassicaceae crops of the fungus such as cabbage and broccoli. The results demonstrate that fusaric acid produced by F. oxysporum f. sp. raphani is non-host-specific toxin and for screening of resistant radish to the fungal pathogen, spore suspension of the fungus without the phytotoxin has to be used.

Pretreatment Characteristics of Potatoes by Soft Steam Treatment (저온스팀 열처리 방식에 따른 감자의 전처리 특성)

  • This study aimed to investigate the effects of various soft steam treatments, namely, forced convection-boiler, forced convection-fan, and natural convection, on the pretreatment characteristics of potatoes. In this study, potatoes were exposed to various cooking conditions, including steaming method, treatment time (0-60 min), and temperature (60, 70, $80^{\circ}C$). Then, changes in temperature, cook value, ascorbic acid content, moisture content, and weight loss in the fresh and steam-treated samples were measured and evaluated. The results clearly showed that natural convective steaming was superior to other treatments in terms of heating characteristics, cook value (FC-b: $46.4{\pm}1.7$, FC-f: $21.8{\pm}1.1$, NC: $52.1{\pm}1.9min$ at $80^{\circ}C$), ascorbic acid content (FC-b: $36.5{\pm}2.7$, FC-f: $28.5{\pm}2.9$, NC: $48.2%{\pm}2.5%$ at $80^{\circ}C$), moisture retention (FCb: $74.6{\pm}0.8$, FC-f: $71.5{\pm}0.5$, NC: $77.6%{\pm}0.4%$ for 60 min at $80^{\circ}C$), and weight loss (FC-b: $13.9{\pm}0.8$, FC-f: $15.6{\pm}0.6$, NC: $10.6%{\pm}0.7%$ for 60 min at $80^{\circ}C$) for thermally processed potatoes.

Effect of Irradiation on Physicochemical Characteristics of Dry-Cured Ham During Storage (감마선 조사가 건조발효햄의 저장기간 중 이화학적 특성에 미치는 영향)

  • This study was carried out to investigate effect of irradiation doses on quality of vacuum-packed dry-cured ham during storage. Dry-cured pork slices were vacuum-packaged in nylon/ polyethylene bags. Vacuum-packed dry curd pork was stored at $10{^{\circ}C}$ for 12 weeks after irradiation (0 kGy, 2.5 kGy, 5 kGy, 7.5 kGy and 10 kGy) and physicochemical properties of dry curd porks, such as meat color surface, thiobarbituric aicd reactive substances, volatile basic nitrogen, pH, microbial (total plate count, Lactobacillus) and sensory properties (color, flavor, overall acceptability) were investigated. Irradiation treatment decreased a*-value, TPC and Lactobacillus. Irradiation dose significantly (p<0.05) increased irradiation flavor and TBA value. However, irradiation dose decreased redness and sensory color in dry-cured ham. The quality of dry-cured ham was affected by the irradiation dose. The results suggested that dry-cured ham irradiated at 5 kGy dose may show optimum quality characteristics of the products as well as microbial safety.
